Objective Proliferation and apoptosis play a major role in the development of tumor cells, and the intranuclear transcriptional factor c-fos is significantly up-regulated in the primary hepatocellular carcinoma and involved in early carcinogenesis. The purpose of the present study is to investigate the apoptotic effect of c-fos antisense oligodeoxynucleotide(ASO) on human hepatocellular carcinoma HepG2 cells and the participation of Caspase3 in this process. Methods Cell culture,Hoechst 33258 staining,real-time PCR and Western blotting were used in present study.Cultured HepG2 cells were divided into 3 groups: 1) control group: cultured with 10?l saline;2) sense oligodeoxynucleotide(SO,used as a negative control) treated group: co-cultured with 10?l SO(5?g/?l);3) ASO treated group: co-cultured with ASO 10?l(5?g/?l).The subsequent experiments were performed 1h after cultivation for each group.Hoechst 33258 staining was performed to detect the apoptosis by observing the staining of nuclear chromatin.Real-time PCR and Western blotting were used respectively to detect the expression of Caspase 3 at mRNA and protein levels after different treatments. Results Hoechst 33258 staining revealed that the nuclei of HepG2 cells showed diffuse and adqulis fluorescence in control and SO-treated groups, while dense and dark fluorescence was observed in ASO-treated group, which indicated that c-fos ASO had significantly induced apoptosis of HepG2 cells. The expression of Caspase 3 in ASO group was enhanced both at mRNA and protein levels compared to that in control groups. Conclusions C-fos ASO significantly induces apoptosis in human hepatocellular carcinoma HepG2 cells,as shown by Hoechst 33258 staining and higher expression of Caspase 3 mRNA and protein. Moreover,Caspase 3 activation is involved and probably plays an important role in c-fos ASO-induced apoptosis of HepG2 cells.

Objective To analyze the radiation doses to the head, body, and tail of the hippocampus in intensity-modulated radiotherapy(IMRT)for nasopharyngeal carcinoma(NPC). Methods Ten NPC patients treated with IMRT were selected, and the head, body, and tail of both hippocampi were delineated on T1-weighted images. The doses to the hippocampus were then analyzed. WAIS-CR speech test results were tested by paired sample t-test. Results The mean doses to left and right hippocampi were 1 147±976 cGy and 1 011±602 cGy, respectively. The mean doses to the head, body, and tail of the left hippocampus were 1 739± 1 317 cGy, 890± 982 cGy, and 547± 688 cGy, respectively(P=0.042);the mean doses to the head, body, and tail of the right hippocampus were 1 691±942 cGy,744±483 cGy,and 531±603 cGy,respectively(P=0.002).The dose to the hippocampus decreased from the head to the tail, and the irradiated volume also decreased as the dose varied. Conclusions The dose to hippocampus decreases from the head to the tail in NPC patients treated with IMRT,which is worthy of attention.

Objective To analyze the exposed dose of hippocampus(HC)of T3,T4nasopharyngeal carcinoma patients treated with intensity modulated radiotherapy(IMRT). Methods The bilateral HCs were delineated and were divided into head(HH),body(HB)and tail(HT)for 62 nasopharyngeal carcinoma patients treated with IMRT.The dose parameters of HC were then analyzed. Results The mean dose of left and right HC was(1 127±704)cGy,(1 173±762)cGy. The mean dose of left HH,HB and HT was(1 732±1029)cGy,(820±632)cGy,(423±366)cGy(P=0.000);while the mean dose of right HH, HB and HT was(1 985±1101)cGy,(837±531)cGy,(432±343)cGy(P=0.000).The exposed dose and the volume exposed in different dose of HH were obviously higher than those of HB and HT.The dose parameters of HH,HB and HT decreased in turn. The involvement of sphenoid sinus, ethmoid sinus and cavernous sinus correlated with high exposed dose of HC. Conclusions The exposed dose of HH,HB and HT was different in nasopharyngeal carcinoma patients treated with IMRT.The exposed dose of HH was the highest, which should be emphasized especially. The involvement of sphenoid sinus, ethmoid sinus and cavernous sinus suggest high exposed dose of HC.

ObjectiveTo investigate the distribution of traditional Chinese medicine (TCM) syndrome types and influencing factors of protein-energy wasting (PEW) in chronic kidney disease (CKD) undergoing maintenance hemodialysis (MHD). MethodsAccording to diagnostic criteria, 164 patients with MHD were divided into PEW group and non-PEW group. The clinical data of all patients were collected, including general information such as gender, age, height and weight, disease characteristics such as course, cormobidity, and haemodialysis duration, laboratory indicators such as blood routine, liver function, renal function, electrolyte, blood lipid, grip strength, and the four examinations. Logistic regression analysis was used to find the influencing factors of PEW by taking the clinical indicators with significant differences between the two groups (P<0.05) as the independent variables, diagnosis of PEW as the dependent variable, and normal values as the reference. ResultsOut of 164 patients with MHD, there were 96 (58.5%) cases in PEW group and 68 cases (41.5%) in non-PEW group. Compared to the non-PEW group,PEW group had increased age, ratios of bedrest, deep vein preservation, edema, and low grip strength, percentages of comorbidities type 2 diabetes, cardiovascular and cerebrovascular diseases, infections and anemia, and levels of alanine aminotransferase and permine amin aminotransferase, as well as decreased body mass index, self-care ratio, internal arteriovenous fistula, red blood cell count, hemoglobin, serum total protein, serum albumin levels (P<0.05). The PEW group had significantly higher frequency of poor appetite and digestion, abdominal distension, fear of cold and preference of warmth, weak breathing and fatigue, poor appetite, oliguria, nausea and vomiting than non-PEW group (P<0.05). The incidence of both yin and yang deficiency syndrome and damp-turbidity syndrome were significantly higher in the PEW group than the non-PEW group, while that of liver-kidney yin deficiency syndrome and stirring of wind syndrome were lower (P<0.05). Logistic regression analysis showed that low BMI (<22 kg/m2), inability to take care of oneself, low grip strength, low serum albumin (<38 g/L), infection, older age, fear of cold and cold limbs, and poor appetite were the risk factors of PEW in patients undergoing MHD (P<0.05). ConclusionThe root syndrome of MHD-PEW patients is both yin and yang deficiency, concurrent with damp-turbidity syndrome. Low BMI, low serum albumin, infection and older age may be the influencing factors of PEW in patients undergoing MHD.
