Objective:To investigate dopaminergic neuron injury and ?-synuclein expression of mouse exposed chronically to low dose of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP). Methods:The C57BLmice received bake hypodermic injections of 25 mg/kg MPTP twice a week for 5 consecutive weeks. Abnormal behavior was observed during the treatment by using herbicide. The expression of throsine hydroxylase(TH)and ?-synuclein was determined by immunohistochemistry. The dopaminergic neurons apoptosis was determined by TdT-mediatd dUTP-biotin nick end labeling (TUNEL). Results:As compared with the controls of mice,those treated with MPTP displayed markedly hypoactive behavior. Immunohistochemical staining showed that the numbers of TH stain cells was decreased significantly after treatment with MPTP,and the density of ?-synuclein stain was increased significantly after treatment with MPTP. Apoptosis displayed after treatment with MPTP. Conclusion:Chronical injection with MPTP might induce abnormal behavior. apoptosis and continual aggregation of ?-synuclein in dopaminergic neurons of substantial nigra.
2.Study on inhibitory elements of a putative PrfA-dependent promoter ParoA2 in Listeria monocytogenes

Objective To investigate the relationship between PrfA-dependent promoters and PrfA regulation. Methods LacZ reporter gene fusions used to investigate the inhibitory elements for PrfA-dependent transcription were carried on two promoters of Listeria monocytogenes: a PrfA-dependent promoter of the phospholipase gene pica (PplcA) and a putative promoter of the aroA gene (ParoA2) which contains a similar PrfA-binding site (PrfA-box) and a similar-10 box as PplcA but does not function as PrfA-dependent promoter. A series of hybrid plcA-aroA promoters by exchanging corresponding sequence elements of these two "promoters" were constructed and incorporated into upstream of a promoterless lacZ gene. The variant promoter-lacZ transcriptional fusions were then electroporated into L. monocytogenes wild-type strain P14, prfA mutant P14a and prfA deletion mutant A42, respectively. The expression level of PrfA is the highest in the P14a and the lowest in A42. The corresponding transcription activities of hybrid promoters were measured by the β-galactosidase assay. Results The two critical elements of PrfA-dependent promoters, the PrfA-box and the-10 box, can be functionally exchanged as long as the distance in between is maintained 22 or 23 bp. However, the interspace sequence and the sequence downstream of the -10 box of ParoA2 were strongly inhibitory for PrfA-dependent transcription. Conclusion Downstream sequence together -10 box of ParoA2 might fold into a hairpin structure when present in a single stranded DNA and possibly block the formation of the transcriptional initiation open complex, hence, inhibit the PrfA-dependent transcription from ParoA2.

Henoch Sch?nlein purpura( HSP) is a vasculitis of primary systemic vascular lesions of aller-gic diseases in children. Henoch Schonlein purpura nephritis( HSPN) is the most common allergic purpura sec-ondary renal disease,hematuria and(or)proteinuria of allergic in purpura the course(the most six months) can make diagnosis. The clinical manifestations varyies in severity,and the incidence has increased year by year,the incidence may be related to infections, allergies, immune abnormalities, immune damage and so on. So raising awareness of children nephritis,early diagnosis,early treatment has a great impact on the prognosis of children.
4.The application of problem-based learning in clinical probation of gynecology and obstetrics

ObjectiveTo investigate the teaching effect of problem-based learning(PBL) in clinical probation of gynecology and obstetrics.Methods48 students who majored in obstetrics and took internship in Xiamen Maternity and Child Care Hospital were selected as study subjects and were randomly assigned to control group and observation group.Traditional teaching pattern and PBL were performed in the above-mentioned groups respectively.The score of final test on maternity nursing and self-evaluation on capacity advancement were compared between two groups.ResultsThe score of observation group were significantly higher than control group (P<0.05); Similarly,the self-evaluation of observation group after internship was also elevated with significance (P<0.05) when being compared with control group.ConclusionEvidence showed that PBL was capable of improving the knowledge of maternity nursing,and it was also been proved effective in fostering the ability of self-learning,communication,and solving problem.Therefore,the PBL was appropriate to be promoted in the internship of nursing education.
5.Insulin receptor substrate expression and insulin resistance in intrauterine growth retarded rats

