Objective To analyze the effect of ATAD2 on glioma cell lines.Methods ATAD2 level in U87MG, U251 and normal human astrocytes was detected by RT-PCR and Western blot.U87MG and U251 cells were divided into four groups: control, mock (lipofection control), ATAD2 siRNA (transfected with ATAD2 siRNA) and ATAD2 overexpression (transfected with ATAD2).Cell proliferation, migration and invasion were respectively measured by MTT and Transwell assay.The level of E-cadherin, N-cadherin and Vimentin was measured by Western blot.Results ATAD2 was highly expressed in U87MG and U251 cells.Compared with control, cell proliferation, invasion, migration, N-cadherin and Vimentin expression were decreased by ATAD2 siRNA, while they were promoted by ATAD2 overexpression (P<0.05).E-cadherin expression was upregulated by ATAD2 siRNA and it was inhibited by ATAD2 overexpression (P<0.05).Conclusions ATAD2 participants in human glioma cells metastasis via epithelial-mesenchymal transition (EMT).

Immunosenescence is a process characterized by alterations and progressive deterioration of the structure and function of the immune system.Manifestations of immunosenescence include damage to both innate and adaptive immunity,reduction of immune responses to antigen stimulation,quantitative and functional changes of immune cells and alterations of cytokine activities,all of which may result in attenuated responses to vaccines,enhanced chronic inflammatory reactions,an elevated risk of infections and an increased susceptibility to cancer among old individuals.In recent years,it has been increasingly recognized that oxidative stress plays a vital role in immunosenescence.This review is focused on the influences of oxidative stress on the structure,function and generation of immunecells and the major mechanisms underlying immunosenescence.

Objective To evaluate effect of bone grafting combining with titanium mesh and/or reinforcement cup in reconstruction of acetabular bone defect in total hip arthroplasty (THA).Methods From January 2008 to November 2011,32 patients,including 23 males and 9 females,aged from 51 to 76 years (average,66 years),underwent THA with acetabular defect reconstruction by using bone grafting combining with titanium mesh and/or reinforcement cup.There were 6 cases of primary THA and 26 cases of revision THA.Twelve cases (Paprosky Ⅱ B) and 7 cases (Paprosky Ⅱ C) underwent impaction bone grafting with titanium mesh; 6 cases (Paprosky Ⅱ C) underwent impaction bone grafting with reinforcement cup; 6 cases (Paprosky Ⅲ A) underwent impaction bone grafting and structural bone grafting with winged reinforcement cup; 1 case (Paprosky Ⅲ A) underwent double-layer impaction bone grafting with titanium mesh and reinforcement cup.Harris score and Gill classification were used to evaluate clinical and radiological results,respectively.Results All cases (32 hips) were followed up for an average of 22 months (range,12 to 25months).One year postoperatively,Harris score improved from preoperative 44.00±11.71 to postoperative 78.41 ±9.32.Twenty-four cases were excellent,4 good,4 fair,and the excellent and good rate was 87.5％.Three cases occurred mild displacement of acetabular rotation center; one case occurred dislocation.There was no loosing,subsidence,and bone resorption in other 28 cases at final follow-up.ConcLusion Bone grafting with titanium mesh and/or reinforcement cup is effective in reconstruction of acetabular bone defect in THA,which can improve the stability of acetabular cup.

Objective: To investigate the anticancer action of taurine and its potential mechanisms. Methods: 101 weanling female SD rats were randomly divided into four groups. They were:(1)control group (CL):fed a basic diet;(2)high fat group(HF):fed a high fat diet (10% lard);(3)taurine group (Tau):fed a taurine diet (5% taurine);(4)taurine and high fat group(TH):fed a mixed diet (5%taurine and 10% lard). 10mg DMBA were given to each rat by gavage at age of 6 weeks. Then the rats were fed assigned diet for 26 weeks. By 5 weeks after DMBA administration the rats were palpated once weekly and the time, size, location of each tumor were recorded. Objective:To observe the effect of Gemcitabine ( GEM) on the viability and apoptosis of non-small cell lung cancer HCC827 in vitro. Methods:The cell viability,apoptosis and cell cycle of HCC827 cells induced by Gemcitabine were detected with cell counting kit-8 assay (CCK-8),Annexin V-FITC/PI staining and flow cytometry. The expression of Bcl-2 protein of cells treated with GEM was examined by Western blot assay. Results: There was significant inhibition effect on HCC827 cells treated with 0. 1-1 000 ng/ml of GEM,which can promote the occurrence of HCC827 cell apoptosis and arrest cell in the S phrase. The apoptosis induced by GEM was accompanied with the down regulation of Bcl-2 protein. Conclusion: GEM can inhibit the cell viability and induce the HCC827 cell apoptosis and S phrase arrest. Its cell dead type was apoptosis,which was related with the expression of Bcl-2 protein.

