1.Study on the relationship between matrix metalloproteinase-9, tissue inhibitors of metalloproteinase-1 and ;missed abortion

Objective To study the differential expression of matrix metalloproteinase-9(MMP-9) and tissue inhibitors of metalloproteinase-1(TIMP-1) in serum and decidua tissue from missed abortion and normal first trimester pregnancy,and to explore their function in early pregnancy .Methods The expression of MMP-9,TIMP-1 in serum was determined using enzyme-linked immunosorbent assay ( ELISA) ,and in decidua by immunohistochemical method . Results The MMP-9 serum level from missed abortion women was higher than that from normal first trimester preg -nancy women [(154.60 ±28.69)ng/mL vs.(135.75 ±18.35)ng/mL,t=3.50,P=0.001].The TIMP-1 serum level from missed abortion women was lower than that from normal first trimester pregnancy women [(253.03 ± 29.90)ng/mL vs.(323.15 ±33.62) ng/mL,t=9.86,P=0.000].The MMP-9 positive expression rate in decidua tissue was higher in missed abortion women than that in normal first trimester pregnancy women (82.5%vs.42.5%,χ2 =13.65,P=0.000).The TIMP-1 positive expression rate in decidua tissue was lower in missed abortion women than that in normal first trimester pregnancy women (87.5%vs.62.5%,χ2 =6.67,P=0.010).Conclusion The expression of MMP-9 and TIMP-1 in serum and decidua may be assosiated with missed abortion .
2.Massive ascites-related intraabdominal hypertension in abdominal malignances

Objective To study the effects of chronic intra-abdominal hypertension (IAH) on the functions of vital organs. Methods From January 2004 to January 2008, 30 chronic massive ascites related IAH patients were enrolled including 12 colonic cancer cases, 7 pancreatic cancer cases, and 11 gastric cancer cases. Control group included 30 cases of gastrointestinal carcinoma without ascites hence no IAH. The specific malignances in this group were colonic cancer in 15 cases, pancreatic cancer in 8 cases, and gastric cancer in 7 cases. Age, gender and cancer stage were matched between the two groups. Symptoms in digestive, respiratoy and circulatory system and liver and kidney functions were recorded and KPS scores were compared respectively. Results The symptoms of abdominal distention in chronic IAH group and control group were presented respectively by 15 cases(50%) and 5 cases (17%) (P < 0.01). Symptoms of heart-throb or hypotension in the two groups by 10 cases (33%) and 3 cases (10%) respectively(P<0.05). Symptoms of asthma or chest distress in the two groups by 9 cases(30% ) and 1 cases (3%) respectively (P< 0.01). Liver function were abnormal in 16 cases (53%) and 6 cases (20%) respectively (P < 0.01 ), kidney functions were abnormal in 14 cases (47%) and 1 case( 3% ) respectively ( P <0.01 ) . There were 22 cases (73 %) and 7 cases (23 %) respectively ( P < 0.01 ) in the two groups with KIS score being lower than 40. Conclusions Chronic IAH causes malfunctions of vital organs deteriorating patients general status and even leading to abdominal compartment syndrome.
3.The dynamic changes of serum TNF-α,IL-1,IL-10 and HMGB-1 during tuberculosis disease progres-sion

Objective To investigate the dynamic changes and clinical significanes of TNF?α,IL?1,IL?10 and HMGB?1 in patients with tuberculosis. Methods Fifty?nine patients with active tuberculosis were enrolled in this study. The patients received the standard chemotherapy and the curative effect of the patient was evaluat ed according to the outcome classification in our country. The peripheral blood serum from the patients during the treatment and 12 healthy volunteers were collected. The concentrations of serum TNF?α,IL?1,IL?10 and HMGB?1 were detected by ELISA tests. Results Significant decreases of the concentrations of serum TNF?α,IL?1 and HMGB?1 were shown in patients from the initial treatment,to retreatment and extra?pulmonary tuberculosis (P<0.01),and the serum IL?10 was increased significantly (P<0.01) along with the chemotherapy and improvement of the disease. At the beginning of the treatment ,the levels of 4 kinds of inflammatory factors had no significant differences between the completed and uncompleted treatment patients (P>0.05). At the end of the treatment , the levels of serum TNF?α,IL?1 and HMGB?1 of the uncompleted treatment group were significantly higher than the normal levels (P<0.01) ,but the level of IL?10 was significantly lower than the normal level (P<0.01). Conclusion The dynamic testing of the inflammatory cytokines contributes to evaluation and judgement of the curative effect and the condition of patients with the active tuberculosis.
4.Survey on Life Quality of Parents of Children with Mental Retardation and Nursing Countermeasures

Objective To explore the effect of specific nursing on quality of life of the parents of children with mental retardation (MR). Methods 160 parents of children with MR and 160 parents of normal children were evaluated with WHO Quality Of Life-BREF (WHO-QOL-BREF). The children with MR were divided into observation group (n=80) and control group (n=80). The control group received the traditional nursing care, and the observation group accepted specific nursing care. The scores of WHOQOLBREF were compared. Results The score of WHOQOL-BREF was lower in the MR group than in the normal group (P<0.01). The score decreased with the aggravation of MR (P<0.001). The score of WHOQOL-BREF was higher in the observation group than in the control group (P<0.01). Conclusion Specific nursing should be given to the parents of children with MR, which can promote their quality of life.
5.Effect analysis on fiberbronchoscope management under life cycle management mode

