1.DETERMINATION OF FOLIC ACID IN INFANT FORMULA MILK POWDER BY SOLID-PHASE EXTRACTION-REVERSED PHASE HIGH-PERFORMANCE LIQUID CHROMATOGRAPHY

Objective A quick and sensitive method was developed for determination of folic acid (FA) in infant formula milk powder. Method Perchloric aicd was added into infant formula milk powder to extract FA in ultrasonic wave condition. After solid-phase extraction (SPE) with C18 material as plug and reaction with potassium permanganate, FA was determined by reversed phase high-performance liquid chromatography (RP-HPLC). pH 5.0 and 0.1 mol/L sodium dihydrogen phosphate were used as mobile phase. the flow rate was 1 ml/min. BDS C18 was used as separating column and column temperature was 30 ℃. Results The linear range of the method was 0.005-1.5 ?g/ml. The average recovery was 91.7%- 98.3%. The relative standard deviation (RSD) was 2.5%. The limit of detection was 0.005 ?g/ml, and the limit of quantification was 0.017 ?g/ml. Conclusion The method has following advantages: simple, precise, accurate and easy to practice.
2.Correlation analysis of asthma and prognosis in 148 children with low immune function

Objective To analyze the effect of immune function on the condition and prognosis of asthma in children with asthma. Methods A total of 148 children with asthma diagnosed in Qinghai women and children's Hospital from January 2018 to January 2021 were included in the analysis, the immune function of the children was determined, and the information of all children was followed up for 6 months after treatment; compared The condition and follow-up prognosis of children with immunocompromised and normal immune function were analyzed and discussed, and the correlation between the expression levels of immunoglobulins (IgG, IgA, IgM) and the condition and short-term recurrence prognosis (6 months) of children was analyzed and discussed, so as to guide Prevention and clinical work. Statistical analysis was done using SPSS19.0. Results The average age of 148 children with recurrent respiratory tract infection in the study was (8.94±3.65) years old, including 70 male children. The condition of the children was evaluated and classified into mild/severe cases: 148 children in this study included mild cases. There were 98 cases and 50 severe cases. There were more males and lower BMI levels in severe children (P<0.05) . The levels of IgG, IgA and IgM in children were all lower in severe children (P<0.05) . The follow-up found that the proportion of relapses in critically ill children was higher (P<0.05). Comparing the levels of IgG, IgA, and IgM in mild and severe children, the average levels of IgG, IgA, and IgM in severe children were lower than those in the mild group, and the difference was statistically significant (P<0.05); recurrence within 6 months of follow-up Prognostic evaluation showed that 19 of the 148 children had relapse, and the levels of IgG, IgA, and IgM in severe relapsed children were significantly lower than those without relapse (P<0.05). Analysis of the relevant factors potentially affecting the prognosis of recurrence showed that gender (female) (OR=1.726) , BMI level (weight loss) (OR=1.613) , IgG expression level factor (low expression) (OR=1.898) , IgA expression Level factor (low expression) (OR=3.509) , IgM expression level factor (low expression) (OR=3.217) and disease factor (severe) (OR=3.619) were potential risk factors, which would increase the risk of poor prognosis. Conclusion The asthma attack in children with immunocompromised immune function is relatively severe, and the short-term recurrence probability is higher, which deserves clinical attention and preventive intervention.

4.Hypoxia at high altitude induces inflammation in the spleen of mice through NOD⁃like receptor signaling pathway

