1.Helicobacter pylori infection and idiopathic thrombocytopenic purpura in children

Idiopathic thrombocytopenic purpura(ITP) is characterized by platelet destruction which is mediated by auto-antibodies. Recent researches show that helicobacter pylori infection is correlated with part of FTP in adult. Eradication of helicobacter pylori infection has brought dramatically platelet elevation. It was reported that cross reaction played a very important part in the helicobacter related FTP, but the exact mechanism is still unknown, whether the FTP in children needs helicobacter pylori eradication is under debate.

3.Effects of acidic reperfusion on ischemia-reperfusion injury in isolated rat teart: role of PDK/Akt signaling pathway

Objective To investigate the effects of acidic reperfusion on ischemia-reperfusion injury in isolated rat heart and the role of PI3K/Akt signaling pathway.Methods Seventy adult SD rats weishing 450-550 g were randomly divided into 7 groups(n=10 each):group Ⅰ ischemia-repedusion injury(X/R);groupⅡ ischemic post eonfioning(IPC);group Ⅲ acidic reperfusion(H+);group Ⅳ ischemie postconditioning+alkalotic nsperfusion(IPC+OH-);group Ⅴ alkalotic reperfusion(OH-);group Ⅵ acidic repeffusion+wortmannin (H+wort)and group ⅦVl wortmannin(wort).The animals were anesthetized with intraperitoneal(IP)urethane. Their chests were opened and hearts were excised and passively perfused in a Lsngendorff apparatus with KrebsHenseleit buffer(KHB)saturated with 95%O2-5%O2 at 37℃.The isolated hearts were made globally ischemic for 30 min followed by 120 rain reperfusion.Group II (IPC)WaS subjected to 6 cycles of 15 rain reperfusion and 15 min ischemia at the beginning of reperfusion. In group H+ and OH- (group Ⅲ ,Ⅴ ) the isolated hearts were perfused with KHB of pH 6.9 and 7.8 for 3 rain respectively at the beginning of reperfusion. In group IPC + OH( Ⅳ ) the isolated hearts were perfused with KHB of pH 7.8 for 3 rain followed by IPC. In group H + wort and wort ( Ⅵ,Ⅶ ) the isolated hearts were perfused with wortmannin 100 nmol/L in KHB of pH 6.9 and 7.4 for 3 min respectively at the beginning of reperfusion. LVEDP and ± dp/dtmax were measured before ischemia and at 30 min of reperfusion. Coronary effluent was collected for determination of NO concentration at 30 min of reperfusion. Lnfart size was measured at 120 rain of reperfusion. Results The LVEDP was decreased, ±dp/dt was increased, the infarct size was smaller and the NO concentration in coronary effluent was higher in group IPO and H+ compared with group I/R. Wortmannin abolished the protective effect of acidic reperfusion on isolated rat heart. Conclusion PI3K/Akt signaling pathway may be involved in the protective effects of acidic reperfusion on myocardium against ischemia-reperfusion injury.
4.Experiences Treatment of Lung System Diseases Caused by External Pathogens with Traditional Chinese Medicine

In broad definition,diseases caused by exogenous pathogenic factors involve with respiratory system,digestive system,urinary system,and nervous system;While concerned with internal medicine of respiration,they are common cold,upper respiratory tract infection,acute tracheitis and bronchitis.Clinically,we call diseases that caused by external pathogens,showing symptoms of respiratory system in general,as FeiXi Waigan Bing(lung system diseases caused by external pathogens).In curing these diseases,we integrated traditional Chinese and western medicine and got satisfactory clinical effect.This demonstrated the advantages and features of Traditional Chinese medicine in curing lung system diseases caused by external pathogens.
5.Comparative study of cardiac muscle structure between two models of chronic cor pulmonale induced by hypoxia and active immunization of M2-muscarinic receptor

