1.Optimization of the Extraction and Purification Technology of Resveratroi from Polygonum Cuspidatum by Orthogonal Experiment

OBJECTIVE:To optimize the extraction and purification technology of resveratrol from Polygonum cuspidatum. METHODS:By alkali extraction and acid precipitation,an orthogonal experiment was conducted to optimize the extraction and purification technology taking the ratio of the CaO to the raw material,the ratio of solid material to the extraction solvent,the kinds of acid and the extraction time as factors and the weighted mean of the content and yield as comprehensive score index.RESULTS:The optimal extraction and purification technology was as follows:the ratio of CaO to the solid material was 9:100;the ratio of solid material to solvent was 1:8;the acid was hydrochloric acid and the extraction time was 45 min.CONCLUSION:The optimized extraction and purification technology of resveratrol from Polygonum cuspidatum is characterized by energy source - saving and simple in operation,and it is suitable for industrial production.
2.Clinical significance of human telomerase RNA component and C-myc gene expression in cervical cancer and precancerous lesions

Objective To investigate the expression level of human telomerase RNA component (hTERC) and C-myc in cervical lesions.Methods Fluorescence in situ hybridization (FISH) was applied to detect hTERC and C-myc expression in 62 cases of cervical tissue including 35 cases of cervical intraepithelial neoplasia,8 cases of invasive cervical cancer,19 cases of inflammation as controls.Results The positive rates of hTERC on chromosome 3 in chronic cervicitis organizations,CIN Ⅰ,CIN Ⅱ-Ⅲ and cervical cancer were 5.3 % (1/19),16.7 %(2/12),87.0 % (20/23),87.5 % (7/8) (x2 =36.299,x2 =40.237,P <0.01).The positive rates of C-myc in chronic cervical inflammation organization,CIN Ⅰ,CIN Ⅱ-Ⅲ and cervical cancer were 5.3 % (1/19),8.3 % (1/12),78.3 % (18/23),62.5 % (5/8) (x2 =30.200,x2 =34.224,P <0.01 ).The differences among groups were statistically significant.hTERC had a positively correlation with C-myc expression (r =0.514,P < 0.01).Conclusion The expression of hTERC and C-myc on chromosome 3 is closely related to cervical cancer progression.
3.The Treatment of Tracheal Stricture and Tracheal Fistula by Means of the Covered Tracheal Stent

Objective The covered tracheal stent in shape of a straight tube or a fork-like tube was placed clinically to treat tracheal stricture and tracheal fistula caused by malignant tumour in order to maintain the normal function respiration and to keep the fistula closed.Methods The stent was made of stainless steel in shape of "Z"covered with membrane partially or entirely.An instrument with tube in three sheathes was used to place the stent in position.Results The tracheal stents were placed successfully in 9 cases at first time and the esophageal stents were placed for 6 cases among them in the meantime.Dyspnea of patiants was remarkably alleviated.The tracheal fistula of them was closed effectivelly.Follow-up for 4~12 monthes all cases demonstrated no adverse effects.Conclusion The application of the covered stent is a safe effective method to treat the tracheal stricture and tracheal fistula.
4.Application value of focused assessment with sonography for trauma in treatment of emergency thoracic and abdominal closed injury

