3.Effect of Huangshao Yinxue Granula on psoriasis-like animal models of animals

AIM:To study whether or not Huangshao Yinxue Granula(HYG) has the therapeutic effect on experimental psoriasis. METHODS: The mice were treated with HYG 3.125,6.25,12.5,and 25 g/kg(i.g.) for 1 or 2 weeks.We studied the effects of HYG on mitosis in mice vagina epithelium,the formation of granular layer in scale epidermis of mice tails,the changes in CD3, CD4 and CD8 factors of the serum in psoriasis-like mice and guinea-pigs. RESULTS: HYG could inhibit the mitosis in mice vagina epithelium,promote the formation of granular layer in scale epidermis of mice tails.It could also diminish the value of serum CD4/CD8 of mice to regulate abnormal immune reaction.HYG had evident therapeutic effect on the ear skin damaged with 5% propanolol in psoriasis-like guinea-pigs. CONCLUSION: HYG has therapeutic effect on psoriasis-like animal models.
4.The correlation studies of the Lp-PLA2,hs-CRP and D-dimmer in patients with coronary heart disease

Objective To study the correlations among the lipoprotein associated phospholipase A2 (Lp‐PLA2) ,hypersensitive C‐reactive protein(hs‐CRP)and D‐dimmer in patients with the coronary heart disease .Methods 260 cases of patients with coronary heart disease(CHD) were enrolled in the study as observation group and 260 healthy people from the physical examinations were recruited as control group .The concentrations of Lp‐PLA2 ,hs‐CRP ,D‐dimer and other biochemical indicators in blood sera of the two groups were detected ,using the Spearman correlation analysis to test the relationship between those indicators .Results The Lp‐PLA2 ,hs‐CRP ,D‐dimer levels of the observation group were obviously higher than those of control group .The Lp‐PLA2 level of the observation group was positively correlated with the cardiac function index(r=0 .873 ,P<0 .05) .hs‐CRP was positively cor‐related with cardiac function index(r=0 .782 ,P<0 .05) .D‐dimmer was positively correlated with the functional grades of heart(r=0 .674 ,P<0 .05) .Conclusion Lp‐PLA2 ,hs‐CRP and D‐dimer could be important indicators for the detection of coronary heart disease .
5.Content Determination of Astragaloside Ⅳ in Qingshen Jianfei Tablets by HPLC-ELSD

Objective To establish a HPLC-ELSD method for determining the content of astragaloside Ⅳ in Qingshen Jianfei Tablets. Methods Stationary phase was C_(18) column (4.6 mm?250 mm, 5 ?m), mobile phase was acetonitrice-water (35 : 65). The evaporation temperature was 120 ℃, the pulverization temperatire was 80 ℃, the flow rate was 1.6 mL/min and column temperature was 35 ℃. Results The standard curve was linear within the range of 1.03~8.24 ?g, r=0.9999. The average recovery was 99.35% with RSD = 1.36% (n = 6). Conclusion The established method is simple and accurate, with good reproducibility and high precision, and suitable for the determination of astragaloside Ⅳ in Qingshen Jianfei Tablets.
6.Correlation between different types of malocclusions and body image issues in college students

Objective:
To explore the correlation between malocclusion and body image issues in college students.
Methods:
A total of 1 851 students in three universities in Jingmen were selected by using stratified cluster sampling method. Angle s classification of malocclusion was used to determine the number of three types of malocclusions. Body image issues were self reported and its relationship with different types of malocclusions was explored.
Results:
The proportions of Classes Ⅰ, Ⅱ and Ⅲ malocclusion in college students with malocclusion were 71.21%, 16.32%, and 12.47%, respectively. The detection rates of body image issues among students with Classes Ⅰ,Ⅱ and III malocclusions were 36.64%, 54.78% and 65.83%, respectively. No significant difference were found in the detection rates of sexual organ issues and gender issues in college students with different types of malocclusions( χ 2= 0.75, 0.53, P >0.05). There were significant differences in the detection rates of appearance troubles (27.59%, 33.12%, 50.83% ) and stature troubles ( 24.09% , 31.21%, 44.17%) in students with Classes Ⅰ, Ⅱ and Ⅲ malocclusions( χ 2=5.62, 2.89, P <0.05).
Conclusion
The prevalence of body image issues in college students increases with severity of malocclusions. Appearance and stature troubles are issues mostly concerned among college students. Psychological evaluation for students with Class Ⅲ malocclusion should be especially emphasized when administrating orthodontic treatment.
7.Effects of Xuanfeitongfufang Decoction on Expressions of MD-2 and NF-κB in Lung of Rats ;with Acute Lung Injury Caused by Lipopolysaccharide

