1.Protective effects of amitriptyline on glutamate-induced neurotoxicity in cerebral neuronal cultures

AIM: To study the protective effects of amitriptyline (Ami) on glutamate induced neurotoxicity in cultured rat cerebral cortical neurons. METHODS: Cortical neurons of fetal rat were cultured in vitro. The protective effects of Ami on glutamate induced neurotoxicity were observed. RESULTS: Exposure of rat fetus cerebral cells to Glu medium developed a neurotoxicity, expressed in the increase of LDH, NO and MDA, and the decrease of activity of SOD,as well as the development of morphological injury. Ami (10 -6 ,10 -7 , 10 -8 mol?L -1 ) significantly protected neurons against above demage. CONCLUSION: Ami can prevent rat cerebral neurons injury from the toxicity of Glu.

Objective: To study the anti-inflammatory, analgesic and immunosuppressive action of Niuhuang Qianjin San (NQS). M ethods: The anti-inflammatory action of NQS was studied by the methods of rat pa w swelling and mouse ear swelling and the analgesic action by mouse body torsion method and K+ subcutaneous penetration method in rats.Immunosuppressive effec t was evaluated by peripheral T lymphocyte percentage, splenic lymphocyte transfor mation ratio, phagocytic function of intraperitoneal magocytes, the activity of delayed hypersensitivity, serum hemolysin level and the function of antibody-yie lding cells in mice. Results: NQS could obviously alleviate inflammation, increa se codeine's analgesic action and suppress immunity. The difference were signif icant (P<0.01) as compared with normal saline contr ol group.Conclusion:NQS exerts certain anti-inflammatory,analagisic and immunosu ppressive action.

4.Study on expression of SENP1 and c-myb gene in patients with acute lymphoblastic leukemia

Objective To investigate the SENP1 and c-myb gene expression and their correlations in bone marrow specimens in the patients with acute lymphoblastic leukemia (ALL ) to provide the basis for expounding the role ,mechanism and prognosis of SENP1 and c-myb in ALL .Methods 31 patients diagnosed with ALL (22 cases of B-ALL ,1 case of T-ALL and 8 cases of uncate-gorized ALL ;6 cases in the low/medium risk group ,25 cases in the high risk group) and 31 patients with proliferative bone marrow and hyperplastic anemia diagnosed by the morphology were taken as the control group .The real-time PCR and immunocytochemical staining(SP method) were adopted to detect the mRNA and protein expressions of SENP1 and c-myb in the bone marrow specimens of the ALL patients and the control group .Results The expression of SENP1 and c-myb were both increased in the bone marrow specimens and smears of ALL patients ,which showed the statistical difference compared with the control group (P< 0 .05) ,the Pearson correlation analysis found that the high expression of SENP1 and c-myb had correlation .The expression of SENP1and c-myb in the low/medium risk group were lower than that in the high risk group ,but the difference had no statistical significance . Conclusion The high expression of SENP1 and c-myb exists in the bone marrow specimens of the ALL patients ,SENP1 and c-myb could possibly have the correlation with the occurrence and development of ALL ;but now the differences of SENP1 and c-myb ex-pression among different risk groups of ALL patients are yet to be proven .
5.Study on the Preparation Process of Xiangsha Liu jun Granule

OBJECTIVE: To optimize the preparation process of Xiangsha Liujun Granule. METHODS: By the orthogonal design. taking the rates of recovery of volatile oil, Ginsenoside Re and qualified granules as the indexes, we optimized the oil extracting conditions, the decocting conditions and the conditions of granulation by spayer. RESULTS: The optimum oil ex- tracting conditions for four kinds of herbs(Pericarpium Citri Reticulatae etc.) were that water was 30 times of herbs in weight, soaking time was 3 hours and extracting time 15 hours. The optimum decocting conditions for four kinds of herbs(Fuling etc.) were that water was 15 times of herbs in weight, soaking time was 3 hours and decocting time 4 hours, herbs were decocted twice. The optimum conditions of granulation by sprayer were that the fliud extract was 2 times of Dextrin in weight, the rel- ative density of the fliud extract was 1. 30(measured at 25℃), the voltage of sprayer was 120 volts, the period of shaking bag was 65 times(shaking bag back and forth twice in each period). CONCLUSION: The results of the experiment make it clear that the preparation process of Xiangsha Liu jun Granule is stable and feasible.
6.The signification of p57kip2immunostaining to distinguish molar gestations from hydropic abortions

Objective To study the signification of p57kip2 immunostaining to distinguish molar gestations from hydropic abortions.Methods To observe the p57kip2 immunohistochemical expression in 58 cases of histological hydropic villi which were divided into complete mole(18 cases) ,partial mole(19 cases),hydropic abortion(11 cases)and undetermined hydropic abortion or molar gestations(10 cases) and in 3 normal placentas.Results Normal villi,partial mole and hydropic abortions show positive staining for p57kip2, which expressed in the nuclei of cytotrophoblast and villous mesenchyme,and complete moles show complete absence of staining in the cytotrophoblast and villous mesenchyme.According to the comparison of the diagnosis based on morphology and the one based on p57kip2 stai-ning,the later whose sensitivity is 96% (46/48) confirmed the earlier diagnosis in 58 cases studied and the later conformed to the earlier very well.Conclusion The immunohistochemical staining for p57kip2 is a valuable diagnostic mean to distinguish molar gestations from hydropic abortions and is a sensitive and specific marker for the diagnosis of complete mole.
7.Comparative Study Between AngioLight System and Intravascular Ultrasound for Coronary Diameter Measurement in Experimental Pigs

