Objective To observe the effects of enzymolysis of human B like antigen by ? galactosidase treatment on the morphology and function of cynomolgus monkey red blood cell (RBC) and to evaluate the safety of the enzyme treated RBC in animal transfusion. Methods RBC with human B like antigen was treated by recombined ? galactosidase and the effect was evaluated by absorption elution test. Meanwhile, morphology and function of the treated RBC were examined and compared with pre treatment RBC. ? galactosidase treated RBC was labeled with FITC and then infused to blood group A cynomolgus monkeys. Flow cytometry was used to detect the survival of donor RBC in recipient’s blood. Blood chemistry and urinalysis of recipients were performed before and after transfusion. Results The human B like antigen of cynomolgus monkey RBC was obliterated by ? galactosidase treatment and the treated RBC maintained normal morphology and function. Survival at 24 h after transfusion was 84.6% and 68.1%. T 1/2 were 7d and 8d versus 13d in the controls. There were no change in blood chemistry and urinalysis of the recipients. Conclusion Enzymolysis of cynomolgus monkey RBC does not affect the cellular function and morphology. Transfusion of the enzyme treated human B like RBC into human A like cynomolgus monkeys is safe.

Activated microglia function after pathologic event has been extensively investigated .In recent decades , it has be-come evident that resting microglia cells are highly dynamic surveillants .Microglia could influence synaptic development and connectiv-ity in the normal and developing brain .Microglia dysfunction results in behavioral deficits , indicating that microglia are essential for normal brain function .Here we summarize the functions of microglia in the physiological conditions , its effects on neural development , the formation of synapses and the regulation of the human′s behavior .

Objective:To construct NBS1 microRNA expressing eukaryotic recombinants,and identify biological activity of recombinants in Hela cell after transfection.Methods:According to sequence of NBS1mRNA,the NBS1 pre-microRNA was designed and synthesized,then cloned into the GFP reporter pcDNA6.2-GW/EmGFP-miR vector and transfected into Hela cell line.To detect integrity of inset fragment through colony PCR and sequencing analysis.The biological activity of recombinants through identify interference efficiency of NBS1 microRNA recombinants by way of Real-Time PCR and Western blot were determined.Results:Sequences of inset fragment in four microRNA expressing recombinants were correct.NBS1 mRNA and protein expression of four microRNA recombinants were decrease,which is the lowest in the NBS1mi-2 group.Conclusion:Four NBS1-targeting microRNA expressing recombinants all have biological activity in Hela cell line,and NBS1mi-2 recombinant has the most interference efficiency.The microRNA expressing plasmid which were successfully constructed and lay foundation for the studies on the tumor gene therapy of microrna targeting NBS1.

Objective To observe the chemical camouflage effects of methoxy polyethylene glycol(mPEG) on like human B antigens on Rhesus monkey red blood cell(rRBC) and to study the safety of transfusion with mPEG modified monkey RBC(mPEG rRBC).Methods rRBC were modified with 3mg/ml mPEG. The rRBC blood group and the effects of mPEG modification on blood conversion were assayed by absorption and elution methods.The structural and functional changes of mPEG rRBC were also tested.The survival fraction of mPEG modified like B rRBC in like A recipient monkey were examined by flow cytometry.The transfusion effects of mPEG modified rRBC on recipient's blood and urine were also observed.Results mPEG have been covalently bound to the surface of Rhesus monkey RBC (like human B blood group),mPEG could cover rRBC like B antigen.Transfusion of mPEG modified like B rRBC to like A Rhesus monkey had no harmful effects on the recipient.Conclusion The present study suggests that transfusion of mPEG modified RBC could be safety in experimental animals

Objective To investigate the hand hygiene status of urban and rural residents in Hangzhou City of Zhejiang Province to provide scientific base for health promotion.Methods Urban residents,rural residents,personnel in government institutions,urban students,rural students and enterprise workers were enrolled in this investigation.By using constant volume sampling method,fifty individuals in each group were recruited from 13 districts of Hangzhou City,and a total of 3 337 people filled in the questionnaire,which included demographic characteristics,knowledge of hand hygiene and hand washing.Logistic regression analysis and x2 test were used for data analysis.Results Correct hand-washing was found in 35.84％ urban and rural residents (urban vs.rural:41.45％ vs.27.06％).Awareness rate of hand hygiene was 35.06％.The influencing factors of proper hand washing were location,gender,age,educational level and understanding of hand hygiene.The influencing factors of awareness of hand hygiene were location,gender,age and educational level.Conclusion Hangzhou citizens had a relatively lower rate of proper hand washing,and significant difference was found between urban and rural residents; the study suggested that para-urben residents need to improve their knowledge and behavior of hand hygiene.Meanwhile,the students urgenty need to improve their hand hygiene