Objective To investigate the effects of Xuanfeitongfufang Decoction on expressions of MD-2, NF-κB protein and its mRNA in lung of the rats with acute lung injury caused by LPS. Methods Wistar rats were randomly divided into normal group, model group, dexamethasone group and Xuanfeitongfufang Decoction large-, medium-, small-dose group, each group had eight rats. The ALI rat model was established by LPS tail-intravenous injection (6 mg/kg). The rats in Xuanfeitongfufang Decoction groups were pretreated by Xuanfeitongfufang Decoction (15.12, 7.56, 3.78 g/kg) for 3 days before LPS induced ALI. The rats in dexamethasone group were pretreated by dexamethasone (5 mg/kg). MD-2, NF-κB protein and its mRNA were measured by immunohistochemistry and PCR. The histopathology of the lung injury was observed by light microscope. Results Compared with normal group, the expression of MD-2, NF-κB protein and its mRNA were obviously increased in model group (P<0.01). Compared with model group, the expression of MD-2, NF-κB protein and its mRNA were obviously decreased in Xuanfeitongfufang Decoction groups and dexamethasone group (P<0.01), and there was no obvious difference between Xuanfeitongfufang Decoction groups and dexamethasone group. Light microscope observation indicates that there were large areas of pulmonary hemorrhage and necrosis in model group. While in Xuanfeitongfufang Decoction group and dexamethasone group, the pathological manifestations were much more ameliorated than those of the model group. The lung bronchiale inflammation appeared occasionally, and the edema was lightly. Conclusion Xuanfeitongfufang Decoction can lessen the injury of lung tissue and has protective effects on rats with ALI, the mechanism is possibly related to the inhibition of the expressions of MD-2 and NF-κB protein and its mRNA in injured lung tissues.

9.Change in expression of vascular endothelial growth factor in serum and wound tissue of rats with electrical burns.

OBJECTIVETo observe the change in expression of vascular endothelial growth factor (VEGF) in serum and wound tissue of rats with electrical burn (EB), and to explore its regulation mechanism in the pathological changes of EB.
METHODSSixty-four SD rats were divided into normal control group (n = 8) and EB group (n = 56) according to the random number table. Eight rats in EB group were sacrificed at post injury hour (PIH) 6 and on post injury day (PID) 1, 3, 7, 14, 21, and 28, to collect wound muscle tissue and serum samples. Histopathological changes in wound tissue were observed with HE staining. The serum content of VEGF was determined with double-antibody sandwich enzyme-linked immunosorbent assay. The expression level of VEGF in wound tissue was determined with Western blotting. VEGF expression intensity in wound tissue was observed with immunohistochemical staining. The microvessel density (MVD) was calculated. The correlation between VEGF expression intensity and MVD was analyzed. Muscle tissue of calf and serum of the rats in normal control group without any treatment were collected for above-mentioned observations and determinations. Data were processed with one-way analysis of variance and Spearman hierarchy correlation analysis, and LSD-t test was applied for paired comparison.
RESULTS(1) In EB group, breakage of muscle fiber, heavy infiltration of inflammatory cells, and obvious tissue edema were observed at PIH 6 and on PID 1; new vessels were observed on PID 3; amount of granulation tissue and number of new vessels were found to be increased on PID 7. (2) In EB group, the serum level of VEGF was (43 ± 11) pg/mL at PIH 6, (44 ± 11) pg/mL on PID 1, and (74 ± 27) pg/mL on PID 14, which were all significantly higher than that in normal control group [(15 ± 9) pg/mL, with t values from 4.001 to 5.724, P values all below 0.01]. (3) The protein expression level of VEGF of wound tissue in EB group was higher than that in normal control group (0.21 ± 0.09) at each time point. The protein expression level of VEGF in EB group peaked on PID 7 (0.63 ± 0.13, t = 4.965, P < 0.05). (4) In EB group, strongly positive expression of VEGF was observed in inflammatory cells at early stage and in new vascular endothelial cells at late stage. (5) The expression intensity of VEGF was positively correlated with MVD in wound tissue on PID 3, 7, 14, 21, and 28 in EB group (r(s) = 0.834, P < 0.01).
CONCLUSIONSDifferent expression levels of VEGF were observed in serum and wound tissue of rats at various stages after EB, and they were closely correlated with different stages of fluid exudation and wound healing process after EB.

10.Prolongation of islet allograft survival in rats by cotransplantion with xenogenetic neonatal porcine Sertoli cells

Objective To investigate whether cotransplant with xenogenetic neonatal porcine Sertoli cells (NPSCs) could prolong rat islet allograft survival and its mechanisms.Methods 1500 islets equivalent quantity (IEQ) and 1×10~7 NPSCs were implanted under renal capsule of diabetic Wistar rats.Islets implanted alone were used as control group (n=6);islets co-transplanted with NPSCs under left renal capsule of recipients served as experimental group (n=6);meanwhile,islets and NPSCs implanted into the different sides of kidneys were used as another control grouP(n=4).Blood glucose level was measured everyday.The graft-bearing kidneys at the time of rejection were Results Co-transplantation with NPSCs to the same site significantly prolonged islet allograft survival (mean survive time,16.3±1.4 days vs.5.7±1.0 days in islet transplant alone control group,P<0.05).In contrast,transplantation with NPSCs and islets separately did not prolong the islet allograft survival (5.3±0.5 days).HE staining showed plenty of local infiltrated lymphocytes in the transplanted site of the eontrol group.which were demonstrated as mainly CD3+ T cells by immunopathology.The local expression of Bcl-2 was markedly elevated in co-transplantation group as compared with the other 2 groups,while there were no significant differences in the HO-1 expression among these groups.Conclusion Co-transplantation with xenogenic NPSCs can significantly prolong islet allograft survival in rats.The immunoprotective mechanism may be associateel with the inhibition of lymphocyte infiltration and the enhancement of the local expression of protective gene Bcl-2.