Objective: Due to the high cost and complicated imaging interpretation of intravascular ultrasound (IVUS), we conducted a comparative study between AngioLight system and IVUS measurement of coronary diameter to explore the safety and efifcacy of AngioLight performance in experiment pigs.
Methods: There were 3 reference target points established in left anterior descending (LAD), left circumlfex (LCX) and right coronary arteries (RCA) in 3 Chinese mini-pigs. Each coronary branch had①target point, ②proximal target point, which was (10-15) mm up to target point,③distaltarget point, which was (10-15) mm down to target point. The coronary diameters were measured and compared by both IVUS and AngioLight system. Upon stent implantation, the diameters were measured again by both IVUS and AngioLight system at distaltarget point, stent edge of distal target point and proximal target point, stent edge of proximal target point respectively. And results comparison between AngioLight and IVUS were also compared.
Results: In 25 measurements, neither adverse complication nor death were occurred during the study. There were no significant statistical differences between IVUS real-time max diameter and AngioLight catheter max diameter measurements (2.57 ± 0.54) mm vs (2.70 ± 0.49) mm,P=0.09, as well as between IVUS analyzed max diameter and AngioLight catheter max diameter (2.68 ± 0.6) mm vs (2.59 ± 0.42) mm,P=0.37. In addition, good correlations were observed (r= 0.73,P<0.001) and (r= 0.67,P=0.001) respectively.
Conclusion: Angio Light system has good safety and correlation with IVUS for quantitative measurement of coronary diameter in experimental pigs.
8.Nutritional risk assessment and inspiration for patients in chemotherapy after radical gastrectomy

Objective To investigate the nutritional status of patients with chemotherapy after radical gastrectomy and to provide dietary guidance and nutritional support for patients in time. Methods Patients who received chamotherary after radical gastrectomy inthe Gastrointestinal Surgery of the First Affiliated Hospital of Zhengzhou University from September 2014 to March 2016 were included in the study. The nutritional status was assessed by unified training investigators within 48 h after hospitalization. The questionnaires of Scored Patient-Generated Subjective Global Assessment (PG-SGA) and the Chemotherapy Side Effects of Patients with Gastric Cancer Questionnaire were used to investigatethe patients by trained investigators. Results The scores of the nutritional status of patients with different stages of chemotherapy were 6.107 ± 2.947, 6.505 ± 3.134, 8.068 ± 3.748, which was statistically significant (F=10.202, P<0.01). The incidence of anorexia was the highest rate of diet-related symptoms, the rate of early, mid-term and later chemotherapy rates were 34.95%(36/103), 56.31%(58/103), and 75.73%(78/103) respectively. Patients acquired the diet knowledge mainly from medical staff. The proportions of early, middle and later stages of chemotherapy were 92.23% (95/103), 79.61% (82/103), 75.73%(78/103). Conclusions With the increase of chemotherapy times, the nutritional status of patients with radical gastrectomy is getting worse. The results suggested that medical staff should provide an effective basis dietary guidance in time, improving the patients′nutritional status and clinical outcomes.
9.Isolation and purification of human coagulation factor Ⅶ from Cohn fraction Ⅲ paste

Objective To isolate and purify human coagulation factor Ⅶ from Cohn fraction Ⅲ precipitate.Methods The purification procedure of human factor Ⅶ from Cohn fraction Ⅲ precipitate involves dissolving fraction Ⅲ,absorbing factor Ⅶ onto barium citrate and eluting,ammonium sulfate fractionation and DEAE-Sepharose Fast Flow ion exchange chromatography,and Sephadex G-100 gel filtration chromatography.Results 10.1mg purified FⅦ was obtained from 400g Cohn fraction Ⅲ precipitate.The purified FⅦ has a specific clotting activity of 1775.8U/mg and the overall yield of FⅦ specific clotting activity is 17.6% of the starting material.The purity of FⅦ was judged by SDS-PAGE and there was only one protein band on the gel.Conclusion The procedure of purifying Ⅶ from Cohn fraction Ⅲprecipitate is established with satisfactory purity.
10.The nutritional status of radical gastrectomy patients during the late course of chemotherapy and its main influencing factors

Objective To explore the nutritional status of radical gastrectomy patients during the late course of chemotherapy and its main influencing factors, providing reference for the development of targeted nursing intervention. Methods The Scored Patient-Generated Subjective Global Assessment, The Chemotherapy Side Effects of Patients with Gastric Cancer Questionnaire were used to collect clinical data that received complete chemotherapy after radical surgery from September 2014 to March 2016. Results A total of 103 cases finally involved in this study. The incidence of malnutrition in patients during pre-chemotherapy and late course of chemotherapy was 45.63%(47/103) and 73.79%(76/103) respectively, and there was significant difference (χ2=16.97, P<0.05). The nutritional assessment score, the hemoglobin levels and body mass index of patients pre-chemotherapy was (3.64±0.78) points, (124.11± 21.78) g/L, (21.29±2.77) kg/m2 , (8.07±3.75) points, (108.54±11.68) g/L, (20.29±2.68) kg/m2 late course of chemotherapy and there were significant differences(t =-11.74, 6.39, 2.63, all P<0.05). The symptom of depression was at a higher level 65.05%(67/103), patients with anorexia in late course of chemotherapy accounted as much as 75.73%(78/103). Logistic regression analysis showed that educational level, loss of appetite and parenteral nutrition were the main factors which influenced the nutritional status of patients during the late course of chemotherapy. Conclusions With the increase in the number of chemotherapy, the nutritional status of the patient is getting worse. The study suggests medical personnel should pay more attention to the patients with low educational level, give patients and their families targeted diet counseling, correct the mistaken conceptions thus enhance their understanding of dietary knowledge. Strengthening the management of complications during chemotherapy, reducing the adverse factors that affect the patient′s nutritional status to improve the nutritional status of patients.