Objective :
Based on metabolomics and transcriptomics analysis , to explore the molecular mechanism of
spleen inflammation induced by high altitude hypoxia in mice through NOD⁃like receptor signaling pathway .
Methods :
C57BL/6 mice were raised at an altitude of 400 m and 4 200 m respectively , with five mice in each group , and spleen tissues were collected after 30 days . Differential metabolites and differentially expressed genes in key pathways were screened by metabolomics and transcriptome analysis and correlation KEGG enrichment analysis , and the related network interaction diagram of differential metabolites and differentially expressed genes in key pathways was constructed and verified by RT⁃qPCR .
Results :
Metabolomics analysis showed that 133 differential metabolites were screened from in the plain spleen control group (PSC group) and the plateau spleen test group (HST group) , 95 of which were up⁃regulated while 38 of which were down⁃regulated . KEGG enrichment analysis showed that they were mainly involved in NOD⁃like receptor signaling pathway , HIF⁃1 signaling pathway , cholesterol metabolism and other metabolic pathways . The results of transcriptome analysis showed that a total of 4213 differentially expressed genes were identified in PSC group and HST group , including 1947 up⁃regulated genes and 2266 down⁃regulated genes . KEGG was enriched in 173 signaling pathways , including NOD⁃like receptor signaling pathway , MAPK signaling pathway , NF⁃κB signaling pathway and other pathways . Comprehensive analysis showed that the differential metabolites and differentially expressed genes were obviously enriched in NOD⁃like receptor signaling pathway . Therefore , the correlation network interaction map was constructed for the differential metabolites ATP and differentially expressed genes in NOD⁃like receptor signaling pathway . RT⁃qPCR results showed that compared with PSC group , the expression levels of DEGs related to NOD1 and NOD2 (CHUK , TAB3 , MAPK8) in the signaling pathway of NOD⁃like receptor and NLRP1 ⁃CASP1 pathway (NLRP1b , CASP1) in HST group were significantly enhanced . The mRNA expression levels of downstream inflammatory factors IL⁃6 , IL⁃1 β , IL⁃18 , INF⁃γ and TNF⁃α were up⁃regulated and differentially expressed .
Conclusion
Based on the combined analysis of metabolomics and transcriptomics , it was found that hypoxia stimulation at high altitude may affect the NOD⁃like receptor signaling pathway in vivo , and the differential metabolite ATP is positively correlated with the differential key genes in the pathway . ATP mediates the release of downstream inflammatory factors by activating NOD1 , NOD2 pathways and NLRP1 inflammable⁃CASP1 pathways . Inflammatory response occurred in spleen tissue of mice.

Objective:To observe the clinical efficacy and influence of modified Chaihu Shugansan combined with ursodeoxycholic acid tablets on inflammatory factors in treatment of chronic cholecystitis cholelithiasis (stagnation of liver and gallbladder Qi). Method:One hundred and ten patients were randomly divided into control group (60 cases) and observation group (60 cases). Both groups received lifestyle intervention, and oral ursodeoxycholic acid tablets, 50 mg/time, taken in the morning and evening meals. Patients in control group additionally took Yidanshu capsules orally, 4 capsules/time, 3 times/day. Patients in observation group additionally took modified Chaihu Shugansan orally, 1 dose/day. The treatment courses continued 3 months in both groups. 6. Effect of altitude hypoxia on blood-brain barrier after subarachnoid hemorrhage based on phosphatidylinositol 3-kinase/protein kinase B/nuclear factor κB pathway in rats

