1.Experimental study of astrocyte reaction to injury in vitro

Objective To study the occurence, development and regulation of reactive gliosis with astrocyte (Ast) in vitro. Methods Ast was isolated and cultured in vitro and its model of reactive gliosis was established by scratching the cultured astrocytes. The reactivity and rules of Ast to injury was studied by morphological changes, RT-PCR, immunocytochemistry, in situ hybridization and imaging analysis. Results After scratching, the astrocytes showed typical features of reactive gliosis, with the hypertrophic cell body, thickened and lengtheded processes, and enhanced glial fibrillary acidic protein (GFAP) staining. In situ hybridization and RT-PCR analysis confirmed that the expression of GFAP mRNA was markedly increased. These changes occurred 1 d after scratching and reached the peak 5 to 7 d after injuring. Conclusion A model of reactive astrogliosis was successfully established in vitro which showed an active reaction to injury. The characteristics of reactive gliosis parallel that seen in vivo.
2.Changes of macroglia during healing of the brain stabbing injury

The changes of the morphology and percentage of the macroglia surrounding focus of brain stabbing injury (BSI) were observed by immunohistochemistry,immunoinfluorescence stain and flow cytometer(FCM) and the effect of magroglia during glial scar forming were elucidated.The results showed that a large number of GFAP-immunoreactive positive cells were accumulated around the focus of BSI.These cells were hyperplastic,hypertrophic,and emerged swollen cytoplasmic processes.The most marked changes were observed at 1-2 week after BSI.The results of FCM showed that the percentage of GFAP positive cells increased gradually and reached to a peak during 1~2 week after BSI.The peak ratio of GFAP positive was about 46%.However,the changes of morphology and number of GC positive cells were not detected after BSI.The authors believed that astrocyte is the main macroglia during glial scar formatting .The oligodendrocytes is not an active cell during this course.
3.Studies on the respiratory function of platelet mitochondria in patients with Parkinson's disease

Objective To investigate the respiratory function of platelet mitochondria in patients with Parkinson's disease(PD) and explore the assosiation with the pathogenesis of PD.Methods The platelet mitochondria of 35 patients with PD were extracted and the mitochondria respiratory rate and respiratory control rate were measured.Furthermore,the changes of activities of mitochondria complex Ⅰ,Ⅱ,Ⅲ and Ⅳ were observed.Results The platelet counts of patients with PD showed no changes, but the respiratory rates of respiratory state 3 were significantly decreased using the 5 mmol/L glutamate,2.5 mmol/L malate +5 mmol/L pyruvate and 5 mM succinate as substrates. Moreover, the activities of mitochondria complex Ⅰ were obviously reduced and the activites of other mitochondria complex were no changes.Conclusion Patients with PD show exactly abnormal mitochondria respiratory functions, and it might be an important mechanism in the pathogenesis of PD.
4.A Comparative Study on Laparoscopic-Assisted Vaginal Hysterectomy and Vaginal Hysterectomy

Objective To explore the clinical value of laparoscopic-assisted vaginal hysterectomy(LAVH)in large uterus.Methods Retrospective analysis was conducted on clinical data of 94 patients(whose uterus were as big as 10-18 gestational weeks)who received hysterectomy from January 2005 to March 2007,in which 56 cases were performed laparoscopic-assisted vaginal hysterectomy(LAVH group)and 38 cases vaginal hysterectomy(VH group).The operation time,blood loss,postoperative hospital stay,and the incidence of postoperative complications were compared between the two groups.Results Compared with VH group,there were a lower chance of abdominal hysterectomy(0/56 vs 5/38,?2=5.389,P=0.020),a shorter operation time [(149?11)min vs(179?14)min,t=-11.610,P=0.000] and a shorter postoperative hospital stay [(5.8?1.4)d vs(7.3?3.6)d,t=-2.825,P=0.006] in the LAVH group.There were no significant differences in blood loss,morbidity and time to first flatus between the two groups.Conclusions The LAVH extends the indications of VH,ensuring the safety of VH for the uterus bigger than 10 gestational weeks,therefore it is an operative procedure to be recommended.
5.Effect of ferulic acid on the proliferation of, as well as melanin synthesis, tyrosinase activity and expressions of c-kit and ERK proteins in keratinocytes

