1.Feasibility of adipose tissue-derived stem cell sheet fabricated ex vivo on temperature-responsive scaffolds as seed cells for ocular surface reconstruction

Background Development of corneal tissue engineering creates a new therapeutic method for severe corneal diseases.However,ideal seed cells and scaffold for corneal surface reconstruction have not yet been investigated well.Adipose-derived stem cells (ADSCs) are varified to have a self-renewal ability and epithelioid features,and temperature-responsive scaffolds (TRSs) can offer technical support for stem cell sheet.Objective This study was to investigate the characteristics of ADSCs cultured on TRSs and compare these features to typical oral mucosal epithelial cells (OMECs),and therefore to explore the feasibility of reconstruction of ocular surface with ADSCs as seed cells.Methods Self-made TRSs were prepared by adding isopropyl alcohol dissolved poly-Nisopropylacrylamide (PNIPAAm) to each polystyrene tissue culture dish and then irradiating using an election beam.Subcutaneious fatty tissue of rabbit neck was obtained to culture ADSCs,and 4 pieces of oral cavity mucosal tissue were digested and cultured to obtain OMECs.Then the ADSCs and OMECs were incubated on TRSs,and cell morphology,growth rate,detached duration and survival counts were compared between ADSCs and OMECs.The ADSCs sheet and OMECs sheet were stained with hematoxylin and eosin for morphological examination.Immunochemistry was used to observe the expressions of stem-cell biomakers and epithelioid-cell biomakers in the cells.The ultrastructure of cell surface was observed under the scanning electron microscope.Results Self-made TRSs were similar to ordinary culture dish in the transparancy and smoothness.The water contact angle of 4 in 5 samples were >10° with the effective rate upto 80%.A DSCs showed the elongated fusiform in shape,while OMECs showed a cobblestone appearance.The growth cycle,detached duration and cell number of ADSCs were 12-14 days,(46.0 ±9.6) minutes and (7.9 ±1.1)×105/sheet,and those of OMECs were 14-16 days,(91.9 ±10.9) minutes and (45.8 ±26.5)×105/sheet,respectively,showing statistically significant differences in the detached duration and cell counts between ADSCs and OMECs (P=0.002,0.028).Hematoxylin and eosin staining showed that ADSCs sheet comprised only 1-3 layer cells,while OMECs showed 4-5 layer cells.ATP-binding cassette superfamily G member 2 (ABCG2),p63 and cytokeratin 12 (CK12) were positively expressed in both ADSCs sheet and OMECs sheet.Closely packed cells and typical eithelial microvilli in the cell surface were exhibited in both ADSCs sheet and OMECs sheet under the scanning electron microscope.Conclusions Self-made TRSs can be used as scaffold of ADSCs.The ADSCs sheet on the TRSs appears to have a good cell vitality and therefore is a new seed source of ocular surface reconstruction.
2.Effects and mechanisms of exogenous nesfatin-1 injection in the central nucleus of the amygdale on exploratory behavior in rats

Objective To explore the effects and mechanisms of nesfatin-1 injection in the central nucleus of the amygdale(CeA) on exploratory behavior in rats.Methods Fifty rats were divided into 5 groups randomly : nesfatin-1 low dose group, nesfatin-1 high dose group, SHU9119 group, SHU9119+nesfatin-1 low dose group and control group.Drugs were administrated via CeA to examine behavioral changes of rats by elevated plus maze model test and nesfatin-1 mRNA expression in CeA of anxiety rats (10 rats).Results Compared with the control group,anxiety behaviors of rats in nesfatin-1 low dose group and nesfatin-1 high dose group were improved remarkably,showing that the open arm entries were significantly reduced (OE, t=4.16-12.87, P<0.01), open arm entry proportion was decreased remarkably (OE%, t=2.39-4.39, P<0.01-0.05), and time proportion in open arm was decreased significantly (OT%, t=5.43-20.55, P<0.01),which presented dose dependent.Compared with the control group, anxiety degree of rats in nesfatin-1 low dose group and nesfatin-1 high dose group significantly enhanced, showing that the rats head dipping reduced obviously (HD, t=6.97-16.73, P<0.01) ,while rearing was increased significantly (RE, t=6.60-13.25, P<0.01) ,which was presented dose dependent.Melanocortin receptor antagonist SHU9119 can partly eliminate nesfatin-1 induce-anxiety (t =2.11-3.08, P< 0.01-0.05).Real-time PCR results showed that nesfatin-1 mRNA expression of anxiety model rats was significantly increased (t =3.40-5.58, P<0.05).Conclusion Amygdala nesfatin-1 can increase anxiety and fear of rats,which may be associated with melanocortin systems.
3.The dynamic changes of serum TNF-α,IL-1,IL-10 and HMGB-1 during tuberculosis disease progres-sion