Insulin receptor substrate(IRS)-1 and IRS-2 expression levels of liver tissues and skeletal muscle in intrauterine growth retarded(IUGR)rats were investigated by RT-PCR and immunohistochemistry.An IUGR animal model was established by maternal nutrition restriction during pregnancy.IRS-2 expression level of liver tissue and IRS-1 expression level of skeletal muscle in IUGR rats at 0 and 3 weeks old were significantly lower than those in normal rats at the same age respectively,and insulin resistance was induced in IUGR,and these findings might be the molecular mechanisms susceptible to metabolic syndrome in IUGR rats.
6.Microsurgical anti endoscopic anatomy of sellar region related to key hole approach

Objective To study the endoscopic anatomy of operative fissures in the sellar region related to pterional key-hole approach and to provide the anatomic basis for endoscope-assisted microneuro-surgery in the region. Methods Fifteen cadaver heads were dissected via pterional key-hole approach and the five operative fissures(Ⅰ、Ⅱ、Ⅲ、Ⅳ、Ⅴ) were studied with both microscope and endoscope, by which the outcomes observed were recorded and compared. Results The basic information obtained by the pterio-nalkeyhole approach was comparable to that of the classic ptefional approach. According to the anatomic guideposts under neuroendoscope, the microstructures of the five fissures and Willis' circle would be well demonstrated. It is better to display the various anatomic structures in sellar region by neuroendoscope rather than microscope, particularly some important microstructures. Angled endoscope can be used to inspec thidden but important structure behind the arteries and nerves. Conclusion According to the anatomic guideposts, an endoscopy can be used to enhance the visible field of an operative microscope related to pterional key-hole approach. The endoscope-assisted microsurgery can reduce complications and injury of the important structures and increase the curative effect on the lesions in the sellar region.
7.Construction and expression of a recombinant Bifidobacterium bifidum vaccine of Schistosoma japonicum

Objective To construct a recombinant Bifidobacterium bifidum (Bb)vaccine[Bb (pGEX-Sj26GST-Sj14-3-3)] of Schistosomajaponicum (Sj) and analyze the expression of the fusion gene Sj26GST-Sj14-3-3 of Sj in Bb.Methods The recombinant plasmid pGEX-Sj26GST-Sj14-3-3 was electroporated into Bb to construct a recombinant Bb (pGEX-Sj26GST-Sj14-3-3) vaccine.Mter induction with isopropyl-β-D-thiogalactoside (IPTG),double restriction enzymes digestion and polymerase chain reaction (PCR) were used to identify the recombinant Bb (pGEX-Sj26GST-Sj14-3-3),expression of the recombinant protein was analyzed and identified by sodium dodecyl sulfonate-polyacrylamide gel electrophoresis (SDS-PAGE) and Western blotting.Results The recombinant plasmid pGEX-Sj26GST-Sj14-3-3 was successfully transformed into Bb identified by double restriction enzymes digestion and PCR.SDS-PAGE analysis showed that the relative molecular mass of the expressed recombinant protein was approximately 67 × 103.The expressed protein could be recognized by the immune sera from rabbits infected with Sj by Western blotting.Conclusions The recombinant Bb (pGEX-Sj26GST-Sj14-3-3) vaccine of Sj is successfully constructed.The fusion gene Sj26GST-Sj14-3-3 can be expressed in recombinant Bb and the expressed target protein shows specific antigenicity.

Methods The ratios of Th1/Th2 cells intestinal mucosa and splenic monocytes (SMC) of mouse with DSS induced murine colitis were detected by FCM with two color intracellular cytokine staining. Results (1) In lamina propria mucosa cell (LPMC) of murine colonic mucosa from the acute model, Th1 cell was (8.90?1.23)%, significantly higher than that in the control( P 0.05). 9.Effect of Community-based Rehabilitation on Typical Hemiplegic Gait Post Stroke

ObjectiveTo observe the effect of the ambulation training on the typical hemiplegic gait in community.Methods50 stroke patients with typical hemiplegic gait were divided into two groups: treatment group was treated with systematic rehabilitation training, 5 times each week for 3 months, and control group accepted self-walking training. The follow-up was performed every week at home. The two groups were assessed by the timed "up & go" test, the Fugl-Meyer Assessment(FMA) and Satisfaction with Life Scale(SWLS).ResultsThe scores of the timed "up & go" test, FMA, and SWLS showed that there weren't significant difference between the treatment group and the control group 1 month after treatment; but the scores in the treatment group were obviously higher than those in the control group 3 months after treatment.ConclusionAmbulation training could improve the typical hemiplegic gait and quality of walking of stroke patients with hemiplegic gait in community.
