1.Investigate the state anxiety and coping style in parents of children with amblyopia

Objective:TO investigate the state of anxiety and coping style in parents of children with amblyopia.Methods:61 parents of children with amblyopia had been researched with the State-Trait Anxiety Inventory(STAI)and Xiao Ji Hua coping style measuring scale.Results:The SAI score of these parents were not different with the normal samples.The SAI score of these parents were not different with the vision of children with amblyopia.In the aspect of using coping strategy,the lower education level of parents always used immature coping strategy.Conclusion:The parents of children with amblyopia felt anxiety in the curing course of amblyopia,but it can keep normal level;Nursing personnel need to pay close attention to the different education level of the parents.
2.Image quality evaluation of 64-slice spiral CT angiography in diagnosis of coronary artery disease

Objective To assessed the image quality and diagnostic value of the coronary artery diseases with 64-slice spiral computed tomography (64-SCT) angiography. Methods A retrospective image analysis of 39 patients who were diagnosed with coronary heart disease by both the 64-SCT and the invasive coronary angiography (CAG). Results In 507 coronary artery segments (13 coronary segments for each patient),1-,2-,3-class image quality by 64-SCT were in 185 (36.5%),236 (46.5%),86 (16.9%) segments respectively,and the heart rate range was respectively 62?5,66?4 and 75?10 per minute. Totally 421/507 (83.1%) segments that were of 1-,2-class image quality,met the requirements of clinical diagnosis,whereas 86/507 (16.9%) segments could not be sufficiently evaluated because of severe calcifications (51/86,59.3%) and/or motion artifacts (35/86,40.7%). Taking CAG as the gold standard diagnostic technique for coronary heart disease,of the lesions≥50% stenosis by 64-SCT (without any exclusion criteria),the overall sensitivity,specificity,accuracy,positive predictive value,and negative predictive value were 89.8%,94.9%,93.3%,88.5%,95.5%,respectively. Conclusion Our results indicate high diagnostic accuracy of coronary artery disease by 64-SCT compared to CAG. 64-SCT is a simple,convenient,safe and reliable,non-invasive effective method for diagnosis of coronary heart disease.
3.Investigation on the use and management of physical examination electronic records

Poor handwriting of physical examination report written by hand made the subjects discard the report at will and resulted in the decrease of the report effectiveness. Therefore, personal electronic files were used in department of physical examination of our hospital since 2006. By contin-uously improving the functionality of the system, automatic transmission and extraction of medical in-formation have been achieved, better health services have been provided and better conditions have been created for teaching and research. Accuracy rate was significantly improved by electronic files compared with paper files, undetected and error detection rate was significantly decreased and satis-faction with the medical examination was significantly improved.
4.Study of the enzyme assay of dimethylarginine dimethylaminohydrolase in rabbit kidney

AIM To establish the method for the enzyme assay of dimethylarginine dimethylaminohydrolase (DDAH) in rabbit kidney and to determine the optimal condition for the assaying. METHODS Five healthy Japanese rabbits weighing 3 0 to 3 5 Kg were killed by air embolism,kidneys were harvested and then were homogenized. Asymmetric dimethylarginine (ADMA) was used as the substrate for DDAH. UV 265 spectrophotometer was applied to determine the amount of the enzymatic product L Citrulline( L Cit).The amount of L Cit produced under different conditions was compared and the optimal condition was screened. The kinetic parameters of ADMA degraded by DDAH were calculated. RESULTS The kinetic parameters of ADMA degraded by DDAH were as follows: K m=(0 28?0 10) mmol?L -1 , V m=(1 36?0 42) mmol?L -1 ?g -1 ?min -1 . The optimal conditions for the enzyme assay of DDAH in rabbit kidney determined in this study follow: The concentration of the enzyme protein was 12 g?L -1 ,the optimal pH of the buffer was 7 4,the final concentration of ADMA was 2 mmol?L -1 ,and the reaction time was 30 min. CONCLUSION The method offered here is easily done. The concentration of the substrate determined in this study was based on the value of Km,thus it was beneficial to the accurate assay of DDAH.
5.Comparison of Stress Reaction Among Laparoscopic, Mini-incision and Open Cholecystectomy

Objective To compare the degrees of stress reaction among laparoscopic cholecystectomy(LC),mini-incision open cholecystectmy(MIOC) and conventional open cholecystectomy(OC).Methods Ninety patients with gallstone were randomly divided into three groups:(LC),(MIOC) and (OC).Blood Thyroxin(T 3),throid-stimulating hormone(TSH),cortisol,insulin,glucose,white blood cell and temperature were measured and/or recorded in the perioperative period.Results The stress rection in LC was significantly lighter than that in OC,but the stress reaction degree in LC and MIOC were basic equality.Conclusion The stress reaction in LC is small than those of OC.
7.Effects of Prinsepia utilis Royle oil on the synthesis of ceramide and expression of ceramidase

