2.Establishment of a Mouse Model of Alzheimer Dementia

Objective To establish a Alzheimer dementia(AD) model in mice. Methods The C57BL/6 mice were lesioned with ibotenic acid in Nucleus basalis of Meynert(NBM). Behavioral tests by eight-arm radial maze were conducted 8 weeks, and immunohistochemical staining of choline acetyltransferase(ChAT), serotonin(5-HT), GAD(GABA), amyloid-?protein (AP) was conducted 12 weeks after NBM lesioning. Results In NBM lesioned mice, the ChAT-positive neurons, serotonin-positive neurons, and GAD-positive neurons in right NBM reduced, and ChAT-positive neurons reduced most evidently. At the same time, the ChAT-positive fibers in prefrontal and parietal cortices decreased significantly, serotonin-positive axons slightly, accompanied by heavily AP co-expression. On the contrary, there was no change of GAD-positive neurons in cortex. The working memory error increased significantly.Conclusion Ibotenic acid lesioning in NBM can provide as a model of AD in that it produces deafferentation of cholinergic system and recent memory disruption.

5.Laparoscopic Cholecystectomy Combined with Endoscopic Retrograde Cholangiopancreaticography for Gallbladder and Bile Duct Stones

Objective To investigate the efficacy of laparoscopic cholecystectomy(LC) followed by endoscopic retrograde cholangiopancreaticography(ERCP) for gallbladder and bile duct stones.Methods A total of 58 patients with both gallbladder and bile duct stones received endoscopic sphincterotomy(EST) in our hospital.After the stones were removed,LC was performed on the patients.Results Of the 58 patients,56 were successfully treated with EST and LC.Two cases were unsuccessful owing to difficulty in intubation during ERCP in one and failure to remove all the stones from the bile duct in the other.These two patients were then cured by LC combined with electronic choledochoscopy.Perioperative complications included bleeding during EST in 1,acute edematous pancreatitis in 2,and mild infection of the puncture site in 1.The overall complication rate was 7.1%(4/56).Retained common bile duct stone was observed in 1 patient,and was successfully removed by subsequent ERCP.Conclusions LC combined with ERCP is a safe and effective minimal invasive method for the treatment of concurrent gallstones and bile duct stones.

Objective To study the mRNA expression of Toll-like receptor (TLR) 2 and 4 in human urinary tract epithelium. Methods The epithelial cells of nephric tubule, pelvis, ureter and bladder were cultured in vitro, and their mRNA expressions of TLR2 and TLR4 were measured by RT-PCR. Results The mRNA expression of TLR2 in nephric tubule, pelvis, ureter and bladder was 0.326, 0.589, 0.602 and 0.735, and that of TLR4 was 0.223, 0.327, 0.379, 0.365, respectively. Objective:To investigate the high risk factors of pelvic lymph node metastasis in patients with type Ⅰ endometrial carcinoma in order to provide the basis for making reasonable operation scope.Methods:Risk factors of pelvic lymph node metastasis were analyzed in 136 cases of type Ⅰ endometrial carcinoma.Univariate analysis was performed with Chi square test or Fisher's exact probability method,and multivariate analysis was performed with a logistic regression mode.Results:The positive rate of pelvic lymph nodes in 136 patients with type Ⅰ endometrial carcinoma was 9.56% (13/136).Univariate analysis showed that histological grade,size of lesion,depth of myometrial invasion and vascular invasion were related to lymph node metastasis(P <0.05);Multivariate Logistic analysis showed that low differentiation,deep muscular invasion,tumor diameter≥2 cm and LVSI were independent risk factors of pelvic lymph node metastasis in patients with type Ⅰ endometrial carcinoma(P <0.05).Conclusions:The rate of pelvic lymph node metastasis is low in type Ⅰ endometrial carcinoma.Patients with low differentiation,deep muscular invasion,tumor diameter≥2 cm and LVSI are more likely to occur pelvic lymph node metastasis in type Ⅰ endometrial carcinoma.
8.ADR Reports:Analysis of 93 Cases in Our Hospital

OBJECTIVE: To study the regular pattern and characteristics of adverse drug reactions(ADR) in our hospital.METHDOS: 93 ADR cases collected from Jan.1,2006 to Dec.31,2006 in our hospital were analyzed statistically in respect of drug categories,Sex and age distribution,routes of administration,organs and systems involved and clinical manifestations etc.RESULTS: Of the 93 ADR cases,the patients aged above 60 showed the highest proportion(47.3%);31.2% were induced by anti-infective drugs;69.9% were induced through intravenous way;and damages of skin and its appendants were the most common manifestations(35.5%).CONCLUSION: Anti-infective drugs should be used rationally to avoid or reduce the occurrence of ADR.

ObjectiveTo detect the content of vascular endothelial growth factor(VEGF)in aqueous humour and in vitreous of patients with proliferative diabetic retinopathy(PDR).MethodsEnzyme linked immunosorbent assay(ELISA)was used,which is one of the basic methods of protein quantitative analysis.ResultsThe VEGF levels in aqueous humour of PDR(mean level 398.6 pg/ml)were higher than those of the controls(mean level 106.5 pg/mlP<0.05).The VEGF levels in vitreous of the four PDR(mean level 1395.2 pg/ml)were higher than those in aqueous humour of the four PDR(P<0.05,Primer t test).The increased VEGF level in vitreous was positively correlated with the level in aqueous humour.In the normal group,the mean value of VEGF in the vitreous was 64.6 pg/ml.In comparison with that of 1140.4 pg/ml in the PDR group,there was a significant difference between the two groups (P<0.05,Primer t test).ConclusionVEGF might play an important role in the pathogenesis of neovascularization of PDR.
10.The effect of Fas gene transfection on bladder cancer cells

Objective To study the effect of Fas gene transfection on bladder cancer cells. Methods Fas cDNA were transduced into bladder cancer cells EJ by DOTAP liposomal transfection regeant. Northern blot,in situ hybrization and flow cytometric evaluation were used to confirm the Fas mRNA and protein expression. The apoptosis and proliferation of EJ cells pre and posttransfection induced by cisplatin were analysed by flow cytometry、DNA ladder and MTT methods. Results Transfection of Fas gene can significantly upregulate the expression of Fas in human bladder cancer EJ cells. Apoptosis and decrease of proliferation were easily induced by cisplatin in the transfected EJ cells. Conclusions Fas system might involve in the development and progression of urogenital malignant tumors. Transfection of Fas gene by lipofectin can significantly upregulate the Fas expression in target cell. The synergistic cytotoxic effect obtainted in EJ cells suggested that combined use of Fas gene transfection and cisplatin may help in the treatment of cisplatin resistant bladder cancer.