Objective To evaluate the application value of focused assessment with sonography for trauma (FAST) in treatment of emergency thoracic and abdominal closed injury.Methods FAST examination was performed in 85 patients with thoracic and abdominal closed injury,to determine whether the thoracic and abdominal cavity,pericardial free effusion and abdominal parenchyma organ injury,and the results were compared with conventional ultrasonography,CT scan and operation results.Results FAST was (2.85 ± 1.15) min,conventional ultrasonography was (15.48 ±5.17) min,there was statistically significant difference (t =21.99,P < 0.05).In 85 patients,10 cases were FAST positive,5 cases of abdominal cavity in that amount or a large number of free effusion,underwent emergency operation,postoperative intra abdominal parenchymal organs were confirmed as rupture and hemoperitoneum; 2 cases of renal rupture with perirenal hematoma,confirmed by the whole abdominal CT examination; 2 cases of thoracic free pneumatosis,effusion,confirmed by thoracic CT examination or closed thoracic drainage; 1 case of prompt hemopericardium,confirmed by the pericardium puncture.Conventional ultrasonography was positive in 11 patients,no statistically significant difference between them (P > 0.05).The sensitivity,specificity,accuracy rate of FAST screening thoracic and abdominal closed injury was 10/12,100.0% (73/73),97.6% (83/85).Conclusions FAST has high sensitivity and specificity for thoracic and abdominal cavity,pericardial free effusion,check with short time,low environmental requirements,without moving the trauma patients,and may be out of sync with the fluid resuscitation and other emergency measures.For large quantities of trauma patients and patients with severe multiple trauma,FAST can quickly,accurately determine the existence of thoracic and abdominal closed injury,and provide guidance for clinical treatment,which has good application value.

6.Effect of PQQ on the hippocampal neurons of aging rat induced by D-galactose

Objective To investigate the effects of pyrroloquinoline quinone(PQQ) on the hippocampal neurons of aged rats induced by D-galactose(D-gal).Methods D-gal was used to induce the model of aging rat,PQQ was administered into rat lateral intracerebroventricle.After 50 days the metamorphosis of hippocampal neurons was observed by H-E and Nissl's staining.The apoptosis rate of hippocampus was tested by flow cytometry.The contents of free radical and C-FOS protern were measured.Results Compared with the control group,the size of the neurosoma was slightly changed,the optical density of Nissl's was decreased,the content of free radical and the apoptosis rate increased markedly in D-gal group.After PQQ injection with D-gal,the size of neurosoma and the optical density of Nissl's were markedly increased,the content of free radical and the apoptosis rate of hippocampus did not change.PQQ improved the expression of C-FOS protern.Conclusion PQQ can slow down the aging progress of hippocamal neurons induced by D-gal.
7.The Clinical Application of Esophageal Stent of the Curved Type

Objective:Since 1996,the esophageal stent of the curved type has been applied clinically by authors in order to solve the defects which the straight tube like stent could not coincide with the shape of the esophageal of the patient.Methods:The curved type stent made of "Z" type stainless steel was implanted after fixing it direction along the selected esophagus.Results:The application of the esophageal stent of the curved type in 29 cases indicated that the stents were coincidence with the folded esophagus,could decreases the pain and discomforts of the patients,and improved the stability of the stents.Conclusion:The esophageal stent of the curved type applied to the folded esophagus is much more effective clinically than the straight tube like type used before. 8.Role and mechanism of Astragalus in alleviating adhesion of neutrophils to human renal tubular cells induced by postasphyxial-serum of neonate

Objective To investigate the role of Astragahs in alleviating adhesion of neutrophils to HK-2 cells induced by postasphyxial-serum of neonate and its signal transduction mechanism. MethodsHK-2 cells were used as target cell. Control group, asphyxia group, Astragalus group were divided in the experiment. The 20% (volume fraction) postasphyxial-serum was used as attacking factor. The following indicators were detected: cellular morphology and neutrophils adhesion were observed with inverted microscope. The activity of myeloperoxidase (MPO) in the cell suspension were determined by biochemistry assay as the indicator for adhesion of neutrophils to HK-2. Intercellular adhesion molecule-1(ICAM-1) were examined by flow eytometer. ResultsThe neutrophils numbers (MPO activity) and ICAM-1 expression in asphyxia group were higher than that of the control group, but dramatically decreased in Astragalus group (P < 0.05). ConclusionsThese data demonstrated that Astragalus could alleviate the adhesion of neutrophils from neonate with asphyxia to HK-2 and it' s intracellular signal transduction mechanism is presumably involved in the inhibition of ICAM-1 expression on HK-2 cellular membrane.