Objective To investigate the dynamic changes and clinical significanes of TNF?α,IL?1,IL?10 and HMGB?1 in patients with tuberculosis. Methods Fifty?nine patients with active tuberculosis were enrolled in this study. The patients received the standard chemotherapy and the curative effect of the patient was evaluat ed according to the outcome classification in our country. The peripheral blood serum from the patients during the treatment and 12 healthy volunteers were collected. The concentrations of serum TNF?α,IL?1,IL?10 and HMGB?1 were detected by ELISA tests. Results Significant decreases of the concentrations of serum TNF?α,IL?1 and HMGB?1 were shown in patients from the initial treatment,to retreatment and extra?pulmonary tuberculosis (P<0.01),and the serum IL?10 was increased significantly (P<0.01) along with the chemotherapy and improvement of the disease. At the beginning of the treatment ,the levels of 4 kinds of inflammatory factors had no significant differences between the completed and uncompleted treatment patients (P>0.05). At the end of the treatment , the levels of serum TNF?α,IL?1 and HMGB?1 of the uncompleted treatment group were significantly higher than the normal levels (P<0.01) ,but the level of IL?10 was significantly lower than the normal level (P<0.01). Conclusion The dynamic testing of the inflammatory cytokines contributes to evaluation and judgement of the curative effect and the condition of patients with the active tuberculosis.
4.Effect of external application of white plaster on cervical radiodermatitis

Objective To explore the effect of externally-applied white plaster on cervical radiodermatitis.Methods Ninety patients with cervical radiodermatitis were equally divided into the observation group and control group according to their admission order.Patients in the control group were treated with furacilin through moist packing as well as routine nursing and the patients in the observation group were given white plaster for external application as well as nursing healthcare education.The two groups were compared in terms of clinical effect,average treatment during,and adverse reactions.Results The total effectiveness rate in the observation group was significantly higher than that of the control group and the average treatment duration was significantly shorter (P<0.05).There were no obvious adverse reactions in both groups.Conchusions The white plaster is clinically effective for the treatment of cervical radiodermatitis.It is advantageous due to shortened curative course and no adverse reactions.
5.Effects of comfort nursing on emotions and comfort of surgical patients

Objective To explore effects of comfort nursing on anxiety, depression and comfort of surgical patients.Methods Comfort nursing was done to 128 surgical patients, including physical comfort, psychological comfort and social comfort and self-designed questionnaire was made to investigate the comfort of patients during operation. When the patients were hospitalized and after operation, patients’physical states were evaluated with self-rating depression scale(SDS)and self-rating anxiety scale(SAS).Results The score of patients’comfort was (6.5±0.2)and the SDS and SAS were lower than that of patients when hospitalized with statistical difference(P<0.05).Conclusion Comfort nursing can reduce anxiety, depression and increase comfort of surgical patients.
6.Clinical efficacy of trimebutine maleate combined with Morita therapy in treatment of patients with irritable bowel syndrome

AIM: To investigate the clinical efficacy of trimebutine maleate combined with Morita therapy in the treatment of patients with irritable bowel syndrome (IBS). METHODS: 108 patients were randomly assigned to two groups. Trimebutine maleate group (n=54) was given trimebutine maleate 200 mg, po, tid, combined with Mortia therapy. The patients were analyzed by self rating anxiety scale (SAS) and self rating depression scale (SDS) before and after the treatment. Control group (n=54) was only given trimebutine maleate 200 mg, po, tid. The course was 4 weeks. RESULTS: The total efficacy rates were 87.04 % and 72.22 % in trimebutine maleate group and in control group, respectively. 7.Spatio-temporal clustering analysis of pulmonary tuberculosis among the elderly in Shaoxing City

