1.Effects of interleukin-1βon MMP-13 expression in rat chondrocytes and its regulation of miR-27b

Objective To observe the effect of interleukin-1β(IL-1β) on expression of Matrix Metalloproteinases 13 (MMP-13) in rat chondrocytes and its regulation of miR-27b. Methods Chondrocytes were extracted from 7 Wistar male rats. Expression of MMP-13 were examined by Western blot at 0 h, 24 h, 48 h after IL-1βstimulation. Differential miRNAs expression profiles were examined by miRNAs microarray. The most obviously down-regulated miRNAs were confirmed by quantitative Real-time PCR. Targeted regulation relationship between miR-27b and MMP-13 was set up by Luciferase re?porter gene experiments. Results Expression of MMP-13 in rat chondrocytes was increased at a timely dependent manner upon IL-1βstimulation(P<0.05);Microarray revealed 36 miRNAs whose expression changed, among which 6(miR-27b, miR-31, miR-26a, miR-26b, miR-23, miR-204)were especially obvious. Real-time PCR confirmed that miR-27b was the one whose expression level were most down-regulated. Transient co-transfection of miR-27b mimics with luciferase expres?sion plasmids resulted in significant repression of luciferase activity in rat chondrocytes (P < 0.05). Conclusion IL-1βstimulation result in down-regulation of miR-27b and up-regulation of MMP-13 expression. MiR-27b and MMP-13 show targeted regulation relationship.

6. One-two punch model for the precision treatment of liver cancer

Liver cancer remains difficult to treat, owing to a paucity of drugs that target critical dependencies; broad-spectrum kinase inhibitors such as sorafenib provide only a modest benefit to patients with hepatocellular carcinoma. The induction of senescence may represent a strategy for the treatment of cancer, especially when combined with a second drug that selectively eliminates senescent cancer cells (senolysis). Here, using a kinome-focused genetic screen, we show that pharmacological inhibition of the DNA-replication kinase CDC7 induces senescence selectively in liver cancer cells with mutations in TP53. A follow-up chemical screen identified the antidepressant sertraline as an agent that kills hepatocellular carcinoma cells that have been rendered senescent by inhibition of CDC7. Sertraline suppressed mTOR signalling, and selective drugs that target this pathway were highly effective in causing the apoptotic cell death of hepatocellular carcinoma cells treated with a CDC7 inhibitor. The feedback reactivation of mTOR signalling after its inhibition is blocked in cells that have been treated with a CDC7 inhibitor, which leads to the sustained inhibition of mTOR and cell death. Using multiple in vivo mouse models of liver cancer, we show that treatment with combined inhibition of CDC7 and mTOR results in a marked reduction of tumour growth. Our data indicate that exploiting an induced vulnerability could be an effective treatment for liver cancer.

8.Use of wire frame tie-over to fix the facial skin grafts

BACKGROUND: Facial tie-over is a conventional method for skin graft fixation, but it needs further improvements due to the unsatisfactory fixation, survival quality, postoperational contraction and appearance. OBJECTIVE: To improve the survival quality of facial skin grafts using the modified wire frame tie-over for the fixation. DESIGN, TIME AND SETTING: An observation trial was carried out at the Department of Burns in Beijing Jishuitan Hospital from January 2003 to December 2007. PARTICIPANTS AND MATERIALS: A total of 23 patients suffered from facial burns and post-burn scar were enrolled from the Department of Burns in Beijing Jishuitan Hospital, 16 males and 7 females, with a mean age of (37?18) years. The wire frame tie-over skin grafts were used in 32 facial wounds, including 8 cheek wounds, 20 eyelid wounds and 4 perioral wounds. The 1.0-1.5 Kirschner wire frame, made of stainless steel, was shaped as the wound outline; the rubber bands and clip were sterilized with ethylene oxide. METHODS: All the patients were grafted by means of wire frame tie-over, and the maximal area of skin graft was 18 cm?10 cm. After the routine skin grafting, the Kirschner wire frame was sutured to the edge of wound, then tie-over was performed using the sutures or the sterilized rubber bands fixed on the wire frame. The wire frame was removed three to four weeks after operation. MAIN OUTCOME MEASURES: The survival of skin grafts was recorded at the removal of tie-over, including survival rate of skin grafts and appearance of crater phenomenon that was prominent at edges and depressed in center; The skin grafts were pictured to observe the early contraction at the removal of wire frames 3-4 weeks later. The patients who were potential for the long-term follow-ups were used to observe the long-term contraction. RESULTS: Among 32 wounds in 23 patients, 31 skin grafts had an excellent skin grafting, except a partial graft necrosis occurred in one case due to insufficient debridement. The skin grafts were flat and intact, no crater phenomenon was found. Three or four weeks after operation, the area of skin grafts maintained in original style, no early contraction was found. One year later, the patients appeared mild contraction of skin graft, and the skin appearance was satisfactory. CONCLUSION: Improved wire frame tie-over method can increase the survival quality of facial grafts and alleviate the graft contraction.
9.Monitoring Instrument of Multiple Physiological Parameters Based on Wireless Internet

Objective To study a monitoring instrument of multiple physiological parameters being capable of wireless long-distance transmission,low in power dissipation and price and small in size.Methods In combination of embedded technology and mobile communication technology,S3C2410 based on ARM9 core was used as main chip,ARM-Linux as embedded operation system and SIM300 as communication module connected to Internet,to complete wireless long-distance transmission of multiple physiological parameters.Results Tele-monitoring system of embedded multiple physiological parameters based on GPRS was realized.Conclusion The instrument is small,easily-expanded and reliable in the process of transmitting data,facilitating tele-monitoring and data sharing.
10.Analysis of tidal breathing flow-volume curves in stable COPD patients

Objective To investigate the severity of airflow obstruction in stable COPD patients and to assess the effects of salbutamol by analysis of tidal breathing flow-volume curves(TBFV).Methods 61 moderate-to-severve COPD patients and 68 normal controls were performed pulmonary function test and TBFV loops in seated position.Measurements of TBFV loops and spirometry were performed at rest after salbutamol 400 ?g inhalation in 26 COPD patients.Results COPD patients exhibited a significant decrease in FEV_1%、FEV_1/FVC、FEF50% and FEF75%.The fraction of exhaled volume to achieve PTEF to V_(TE)(V_(PTEF)/V_(TE))and the fraction of exhaled time to achieve PTEF to T_(E)(T_(PTEF)/T_(E))were lower than 18%[(12.8?2.7)% and(11.6?2.4)%],and much lower than normals(25.0?3.1)% and(29.5?3.7)%.But peak tidal expiratory flow(PTEF)and peak tidal inspiratory flow(PTIF)were decreased slightly.After salbutamol administration,26 COPD patients exhibited a significant increase in TEF50%(21.5?6.8)% versus(15.6?4.2)%,P