Objective To investigate the effect of plateau hypoxia on the blood-brain barrier after subarachnoid hemorrhage (SAH) in rats. Methods Adult male SD rats (n = 78) were randomly divided into 4 groups: sham group (sham), SAH model group (SAH), plateau hypoxia sham group (Hp sham) and plateau hypoxia SAH model group (Hp SAH). The rat model of plateau hypoxia was established through low-pressure simulation chamber (altitude 5000 m), and the SAH model was established by endovascular perforation method. At 24 hours after SAH, neurobehavior score and SAH grade were assessed. The morphological changes of neurons and apoptosis of nerve cells in the CA1 region of hippocampal were observed by the staining of Nissl and TUNEL. The expression of phosphorylated PI3K (p-PI3K), PI3K, phosphorylated Akt (p-Akt), Akt, phosphorylated nuclear factor κB (p-NF-κB), NF-κB, matrix metalloproteinase-9 (MMP-9), occludin and claudin-5 in hippocampal were detected by the method of Western blotting. The expression of occludin and claudin-5 proteins in the CA1 region of hippocampal were observed by immunofluorescent staining. Results At 24 hours after SAH, the neurobehavior score decreased significantly and SAH grade increased significantly in the SAH and Hp SAH group (P< 0.05). Neurobehavior score decreased significantly in the Hp SAH group compared with the SAH group (P < 0.05). In the SAH group, neurons in the CA1 region of hippocampus were atrophied and deformed, the arrangement were disordered, the number of neurons decreased significantly, and the apoptosis of nerve cells increased significantly(P< 0.05). Plateau hypoxia could aggravate the morphological damage of neurons and apoptosis of nerve cells. The expression of p-PI3K/PI3K, p-Akt/Akt, occludin and claudin-5 proteins decreased significantly, while the expression of p-NF-κB/NF-κB and MMP-9 proteins increased significantly in the SAH and Hp SAH group (P< 0.05). The expression of p-PI3K/PI3K and MMP-9 proteins increased significantly in Hp SAH group compared with the SAH group. The expression of claudin-5 protein increased significantly in Hp sham group compared with the sham group (P < 0.05). Immunofluorescent staining showed that the expression of occludin and claudin-5 proteins in the CA1 region of hippocampus decreased in the SAH group. Plateau hypoxia could further decreased the expression of occludin and claudin-5 proteins. Conclusion Plateau hypoxia aggravates blood-brain barrier disruption after subarachnoid hemorrhage in rats through inhibiting PI3K/Akt/NF-κB pathway.

9.Analysis for the blood mineral content of children aged 3 to 12 years in 7 cities and 2 towns in China

Objective:To investigate the status of blood mineral content of children aged 3 to 12 years in Chinese cities and countryside and explore the possible related influencing factors .Methods: The multistage stratified cluster random sampling was used to select one kindergarten and one primary school in seven cities ( Beijing and Guangzhou and so on ) and two towns randomly .Firstly, we selected one bottom class , middle class , top class in one kindergarten and one second grade and fifth grade in one pri-mary school randomly .Then all of the healthful students of these classes were investigated and the blood mineral content of calcium , magnesium, lead, iron, copper, and zinc were detected.Results:In the re-search, 1 842 students were investigated .The means of calcium, magnesium, lead, iron, copper, and zinc were in the standard range .The blood lead content of the boys was higher than that of the girls .The blood mineral content of different ages had statistical significance .The blood calcium and blood copper contents of the preschool children were higher than those of the school children .However , the school children had significantly higher blood lead , iron, and zinc contents in comparison with those of the pre-school children .The incidences of iron deficiency and zinc deficiency were 35.5%and 39.6%, respec-tively .The incidence of iron deficiency and zinc deficiency of different ages had statistical significance , and with the age increasing , the incidence showed a decreasing trend .The incidences of iron deficiency and zinc deficiency of children whose age was more than or equal to 3 years and less than 4 years were up to 47.1%and 64.6%, respectively.The incidence of iron deficiency of the children in the countryside area was significantly higher than those in the first-tier cities.However, the incidence of zinc deficiency of the children in the countryside area was significantly lower than those in the first-tier cities and second-tier cities.Conclusion:The status of iron deficiency and zinc deficiency of children aged 3 to 12 years in Chinese cities and countryside were still serious .We should pay more attention to the nutrition interven-tional research on iron and zinc .
10.The clinicopathologic characteristics of desmoplastic fibroblastoma in neck and nape

OBJECTIVE To describe the clinical, CT or MRI, pathological characteristics of desmoplastic fibroblastoma for helping the diagnosis and treatment. METHODS The clinical data of 4 cases with desmoplastic fibroblastoma were retrospectively analyzed. RESULTS The tumors of all 4 cases were found in the compartment of muscles, and can be removed by conservative excision. The 4 cases were followed up for 38, 7, 6, 3 months respectively, and no local recurrences were found. CONCLUSION Desmoplastic fibroblastoma is a benign fibroblastic neoplasm with distinctive clinical, CT or MRI imaging and pathological characteristics. Optimal management is conservative excision with functional preservation. Needle aspiration cytology and immunohistochemistry are of non-diagnostic value.