Objective:
To investigate the spatio-temporal clustering characteristics of pulmonary tuberculosis (PTB) aged 60 years and above in Shaoxing City from 2019 to 2023, so as to provide insights into prevention and control of PTB among the elderly.
Methods:
Data of PTB cases aged 60 years and above in Shaoxing City from 2019 to 2023 were collected from Tuberculosis Management Information System of Chinese Disease Prevention and Control Information System. The population data were collected from Shaoxing Statistical Yearbook. Vector map information was collected from Shaoxing Geographic Information Public Service Platform. The spatio-temporal clustering characteristics of PTB cases aged 60 years and above were analyzed using global spatial autocorrelation, local spatial autocorrelation and spatio-temporal scanning.
Results:
Totally 3 722 PTB cases aged 60 years and above were registered in Shaoxing City. The average annual registration rate was 61.71/105, showing no significant trend (P>0.05). Totally 2 548 pathogenetically positive cases were registered, with an average annual registration rate of 42.25/105. Spatial autocorrelation analysis showed there was a positive spatial correlation of PTB in 2019 and 2021 (both Moran's I>0, both P<0.05). Shengzhou City showed a high-low clustering, and Keqiao District and Shangyu District showed a low-low clustering. Spatio-temporal scanning analysis showed that a class Ⅰ cluster was located in Shengzhou City, with aggregation time from March 1, 2019 to August 31, 2021. The class Ⅱ clusters were located in Zhuji City, Shangyu City and Keqiao District, with aggregation time from March 1, 2021 to August 31, 2023, from April 1, 2021 to September 30, 2023, and from June 1, 2021 to November 30, 2023, respectively.
Conclusion
The PTB cases aged 60 years and above in Shaoxing City from 2019 to 2023 mainly concentrated in Shengzhou City, where the prevention and control of PTB among the elderly should be strengthened.
8.Practice and exploration of PBL: experience in biochemistry course

According to content of ‘lipid metabolism’,we crafted a effective case for problem based learning (PBL) and practiced it in biochemistry course.In group discussion,students participated in discussion with great enthusiasm and initiative.During the discussion,students got a better understanding about lipid metabolism and clinical application of the knowledge.As teachers,we maily guided students during PBL,and at the same time,we also summed up the experience and explored how PBL be implemented for freshmen and sophomores who lack clinical practice.
9.Clinical application of double incision in phacoemulsification combined with trabeculectomy

Objective Conducting phacoemulsification combined with trabeculectomy through double incision to treat glaucoma and cat-aract,and observing the visual acuity,intraocular pressure before and after the operation and complications of this therapy. Methods Totally 60 patients (78 eyes) with glaucoma and cataract from January 2010 to December 2013 in our hospital were observed. 32 patients (40 eyes) who were given phacoemulsification combined with trabeculectomy through double incision were regarded as the observation group,and the other 28 patients (38 eyes) who were merely given phacoemulsification were regarded as the control group. The visual acuity,intraocular pres-sure and anterior chamber of the two groups were observed and recorded,and they were followed up for 36 months. Results Visual acuity and intraocular pressure of the two groups were improved after operation,but the observation group achieved a better result which showed a significant difference (P< 0. 05). Anterior chamber inflammation occurred in patients of the observation group,and 2 cases were of anterior chamber exudation. The control group,by contrast,has less anterior chamber inflammation. There was no hyphema,shallow anterior chamber, choroidal detachment occurred in both of the two groups. Conclusion Using phacoemulsification combined with trabeculectomy through double incision to treat glaucoma and cataract could receive a good recovery of visual acuity and intraocular pressure. Although,more severe anterior chamber inflammation occurred after operation compared with the control group,it can be alleviated soon after medication. This meth-od is a good therapy which can reduce pain and economic burden of patients from multiple operations. But it requires a strict selection of ap-propriate case.
10.Purification of monoclonal antibody against PGRP and its activity for small cell lung cancer in vitro

Objective To explore the effect of purification on monoclonal antibody (MAb) against PGRP by Protein A-Sepharose affinity chromatography, and to provide some based data for the purification of other antibody using the same method. Methods The ascites which include MAb was purified by Protein A-Sepharose affinity chromatography. The purity and activity of MAb was tested by SDS-PAGE and ELISA. The biological function was identified by flow cytometer and immunohistochemistry. Results The average concentration of protein in ascites before purification is 23.62 mg/ml. Before and after purification, the total protein is 148.79 mg and 146.67 mg, respectively. The recovery coefficient of protein is 98.58%. The concentration of MAb in ascites is 5.21 mg/ml averagely. The MAb purity is more than 95 %. The immunoactivity of purified antibody is higher than that of unpurified antibody. Conclusion The purity of MAb against PGRP purified by Protein A-Sepharose affinity chromatography is very high. The immunoactivity of purified antibody is higher than that of unpurified antibody. So the ProteinA-Sepharose affinity chromatography is a rapid, convenient and reliable method for the purification of MAb Against PGRP.