Objective To compare the similarities and differences in changes of cardiac muscle structure between the rat models of chronic cor pulmonale induced by hypoxia and active immunization of M2-muscarinic receptor under light microscope and electron microscope, and to explore the dependablity of the antibody effects on cardiac architecture. Methods The 48 healthy male Wistar rats were randomly divided into 4 groups: (1) Hypoxia group: the typical model of chronic cor pulmonale was established according to XUE's method; (2) Active immunization group was immunized by M2-muscarinic receptor peptide; (3) Control group was fed in normal condition; (4) Cyclosporin A group: the hypoxia group plus cyclosporin A treatment at the same time. The antibody against M2-muscarinic receptor was detected by SA-ELISA, and the pathological exemplar of cardiac muscle was observed by light microscope and transmission electron microscope. Results (1) Antibody level of M2-muscarinic receptor; the P/N values gradually increased along with the process of experiment, and the max value in active immunization group was 5. 13. At the end of the second week, the value in hypoxia group increased to 2.08, but was still less than in active immunization group (4.66). (2) Under light microscope: in hypoxia group and active immunization group, the hearts displayed significant alterations including disorder of cardiac muscle fiber, necrosis of myocardial cells together with the infiltration of inflammatory cells. There were no differences in light microscopic findings between hypoxia and cyclosporin A group. (3) Under transmission electronic microscope examination in both active immunization group and hypoxia group, the hearts showed s1imilar significant alterations such as focal cardiac muscle fiberlysis, loss of normal muscle fiber banding pattern, mitochondrial swelling and condensation, sarcoplasmic vacuolation and deposition of dense granules in both the sarcoplasmic and muscle fiber. The contour of myocyte was irregular and plasma membranes were discontinuous in some cells. All altered myocytes were fairly widely distributed throughout the myocardium. The interstitium showed edema, deposits of flocculent serum protein, activated fibroblasts and increased amounts of collagen fibers. No obvious alterations were observed in cyclosporin A group. Conclusions The positive rate and the titer of antibody against M2-muscarinic receptor are obviously increased in the rat model of chronic cor pulmonale, which indicates that there is a relationship between the antibody against M2-muscarinic receptor and the pathogenesis of chronic cor pulmonale.
6.Effect of silencing of ATM expression by siRNA on radiosensitivity of human lung adenocarcinoma A549 cells

Objective To investigate the effect of silencing of ataxia-telangiectasia mutated (ATM) expression by plasmid-mediated RNA interference on the radiosensitivity of human lung adenocarcinoma A549 cells.Methods Eukaryotic expression plasmid containing ATM small interfering RNA (siRNA) (pSilencer2.1-ATM),as well as pSilencer2.1-nonspecific,was constructed.Lung adenocarcinoma A549 cells were divided into positive group,negative group,and control group to be transfected with pSilencer2.1-ATM,pSilencer2.1-nonspecific,and no plasmid,respectively.The mRNA and protein expression of ATM was measured by RT-PCR and Western blot,respectively.The change in cell radiosensitivity was observed by colony-forming assay.Cell cycle and cell apoptosis were analyzed by flow cytometry.Results The eukaryotic expression plasmid containing ATM siRNA was successfully constructed.The RT-PCR and Western blot demonstrated that the expression of ATM was down-regulated in the positive group.The sensitization enhancement ratios (D0 ratios) for the positive group and negative group were 1.50 and 1.01,respectively.The flow cytometry revealed that the proportions of A549 cells in G1 and G2/M phases were significantly lower in the positive group than in the control group (51.27% vs 61.85%,P =0.012;6.34% vs 10.91%,P =0.008) and that the apoptosis rate was significantly higher in the positive group than in the control group and negative group (49.31% vs 13.58%,P=0.000;49.31% vs 13.17%,P=0.000).Conclusions Silencing of ATM expression may increase the radiosensitivity of human lung adenocarcinoma A549 cells,probably by affecting the cell cycle and promoting cell apoptosis.
7.Efficacy of three-dimensional conformal radiotheraPy combined with concurrent S-1 for locally advanced esoPhagus carcinoma in the elderly

Objective To investiGate the efficacy of three-dimensional conformal radiotherapy( 3-DCRT)combined with concurrent S-l for locally advanced esophaGus carcinoma in the elderly. Methods Fifty-four elderly patients with locally advanced esophaGus carcinoma were randomly divided into test Group(27 cases treated with 3-DCRT combined with S-l)and control Group(27 cases treated with 3-DCRT alone). DiaGnosis in all cases was confirmed by patholoGy. Results The complete remission rate and overall efficiency rate in test Group were 62. 96% and 85. l9% respectively,and 33. 33% and 59. 26% in control Group(χ2 =4. 75,P=0. 029;χ2 =4. 52,P=0. 033). All 23 patients with dysphaGia in test Group were in remission,and l6 of 22 patients with dysphaGia in control Group were in remission,and there was statistical siGnificant difference between two Groups(χ2 =7. 24,P=0. 007). The one-and two-year survival rates in test Group were 74. l% and 54. 8%respectively,and 55. 6% and 36. l% in control Group(χ2 =3. 9l,P=0. 047;χ2 =3. 98,P=0. 046). Toxicities of Grade 0-Ⅱ in both Groups were similar,and there was no statistical siGnificant difference(χ2 =l. 08,P=0. 299). Conclusion The complete remission rate,overall efficiency rate and one-and two-year survival rates of 3-DCRT combined with concurrent S-l were siGnificant hiGher than 3-DCRT alone in the treatment for the elderly patients with locally advanced esophaGus carcinoma. And ti’s adverse effects were well tolerated.

