1.Inflammatory cytokines and Alzheimer disease
Chinese Pharmacological Bulletin 1987;0(01):-
Inflammatory response clearly occured in pathologically vulnerable regions of the Alzheimers disease (AD) brain. Many proinflammatory mediators, such as IL-1, IL-6,TNF-? and TGF-?, were significantly up-regulated in the pathophysiological process of AD. In addition to their traditional actions as proinflammatory molecules, AD-specific interactions of the cytokines and amyloid ?(A?) may be pathophysiologically relevant, and there was a reciprocal relationship within the cytokines inducing and being induced by A?.

Objective To compare the accuracy of nucleic acid detection and antibody detection for diagnosing human immunode‐ficiency virus(HIV) infection .Methods Retrospectively analysed data of nucleic acid detection and antibody detection from 124 ca‐ses of patients diagnosed with HIV infection from 2005 to 2014 .The positive rates of the two methods were compared respectively in patients with early‐stage of HIV infection(76 cases) and patients with intermediate and advanced stage of HIV infection (48 ca‐ses) .Results In patients with early‐stage of HIV infectionn ,the positive rate of nucleic acid detection (94 .74% ) was higher than that of antibody detection (84 .21% );while in patients with intermediate and advanced stage of HIV infection ,the positive rate of antibody detection(97 .92% ) was higher than that of nucleic acid detection (81 .25% );both had statistically significant difference (P<0 .05) .Conclusion On the early stage of HIV infection ,the accuracy of nucleic acid detection is higher than that of antibody detection ;while on the intermediate and advanced stage of HIV infection ,antibody detection shows better accuracy .

Objective To evaluate the usefulness of contrast enhanced ultrasound in the diagnosis of breast lesions. Methods Contrast enhanced ultrasound was performed after administration of SonoVue in 104 patients scheduled for surgical tumor removal. The enhancement morphology was classified as non, peripheral, homogeneous, regional,or heterogeneous enhancement based on distribution of enhancing areas. All breast lesions enhancement morphology were analyzed, and the diagnostic value of contrast enhanced ultrasound was calculated. Results The prevalence of non enhancement was significantly higher in benign lesions than in malignant ones(P = 0.013),and the prevalence of peripheral enhancement was significantly higher in malignant lesions than in benign ones (P = 6.02 × 10-7). Conclusions Contrast enhanced ultrasound is useful in diagnosis of breast lesions.

Objective Diallyl disulfide ( DADS) has achieved remarkable effects in treatment and research of diverserfied cancers. The article was to explore the effects and the mechanism of DADS on the xenograft growth of human small cell lung cancer ( SCLC) cells in nude mice . Methods A total of 25 nude mice were selected to establish xenograft model of NCI-H446 human SCLC cells. The nude mice bearing with SCLC H446 were divided into 5 groups by random selection:positive control group(DDP 66 mg/kg), negative control group(physiological saline), 20 mg/kg DADS group, 60 mg/kg DADS group and 180 mg/kg DADS group, which is 40.6%, 53.1%and 66.4%, respectively. The growth of xenograft tumor in mice was observed after being treated with differ-ent concentrations of DADS .The morphological changes of the tumors were examined under light microscopy .Phase distribution and apoptosis of xenograft cells were analyzed by flow cytometry ( FCM) . Results The growth of xenograft tumor were inhibited signifi-cantly by DADS, resulting in decreased cell density and cellular atypia .Moreover, xenograft cell cycle was blocked in G 2/M and cell apoptosis rate was enhanced . Conclusion DADS can significantly inhibit the growth of NCI-H446 cells and lead to apoptosis .

Objective To establish a rapid method of real-time fluorescence RT-PCR to quantify Soul virus. Methods The pro-fessional software was adopted to design the primer and the TaqMan-BHQ probe. With artificially synthesized L gene segment as the template of Soul virus, the real-time RT-PCR for detecting Soul virus was researched. Results The Ct value of templates had a good linear relationship with the log value of the template diluted concentration. The standard curve was Y =-3.607X +41.84, r2 =0.998,the PCR amplification efficiency was 108.1%,its lowest detection limit was 53.2 copies/μL.Conclusion Applying the real-time fluorescence RT-PCR by the TaqMan-BHQ probe for detecting nucleic acid of Seoul virus has the characteristics of short time-consuming and high sensitivity.

Objective To observe the curative effect of adult ischemic necrosis of femoral head by intramedullary decompression and arterial pulse pressor infusion. Methods 82 patients with ischemic necrosis of femoral head were treated in the combined treatment of percutaneous intramedully decompression and pulse pressor five medicine combined infusion in target vessels using ZC-Ⅰ-discome.Results By follow-up for 12~48 months, the painful in hip joint was alleviated and joint movement was also improved. The regenerative vessels and bone in femoral head and neek were obvious. Conclusion This therapcutic methods is safe and superior to single interventional medicine treatment for ischemic necrosis of femoral head.

In comparison with the clinical PET scanner,PET scanner for small animals puts forward a higher demand in spacial resolution and sensitivity. This article describes the development of the PET scanner for small animals and compares the characteristics of several kinds of contemporary typical PET scanners for small animals in different aspects based on the development of detectors. Meanwhile,it also introduces the development of the detectors designed for measuring depth of interaction (DTC) to reduce parallax error.

Objective:To construct a recombinant retroviral vector carrying human inducible costimulator (ICOS) gene, screen for CHO cell line stably expressing ICOS protein and to study its biological activity. Methods: ICOS cDNA was obtained from human peripheral blood mononuclear cells (PBMC) through RT-PCR and was cloned into retroviral vector to construct retroviral recombinant pMSCV-ICOS; the latter was then packed and the high-titer virus producing cells were screened. Then CHO cell was infected by this high-titer virus and the stable cell line was screened.CHO-ICOS cells were co-cultured with PBMC (the ratio of CHO-ICOS to PBMC being 11,12,15, and 110) in presence of substimulating dose of anti-human CD3 antibody. The proliferation of PBMC and the CD25 expression on T cells were examined by 3H-TdR incorporation method and flow cytometry,respectively.CHO-pMSCV cells co-cultured with PBMC (11) served as the negative control and PBMC served as blank control. Results: We successfully constructed the retroviral recombinant pMSCV-ICOS and obtained CHO cell line stably expressing ICOS protein.3H-TdR incorporation method and flow cytometry showed that, compared with the negative control group and the blank control group,co-culture with CHO-ICOS cells significantly inhibited the anti-CD3 antibody-induced activation and proliferation of PBMC(P

China Pharmacist 2016;19(3):571-573
Curcumin is the main active component in turmeric, which possesses many pharmacologic effects, including anti-in-flammatory, antioxidant, anti-tumor, anti-atherosclerosis, liver and kidney protection and so on. However, due to its poor bioavail-ability, its clinical application is limited. Therefore, the methods for improving the bioavailability of curcumin were reviewed by refer-ring to a large number of literatures. The bioavailability of curcumin can be improved by different administration routes and various dos-age forms. The review provides theoretical basis and research ideas for the development of new drugs.
