1.Extraction of Alsophila spinulosa(Hook.) Tryon flavonoids by orthogonal experiment and separation by macroporous resin

AIM:To study the optimum extraction process of total flavonoids in Alsophila spinulosa(Hook.) Tryon and technological parameter of macroporous resin. METHODS: The optimum parameters were chosen by ratio of total flavonoids as the assessment index such as static and dynamic adsorption of macroporous resin. RESULTS: The optimum extracting process conditions were as follows: solvent was ethanol,ratio of solid-liquid was 1(∶)12,extraction time was 1 h,temperature was at(80 ?C).The optimum technological conditions of the D104 were: the solution concentract at 1.36 mg/mL,the current velocity of adsorption 2 BV/h,the amount of saturated adsorption at 5 BV,the eluting reagent 70% ethanol,the eluting velocity 3 BV/h,eluent volume at 2 BV. CONCLUSION: The process can separate and concentrate the total flavoionds of Alsophila spinulosa(Hook.) tryon.
2.Tiaohe Ganpi Hexin Decoction in treatment of irritable bowel syndrome with diarrhea: a randomized controlled trial.

Background: The quality of life has been greatly influenced and the cost of medical expenses is very high in patients with irritable bowel syndrome (IBS). The etiology and pathogenesis of IBS are still unclear, and the prevention and treatment of this disease still lack of effective methods. Objective: To explore and analyze the effects of Tiaohe Ganpi Hexin Decoction (TGHD), a compound traditional Chinese herbal medicine for regulating the liver and spleen, on IBS patients with diarrhea. Design, setting, participants and interventions: All 40 IBS patients came from the First Hospital of Jinan University, Guangzhou Red Cross Hospital, and the First Affiliated Hospital of Guangzhou University of Chinese Medicine, and were randomly divided into two groups. Patients in the treatment group (n=20) were given TGHD, while those in the control group (n=20) were prescribed oral pinaverium with a four-week treatment period. Main outcome measures: Traditional Chinese medicine (TCM) syndrome score, total obviously effective rate, disappearance rate of symptoms, and clinical symptom score in the two groups were evaluated before and after four-week treatment. Results: After the treatment, TCM syndrome scores in both groups were decreased (P<0.01), and the TCM syndrome score in the treatment group was significantly lower than that in the control group (P<0.01). There was a significant difference in the total obviously effective rate between the two groups (P<0.01), and the total obviously effective rates in the treatment and control groups were 85%(17/20) and 45%(9/20) respectively. The disappearance rates of abdominal pain, abdominal distention, poor stool output, stool frequency, stool character and mucous stool in the treatment group were higher than those in the control group (P<0.05). The symptom scores of abdominal pain, abdominal distention, poor stool output, stool frequency, stool character and mucous stool in the treatment group were lower than those in the control group (P<0.05). Conclusion: TGHD can significantly improve the clinical symptoms in IBS patients with diarrhea.
3.An analysis of the expression of P53 protein and the radiosensitlvity in patients with advanced maxmary squamous ceH carcinoma

Objective To explore the relationship between the expression of P53 protein and the radiosensitivity in patients with advanced maxillary squamous cell carcinoma.Methods An immunohistochomical method wag used to detect the expression of P53 protein in patients with advanced maxillary squamous cell carcinoma.The follow up time was 2 years.The local recurrence of the patients having been treated with radical surgery and affiliated radiotheraphy were analyzed.Results The overexpression of P53 protein in 26 cages was 65.4% (17/26).In the P53 overexpression group,the local recurrence after systiem therapy wag occurred in one case within 6 months,6 cages between 7~12 months.3 cages between 13~18 months and 2 cases between 19~24 months.In the P53 low expression group,there were no recurrence within 6 and 12 months and one case ocurred within 18 months.5 cages between 19~24 months.The difference of recurrence within 18 months after system therapy between the expression of P53 Wag statistically significant(P<0.05),but it Wag not significant for those within 24 months(P>0.05).Conclusions The expression of P53 protein Wag correlated with the radiosensitivity in patient with advaneed maxillary squamous cell carciaoma,especially for the resid.rod cells in mitosis phase.The affiliated radiotherapy after radical surgery Wag limited effect.
4.Application of continuous femoral nerve blockade combined with epidural morphine on total knee arthro-plasty in elderly patients

Objective To observe the effect of combined continuous femoral nerve blockade (CFNB)with epidural morphine postoperative analgesia on total knee arthroplasty (TKA)in elderly patients.Methods Eighty elderly patients scheduled to undergo TKA were randomly divided into two groups:groups FN and FNM.Groups FNM and FN were received 0.1% morphine 2 ml and normal saline 2 ml through epidural space respectively at the end of surgery.After surgery,all patients were implanted catheter around femoral nerve guided by ultrasound,and were infused 0.2% ropivacaine 25 ml.Continuous infusion of 0.2% ropivacaine for femoral never block was administrated at a rate of 5 ml/h for 48 hours to all patients.VAS scores and bending degree during rest,active and passive exer-cise were recorded at time point of postoperative 24,48,72,96 h.Ambulation status on the fifth day after operation were observed.Satisfaction of hospitalization were graded before hospital discharge. Results Compared with group FN,VAS scores during rest,active and passive excercise were signifi-cantly decrease at 24 h in group FNM (P <0.05 or P <0.01).The bending degree of knee joints in group FNM were significantly greater compared with group FN at 24 h after surgery (P <0.01).Am-bulation status on the fifth day after surgery were significantly better in group FNM compared with group FN (P <0.05).The level of patient satisfaction was much higher in group FNM (P <0.05). Conclusion After TKA,CFNB combined with single epidural morphine injection can provide better analgesia and bending degree with a high level of satisfaction.
5.Establishment of a new canine model of acute hepatic failure

