1.Effect of methylene blue on T-lymphocyte subsets and oxidative stress in rat thymus with paraquat poisoning

[ ABSTRACT] AIM:To illustrate the effect of methylene blue ( MB) on T-lymphocyte subsets and oxidative stress of the thymus in rats with paraquat ( PQ) poisoning.METHODS: Male SD rats ( n=40) were randomly assigned to 4 groups:normal group, control group, low-dose (2 mg? kg-1? d-1 ) MB group and high-dose (4 mg? kg-1? d-1 ) MB group.After 72 h, the pathological morphological changes of the thymus were observed under microscope with HE staining. The SOD activity and MDA content were measured by colorimetry method.The expression of Bcl-2, Bax, CD4 and CD8 was determined by immunohistochemical method.RESULTS:In MB group, the thymus tissue showed good corticomedul-lary demarcation.MDA content decreased and SOD activity increased, indicating that the ability of antioxidation enhanced. Bcl-2 and Bax expression was down-regulated, Bcl-2/Bax ratio increased, and CD4 positive cells increased.CONCLU-SION:MB protects against oxidative damage induced by PQ, and regulates the distribution of T-lymphocyte subsets in the cortex and medulla, thus relieving the persistent body damage.
2.Correlation of pericardial adipose tissue with the severity of coronary artery atherosclerosis in coronary heart disease patients with metabolic syndrome

Objective To assess the volume of epicardial adipose tissue(EAT) and intrathoracic adipose tissue(IAT) and the correlation of EAT,IAT and the EAT/IAT ratio with the severity of coronary artery atherosclerosis in patients with metabolic syndrome(MS) and coronary heart disease(CHD).Methods Ninty-seven MS subjects without coronary atherosclerosis and one hundred and eighteen MS subjects with CHD were enrolled in this study.The volumes of EAT,PAT and IAT were measured using axial data from base to apex traced manually with a dedicated semiautomatic software program-volume.Results (1)Compared with MS subjects without coronary atherosclerosis,MS subjects with CHD had significantly increased age[(60.6± 1 1.4)years vs.(57.9 ± 8.7) years,P=0.001],positive family history of cardiovascular disease(29.7% vs.21.6%,P=0.03),EAT[(98.3±41.4) cm3 vs.(82.2±39.7) cm3,P=0.001],IAT [(171.3±64.1) cm3 vs.(156.2±48.1) cm3,P=0.001] and HbA1c[(7.1 ± 1.8)% vs.(6.6±2.3)%,P=0.02],but siginificantly reduced total cholesterol[TC,(4.9 ± 1.2)mmol/L vs.(5.4 ± 1.0) mmol/L,P=0.003],low-density lipoprotein cholesterol[LDL-C,(3.0±1.1) mmol/L vs.(3.6±1.0) mmol/L,P=0.03] and high-density lipoprotein cholesterol[HDL-C,(1.0 ± 0.4) mmol/L vs.(1.1 ± 0.3) mmol/L,P=0.04].(2) In MS subjects without coronary atherosclerosis,the volumes of EAT,pericardial adipose tissue(PAT) and IAT were associated with gender,body mass index(BMI),waist circumference(WC),diabetes mellitus and hyperlipidemia(all P<0.05); however,in MS subjects with CHD,the volumes of EAT,PAT and IAT were associated with BMI and WC (both P<0.05).(3) The volumes of EAT and IAT were correlated with calcification grades(r values were 0.45 and 0.50,P values were 0.017 and 0.013,respectively) and Gensini score(r values were 0.476 and 0.563,P values were 0.015 and 0.017,respectively) instead of coronary artery calcifacation score(CACS).Moreover,the EAT/IAT ratio was negatively correlated with CACS(r=-0.321,P=0.028).Conclusions Compared with MS subjects without coronary atherosclerosis,MS subjects with CHD have significantly increased EAT and IAT volumes.In our subjects,the volumes of EAT,PAT and IAT were associated with BMI and WC.Only in MS subjects with CHD,the volumes of EAT and IAT were correlated with caleification and Gensini score.
3.Therapeutic effect of early rehabilitation on aged patients with acute myocardial infarction complicated heart failure