Objective To evaluate the skin protection of the 3M transparent dressing joint 3MTM CavilonTM No Sting Barrie Film after peripherally inserted central catheter (PICC).Methods A total of 120 PICC catheter patients with lung cancer (observation time last about 3 months from starting the rearing pipe on the day to pipe-taken) were selected.They were divided into 2 groups by random digits table method with 60 cases each.Control group was given 3M transparent dressings stick in the PICC fixed place skin.Observation group was given 3M transparent dressings paste joint 3MTM CavilonTM No Sting Barrie Film.The incidence of complications in the PICC fixed place skin were observed and compared between 2 groups.Results The skin allergy,phlebitis,skin damage,total complications were 5,4,1,10 cases in control group after PICC catheter and 2,1,3,6 cases in observation group,and there was significant difference in the incidence of the total complications,x2=4.23,P＜0.05.Conclusion Using 3M transparent dressings joint 3MTM CavilonTM No Sting Barrie Film for skin care after PICC catheter,can effectively protect the skin after PICC catheter,reduce the incidence of adverse reactions.

In order to adapt to the demand of culturing high quality talents and to strengthen student's comprehensive quality,pathophysiology department of medical college of Qinghai University carried on the reform on the pathophysiological experiment examination system.After researches and practices in recent years,we established a set of relatively sound examination system including experiment operating and experiment report in normal study,report for designable experiment and experiment skill and theory exam.Investigation results demonstrated that examination system after reform functioned well in training the students' operation ability,observation ability and ability to analyze and solve questions.

Objective:The study aims to screen chemosensitivity-associated proteins in colorectal carcinoma tissues by using two-dimensional gel electrophoresis (2-DE) and mass spectrometry,then identify some differentially-expressed proteins. Methods:The patients with advanced colorectal carcinoma were confirmed by clinical diagnosis. Fresh carcinoma specimens were collected by biopsy and preserved in liquid N2. The tissues were classified into two groups: high sensitivity group (HS) and low sensitivity group (LS) based on drug sensitivity test. The total proteins were extracted and separated by 2-DE. The images were composed, compared, and differentially analyzed to identify the proteins with differential expression in HS and LS groups. Then the differentially-expressed protein spots were incised from the gels and digested by trypsin. The peptide mass fingerprintings (PMF) was acquired after matrix assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS) and the proteins were identified by data searching in the Mascot database. Two proteins with differential expression were detected by Western blotting.Results:The 2-DE spectrum of HS and LS groups were established. Most protein spots were distributed in the area with pH 4-8 and relative molecular weight of (20-100)×10~3. The average number of the protein spots was 842±23 in HS group and 793±19 in LS group,respectively. The mean matching rate was 90.7%. The number of differentially-expressed dots between HS and LS group was 79.00±13.56. Thirty protein dots were selected for mass spectrum and bioinformatic analysis, and 9 proteins were identified. Conclusion:Colorectal carcinoma with different chemosensitivity had differential protein expression profiles. The differentially expressed proteins may be associated with chemosensitivity and could be used for prediction of chemosensitivity of colorectal carcinoma.

Objective To clarify the mechanisms of total saponins of panax ginseng (TSPG) in induction of apoptosis of K562 cells and to provide the theoretical basis and the experimental evidence for its clinical application. Methods The effects of TSPG on apoptosis of K562 cells were studied by morphometry, flow cytometry, morphological observation, and immunocytochemistry. Results The results indicated that TSPG could markedly inhibit the proliferation of K562 cells and induce the apoptosis of K562 cells. Our experiment also showed that after induction by TSPG, the ratio of positive K562 cells with Fas expression increased. Conclusion The mechanisms of TSPG in the induction of K562 cell apoptosis may be related to the increased expression of Fas in K562 cells.
