1.Expression and significance of p57 kip2, Cyclin D1 in human endometrial carcinoma

Purpose To study the effects of p57kip2, Cyclin D1 on the carcinogenesis and progression of endometrial carcinoma. Meth-ods 100 cases of endometrial adenocarcinoma ( EA) , 20 cases of endometrial intraepithelial neoplasia ( EIN) , 20 cases of hyperpla-sia lesion, 20 cases of endometrial proliferative phase were selected. Different endometrial cells ( well-differentiated endometrial cancer cells Ishikawa, moderately differentiated endometrial cancer cells JEC, poorly differentiated endometrial cancer cells KLE and normal endometrial cells ESC) were cultured. Protein expression of p57kip2, Cyclin D1 was measured by immunohistochemical EliVision meth-ods. The expression of p57kip2, Cyclin D1 protein in different endometrial cells was detected by Western blot. Results The highest expression of p57kip2 protein was in EIN, the higher expression was in endometrial proliferative phase, the low expression was in EA and the lowest expression was in hyperplasia lesions, but only the expression of p57kip2 protein in EIN was higher than that in hyperplasia le-sions (P<0.05). The highest expression of Cyclin D1 protein was in EA and the lowest expression of Cyclin D1 protein was in endom-etrial proliferative phase, expression of Cyclin D1 protein increased gradually in hyperplasia lesions and EIN, p57kip2, Cyclin D1 pro-tein expression in the tissue of EA increased along with the histological grade, all present decreasing trend, but only p57kip2 protein ex-pression related to histological grade ( P<0.05 ) . The highest expression of p57 kip2 protein was in KLE and the lowest expression of p57kip2 protein was in ESC, but only the expression of p57kip2 protein in KLE was higher than that in ESC (P<0.05). The expression of Cyclin D1 in JEC, Ishikawa higher than that in ESC (P<0.05). Conclusion p57kip2, Cyclin D1 are involved in the occurrence and development of endometrial carcinoma, Cyclin D1 is an early event in endometrial carcinoma, but there may also be have abnormal p57kip2 protein by synthesized, synergistically. Cyclin D1 promoting endometrial malignant transformation. Combined detection of p57kip2, Cyclin D1 expression in endometrial carcinoma, to predict the prognosis of endometrial carcinoma has a certain clinical signifi-cance.
2.Analysis of IgH gene rearrangement and its significance in non-Hodgkin lymphoma

Objective To explore the significance of detecting IgH gene rearrangement in diagnosis of non-Hodgkin lymphoma. Methods By using family-specific primers mixtures (VH1,VH2,VH3) for IgH FR3A,FR2A,FR1 genes,clonal IgH gene rearrangement was detected by heminested and multiplex PCR in 44 cases of B-NHL,1 undifferentiated lymphoma,15 T-NHL and 5 lymphonode reactive hyperplasia. Results Clonal IgH gene rearrangement was detected in 36 of 44 B-NHL (positive rate 82%) by using FR3A primers,27 of 44 B-NHL (positive rate 61%) by using FR2A primers. By using FR1 family-specific primers mixtures (VH1,VH2,VH3) and JH (JHb,JHc),clonal IgH gene rearrangement was demonstated respectively in 34/33 of 44 B-NHL (positive rate 77%,75%). The total positive rate was 95% (42/44) by combined family-specific primers mixtures (VH1,VH2,VH3) of FR3A,FR2A,FR1 gene. Four of 15 T-NHL revealed clonal IgH gene rearrangement,while 5 LRH had no clonal IgH gene rearrangement. One NHL unclassified by immunohistochemical methods was classified definitely as B cell-derived lymphoma by PCR. Conclusion The detection rate of IgH gene rearrangement can be improved by combined usage of several pairs of primers,especially family-specific primers. Detecting clonal IgH gene rearrangement is significant in the procedure of differential diagnosis for B/T-NHL and reactive hyperplasia.

4.Clinical and Experimental Study on Autoimplantation Technique in the Treatment of Verruca Plana

Objective To apply autoimplantation technique in the treatment of verruca plana, and to investigate the mechanisms of it. Methods The changes of T cell subsets and IL-2 level in the peripheral blood of the patients with verruca plana, in conjunction with histopathological and immunohistochemical features of verruca plana tissue at reactive stage were studied by APAAP and radioimmunoassay, respectively. The tissue specimens were studied by H.E stain and immunohistochemical techniques. Results Significantly decreased CD4+ cell numbers, CD4+/CD8+ ratio and IL-2 level were found before treatment, compared with those in healthy controls (P 0.05). Lymphocytic infiltration, epidermal edema and dissolution were shown in the histopathological study. There were CD45RO+, CD6+, CD4+ and CD8+ cells in the lymphocytic infiltration, but no CD20+ and Mac387- cells. Langerhans cells with positive S-100 protein were observed in basal layer of the epidermis and reaction area. Conclusions T cell-mediated immune responses could be improved in autoimplantation, the warts are rejected mainly by of CD8+ subset cytotoxicity.
