Objective To explore the mechanism of diabetic hepatic fibrosis by observing effects of high glucose and insulin on expressions of transforming growth factor beta 1 (TGF-β1) and tissue inhibitor of metalloproteinase (TIMP-1) mRNAs of hepatic stellate cell (HSC) in rat.Methods Hepatic stellate cell lines in vitro were administrated by different dose of glucose without insulin and glucose with insulin for 72 h,and mannitol was chosen as hyperosmosis control group.Real time fluorescent quantitation polymerase chain reaction (RT-FQ-PCR) was used to determine expressions of TGF-β1 and TIMP-1 mRNAs of HSC in each group.Results Expressions of TGF-β1 and TIMP-1 mRNAs of HSC in each group could be detected and showed no significant difference among groups (P ＞ 0.05).However,in general,expression of TGF-β1 mRNA in glucose with insulin group decreased and TIMP-1 mRNA increased.Conclusions The mRNA expressions of TGF-β1 and TIMP-1 in HSC could not be induced by high glucose alone,high dose of insulin may increase expressions of HSC TIMP-1 and reduce TGF-β1 expression.The main mechanisms of diabetic hepatic fibrosis may not be TGF-β1 pathway but be closely related to TIMP-1 pathway.

Primary hepatic carcinoma is one of the high incidence malignant tumors and the mortality ranked the second in the death caused by malignant tumors in China. The research progress on the epidemiology of primary hepatic carcinoma, the related environmental factors(virus, parasite, bacterium infected, aflatoxin ingested, drinking water pollution, smoking and drinking alcoholic beverages) and genes were reviewed and the mechanism of primary hepatic carcinoma was discussed in the present paper.

Objective To study the changes of colonic motility in chronic stress-induced depression rat model,the association between interleukin 6 (IL-6) and depression and its effects on contraction of colonic smooth muscle in vitro.MethodsThere were 30 healthy adult Wistar rats,of which chronic stress-induced depression model was created in 15 rats and the other 15 rats were in control group.The ratio of sugar consumption to body weight and the percentage of sugar preference were tested in these two rats groups through sugar preference experiments.The behavioral changes of two rats groups were detected by open field test.Colonic motility was evaluated with simulated fecal discharge experiment,and the serum IL-6 level was measured with enzyme linked immunosorbent assay (ELISA).IL-6 in colon tissue at mRNA level was determined by RT-PCR.The contraction ability of rat colonic smooth muscle was observed.The colonic smooth muscle of control group was treated with different concentration of IL-6,the effects of IL-6 on normal colonic smooth muscle and its response to acetylcholine (Ach) were evaluated.Results In model group,the ratio of sugar consumption to body weight was 0.12±0.03,the percentage of sugar preference was 16.17％ ±2.61 ％,total walking distance was (741.54 ± 341.10) cm and total times of upright was 15.69 ± 8.00,which were all significantly lower than those of control group (0.18±0.02,25.54 ％ ± 2.32 ％,1336.20 cm±698.80 cm and 24.87±7.90,respectively) (P=0.041,0.044,0.002 and 0.001).It took (109.78±48.00) min for defecating in model group,which was significantly longer than that of control group (28.00±11.10) min (P=0.002).The IL-6 levels were high both in the serum and colon tissue of model group rats.The response of colonic smooth muscle to Ach was weaker in model group than that of control group (P=0.035).In model group,there were no significant difference in the response of colonic smooth muscle to IL-6 at 200,500 and 1000 pg/ml (P =0.935、0.825、0.766 ).After Ach added,there was significant difference in the response to Ach between IL-6 at 500,1000 pg/ml (R=1.1480±0.1000 and 1.1361±0.1500) and control group only added with Ach (R=1.6100±0.4500) (P=0.000 and 0.004).ConclusionIL-6 level increase in chronic stress-induced depression rat model and causes smooth muscle contraction disorder through reducing the sensitivity of intestinal smooth muscle to Ach.