Objective To compare the curative effects of two different approaches in the surgical treatmentof small breast.Methods Fourty-seven patients with small breast were treated at our department.Twenty-eight sides of patients adopted an approach of prosthesis while the nineteen sides of patients were treated in the approach of autologous fat granules injection.Clinical assessment was done by analyzing operation time,blood loss volume,hospitalization duration and recent and long-term complications in breasts.All the pa-tients were followed up for thirty-two months on average.Results During follow-up period the appearance ofbreast was satisfied.Silicone gel breast prosthesis was well tolerated and the whole operation time was about (35±5 )min,the amount of blood loss was (35±5 ) mL,hospitalization duration was (2±1 ) days,during thefollow-up period prosthesis capsular contracture after hematoma occurred in one patient.The use of autolo-gous fat granules injection to breast augmentation was found limited,and the whole operate time was about (35±5 ) rain,the amount of blood loss was (15±5 ) mL,hospitalization duration was (1±1 ) days.With in-jection(2±l)times.During the follow-up period,one patient had infection,one patient liquefied fat,andone patient the induration.Conclusion Each treatment approach has its own advantages and disadvanta-ges.The approprite one should be selected according to the special situation of patients.

Objective: Study the morphology and kinetics characters in breast diseases by using helical CT,to assess the value of differential diagnosis in benign and malignant breast diseases.Methods: Forty-three focuses in thirty-eight patients were detected by using double row detector CT with histopathologic correlation.Analyzed the characters of morphology,the time-density curve and the enhancement rate of all focuses.Results: Of the 43 focuses,breast cancers were twenty-eight and benign diseases were fifteen. There were eleven pieces and thirty-two masses.Lobulation signs and spicule signs were twelve respectively,among twenty-one malignant masses.One had spicule sign and two had lobulation sign among eleven benign mas-ses.Twenty breast cancers had Ⅲ type time-density curve,eleven benign diseases had Ⅰ type curve,and there were some overlaps on Ⅱ type curve.Enhancement rate(E) reflected the blood kinetics of breast diseases.E 1min and E 3min were better than E 3min in differentiating benign and malignant breast diseases.Conclusion:Lobulation signs and spicule signs are more often found in malignant lesions than in benign lesions.Early enhancement rate and time-density curve present some important information in differentiation.Synthesizing all these specialties can promote the diagnosis accuracy.

Objective To investigate the relative percentage of normal cognitive function (NCF),age associated memory impairment (AAMI),mild cognitive impairment (MCI) and Alzheimer's disease (AD) in the elderly,and the correlation between cognitive function and ApoE genotypes.Methods A total of 2666 elderly people aged ≥65 years (2132 males and 534 females)were divided into 3 groups according age:65-74-year age group (925 cases),75-84 year age group (1054 cases) and 85-100-year age group (687 cases).ApoE genotypes were determined in the controls and patients with AAMI and MCI.The degrees of fundus arteriosclerosis were detected in all subjects except for patients with AD.Results There were 867 cases with NCF,860 cases with AAMI and 782 cases with MCI.The incidence of AAMI was higher in 65-74-year age group than in the other two groups (42.0％ vs.31.1,20.96).The incidences of MCI and AD were higher in 85-100-year age group than in the other groups (42.5％,13.3％).The major degrees of fundus arteriosclerosis were Ⅰ+,Ⅰ-Ⅱ°,Ⅱ in subjects with AAMI (34.7％,x2=10.02,P＜0.01) and were Ⅱ °/ Ⅱ + / Ⅲ° in subjects with MCI (34.9 ％,x2 =23.39,P＜0.001).The APOEε4 allele frequency was significantly higher in patients with MCI than in the controls (x2=8.31,P＜0.05).However,no significant differences in APOEε4 allele frequency were found between patients with AAMI and the controls.Conclusions The incidence of AAMI is highest in 65-74-year age group,while the incidences of MCI and AD are highest in 85-100-year age group.Compared to patients with AAMI,the more serious fundus arteriosclerosis and higher allele frequency of APOEε4 appear in patients with MCI.

BACKGROUND: Total hip replacement has been widely used in the treatment of severer hip diseases in China.However,most of prostheses are designed according to Caucasian osteometric measurements,whether those prostheses are suitable for Orientals is still in dispute.It is emerging to design prostheses for Orientals,there are few reports addressing therapeutic effect after replacement.OBJECTIVE: To follow up the long-term results of total hip replacement using domestic designed dual-assembly total hip prosthesis.METHODS: Totally 30 patients(33 hips)who underwent total hip replacement at the Department of Orthopaedic Surgery,China-Japan Union/Affiliated Hospital of Jilin University,from June 1987 to December 1996,were followed up.The designed dual-assembly total hip prosthesis was produced by Central Iron & Steel Research Institute,which was similar to bipolar prosthesis with a polyethylene acetabular cup.First generation cementing techniques were applied.The average age at the time of the operation was 56.5 years(30-81 years)and the average follow-up period was 8.5 years(5.0-14.4 years).Standard anteroposterior radiographs of the hip and clinical examinations were used to analyze therapeutic efficacy.RESULTS AND CONCLUSION: The total excellent and good rate was 68.6%.The imaging examination showed that there were16 hips(48.5%)had aseptic loosening and 11 hips(33%)had heterotrophic bone formation.Additionally,two cases suffered from infection and one case suffered from dislocation.The utilization of prosthesis achieved certain curative effects.However,the design and cement technology have problems due to limitation of conditions,which should be solved in further experiments.