Objective To evaluate the in vitro effect of ferulic acid on the proliferation of,as well as melanin synthesis,tyrosinase activity and expressions of c-kit and ERK proteins in cultured normal human epidermal melanocytes.Methods Cultured normal human epidermal melanocytes were treated with various concentrations of ferulic acid for different durations,and those remaining untreated served as the control.Then,3-(4,5-dimethylthiazol2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium,inner salt (MTS) assay was performed to estimate cell proliferative activity at 24,48 and 72 hours,sodium hydroxide solubilization method to quantify melanin content in melanocytes at 72 hours,dopa oxidation assay to evaluate tyrosinase activity at 72 hours,Western blot to measure the expressions of c-kit and ERK1/2 proteins at 72 hours.Results Cellular proliferative activity was significantly inhibited in melanocytes treated with ferulic acid of 0.01,0.1 and 1 mg/ml for 24,48 and 72 hours compared with untreated melanocytes (all P < 0.05),and the 72-hour treatment with ferulic acid of 1 mg/ml showed the strongest inhibitory effect.Ferulic acid at 0.01,0.1 and 1 mg/ml all markedly suppressed melanin synthesis and tyrosinase activity,decreased the expressions of c-kit and ERK1/2 proteins in melanocytes,with significant differences in these parameters between ferulic acid-treated and untreated melanocytes (all P < 0.05).Conclusions Ferulic acid could downregulate the proliferation of,as well as melanin synthesis,tyrosinase activity,and expressions of c-kit and ERK proteins in cultured human epidermal melanocytes.
6.Thoracolumbar spinal reconstruction with titanium mesh implantation combined with internal fixation after tumor resection: stability and biocompatibility

BACKGROUND:Clinical resection of thoracolumbar spinal tumor has a great impact on the spinal stability. Positive internal fixation is required clinicaly in order to maintain the spinal stability. The use of titanium mesh implantation can provide a firm internal fixation folowing resection of tumors. OBJECTIVE:To explore the spinal stability undergoing titanium mesh implantation with internal fixation folowing thoracolumbar tumor resection. METHODS:Twenty-four patients with thoracolumbar tumor admitted at the Central Hospital of Suining City from September 2013 to September 2014 were randomly selected and given tumor resection folowed by titanium mesh implantation with internal fixation. After treatment, patients were folowed up for 1-12 months to observe and analyze the neural functional recovery and spinal stability of the patients. RESULTS AND CONCLUSION:Al the 24 patients successfuly completed the operation treatment, and there was no case of death at the end of folow-up. During the folow-up, patient's clinical symptoms and neural function were significantly relieved, and Frankel classification was significantly improved after treatment. Regular X-ray examination showed that there was no change in the position of titanium mesh and anterior internal fixation system. There was also no titanium mesh colapse, internal fixation fracture and loosening, and the spinal stability was stil excelent. These findings indicate that patients were not changed, and did not appear because the amount of loose, good spinal stability. Resection of thoracolumbar tumors showed that the thoracolumbar spinal reconstruction with titanium mesh implantation combined with internal fixation folowing tumor resection can obtain good clinical effect and excelent biocompatiblity.
7.Prediction of the bioequivalence of different crystal forms of rifampicin based on physiologically based pharmacokinetic model

The physiologically based pharmacokinetic (PBPK) modeling strategy was adopted to predict the pharmacokinetic behavior of crystal forms I and II of rifampicin in humans, which was used to determine whether the two were bioequivalent.After conducting studies in vitro of the two crystal forms, a rat PBPK model was established based on the pharmacokinetic data of intravenous administration in rats.The model was optimized by the pharmacokinetic data of oral administration in rats.Species were extrapolated to healthy humans, and the extrapolation model was used to predict such pharmacokinetic behaviors as the drug-time curve, absorption site, and absorption amount of the two crystal forms of rifampicin in healthy humans.The prediction results of the healthy human model showed that the cmax of form I and form II rifampicin were 8.42 and 10.35 μg/mL, tmax were 0.40 and 0.32 h,and AUC0-t were both 62.90 μg·h/mL.According to the prediction results of absorption, neither crystal form I nor crystal form II rifampicin was absorbed in the stomach, yet both were completely absorbed in the intestinal tract, with both the absorption site and the absorption amount were basically the same.The pharmacokinetic parameters of both crystal forms I and II of rifampicin were very close, which could indicate bioequivalence.
8.Pharmacokinetics and in vitro-in vivo correlation of progesterone suspension injection

