AIM:To introduce and evaluate two methods for sample size calculation in doseresponse research.METHODS:The methods used for sample size calculation in dose-response research include Cochran-Armitage trend(CAT) test proposed by Jun-mo Nam and the Unified contrast(CUC) method proposed by Chang. This paper takes the example of binary data, uses the two methods to calculate the required sample size and evaluate the power by simulation under the assumption that the response rate had a linear relationship with dosage under the logit scale.RESULTS:When the probability of response followed a linear trend on the logit scale, the sample sizes estimated from the two methods are approximately equal in various parameter's settings. The simulated power of CAT test was close to the expected one, however, the simulated power of CUC method was obviously affected by the values of contrast coefficient,if a contrast whose coefficients parallel the expected response, the CUC method was more powerful. CONCLUSION:When the probability of response follows a linear trend over dosage on the logit scale, the two methods reach almost the same result,in such circumstance, the Chang's unified contrast method will obtain an increasing power.

AIM: To aim to seek the best operation conditions of the inorganic ceramic membrane in the separation of effective matters from the homogenate of earthworm. METHODS: The homogenate flux was adopted as index,to select the membrane material, filter pressure、homogenate temperature and concentration. RESULTS: The experiment showed that the filter flux had a significant positive correlation with pressure difference and liquid temperature, and negative correlation with liquid concentration. CONCLUSION: In the separation of earthworm homogenate,0.2 ?m Al_2O_3 and 50 nm ZrO_2 film pipe were respectively selected as primary and secondary separating unit on the condition of 0.15 mPa pressure,50 ℃ and 33.%(w/w) homogenate concentration.

This study was aimed to explore an effect evaluation method of traditional Chinese medicine (TCM) in the treatment of viral pneumonia. Association rules were used to analyze the therapeutic effect of heat, stagnation, and phlegm related syndromes from 297 respiratory syncytial virus (RSV) pneumonia children before and after treatment. The results showed that through the comparison of the frequent itemset of heat, stagnation, and phlegm related syn-dromes, in part of the time, the support degree of the experimental group was obviously lower than that in the con-trol group (P < 0.05). The frequent itemset of the experimental group disappeared 1~3 d earlier than the control group, which indicated the effect in the experimental group was better than the control group. It was concluded that association rules can be used for evaluating therapeutic effect of TCM in treatment of viral pneumonia. The effect evaluation method based on association rules of symptoms is more in line with the syndrome differentiation of TCM.

OBJECTIVETo study the regulation trend of resveratrol on TNF-alpha, IL-1beta, IL-6 expressions in bronchoalveolar layage fluid (BALF) of RSV-infected BALB/c mice at different time points.
METHODRSV-induced BALB/c mice were orally administered with resveratrol. Their BALFs were collected at 24, 72 and 144 h after the first nasal drip with RSV to detect the level of TNF-alpha, IL-1P3, IL-6 by EILSA.
RESULTThe expression of TNF-alpha, IL-1Pf and IL-6 in BALF increased significantly compared with the normal group (P <0. 01) after 24 hours of RSV infection, while the expression of TNF-alpha (P < 0.01), IL-1beta (P < 0.05), IL-6 (P < 0.01) in the resveratrol group decreased notably compared with the model group. After 72 hours of infection with RSV, although the expression of TNF-alpha (P < 0.05), IL-1beta (P < 0.01) and IL-6 (P < 0.01) in BALF in model group were higher than those in the normal group, they were much more lower than at 24 h. The expression of IL-1beta and IL-6 (P < 0.05) in the resveratrol groups were down-regulated significantly, but no difference had been shown in TNF-alpha expression compared with the RSV infection group. After infection with RSV for 144 h, the expression of IL-1beta (P < 0.01) and IL-6 (P < 0.05) in BALF in the model group were higher than those in the normal group, but there was no difference in the secretion of TNF-alpha. The expression of TNF-alpha, IL-1beta and IL-6 showed also no remarkable difference between the resveratrol groups and the RSV infection group.
CONCLUSIONResveratrol can inhibit the over expression of inflammatory factors TNF-alpha, IL-1beta, IL-6 in bronchoalveolar lavage fluid of RSV-induced BALB/c mice and keep them at a low level with the passing of infection time.

