1.CCl_4 promotes the proliferation and expression of TGF?1 in rat hepatic stellate cells
Basic & Clinical Medicine 2006;0(02):-
Objective To investigate the molecular mechanism of CCl_(4) inducing fibrosis in hepatic stellate cells. Methods The expression of Transforming growth factor-?1(TGF-?1) at mRNA and protein level were detected by RT-PCR method and Western blot analysis. The proliferation and cell cycle distribution were detected with MTT method and flow cytometry. Results After treatment by(10 mmol/L) CCL_(4) for 24 hours, the mRNA and protein expression of TGF-?1 in CCl_(4) group sharply increased. In the meantime, the proliferation of CCl_(4) group cells was markedly increased. There were less G_(0)/G_(1) phase_() cells and more G_(2)/M phase cells in CCl_(4) group than those in control group. Conclusion Suitable CCl_(4) may increase the expression of TGF-?1 and the proliferation of hepatic stellate cells, which is one of mechanisms leading to hepatic fibrosis.

Objective To investigate the predictive value of peripheral leukocyte and its subtypes count on cerebral-cardiovascular events in patients with unstable angina pectoris. Methods From December 2000 to January 2003,141 patients(aged 41-84 years) with unstable angina pectoris referred for coronary angiography were enrolled and their peripheral leukocyte and its subtypes were counted. The major adverse cerebral-cardiovascular events rate of these patients during 46.2?6.6 months of follow-up were assessed. Results A total of 55 cerebral-cardiovascular nonfatal events and 8 deaths were observed. After adjusting the risk factors of diabetes mellitus, smoking, hypertension, hyperlipidemia, prior myocardial infarction, family history, and blood fibrinogen and uric acid level, leukocyte count was positively associated with cerebral-cardiovascular events(adjusted relative risk 1.31,95% CI 1.15 to 1.49).The monocyte count in patients with cerebral-cardiovascular events was higher than that in patients without cardiovascular events(P

Objective To compare the clinical effect and safety evaluation of three different dose regimens for treating children with viral encephalitis. Methods Totally 126 cases treated in Xi'an Central Hospital from January 2010 to December 2015 were randomly divided into observation group 1 (ganciclovir combined with gangliosides,42 cases),observation group 2 (ganciclovir combined with gamma globulin,43 cases),and control group (39 cases). The clinical effect and levels of NSE,inflammatory cytokine were compared in the three groups. Results The total effective rate in observation group 1 was 95.24% and that of observation group 2 was 93.02%,which were significantly higher than that of control group (79.48%).The disappearance time of headache, fever, convulsions, clouding of consciousness, meningeal irritation sign, cerebrospinal fluid abnormalities, and length of stay in observation groups (both 1 and 2)were significantly shorter than those in control group (P < 0.05);After therapy, the levels of NSE in three groups were obviously decreased compared with those before therapy (P < 0.05),and those in observation group were significantly lower than the control group (P < 0.05);the levels of inflammatory cytokine in all three groups were obviously decreased compared with those before therapy (P < 0.05),and that of observation group 1 had no statistical difference with the normal group, whereas that in control group was significantly higher than the normal group (P < 0.05).Conclusion Ganciclovir combined with gangliosides as well as ganciclovir combined with gamma globulin were both effective methods in treating children with viral encephalitis and could decrease levels of inflammatory cytokine.Ganciclovir combined with gangliosides could effectively repair nerve damage, which deserves clinical expansion.

Objective To investigate the effect of intratumor injection of 131 I 3H11 for gastric cancer (GC). Methods 16 patients with GC subjected to endoscopic intratumor injection of 131 I 3H11 as a treatment group; 6 GC patients with FAM chemotherapy as a control group. Histological examination of the postoperative specimens of the two group were comparated. Results In treatment group, 75.0%(12/16) of cases were found to have morphological changes with karyopyknosis, karyorrhexis, coagulation of cytoplasm, and invasion of lymphocyte in mesochyma. Most of these changes were medial; but in control group no obvious morphological change was found. Conclusions The results suggest that GC subjected to endoscopic intratumor injection of 131 I 3H11 preoperatively has promising application in the clinic.

The safflower floret is a traditional Chinese medicine used to promote blood circulation and remove obstruction in the channels. The spines on its bracts are considered a handicap when manual harvest is involved. In this study, cDNA-SRAP was used to systematically investigate which genes are associated with the spines. Sixty pairs of possible primer combinations were used on two cDNA pools representing spininess and spinelessness. Six transcript-derived fragments were identified, of which two with low recombination were sequenced successfully and named as GPY-1 and GPY-2. By using the RACE method, the full-length cDNA of GPY-2 is cloned and named as CTL-spn. The full-length cDNA of CTL-spn was 1 679 bp long with a 1 524 bp ORF encoding a 508 aminoacid protein. The deduced amino acid sequence of the CTL-spn gene shared a high homology (97%) with other known ATP synthase CF1 alpha subunits. Semiquantitative RT-PCR analysis revealed that the mRNA of GPY-1 and GPY-2 accumulated in only spiny lines. Considering the important role of ATP synthase CF1 alpha subunit in plants, it may directly take part in the formation process of spininess and enhancing resistance reaction of spiny safflower. Also, our results provide the important insights for breeding spineless cultivars of safflower.

Objective To observe the extracellular content of dopamine (DA)and expression of D1 receptors in hippocampal dentage gyrus (DG)in the model rats with vascular dementia (VD),and to investigate the relationship between them. Methods 12 male SD rats were randomly divided VD group and sham-operation group, and the VD model was prepared by permanent bilateral carotid occlusion. The extracellular content of DA in the DG was determined by in vivo microdialysis and HPLC,and the expression of D1 receptors was measured by immunehisto-chemistry.Results The DA content in the DG of the rats in VD group was lower than that in sham-operation group (P <0.05).The number of D1 receptor-positive cells in the DG hilus in VD group was increased compared with sham-operation group (P < 0.05),whereas the expression of D1 receptor in DG granule cell layer did not change (P > 0.05).Conclusion The DA content in the hippocampal DG is decreased in the VD rats, and its function may be compensated by the up-regulation of D1 receptors in the DG hilus.
