2.Effect of different concentrations of calpain inhibitor ALLN on proliferation and apoptosis of C2C12 myoblasts

Objective To explore the effect of different concentrations of ALLN on proliferation and apoptosis of C2C12 myoblasts.Methods After intervention with Ca2+ and ALLN,methyl thiazolyl tetrazolium and flow cytometry were used to determine the effect of Ca2+ and ALLN on the proliferation and apoptosis of C2C12 cells,respectively.The morphological changes of C2C12 myoblasts were observed using Giemsa staining.Results The absorbance of Ca2 + group was significantly lower than that of the control group (P <0.05).After 6,12,24,36 hours of intervention,the absorbance in ALLN groups 1 to 7 (cultured in serum-free media containing 16 mmol/L Ca2+ and ALLN at final concentrations of 3.125,6.25,12.5,25,50,100,200 μmol/L) were all significantly higher than that in the 16 mmol/L Ca2+ group (after 6 hours:0.449±0.024,0.472±0.022,0.513 ±0.008,0.540±0.014,0.588±0.016,0.607±0.030,0.700±0.020 vs.0.355 ±0.012,all P =0.000; after 12 hours:0.407 ±0.007,0.414 ±0.006,0.434 ±0.004,0.441 ±0.003,0.460 ±0.010,0.484 ± 0.006,0.525 ± 0.006 vs.0.368 ± 0.027,all P =0.000; after 24 hours:0.436±0.005,0.431 ±0.015,0.441 ±0.006,0.459 ±0.013,0.527 ±0.009,0.581 ±0.005,0.599 ±0.011 vs.0.386 ± 0.007,all P =0.000 ; after 36 hours:0.464 ± 0.022,0.460 ± 0.018,0.461 ± 0.007,0.434 ± 0.020,0.454 ± 0.028,0.479 ± 0.006,0.524 ± 0.011 vs.0.379 ± 0.011,all P =0.000),while no significant differences were observed after 48-72 hours of intervention.After treatment for 36 hours,the apoptosis rate in ALLN 10,50,100,and 200 μmol/L groups were (6.00 ± 1.20) %,(5.02 ± 1.13) %,(4.89±1.11)%,and (2.71 ± 1.15)%,all significantly lower than that in the Ca2+ group [(13.70 ±2.30)%] (all P =0.000).Giemsa staining showed apoptotic morphological changes in the Ca2+ group,which were obviously alleviated in the ALLN group.Conclusions Ca2+ at a concentration of 16mmol/L can induce apoptosis of C2C12 cells.In contrast,ALLN can inhibit cell apoptosis and promote proliferation in a time-and dose-dependent manner.
3.Therapeutic Observation of Moxa Stick Moxibustion for Diabetic Gastroparesis Due to Deficient Cold of Spleen-stomach

Objective To observe the clinical efficacy of moxa stick moxibustion in treating diabetic gastroparesis (DGP) due to deficient cold of spleen-stomach, and to discuss the action mechanism.Method Seventy DGP patients were randomized into a treatment group and a control group, 35 cases each. The treatment group was intervened by moxa stick moxibustion, while the control group was given oral administration of Domperidone tablets, 7 d as a treatment course, totally for 2 courses. The DGP clinical symptoms evaluation scale, gastric emptying time and biochemical indexes (contents of glycosylated hemoglobin and serum motilin) were compared before and after the treatment, and the clinical efficacies were compared between the two groups.Result After the treatment, the DGP clinical symptoms evaluation scales, gastric emptying time, and biochemical indexes were changed significantly in both groups (P<0.05). The DGP clinical symptoms evaluation scales, gastric emptying time, and biochemical indexes in the treatment group were significantly different from those in the control group after the treatment (P<0.05).Conclusion Moxa stick moxibustion can improve the quality of life of DGP patients due to deficient cold of spleen-stomach, which is possibly related to the improvement of the glycosylated hemoglobin, serum motilin and gastric emptying time.
4.Upper airway morphology and function study of middle-age and old women with OSAHS

20/h. In the regression equationthat AHI was as the dependent, only volumetric change rate of velopharynx had statistic significance. CONCLUSION Middle-age and old women with OSAHS have obvious anatomy abnormalities which are the morphological foundation of the OSAHS. Higher compliance of upper airway plays an important role in the OSAHS pathogenesis in middle-age and old women.
5.The studies of cobalt nanoparticles on the toxicity and biological activity of osteoclast

Objective To explore the toxicity and biological activity of cobalt nanoparticles (CoNPs) on osteoclasts,and to analyze the relationship between cobalt nanoparticles and osteolysis.Methods From November 2014 to July 2015,RAW264.7 cell was induced to osteoclast-like cell by LPS.Different concentrations of cobalt nanoparticles and cobalt chloride were added,and the cell morphology was observed under a microscope.24 h after induction on RAW264.7,cells were grouped into cobalt nanoparticles group (10,20,50,100 μmol/L),cobalt chloride group (10,20,50,100 mol/L) and control group.MTT assessment and Q-PCR were performed at 2 h,4 h,8 h,24 h,48 h post-treatment.Results With the increase of concentration (10,20,50,100 μmol/L) and the action time (2 h,4 h,8 h,24 h,48 h),the inhibition rate of cobalt nanoparticles and cobalt chloride on osteoclast like cells was significantly increased,and the inhibition rate of cobalt nanoparticles was higher.With different concentrations (10,20,50,100 μmol/L) of CoNPs and cobalt chloride,the relative expression of CAⅡ,Cat K gene mRNA expression decreased compared with the control group,when the concentration of CoNPs was in the range of 10-50 μ mol/L,the relative expression of CAⅡ and Cat K was increased,which was reduced in cobalt chloride group.Conclusion Different concentrations of cobalt nanoparticles and cobalt chloride can inhibit the proliferation and differentiation of osteoclasts,and cobalt nanoparticles is more pronounced,when the concentration of cobalt nanoparticles was 10-50 μmol/L,the relative expression of osteoclasts CAⅡ,Cat K increaseed,which was suppressed at the same concentration of cobalt chloride.
6.Chemical constituents from the leaves of Anisopus mannii and the melanogenesis inhibitory activities

