China Biotechnology 2006;0(01):-
HIV-1 integrase enzyme is a 32kDa protein encoded by HIV pol gene. It is responsible for integration of viral cDNA into host chromosomal DNA, which is indispensable for HIV replication.Since there was no functional equivalent for this enzyme in human cells, inhibition of integrase will bring little side effect to human body. Thus HIV integrase has become an attractive and rational target for therapy of AIDS after reverse transcriptase and protease. The Recent research on HIV-1integrase structure, inhibitors and new therapeutic method target at HIV integrase was reviewed.

Objective:Investigate the relation between the phosphorylation of FOXO1 and the apoptosis and the proliferation of lymphoma cells and to clarify its specific mechanism. Methods:The lymphoma cells Namalwa and Jurkat were treated with PI3K inhibitor wort mannin or etoposide or Wortmannin plus etoposide for different times-pan and at different concentration. The inhibition rates for cell growth of lymphoma cells were examined by XTT assay.Apoptosis were detected by flow cytometry. The expressions of p-Akt,p-FOXO1,FOXO1 and Bim were determined by Western blot analysis. Results:Wortmannin induced apoptosis of Jurkat cells and Namalwa cells and inhibited their survival effectively. The growth inhibition rate and the apoptosis rate of lymphoma cells induced by Wortmannin plus etoposide were higher than those induced by etoposide alone. After treated with Wortmannin, phosphorylation of FOXO1 remarkably reduced and bim markedly increased. Conclusion:The dephosphorylation of FOXO1 inhibits proliferation of Jurkat cells and Namalwa cells, promotes their apoptosis and enhanced the sensitivity of Non-Hodgkin lymphoma cells to etoposide.Bim activated by FOXO1 promotes cell apoptosis.

7. Efficacy and safety of bevacizumab combined with chemotherapy in treatment of EGFR wild type or EGFR-TKI resistant lung adenocarcinoma with brain metastases
Tumor 2016;36(10):1139-1145
Objective: There is no standard chemotherapy regimen for patients with wild type epidermal growth factor receptor (EGFR) and EGFRtyrosine kinase inhibitor (TKI) resistant lung adenocarcinoma with brain metastases. This study aims to observe the efficacy and toxicity of chemotherapy combined with bevacizumab in wild-type EGFR and EGFR-TKI resistant lung adenocarcinoma patients with brain metastases. Methods: A total of 10 lung adenocarcinoma patients with brain metastases between January and December 2015 received bevacizumab and chemotherapy. The response was evaluated by Response Evaluation Criteria in Solid Tumors version 1.1 (RECIST v1.1) every 2 cycles and the toxicity was evaluated by National Cancer Institute-Common Toxicity Criteria version 3.0 (NCI-CTC 3.0) every cycle. Results: Of the eight patients who could be evaluated for response, the overall response rate was 25% (2/8) and the disease control rate was 75% (6/8). And for intracranial lesions, the response rate was 12% (1/8) and the disease control rate was 87% (7/8). The most commonly adverse events induced by bevacizumab were hypertension, proteinuria and bleeding, which were mostly degrees I-II. Conclusion: Bevacizumab combined with chemotherapy is an effective and safe treatment method for EGFR wild type and EGFR-TKI resistant lung adenocarcinoma patients with brain metastases.

Objective To assess the long efficacy and safety of Lamotrigine(LTG) monotherapy and add-on therapy different types of epileptic seizures in children. Methods According to the classification of the 1981 and 1989 International Union of Antiepileptic for epileptic seizure,a total of 124 cases with epilepsy were included in the study and divided into non-refractory group with 93 cases and refractory group with 31 cases.LTG treatment only or add-on were used.Original drug dosage was not changed and LTG was added slowly and carefully untill the terrible side effect appeared. The average monthly seizure frequency with baseline in the last 3 months was compared and the side effect was observed. Results Total efficiency was 72.6%,control rate was 51.6%.Total efficiency and control rate of the non-refractory group was 81.0% and 61.3%,which was significantly higher than those of refractory group(48.4%,22.6%).Total efficiency and control rate of the combination group with LTG and valproate sodium(VPA) was 78.4% and 54.5%,which was significantly higher than those of the group of LTG only(61.0%,44.0%).Clinical results was different significantly between the course of the observation period within and over 5 years(P
9.Systematic review on the rate for injury incident among elementary and middle school students in China.
Chinese Journal of Epidemiology 2007;28(9):881-885
OBJECTIVETo obtain the incidence rate of injury in primary and middle school students in China, so as to provide basic information for the development of prevention and control strategy.
METHODSSystematic review was applied to summarize the published data. After strict inclusion and exclusion, 42 eligible articles were included for analysis.
RESULTSA total number of 259 994 subjects was studied with 79 995 persons, 127 978 person-time involved in different kinds of injuries with the total incidence rate of 30.77% by person and 49.22% by person-time, respectively. The incidence rates of injury were not statistically significant between primary and middle schools (t = -1.752, P = 0.110) or at different places (F = 1.890, P = 0.120). However, significant differences were found between boys and girls (t = 5.998, P = 0.000). The top three injuries were caused by fall, hurt during sports activities and transportation vehicles.
CONCLUSIONResults from the systematic review indicated that the majority of injuries were fall, hurt during sports activities and transportation vehicles among primary and middle school students in China. In order to reduce the incidence of injury, health education and improvement of environment should be particularly strengthened in the future.
