Objective:Investigate the relation between the phosphorylation of FOXO1 and the apoptosis and the proliferation of lymphoma cells and to clarify its specific mechanism. Methods:The lymphoma cells Namalwa and Jurkat were treated with PI3K inhibitor wort mannin or etoposide or Wortmannin plus etoposide for different times-pan and at different concentration. The inhibition rates for cell growth of lymphoma cells were examined by XTT assay.Apoptosis were detected by flow cytometry. The expressions of p-Akt,p-FOXO1,FOXO1 and Bim were determined by Western blot analysis. Results:Wortmannin induced apoptosis of Jurkat cells and Namalwa cells and inhibited their survival effectively. The growth inhibition rate and the apoptosis rate of lymphoma cells induced by Wortmannin plus etoposide were higher than those induced by etoposide alone. After treated with Wortmannin, phosphorylation of FOXO1 remarkably reduced and bim markedly increased. Conclusion:The dephosphorylation of FOXO1 inhibits proliferation of Jurkat cells and Namalwa cells, promotes their apoptosis and enhanced the sensitivity of Non-Hodgkin lymphoma cells to etoposide.Bim activated by FOXO1 promotes cell apoptosis.

HIV-1 integrase enzyme is a 32kDa protein encoded by HIV pol gene. It is responsible for integration of viral cDNA into host chromosomal DNA, which is indispensable for HIV replication.Since there was no functional equivalent for this enzyme in human cells, inhibition of integrase will bring little side effect to human body. Thus HIV integrase has become an attractive and rational target for therapy of AIDS after reverse transcriptase and protease. The Recent research on HIV-1integrase structure, inhibitors and new therapeutic method target at HIV integrase was reviewed.
6. Efficacy and safety of bevacizumab combined with chemotherapy in treatment of EGFR wild type or EGFR-TKI resistant lung adenocarcinoma with brain metastases
Tumor 2016;36(10):1139-1145
Objective: There is no standard chemotherapy regimen for patients with wild type epidermal growth factor receptor (EGFR) and EGFRtyrosine kinase inhibitor (TKI) resistant lung adenocarcinoma with brain metastases. This study aims to observe the efficacy and toxicity of chemotherapy combined with bevacizumab in wild-type EGFR and EGFR-TKI resistant lung adenocarcinoma patients with brain metastases. Methods: A total of 10 lung adenocarcinoma patients with brain metastases between January and December 2015 received bevacizumab and chemotherapy. The response was evaluated by Response Evaluation Criteria in Solid Tumors version 1.1 (RECIST v1.1) every 2 cycles and the toxicity was evaluated by National Cancer Institute-Common Toxicity Criteria version 3.0 (NCI-CTC 3.0) every cycle. Results: Of the eight patients who could be evaluated for response, the overall response rate was 25% (2/8) and the disease control rate was 75% (6/8). And for intracranial lesions, the response rate was 12% (1/8) and the disease control rate was 87% (7/8). The most commonly adverse events induced by bevacizumab were hypertension, proteinuria and bleeding, which were mostly degrees I-II. Conclusion: Bevacizumab combined with chemotherapy is an effective and safe treatment method for EGFR wild type and EGFR-TKI resistant lung adenocarcinoma patients with brain metastases.

The relationship of nutrients and microorganisms in soils with polyphenols and total flavonoids of Houttuynia cordata were investigated by measuring nutrients, enzyme activity, pH, concentrations of microbe phospholipid fatty acids (PLFAs) in soils, and determining concentrations of polyphenols and total flavonoids of H. cordata. The research is aimed to understand characteristics of the planting soils and improve the quality of cultivated H. cordata. The soils at different sample sites varied greatly in nutrients, enzyme activity, pH, microbic PLFAs and polyphenols and all flavonoids. The content of total PLFAs in sample sites was following: bacteria > fungi > actinomyces > nematode. The content of bacteria PLFAs was 37.5%-65.0% at different sample sites. Activities of polyphenol oxidease, concentrations of available P and content of PLFAs of bacteria, actinomyces and total microorganisms in soils were significantly and positively related to the concentrations of polyphenols and total flavonoids of H. cordata, respectively (P < 0.05) . The Content of fungi PLFAs in soils was significantly and negatively related to concentrations of polyphenols and total flavonoids of H. cordata, respectively (P < 0.05). This study provides evidence that effectiveness of the soil nutrient, which may be improved due to transformation of soil microorganisms and enzymes to N and P in the soils, was beneficial to adaptation of H. cordata adapted to different soil conditions, and significantly affects metabolic accumulation of polyphenols and flavonoids of H. cordata.

Objective To retrospectively analyze the value of postoperative adjuvant therapy in the treatment of stageⅢthoracic esophageal squamous cell carcinoma ( ESCC) . Methods From 2008 to 2011, a total of 395 patients with stageⅢthoracic ESCC undergoing radical resection were enrolled as subjects. In those patients.97 received surgery alone (S).212 postoperative adjuvant chemotherapy (POCT),and 86 postoperative radiotherapy (PORT).Comparison of categorical data was made by chi?square test. The survival rates were calculated by the Kaplan?Meier method. The log?rank test was used for between?group comparison and univariate analysis. Results All patients were followed up for at least 3 years.125 cases were followed up for at least 5 years. The 5?year overall survival ( OS) rates in patients treated with S,POCT and PORT were 17. 1%,29. 2% and 36. 4%,respectively (P=0. 000).POCT and PORT could mainly increased OS in patients of males.upper?and middle?segment, severe ahhesion at surgery.well?or middle?differentiation, stageⅢa andⅢb(P=0. 000?0. 049);whenever ages.tumor lesion,two?/three field esophagectomy.and the number of removal lymph nodes. PORT could improved OS also (P=0. 001?0. 047).POCT could also improve OS in patients of ages≤60, tumor lesion<6 cm and removal lymph nodes<10 ( P=0. 002?0. 049 ) . The 5?year progression?free survival (PFS) were 19. 0% with S,28. 8% with POCT,36. 4% with PORT,respectively (P=0. 012).PORT could improve PFS (P=0. 012);especially for patients of males, ages ≤60,upper?and middle segment ESCC,tumor lesion ≥6 cm,severe ahhesion at surgery, removal lymph node<10 and ≥10,well or middle differentiation, stageⅢa andⅢb(P=0. 001?0. 042).But POCT could not increased PFS (P=0. 119) . Conclusions In the treatment of patients with stage Ⅲ thoracic ESCC undergoing radical resection, both POCT and PORT can improve the OS rate, particularly in patients with stage Ⅲa or Ⅲb middle and upper thoracic ESCC, severe adhesion formation during surgery. and moderately or well differentiated squamous cell carcinoma. The DFS rate is improved in patients treated with PORT,but not in those treated with POCT.