Objective: To explore the therapeutic effect of early rehabilitation on aged patients with acute myocardial infarction (AMI) complicated heart failure (HF). Methods: According to number table method, a total of 168 AMI cases were randomly divided into early rehabilitation group (n=84, received early rehabilitation protocol) and routine treatment group (n=84, received traditional treatment). Incidence rate of complication, bicycling test and walking test result were compared between two groups at the end of rehabilitation program. Results: There were no significant difference in angina pectoris after myocardial infarction, re-infarction, dead cases during hospitalization and left ventricular ejection fraction (LVEF)etc. between two groups (P>0.05 all). Compared with routine treatment group, there were significant rise in completion rates of bicycling test (77.6% vs. 95.0%) and walking test (65.4% vs. 95.8%) at the end of program in early rehabilitation group, P<0.01 both. Incidence rates of complications caused by long-term lying in bed in early rehabilitation group were significantly lower than those of routine treatment group (P<0.05 or P<0.01). 4.The moderating and mediating role of social support in the relationship between Caregiver Burden and Negative Affect in Caregivers of cancer patients

Objective To explore the moderating and mediating role of social support in the relationship between caregiver burden and Negative Affect in Caregivers of cancer patients. Methods A total of 407 caregivers of cancer patients in tertiary hospital in Xinjiang were recruited by cross-sectional survey method. Caregives were investigated using Caregiver Burden Scale for Cancer patients, Positive and Negative Affect Schedule, theSocialSupport Rating Scale. Results Social support mediated the relationships between caregiver burden andnegative effect, which accounted for 6.41% of the total variance. The interactive effects of caregiver burden× social support on thenegative affect was significant(β=0.003, t=2.782, P<0.01). Conclusions Social support plays a mediatingand moderating role on the relationships between caregiver burden and negative effect.

7.Protein expression levels of Nogo-A,Adropin,ZO-1 and ANGPTL4 in serum of patients with acute cerebral hemorrhage and their predictive value for prognosis

Objective To explore the protein expression levels of serum neurite outgrowth inhibitor-A(Nogo-A),energy balance related protein(Adropin),zonula occludens-1(ZO-1)and angiopoietin-like protein 4(ANGPTL4)in patients with acute cerebral hemorrhage and their predictive efficacy for prognosis.Methods A total of 148 patients with acute cerebral hemorrhage treated in the hospital from April 2020 to May 2022 were selected as the study group,and another 150 healthy people who came to the hospital for gen-eral physical examination during the same period were selected as the control group,and a retrospective analy-sis was conducted.The protein expression levels of Nogo-A,Adropin,ZO-1 and ANGPTL4 in serum of the two groups were detected by enzyme-linked immunosorbent assay(ELISA)and compared.Spearman correla-tion coefficient was used to analyze the correlation between serum Nogo-A,Adropin,ZO-1,ANGPTL4 protein expression levels and the occurrence of acute cerebral hemorrhage.Receiver operating characteristic(ROC)curve was drawn to analyze the diagnostic efficacy of the combined detection of the above four indicators for a-cute cerebral hemorrhage.In addition,patients in the study group were followed up for 6 months,and were di-vided into good group(91 cases)and poor group(57 cases)according to the prognosis.Serum protein expres-sion levels of Nogo-A,Adropin,ZO-1 and ANGPTL4 were compared between the two groups.ROC curve was used to verify the predictive efficacy of the combined detection for the prognosis of these patients.Results Compared with the control group,the protein expression levels of Nogo-A,ZO-1 and ANGPTL4 in the study group were significantly increased,while the protein expression level of Adropin was significantly decreased(all P<0.05).Spearman correlation coefficient analysis showed that serum Nogo-A,ZO-1,ANGPTL4 pro-tein expression levels were positively correlated with the incidence rate of acute cerebral hemorrhage,while Adropin protein expression level was negatively correlated with the incidence rate of acute cerebral hemor-rhage(all P<0.05).ROC curve verification showed that the combined detection of Nogo-A,Adropin,ZO-1 and ANGPTL4 protein expression levels had high diagnostic efficacy in the evaluation of acute cerebral hemor-rhage,with the sensitivity and the specificity of 92.57%and 90.67%,respectively.Compared with good group,the protein expression levels of Nogo-A,ZO-1 and ANGPTL4 in poor group were significantly higher,while the protein expression level of Adropin was significantly decreased(all P<0.05).ROC curve showed that compared to single detections,the area under the curve of the combined detection of protein expression levels of Nogo-A,Adropin,ZO-1 and ANGPTL4 was larger(P<0.05),with the sensitivity of 84.21%and the specificity of 89.01%.Conclusion The protein expression levels of Nogo-A,Adropin,ZO-1 and AN-GPTL4 are abnormal in patients with acute cerebral hemorrhage,and the protein expression levels of Nogo-A,Adropin,ZO-1 and ANGPTL4 are closely related to the development of acute cerebral hemorrhage.The com-bined detection of the four indicators could not only assist clinical accurate judgment of acute cerebral hemor-rhage,but also have important clinical value in predicting the prognosis of patients.
8. The function of exosome-derived miRNAs in gastric cancer