Abstract: To investigate the in vitro release, in vivo pharmacokinetics, and the in vitro-in vivo correlation of progesterone suspension injection, self-made progesterone suspension injection was taken as an example. The in vitro release curves of three different particle sizes of progesterone suspension injections were measured using paddle method and dialysis bag method. The in vivo pharmacokinetic characteristics of self-made progesterone suspension injection was studied on SD rats. The plasma concentration of self-made progesterone preparation was detected after intramuscular injection, and correlated with the in vitro release profiles obtained by the dialysis bag method after processing by Wagner-Nelson method. The results showed that when the in vitro release of three different particle sizes of progesterone suspension injections was measured by the paddle method, more than 85% was rapidly released within 20 min, while 85% cumulative release was reached at 40 h, 84 h and 120 h by dialysis bag method, respectively. The release rate obtained by the dialysis bag method was basically consistent with the in vivo release trend, with a correlation coefficient of >0.95, indicating a strong in vivo and in vitro correlation. This study provides some reference for the establishment of the in vitro and in vivo correlation of long-acting suspension injection.
9.An instrument for inducing continuous passive motion after fracture of the tibial plateau in rabbits

Objective To design and develop intelligent rehabilitation equipment for administering continuous passive motion (CPM) of a rabbit's knee joint after tibial plateau fracture.Methods The equipment constructed had three main parts:the core machinery,electronic control and a control program designed based on bionics principles.Twenty six-month-old New Zealand White male rabbits were randomly divided into sedentary (SED) and CPM groups after their knees had been fractured.The rabbits in the CPM group were given 30 min of early joint rehabilitation once a day for 4 weeks using the CPM equipment,while those in the SED group were kept in their cages and allowed free activity without any special exercise program.The body weight,range of motion and swelling of the affected knee joint were measured before the fracture and on the 3rd,7th,14th,21st and 28th days after the fracture.On the 28th day after the fracture the pathological structure of the articular cartilage on the operative side was observed under a light microscope.Results The equipment ran safely and reliably,and drove the rabbits to move synchronously.It could accurately and conveniently adjust the knee flexion angle,movement speed and movement time.The intelligence of the equipment met the experimental requirements.On the 3rd day after the operation the average range of motion in the joints of both groups had changed significantly compared to that before the fracture.On the 28th day after the fracture the average degree of swelling and range of motion in the CPM group were significantly different from those of the SED group.On the 28th day,deformity and the smoothness of the fracture line in the CPM group were superior to those in the SED group.Moreover,the dominant tissues in the defect area of the CPM group were mainly hyaline cartilage while those in the SED group were mainly repair fibrocartilage.The defect area and its adjacent articular cartilages,chondrocyte regeneration and arrangement,layers of cells and subchondral tidal line recovery of the CPM group were better than in the SED group on average.Conclusion The equipment for knee joint manipulation is convenient to use,reliable and effective for the early rehabilitation of tibial plateau fracture,at least in rabbits.It promotes remodeling of the fracture and cartilage repair after tibial plateau fracture,and also improves range of motion in the knee and reduces swelling.
10.Development and application of intelligent management system for documents and records of regional medical laboratory cen-ter

Objective To develop an intelligent management system of documents and records that meets the requirements of ISO 15189 quality management system applied to regional medical laboratory center.Methods Based on the relevant terms of ISO 15189 document and record,an intelligent management system for laboratory documents and records with B/S architecture was established by the help of computer technology.Results The document and record management system stored the quality system documents required for ISO 15189 review as well as the external documents,such as laboratory-related laws and regulations,standard guidelines and expert consensus,which can be accessed by users through a computer or mobile phone.The multi-process control mode was adopted to author-ize the special personnel to complete the compiling,review,approval and,release of the files according to the type and attribute of the documents.The management system of records established 9 special records,319 form records and 20 process records to support the ef-fective operation of the quality system.Conclusion The established intelligent management system for documents and records has fully met the requirements of ISO 15189 on documents and records management,realized the sharing and interconnection of the documents in the given region.It is convenient for the employees to consult the documents and fill in the records in a timely and efficient manner.