By analyzing the epidemic characteristics of influenza during the COVID-19 epidemic in Jiangsu Province from 2020 to 2022, it found that 90 721 influenza-like case samples were collected in Jiangsu Province from 2020 to 2022, of which 6 732 were nucleic acid-positive samples, with an average positive detection rate of 7.4% in three years. The annual positive detection rate presented a U-shaped distribution, with positive detection rates of 4.4%, 3.2% and 14.7%, respectively, with statistically significant differences ( χ2=12 126.00, P<0.001). During the seasonal peak period of influenza from 2020 to 2022, there was a significant decrease in the intensity of the two influenza activity peaks that occurred before the virus became fully prevalent in the population. The first peak occurred from January to February 2020, and the positive detection rate of influenza nucleic acid decreased from 54.4% (317/583) in the third week to 2.1% (12/584) in the eighth week, with a statistically significant difference ( χ2=394.49 , P<0.001) . The second occurred in December 2022, and the positive detection rate of influenza nucleic acid decreased from 14.9% (90/605) at the 49th week to 1.9% (11/572) at the 52nd week, with a statistically significant difference ( χ2=62.88, P<0.001). The influenza epidemic in Jiangsu Province from 2020 to 2022 had obvious seasonal characteristics, and the distribution differences of influenza virus-positive cases in each month were statistically significant ( χ2=858.00, P<0.001), with two epidemic peaks each year: winter, spring (December to March of the following year), and summer, and autumn (July to November). The epidemic strains were the B-V strain and seasonal H3 strain, respectively. There was a statistically significant difference in the positive detection rate of influenza cases detected in different age groups ( χ2=60.00, P<0.001). The age group between 5 and 14 years old had the highest influenza-positive detection rate (10.4%), while the age group≥60 years old had a relatively low influenza-positive detection rate (5.1%). The positive detection rate decreased with the increase in the age group ( Z trend=12.82, P<0.001).

Allergic rhinitis is a common allergic disease in children. Its pathogenesis is complex and it is difficult to achieve radi-cal cure or effective and stable long-term treatment goals.Chinese medicine has obvious advantages in preventing and treating allergic rhinitis in children due to its wide range of targets,long-lasting effects and few adverse reactions. This paper proposes that the onset of allergic rhinitis is mostly caused by the dysfunction of the lung, spleen and kidney, the external wind triggering the latent wind, and the combination of the two winds.A staged prevention and treatment strategy of Chinese medicine should be adopted, which includes dispersing external wind, suppressing latent wind, and promoting lung-qi and clearing nasal orifice during the attack period to treat its symptoms, and preventing external wind, calming down latent wind, and regulating and tonifying the lung, spleen, and kidney during the remission period to treat its root cause;meanwhile, attention should be paid to avoiding the adverse effects of congenital endowment factors and the induction of acquired environmental factors, strengthening the body's health to protect against the evil wind, preventing the transformation of existing diseases and the recurrence of allergic rhinitis in children at all stages.

Objective One H5N6 subtype avian influenza virus,A/environment/ Zhenjiang/C13/ 2013,was isolated from environment in Zhenjiang in 2013 during epidemiological surveillance and was identified by HA/HI test and specific RT-PCR test.H5N6 AIV was subjected to genome sequencing to investigate the genomic composition. Methods The next-generation sequencing (NGS) Illumina MiSeq Platform was used to obtain complete genome sequence information from influenza virus isolates.MEGA6.0 software was used to align the eight fragments of the virus. The phylogenetic and molecular characteristics of the isolate were analyzed by Neighbor-Joining method. Results Blast searches demonstrated that the NA gene belonged to the same clade with H6N6 viruses currently circulating in China, whereas the other seven genes were found to be more similar to those of eastern Asian H5N1 AIV strains gene. The amino acid motif of cleavage site between HA1 and HA2 was PLREKRRKR ↓ GLF,which was the typical characteristics of the HPAIVs.The receptor binding site in the viral HA gene possesses the residues Gln 226 and Gly 228 (H3 numbering),indicating an avian-like receptor binding specificity. Conclusion One H5N6 subtype avian influenza virus was isolated from environment, phylogenetic analysis showed that the NA gene belonged to the same clade with H6N6 viruses. The other seven genes belonged to H5N1 subtype. It suggests that strengthen the continuous surveillance of avian influenza A viruses is necessary.
