5.Clinical value of ultrasonography in detection of median nerve in diabetic patients

ObjectiveTo explore the clinical value of ultrasonography in the detection of median nerve in diabetic peripheral neuropathy (DPN).MethodsMedian nerve was detected in 70 healthy adults (control group) and 43 patients with type 2 diabetes mellitus (diabetes mellitus group) by color Doppler ultrasonography.The type 2 diabetes mellitus patients were divided into two groups according to the duration of the disease:< 10 years group (26 cases) and ≥ 10 years group( 17 cases).And they were divided into two groups according to the random plasma glucose (RPG):RPG < 15 mmol/L group(28 cases) and RPG≥ 15mmol/L group ( 15 cases).The nerve cross section area (CSA) was taken at the level of the pisiform.Results The median nerve in diabetes mellitus group had decreased echogenicity and increased color in it.The CSA of the median nerve in diabetes mellitus group was significantly higher than that in control group [ ( 10.3 ± 2.1 )mm2 vs. (8.3:± 1.7) mm2 ] (P < 0.01 ).The C SA of the median nerve in ≥ 10 years group was significantly higher than that in <10 years group [ ( 12.3 ± 2.0) mm2 vs.( 10.2 ± 2.6) mm2] (P < 0.01 ).The CSA of the median nerve in RPG ≥ 15 mmol/L group was significantly higher than that in RPG < 15 mmol/L group [ ( 11.7 ± 1.8) mm2 vs.(9.6 ± 2.7 ) mm2 ] (P < 0.01 ).ConclusionColor Doppler ultrasonography examination may reveal morphology of median nerve and is useful for the diagnosis of DPN.
6.Enhancive Effect of TNF-a on Transfection Efficiency of Ad-mediated Multidrug Resistance Gene1 (MDR1) in Mononuclear Cells of Mouse

Objective: To investigate the enhancive effect of TNF-α on transfection efficiency of adenovi-rus in mononuclear cells of mouse and the enhancive capability of protection of bone marrow by MDR1. Meth-ods: Before the mononuclear cells of mouse were infected by recombinant adenovirus encoding human MDR1 gene, they had been pretreated by TNF-α. Transfection efficiency of adenovirus was monitored by fluo-rescence microscopy, immunohistochemistry and flow cytometry (FCM). mRNA and protein levels of MDR1 in the mononuclear cells of mouse were detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot before and after treatment of TNF-α. Results: After treatment of TNF-α, tansfection rates of adenovirus were obviously increased in the treated group (26.26%) compared with the untreated group (11.96%). mRNA levels and protein levels of MDR1 were obviously increased in the treated group compared with the untreated group. Conclusion: TNF-α could enhance transfection efficiency of adenovirus in mononu-clear cells of mouse and enhance capability of protection of bone marrow by MDR1.
8.Sheng's acupuncture manipulation at bone-nearby acupoints and the academic thoughts.

Sheng's acupuncture manipulation at bone-nearby acupoints is a set of needling manipulation of the chief physician of TCM, SHENG Xie-sun, summarized through his over 50 years clinical experiences and on the basis of Internal Classic. Regarding this manipulation, on the premise of acupoint selection based on syndrome differentiation, the acupoints close to bone are possibly selected and punctured, with the needle tip toward bone edge, and followed by the technique to achieve reducing purpose. Clinically, the significant immediate analgesia can be achieved in pain disorders such as headache and toothache. Professor Sheng thought, corresponding to the location of needle insertion and needling depth, the tissue layers of needle tip passing through should be considered specially. The site of needle insertion should be changeable so as to ensure the needle tip reaching the bone. This manipulation for analgesia provides a certain guide for acupuncture study, especially for the mechanism study on acupuncture analgesia.

10.Role of FPRL1 in the chemotaxis of polymorphonuclear leukocytes stimulated by LPS and its regulation by polydatin

AIM: To investigate the chemotaxis mechanisms of polymerphonuclear leukocytes (PMNs) under the stimulation of LPS and its regulation by polydatin (PD). METHODS: Chemotactic chamber assay was used to investigate the regulatory role of formyl peptide receptor like-1 (FPRL1) in the chemotaxis of PMNs in response to LPS and PD. RESULTS: LPS increased the secretion of PMN chemotactic factor and up-regulated the function of formyl peptide receptor like-1. PD did not obviously affect the chemotactic factor secretion in normal PMN and the function of formyl peptide receptor like-1, but it significantly reduced the rise of chemotactic factor excretion in PMNs caused by LPS stimulation and PMNs transfected with formyl peptide receptor like-1. CONCLUSION: FPRL1 may mediate LPS-induced PMN chemotaxis, and PD regulates PMN chemotaxis by down-regulating the secretion of chemokine and the function of FPRL1 and may play a crucial role in the treatment of inflammation.