Objective To explore the early surgical treatment for sleep apnea in children with the influence of mental development,provide important basis for children with sleep apnea early surgery.Methods 40 children with sleep apnea underwent bilateral amygdala and/or adenoidectomy (observation group).The Chinese wechsler intelligence scale for children (C-WYCSI) was used for preoperative and postoperative 6 months of intelligence tests,including language IQ and operation IQ test two parts,a total of 11 points.Operation IQ but also contain visual analysis test(VA) and two categories,including geometry test (GD).According to the 1 ∶ 1 matching selection of the same gender,and age(± 3 months),family condition was quite healthy check-up preschool children comparing 40 cases as control group.Results Observation group preoperative total intelligence quotient (IQ),verbal IQ,IQ operation was 82,81,83,respectively,which were lower than those of the control group 101,101,101 (t =4.131,3.952,3.842,all P ＜ 0.05).Postoperative total intelligence quotient(IQ),verbal IQ and operation IQ 98,98,99,respectively,were in normal range,with no statistically significant difference were observed in the control group (all P ＞ 0.05).Conclusion Children with sleep apnea early surgical treatment can improve children mental development.Children with sleep apnea early,breathing and surgical indications should be early surgical treatment.

Objective; To explore the possible effect of hyperthermia combined with Prog on a human tongue cancer cell line Tca8113 and drug-resistance cell line Tca8113/BLM in vitro. Methods;The effect of hyperthermia combined with Prog on Tca8113 and Tca8113/ BLM cells were determined by MTT. The concentration of ADM, the expression of P-gp and MRP were detected by flow cytometry (FCM). Results;Compared with Prog or hyperthermia, the hyperthermia at 41 X. for 1 h combined with Prog showed obvious inhibitory effect in Tca8113 and Tca8113/BLM cells(P<0.01). The concentration of ADM in Tca8113 and Tca8113/BLM cells increased obvi-ously(P<0.01). Expression levels of P-gp and MRP in Tca8113 and Tca8113/BLM cells decreased significantly. Conclusion:The hyperthermia combined with Prog can increase the intracellular concentration of ADM in Tca8113 and Tca8113 /BLM cells, which reverse the drug-resistance of Tca8113/BLM cells to BLM and down-regulate expressions of P-gp and MRP.

Objective To evaluate the clinical significance of determination for serum insulin like growth factor binding protein 3 (IGFBP 3) protease activity. Methods Serum IGFBP 3 protease activity in normal persons, parturient pregnant women, acromegalic patients and patients with growth hormone deficiency (GHD) was determined by Western blot technique. Results Serum IGFBP 3 protease activity in parturient pregnant women, acromegalic patients,normaladultsanduntreatedGHDpatientswere (75.2?2.2)%, (68.6?3.9)%, (22.7?3.1)% and (13.4?4.2)% respectively. Serum IGFBP 3 protease activity in GHD increased to (29.0?6.8)% after treatment with recombining human somatropin (rhGH) for six months. Conclusions Serum IGFBP 3 protease activity is modulated by GH and affects serum levels of members of GH IGF Ⅰ axis.

Objective To investigate the diagnostic value of contrast-enhanced ultrasound endoscopy(CE-EUS) for the upper gastrointestinal tract and adjacent lesions.Methods One hundred and eighty samples of upper gastrointestinal tract and adjacent lesions in the First Affiliated Hospital of Guangxi Medical University from June 2014 to March 2015 were collected and performed CE-EUS.The preliminary diagnosis was taken by observing the features of various kinds of solid mass,which was conducted the contrastive analysis with the results of fine needle aspiration cytology(EUS-FNA) and (or) histopathology, biliary brush cytology smear results,operative pathologic findings,and CT,MRI scans and follow-up results.Results In 180 cases of gastrointestinal tract and adjacent placeholder,169 cases of CE-EUS were in line with the final results,while 11 cases were not consistent,and the diagnostic consistency rate was 93.9%.Conclusion CE-EUS can guide EUS-FNA puncture for increasing its positive rate..

50 000 bp)was extracted from frozen gastric cancer tissue and their corresponding normal samples and used for RLGS assay.The genome DNA was digested by methylation-sensitive restriction enzyme Not Ⅰ, and labeled by radioisotope ~(32)p,then separated by two-dimensional electrophoresis and autoradiography. Experimental conditions for each step were optimized step by step.DNA fragment sequences for the dots on scanning profile were identified according to the human RLGS database.Results RLGS assay was set up and used to get the RLGS profiles of the representative tested samples successfully.These profiles can display more than 1 200 available dots averagely,the profiles of high quality DNA sample can display more than 1 800 dots which is the average level at an excellent RLGS lab,discrepant dots which had weaken or enhanced signals and their sequence information were obtained.The result can be reproduced.Conclusion The RLGS assay is established,stabilized for detection of DNA methylation of tissue samples.