Objective To establish a new model of acute hepatic failure in canines for bioartificial liver treatment research . Methods Acute hepatic failure was induced by end to side portocaval shunt combined with common bile duct ligation and transection. The survival rate, liver function, ammonia, Fischer's ratio (BCAA/AAA) and pathological change of injury liver were investigated. Results The ammonia, ALT, AKP, total bilirubin and PT were elevated dramatically and Fischer's ratio declined significantly 14 days after the operation. The survival rate of the models decreased with time 14 days after the operation. Pathological examination of liver tissues revealed evidence of cholestasis, inflammation with accumulation of neutrophils, degeneration and necrosis of hepatocytes.Conclusions End to side portocaval shunt combined with common bile duct ligation and transection can produce a satisfactory acute hepatic failure model in canines, which could be used in the study of bioartificial liver.
7.Epidemiological analysis of malaria situation in Wenzhou City in 2015

Objective To analyze the characteristics of malaria prevalence in Wenzhou City,so as to provide the evidence for improving the comprehensive control of malaria. Methods The epidemiological data of malaria cases in Wenzhou City in 2015 were collected and analyzed with the descriptive epidemiological methods. Results Totally 24 imported malaria cases were reported in Wenzhou City in 2015 with the incidence of 0.26 per 100000 and no case was dead. Plasmodium vivax,P. fal-ciparum,and P. ovale were identified in 1(4.17%),20(83.33%),and 3(12.50%)cases,respectively. The cases reported in Rui'an,Cangnan and Lucheng counties/districts accounted for 70.83%(17/24)of the total cases in Wenzhou City. The cases were mostly concentrated in male young adults and 23 cases(95.83%)were imported from Africa. Conclusion Malaria epi-demic situation in Wenzhou City is relatively stable and no local malaria cases were reported in 2015. However,the control work of imported malaria should be strengthened.
8.A Preliminary Study on Gastric Electrical Activity in Patients with Gastrointestinal Stromal Tumor

Objects: To investigate the behavior of gastric electrical activity in patients with gastrointestinal stromal tumor(GIST) to identify the influences of GIST on the normal gastric electrical activity. Methods: The electrogastrogram (EGG) parameters of 27 patients with gastric GIST (GIST group) and 30 healthy volunteers (control group) were detected by the multi-channel electrogastrogram and the data were analyzed. Results: The values of postprandial mean frequency (MF), mean amplitude (MA) and the percentage of normal slow wave (N%) were increased, and the percentage of bradygastria (B%) was decreased than those of the fasting in control group(P < 0.05 or P < 0.01). However, those postprandial parameter changes were not found in GIST group(P > 0.05). Compared with control group, the fasting MF and MA increased, the fasting N% of lead 1, 3, 4 and postprandial N% decreased, both percentages of fasting and postprandial tachygastria (T%) increased in GIST group (P < 0.01). The tachygastria incidence was significantly higher in GIST group than that of control (66.7% vs 3.3%, P <0.05). Conclusion: The gastric electrical activity was affected by the existence of GIST. The abnormal gastric electrical rhythm displayed mainly as tachygastria.

ObjectiveTo explore the clinical significance of fms-like tyrosine kinase3(FLT3) expression in evaluation prognosis of acute myeloid leukemia(AML) prognosis.Methods50 patients with AML were selected.AML patients with normal karyotype were 20 cases,the abnormal karyotype were 30 cases.3ml bone marrow before themotherapy was aspirated respectively,and the FLT3 gene expression in leukemia cells was detected with polyenzyme chain react(PCR).ResultsThe FLT3 expression rate in AML patients with normal karyotype was 5.0%,and was 26.7% in AML patients with abnormal karyotype,33.3% in AML patients with refractory-relapse,and 4.5% in AML patients with continue remission.The FLT3 expression rate was related with high leukemia cells percentage in bone marrow and high blood cells count in peripheral blood,and was not related with Franch America British(FAB) classification.The free-disease survival(FDS) and overall survived(OS) was shorter in FLT3 expression AML patients than that in no FLT3 expression AML patients.There was a statistical significance between the former and the latter( x2 =4.17,P <0.05 ).Conclusion FLT3 was a kind of worse factor in AML patients prognosis,and could guide clinical individual treatment in AML.
10.The development of a bioartificial liver and its application in acute liver failure patients

Objective Using polysulfon fibers, a new bioartificial liver was developed. This study was to evaluate the efficacy of this bioartificial liver in the support of a disfunctioned liver. Methods Hepatocytes were procured from swine using Seglen′s methods. The bioartificial liver was constructed based on polysulfon bioreactor with a procurement of 10 10 hepatocytes, and was applied in 12 acute liver failure patients for 14 sessions.