The phytochemical studies on the leaves of Anisopus mannii led to the isolation of seven compounds by silica gel, ODS, DIAION HP-20, Sephadex LH-20 colunmn chromatographer, their structures were elucidated on the basis of the spectroscopic analyses(NMR, HRMS)and the comparisons with the literatures as 3β-acetoxylup-20(29)-ene(1), 1-acetoxy-2-isopropyl-1-tridecene(2), rutin(3), 3, 6′-diferuloylsucrose(4), 3-O-β-D-glucopyranosyl-3-β-hydroxyolean-12-en-28-oic acid 28-O-[α-L-rhamnopyra- nosyl-(1→2)-β-D-glucopyranosyl] ester(5), conduritol A(6), hoyacarnoside I(7). Meanwhile, the isolated compounds were evaluated for their inhibitory activities against melanogensis in B16 melanoma cells, as the results, all compounds exhibited melanogenesis-inhibitory activity and compound 5 showed a strongest activity(Melanin content: (27. 4±3. 5)%, Cell Viability: (54. 9±5. 6)% with a concentration of 30 μmol/L)which could be further developed.

Objective To analyze the application of multi‐slice spiral CT and 3D airway reconstruction technique on obstruc‐tive sleep apnea syndrome (OSAS) of pharyngeal soft tissue and upper airway stenosis .Methods Selected 92 patients with OSAS in our hospital as the observation group and another 92 healthy persons were selected as control group ,all patients achieved multi‐slice CT and 3D airway reconstruction technical inspections ,counted and compared their pharyngeal soft tissue and upper airway stenosis .Results Airway stenosis of tongue back ,and hypopharynx retro‐palatal of control group were more narrow compared with the control group(P<0 .05);the observation group′s pharyngeal soft tissue was more thick than the control group ,so as the length and width(P< 0 .05) .Conclusion Multi‐slice spiral CT and 3D airway reconstruction technique can effectively share pharyngeal soft tissue and upper airway stenosis of patients with OSAS .
8.2019 Novel Coronavirus (COVID-19) Pneumonia: Serial Computed Tomography Findings

From December 2019, Coronavirus disease 2019 (COVID-19) pneumonia (formerly known as the 2019 novel Coronavirus [2019-nCoV]) broke out in Wuhan, China. In this study, we present serial CT findings in a 40-year-old female patient with COVID-19 pneumonia who presented with the symptoms of fever, chest tightness, and fatigue. She was diagnosed with COVID-19 infection confirmed by real-time reverse-transcriptase-polymerase chain reaction. CT showed rapidly progressing peripheral consolidations and ground-glass opacities in both lungs. After treatment, the lesions were shown to be almost absorbed leaving the fibrous lesions.

10.Changes in sleep and psychological characteristics of patients with insomnia under exposure to noise pollution

Objective Exposure to nocturnal traffic noise at the same levels can produce different effects on sleep and psychological health in different populations. This study compared the electroencephalographic characteristics during sleep of people with insomnia and those with normal sleep quality under the same levels of nocturnal traffic noise exposure to elucidate the different effects of noise pollution on different populations,and to provide individualized precise treatment plans for people with insomnia who are sensitive to noise pollution. Methods We selected people who lived near highways,railways,and aircraft routes for more than six months where noise levels were relatively fixed(50-60 decibels,10-15 seconds per time) during the nighttime sleep period (22:00-6:00). We included 26 people with normal sleep quality and 30 people with insomnia based on the Pittsburgh Sleep Quality Index. The Self-rating Anxiety Scale (SAS) and Self-rating Depression Scale (SDS) were administered. Polysomnography (PSG) was used to monitor the spindle density index during sleep. The sleep quality and psychological status of the two groups were compared. Results There were no significant differences in demographic characteristics between the two groups under the same nocturnal noise exposure conditions. The incidence of anxiety and depression was significantly lower in the people with normal sleep quality than in those with insomnia(P<0.01). Among the 26 people with normal sleep quality,5(19.23%) had mild anxiety,and no one had moderate to severe anxiety or depression. Among the 30 people with insomnia,6(20.00%) had mild anxiety,15 (50.00%) had moderate anxiety,and 4(13.33%) had severe anxiety;3(10.00%) had mild depression,9(30.00%) had moderate depression,and 3(10%) had severe depression. PSG showed that the sleep spindle index of the normal sleep group was significantly higher than that of the insomnia group(25.43 vs 16.60,P<0.05),with no significant differences in the average duration of sleep spindles during sleep between the two groups (P>0.05). Conclusion Under the same levels of nocturnal traffic noise exposure,people with insomnia have a higher incidence of anxiety and depression than people with normal sleep quality. Patients with insomnia with a reduced spindle density index have worse sleep quality under exposure to noise pollution.