9.Correlation between serological indexes with disease severity in patients withcerebral infarction and analysis of prognostic influencing factors

Objective To investigate the relationship between serum levels of D-dimer(DD),hs-CRP and IL-6 with the infarction severity and prognosis in the patients with cerebral infarction(CI),and to analyze the influencing factors of prognosis for screening the independent risk factors affecting prognosis.Methods Eighty-one patients with CI in our hospital from January 2014 to June 2015 were selected as research subjects.All patients were treated with the CI routine treatment after admission.On admission,the CI severity was assessed by NIHSS,the serum levels of DD,hs-CRP and IL-6 were detected.After discharge,following up lasted for 12 months,the prognosis was assessed by MRS scale.The correlation between DD,hs-CRP and IL-6 with the infarction severity and prognosis was analyzed.The prognostic influencing factors were analyzed by adopting the univariate analysis and multivariate Logistic regression analysis.Results On admission,the levels of DD,hs-CRP and IL-6 in the severe group were significantly higher than those in the moderate group(q=5.736,7.772,4.757,P<0.05) and the mild group(q=11.094,16.920,12.255,P<0.05),while the moderate group was significantly higher than the mild group(q=5.783,9.870,8.049,P<0.05).Pearson′s analysis showed that the levels of DD,hs-CRP and IL-6 were positively correlated with the CI severity(r=0.603,0.457,0.394,P<0.05).After following up for 12 months,the levels of DD,hs-CRP and IL-6 in the good prognosis group were significantly lower than those in the poor prognosis group(t=4.775,3.101,2.910,P<0.05).Pearson′s analysis showed that DD,hs-CRP and IL-6 levels were positively correlated with poor prognosis(r=0.584,0.513,0.432,P<0.05).Multiple Logistic regression analysis showed that infarct volume,NIHSS score,DD,hs-CRP and IL-6 were the independent risk factors for poor prognosis in the patients with CI(P<0.05).Conclusion The infarct volume,NIHSS score,DD,hs-CRP and IL-6 are the independent risk factors for poor prognosis in the patients with CI,which provide a certain reference for judging the prognosis in CI patients.
10.Effect of ERG6 Gene Extra Copies on Aspergillus fumigatus

To investigate the role of ERC,6 gene in the growth rate and antifungal susceptibility,Aspergillus fumigatus strain with extra copies of ERG6 gene was constructed.Open reading frame (ORF) of putative ERG6 gene was searched in A.fumigatus genome.A PCR fragment, ERG6 ORF sandwiched by its flanking sequences (about 1 kb respectively),was amplified and was then subcloned into vector pRG-AMA1- NotI to produce a recombinant plasmid pERG6,which was further transformed into uracil auxotroph A.fumigatus strain AF293.1 to produce the transformant AF-pERG6.In the same time,empty plasmid pRG-AMA1-NotI was also transformed into A.fumigatus strain AF293.1 to produce the transformant AF-empty as a control.Radial growth of the transformants was tested on minimal medium (MM) and YAG medi- um.Antifungal susceptibilities of these resahing transformants,AF-pERG6 and AF-empty,to the common antifungal agents were performed by using both disk diffusion and broth microdillution methods.A.fumigatus genome contains a ERG6 gene,of which the ORF size is 1 256 bp.Comparing to Candida albicans and Sacchromyces cerivisiae sterol methyltransferase,A.fumigatus Erg6p had 57% and 50% identity, and had 70% and 63% similarity in amino acid sequences,respectively.Radial growth of transformant AF-pERG6 was slower than that of transformant AF-empty.The antifungal susceptibilties of transformant AF-pERG6 to the antifungal drugs itraconazole,voriconazole,terbin- aline,amphotericin B,caspofungin and grisefolvin were same to that of transformant AF-empty.In A.fumigatus,extra copies of ERG6 gene have no effect on antifungal susceptibilities to itraconazole,voriconazole,terbinafine,amphotericin B,caspofungin and grisefolvin.Radial growth of A.fumigatus harboring extra copies of ERG6 gene becomes slower compared to the control.
