2.Dual role of dendritic cells in the pathological mechanism of atherosclerosis
International Journal of Surgery 2013;40(12):831-834
Atherosclerosis is a chronic inflammatory immune disease, which is one of the causes of human lower limb disability.Dendritic cells are the most powerful professional antigen presenting cell in immune system, and dendritic cells have the dual role of immunogenicity and tolerability. As a professional antigen presenting cell, the immunogenicity of dendritic cells present self abnormal antigen and course immune system disorder, which may be one role of dendritic cells participate in atherosclerosis. However, the tolerability of dendritic cells can course the generation of immune tolerance, which makes dendritic cells exert a regulatory role in atherosclerosis. This paper makes an introduction about how dendritic cells are involved and play a regulatory role in the pathogenesis of atherosderosis.
3. Association between variable number of tandem repeat polymorphism of thymidylate synthase gene and efficacy of pemetrexed in the treatment of advanced non-small cell lung cancer
Tumor 2012;32(7):539-545
Objective: To investigate the association between variable number of tandem repeat (VNTR) polymorphism in 5'-untranslation region (5'-UTR) of thymidylate synthase (TS ) gene and the efficacy of pemetrexed-based treatment in non-small cell lung cancer (NSCLC). Methods: Peripheral blood samples were collected from 90 patients with advanced NSCLC and the genomic DNAs were extracted from these blood samples. The VNTR polymorphism in 5'-UTR of TS gene was detected by fluorescent labeled PCR and multiplex short tandem repeat polymerase chain reaction in combination with capillary electrophoresis. Results: The homozygosis (3R/3R and 2R/2R) and heterozygosis (2R/3R and 3R/4R) of VNTR polymorphism in 5'-UTR of TS gene were not associated with the efficacy of pemetrexed. The progression-free survivals, disease control rates and the objective response rates in patients harboring homozygosis (3R/3R and 2R/2R) and heterozygosis (2R/3R and 3R/4R) were 112 and 94 d (P = 0.549), 64.3% and 67.6% (P = 0.745), and 12.5% and 8.8% (P = 0.591), respectively. The patients harboring 2R alleles had similar clinical outcomes as compared with those harboring 3R alleles (progression-free survival: P = 0.518; disease control rate: P = 0.631; objective response rate: P = 0.541). Conclusion: TS gene VNTR polymorphism may not be associated with clinical outcomes of advanced NSCLC treated with pemetrexed. Copyright © 2012 by TUMOR.

The method of gene transfer into corneal endothelium was investigated to provide a foundation for the study of TGF-beta 1 gene transfer to inhibit corneal graft rejection. Two days after direct injection of pMAM TGF-beta 1 mediated by liposome into the anterior chamber of rabbits, one half of corneas were made into paraffin slides and the endothelial layer was carefully torn from the other half to make a single layer slide of endothelia. By means of immunohistochemical technique, the plasmid pMAM TGF-beta 1 expression product TGF-beta 1 in the endothelia was detected. Specific TGF-beta 1 expression was positive in the endothelia on both the paraffin slide and the single layer slide. The results showed that by direct injection into the anterior chamber, foreign plasmid DNA could be transferred into the endothelia and its expression was obtained. This may provide a foundation for further study on TGF-beta 1 participating in local induction of corneal immune tolerance.

Journal of Applied Clinical Pediatrics 2006;0(16):-
Objective To explore the atopic condition of children with stable asthma and their parents by detecting total IgE concentration in serum and collecting history. Methods Fifteen children with asthma in remission stage and 24 parents and 40 normal children were involved in this study. The concentrations of serum total IgE were measured with UniCAP Pharmacia system. The history including eczema and allergic rhinitis and asthma of children and parents were collected. Skin test with 10 kinds of inhalant allergens were taken in children with asthma. Results Of 15 children with asthma, 13 cases (86.7%)had eczema in infant stage and allergic rhinitis, 13 parents(86.7%)had allergic rhinitis or asthma. There was significant difference in the stable asthmatic children compared with parents and normal control group increased amounts of serum total lg IgE(F=68.42 P=0). There was significant difference in serum total lg IgE in patients group(2.43?0.73)kU/L compared with that in normal control group(0.72?0.54)kU/L (q=14.176 P0.05). Twelve children were positive in skin test, which were mainly dermatophagoides pteronyssinus and dermatophagoides farinae. Conclusions There is obvious congregate phenomena of atopy in the family of asthmatic children. The continuous high IgE concentration was associated with immune pathological mechanisms of atopic diseases.