Objective To evaluate the effects of Prinsepia utilis Royle oil (PURO) on the synthesis of ceramide and expression of acid ceramidase N-acylsphingosine amidohydrolase 1 (ASH1),and to explore the mechanisms underlying its moisturizing and skin barrier-repairing effects.Methods Keratinocytes from human foreskin tissue were classified into 2 groups to be cultured in keratinocyte-serum free medium (K-SFM) with or without the presence of PURO.Enzyme linked immunosorbent assay (ELISA) was performed to measure the level of ceramide in the culture supernatant of keratinocytes at 0,3,8,24 and 48 hours.The back of nude mice was divided into 4 areas,i.e.,test area,matrix area,blank control area and negative control area.Acetone and ether were used to destroy the epidermal barrier in the test,matrix,and blank control areas,then,the former 2 areas were topically treated with emulsions containing 1% PURO and matrix,respectively,and the blank control area remained untreated.The epidermal barrier remained intact and untreated in the negative control area.Noninvasive methods were used to determine transepidermal water loss (TEWL),epidermal moisture content and skin lipid content in these areas on day 0,1,3,and 7.Skin tissue was obtained from these areas on day 0 and 7 followed by an immunohistochemical study for the quantification of ASH1 expression.Results The level of supernatant ceramide increased with time in the PURO-treated keratinocytes,which was significantly higher at 24 hours and 48 hours than at 0 hour (1.3817 ± 0.100 and 1.3737 ± 0.047 vs.0.7630 ± 0.143,both P < 0.05).The supernatant ceramide was also elevated in the PURO-treated keratinocytes compared with untreated keratinocytes at 24 and 48 hours (both P < 0.05).Noninvasive skin tests showed a gradual decrease in the TEWL,but an increase in the epidermal moisture content and skin lipid content with time in the 3 epidermal barrier-destroyed areas.As far as the test area was concerned,TEWL value was significantly lower on day 3 and 7 than on day 0 (10.85 ± 0.64 and 8.01 ± 0.58 vs.12.65 ± 0.71,both P < 0.05),while a significant increment was observed in the skin lipid content on day 3 and 7 compared with day 0 (29.14 ± 0.40 and 31.30 ± 0.88 vs.27.02 ± 0.65,both P < 0.05),as well as in the epidermal moisture content on day 1,3 and 7 compared with day 0 (13.98 ± 0.28,15.00 ± 0.38 and 15.86 ± 0.18 vs.11.74 ± 0.62,all P< 0.05).On day 7,there was a statistical decline in TEWL value,but an elevation in epidermal moisture content,skin lipid content and ASH1 expression in the test area compared with the matrix area and blank control area (all P < 0.05).Also,the expression of ASH1 was upregulated on day 7 compared with day 0 in the 3 barrier-destroyed areas (all P < 0.05).Conclusion PURO may exert skin-moisturizing and barrier-repairing effects by enhancing the synthesis of ceramide and expression of acid ceramidase ASH1.
8.Dysfunction of epidermal barrier in psoriasis

ObjectiveTo seek experimental evidence of epidermal barrier dysfunction in psoriasis,and to provide a basis for adjuvant therapy of psoriasis.MethodsPhysiometric methods were used to determine the value of sebum content,transepidermal water loss(TEWL) and water content of stratum corneum in 60 patients with psoriasis and 48 normal human controls.The ultrastructure of lamellar bodies was observed with transmission electron microscopy,and the expression of acid ceramidase in normal skin and psoriatic lesions was detected by using immunohistochemical techniques.ResultsCompared with the normal skin,TEWL value was increased(P < 0.01),but water content of stratum corneum decreased(P < 0.01 ) in psoriatic lesions,and sebum content was similar between normal skin and psoriatic lesions.As electron microscopy showed,lamellar bodies in keratinocytes were reduced in number with a disorganized arrangement and irregular size in psoriatic lesions.The expression of acid ceramidase also decreased in psoriatic epidermis.Conclusions The function of epidermal barrier in psoriasis is impaired,and to restore epidermal barrier function and enhance hydration may serve as an important adjuvant therapy of psoriasis.

Objective To investigate the application of ultrasound visualization in instantly evaluation and supplement therapeu-tics in the treatment of uterine fibroids and adenomyosis with high intensity focused ultrasound (HIFU ) .Methods 57 patients with 67 uterine fibroids and 31 patients with 41 adenomyosis were treated with JC-200 HIFU treatment system and monitored the blood flow change in the lesion with B-ultrasound .Evaluated the curative effect with ablation ratio and ablation ratio after supplement therapeutics .Results The average ablation ratio of 57 uterine fibroids was(84 .6 ± 16 .1)% and the increased to(87 .0 ± 10 .7)% af-ter supplement therapeutics to 9 lesions with blood flow in the border of all .The changes were no significance(P>0 .05) .The aver-age ablation ratio of 31 adenomyosis was(62 ± 22 .7)% and increased to(74 ± 14 .7)% after supplement therapeutics to 11 lesions with blood flow in the border of all .The changes were statically significance(P<0 .05) .Conclusion Ultrasound visualization could be used to evaluate the area and extent of ablation with HIFU therapy ,it can clear lesions remaining parts and guiding the supple-ment therapeutics to improve the ablation ratio .Ultrasound visualization provided an evidence of therapeutics in the early period .
10.Effects of Panax notoginoside on the expression of TGF-β1 and Smad-7 in renal tissues of diabetic rats.

In order to explore the effects of Panax notoginoside (PNS) on the expression of transforming growth factor β1 (TGF-β1) and Smad-7 in renal tissues of diabetes, a rat model of diabetic nephropathy was set up by intravenous injection of streptozotocin (STZ). Wistar rats were randomly divided into normal group, diabetic control group, group treated by PNS at low-dosage (PL), group treated by PNS at high-dosage (PH) and group treated by catopril (C), respectively. Fasting blood glucose (FBG), renal index, endogenous creatinine clearance rate (C(Cr)) and urinary albumin (UAlb) in 24 h were examined after 6 weeks. Meanwhile, the expressions of TGF-β1 and Smad7 in renal tissues were immunohistochemically dectected. At the end of the sixth week, FBG, renal index, C(cr), UAlb were all elevated significantly in control group (P<0.01). The expression of TGF-β1 protein was increased while Smad7 protein decreased in renal tissue (P<0.01). However, the treatment with PNS reversed the aforementioned changes in renal tissues of diabetic rats. These results indicate that PNS possess a protective effect on the kidney of diabetic rats and it might protect kidney by inhibiting the expression of TGF-β1 protein and enhancing the expression of Smad7 protein.