Gastric cancer, a common malignant tumor in digestive system with high morbidity as well as mortality rate, is insidious at the onset and lack of effective treatments so far. A growing number of studies have shown that exosome-derived miRNAs play an important role in the occurrence and development of gastric cancer. Autocrine exosome miRNAs from gastric cancer cells regulated tumor growth, recurrence, metastasis and drug resistance, etc. Moreover, exosomal miRNAs in the tumor microenvironment can be delivered into cancer cells to facilitate intercellular communication, thus affecting the progress of gastric cancer. Due to exosomes, which were released into circulation from tumor cells, contain abundant, specific and stable miRNAs, exosome-derived miRNAs have a great potential to be used as novel diagnosis biomarkers and treatment targets of gastric cancer.(
9.Analysis of the efficacy and safety of modular flexible ureteroscopic lithotripsy for patients with diabetes and kidney stones

Objective:To investigate the efficacy and safety of modular flexible ureteroscopic lithotripsy (M-FURL) in treatment of diabetic patients with kidney stones.Methods:Sixty patients with diabetes and kidney stones admitted to the Department of Urology, Xindu District People’s Hospital of Chengdu were selected as the observation group, and 78 patients with diabetes without kidney stones who were treated in our hospital during the same period were selected as the control group. M-FURL was performed to compare the operation of the two groups of patients.Results:Compared with the control group, fasting blood glucose and blood glucose level at 2h after meals in the observation group were significantly increased ( P<0.05) ; and the ratio of uric acid stones and calcium oxalate stones in the observation group was significantly higher than that ine the control group ( P<0.05) ; The two groups of patients were treated with M-FURL, and there was no statistically significant difference in stone clearance rate, total stone clearance rate, or complication rate ( P>0.05) ; At the same time, the difference in operation time and intraoperative blood loss between the two groups had no statistical significance ( P>0.05) ; while the hospital stay of the observation group was significantly longer than that of the control group ( P<0.05) . Conclusions:Patients with kidney stones and diabetes are more likely to have calcium oxalate stones and uric acid stones. M-FURL treatment in patients with diabetes and kidney stones is ideal and safe. Combining diabetes will not affect the effect of surgery.

Objective To explore the mechanism of Dahuanglinxian Capsule for intervening gallstone formation by regulating the expression levels of ABCB11 and ABCC2 mRNA and protein.Methods Forty male C57BL/6 mice were divided into the normal group(group N),model group (group M),ursodeoxycholic acid group (group U) and Dahuanglinxian Capsule treatment group (group D),10 cases in each group.The group N was fed with normal diet,while the group M,U and D were fed with lithogenic fodder for 8 weeks.Meanwhile the group U and D were given the medication intervention,once daily,for continuous 8 weeks of gavage.After successful modeling,mRNA and protein expression levels of ABCB11 and ABCC2 were detected by RT-PCR and immunohistochemistry.Results Compared with the other three groups,the expressions of ABCB11 and ABCC2 mRNA gene and protein in M group were significantly reduced(P<0.01);while,there was no statistical difference in the expressions of ABCB11 and ABCC2 mRNA and protein between the group D and N(P>0.05).Conclusion Dahuanglinxian Capsule can prevent the gallstone formation by regulating the expression of ABCB11 and ABCC2 mRNA and protein.