Objective To ensure the quality of disinfection and sterilization of the fiberbronchoscope in the sterilesupply center through the implementation of whole life cycle management.Methods Using the historical comparative method,the fiberbronchoscopes managed by the departments were enrolled into a control group from February 2013 to March 2014,and the ones undergoing life-cycle management in the sterile supply center were involved in an observation group from April 2014 to May 2015.The two groups were divided according to the management subject and mode,and compared from the aspects of treatment flow,process quality control and record tracing validity.All the data were input with Excel sheet,andanalyzed statistically with SPSS 17.0.Results The observation group and the control group had the numbers of positive results in internal cavities,positive results in external surface,damaged or irrationally-placed package,invalid data tracing and non-standardized storage being(0,1.11±0.15),(0,1.09±0.20),(1.07±0.13,2.75±0.22),(0.57±0.03,1.53±0.31) and (0.13±0.07,0.95±0.21) respectively,and the differences between the two groups were significant (P<0.05).The turnover of the fiberbronchoscope was enhanced significantly in the observation group when compared with that in the control group.Conclusion Life cycle management contributes to strengthening quality control of the treatment of polluted fiberbronchoscope,and decreases the incidences for hospital infection.

Objective: To evaluate the effects of the modified fixed r everse Twin-block appliance(TBA) combined with maxillary protraction appliance( MPA) in the treatment of early Angle Ⅲ skeletal anterior crossbite. Met hods:TBA combined with MPA was used in 15 growing subjects with early An gle Ⅲskeletal malocclusion (group TBA-MPA). Another 15 subjects with the same type of malocclusion were treated by MPA(group MPA). The effects of treatment we re studied by cephalometric measurments.Results:3-5 months afte r treatment SNB, B-Vert T, Po-Vert T,NPo-FH,Co-Po and 1-NB in group 1 were decreased more than those in group 2 (P0.05).Conclusion:Angle Ⅲ skeletal ante rior crossbite can be corrected successfully with reverse TBA combined with maxi llary protraction appliance by mandible retrusion and maxilla forward growth.
8.Purification of monoclonal antibody against PGRP and its activity for small cell lung cancer in vitro

Objective To explore the effect of purification on monoclonal antibody (MAb) against PGRP by Protein A-Sepharose affinity chromatography, and to provide some based data for the purification of other antibody using the same method. Methods The ascites which include MAb was purified by Protein A-Sepharose affinity chromatography. The purity and activity of MAb was tested by SDS-PAGE and ELISA. The biological function was identified by flow cytometer and immunohistochemistry. Results The average concentration of protein in ascites before purification is 23.62 mg/ml. Before and after purification, the total protein is 148.79 mg and 146.67 mg, respectively. The recovery coefficient of protein is 98.58%. The concentration of MAb in ascites is 5.21 mg/ml averagely. The MAb purity is more than 95 %. The immunoactivity of purified antibody is higher than that of unpurified antibody. Conclusion The purity of MAb against PGRP purified by Protein A-Sepharose affinity chromatography is very high. The immunoactivity of purified antibody is higher than that of unpurified antibody. So the ProteinA-Sepharose affinity chromatography is a rapid, convenient and reliable method for the purification of MAb Against PGRP.
9.Effects of fosinopril and losartan on renal Klotho expression and oxidative stress in spontaneously hypertensive rats

Objective To explore effects of fosinopril and losartan on renal Klotho expression and oxidative stress in spontaneously hypertensive rats (SHR) and the mechanisms underlying the protection against renal damage. Methods Fifteen male SHRs (22 weeks old) were randomly divided into 3 groups (n=5 in each group): a SHR group, a fosinopril group [10 mg/(kg?d)], and a losartan group [50 mg/(kg?d)]. Age-matched Wistar-Kyoto (WKY) rats were chosen for a control group. Eight weeks later, tail arterial pressure, 24 hours urinary protein (Upro),urinary N-acetyl-β-D-glucosaminidase (NAGase) were measured. Renal pathological changes were examined under light microscopy by HE staining. The renal mRNA and protein expression of Klotho were determined by RT-PCR, immunohistochemical staining or Western blot. The levels of total antioxidant capacity (TAOC), malondialdehyde (MDA), Cu/Zn superoxide dismutase (Cu/Zn-SOD), Mn superoxide dismutase (Mn-SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) were determined.Results The typical pathological characteristics of hypertensive renal damage were observed in the kidney of the SHR group.Compared with the SHR group, the systolic pressure, Upro, and urinary NAGase, the content of MDA and renal pathological damage was reduced while the renal Klotho expression and activities of TAOC, Cu/Zn-SOD, CAT, and GSH-Px were increased (P<0.05 or P<0.01) in the fosinopril or losartan group. There was no significant difference in renal Mn-SOD level among the 4 groups (P>0.05). Conclusion Fosinopril and losartan can exert protection against hypertensive renal damage through upregulating Klotho expression as well as reducing oxidative stress.
10.The optical microscopic observation on the effects of rats of hyperthermia combined with trauma through topical oxygen therapy

Objective Through the optical microscopic observation to investigate the pathological influence of topical oxygen therapy on wounds of rats' muscles in hot and humid environment. Method The rat traumatic models were established. Twenty-four rats were randomly divided into four groups, including normal envi-ronment comparison group (NEC group, n=6), normal environment oxygen therapy group (NEO group, n=6). hot and humid environment comparison group (HHE group, n=6), hot and humid environment oxygen therapy group (HHO group, n=6).Then all rats were sampled to observe the changes of rats' muscle under optical microscope. Results Pathologic histology of muscle cells were obviously changed in HHE group. In these rats. disorganized myofibrillae with some loss of myofilaments. The damage of the muscle fibers in HHO group were better than that in HHE group. The damage of muscle ceils in NEC group were slighter than that in HHE group, the muscle of rats in NEO group are all normal. Conclusion In hot and humid en-vironment pathologic histology changes of wounds were serious, topical oxygen therapy could relieve muscle tissue ultrastructures change.
