1.Dopamine content in the stratum of Parkinson’s disease rats after transplantation of tyrosine hydroxylase-modified human umbilical cord blood mesenchymal stem cells

BACKGROUND:Stem cel therapy is superior to drug therapy for recovery of patient’s physiological mode, and cel transplantation therapy is becoming a trend.
OBJECTIVE:To observe the changes in dopamine content in the stratum of Parkinson’s disease rats after transplantation of tyrosine hydroxylase-modified human umbilical cord blood mesenchymal stem cel s.
METHODS:After identification by enzyme digestion, pEGFP-C2-TH plasmid was transfected into the fourth generation of human umbilical cord blood mesenchymal stem cel s by electroporation method, and then transfected cel s were injected into the right cerebral ventricle of Parkinson’s disease rats (experimental group). PBS injection was performed in the control group. Migration of dopamine in the brain tissue of rats was observed, and the content of dopamine was detected by high performance liquid chromatography.
RESULTS AND CONCLUSION:At 8 weeks after cel transplantation, the cel s gradual y migrated to the ventricles;after 12 weeks, the cel s migrated to the cortex, and expressed tyrosine hydroxylase antigen. Meanwhile, the content of dopamine was significantly higher in the experimental group than the control group (P<0.05). These results reveal that the intraventricular transplantation of tyrosine hydroxylase-modified human umbilical cord blood mesenchymal stem cel s has obvious therapeutic effect on Parkinson’s disease rats.
2.Thrombospondin 2 and tumors

Thrombospondin (THBS) 2, as a member of the THBS family, is expressed in a variety of tumor cells and has important significance in the development and metastasis of tumors. In recent years, studies have reported that the specific role of THBS2 in different tumors is different, and it participates in a variety of biological processes, such as cell apoptosis, wound healing, angiogenesis and inflammation. At present, the role of THBS2 in tumorigenesis, development, and prognosis is not completely clear. Exploring the abnormal expression and mechanism of THBS2 in tumors is expected to provide a new method for tumor diagnosis and treatment.

Objective To investigate the antitumor effect of specific cytotoxic T lymphocytes induced by renal cell carcinoma multiple antigen peptide dendritic cell (DC) vaccine in vitro. Methods Dendritic cells were induced by hGM-CSF,hIL-4 from blood.Peptide of renal cell carcinoma cell line (RCC786-0)was got by citrate-phosphate buffer elution.Peripheral blood mononuclear cell was cultured.Multiple antigen peptide DC cell vaccine was obtained by acid-eluted peptide pulsed DC.The tumor antigen specific CTL was generated from activated T cell by vaccine.Killing activity of the tumor antigen specific CTL was activated by vaccine. Results Antigen peptide DC cell vaccine could show a strong cytotoxic activity of CTLs(31.93±5.05%),which was much higher than control groups(5.88±2.26%,8.03±6.70%,9.70±2.09%,9.35±3.58%). Conclusion Renal cell carcinoma antigen peptide DC vaccine could show a high antitumor effect in vitro.
4.The evolvement of the action and the application of Astragalus and its development

Based on that study, its beneficial to reasonably apply Astragalus in the clinical application, further to deeply find and succeed the potential and the loosed action and the applied experience of Astragalus. That study provides the theory foundation for further expanding the applied scope of Astragalus, also presents a kind of train of thought for the research and develop of new Chinese herbs.
5.Experimental study of intermittent ischemia affects surviving time limit of skeletal muscle in dog

Objective To study proloning the surviving time limit of servered skeletal muscle through intermittent ischemia Methods We use canine gracilis as experimental object to observe its changes by"4C" (colour,capillary bleeding,contractility and consistency of muscle),succinic,dehydrogenase and electron microscope Result (1) At normal temperature the ischemic time limit of skeletal muscle was 7 hours (2) At low temperature (4℃),the surviving time limit of skeletal muscle was 15 hours.(3) However, intermittent ischemia, the surviving time limit of skeletal muscle was 25 hours. Conclusion Intermittent ischemia can prolong the surviving time limit of skeletal muscle obviously So, intermittent ischemia may be used as a new better method of servered limb perservation
6.Detection of pathogenic bacteria and drug resistance analysis on children with pulmonary infection in ICU and respiratory ward

Objective To investigate the distribution and drug resistance of bacteria infection in children with pneumonia between pediatric intensive care ward and respiratory ward,in order to guide clinical treatment.Methods From 2010 January to 2014 January,two-hundred and sixty cases of pneumonia in hospitalized pediatric intensive care ward were included in the severe group,300 patients from respiratory ward into the common group,severe group were divided into the mechanical ventilation group (126 cases),non mechanical ventilation (134 cases),mechanical ventilation were divided into invasive ventilation group (94 cases),noninvasive ventilation group (32 cases).Conventional sputum specimens were detected,bacterial culture,identification and drug resistance of pathogenic bacteria ratio were done in each case,to compare the pathogenic bacteria and drug resistance rate of different groups.Results Both severe group and common group,gram negative bacteria were significantly higher than those of gram positive,But in the severe group,gram negative bacteria ratio was significantly higher than the common group (P < 0.05),while the positive bacteria ratio was significantly lower than that of common group (P < 0.05).Stenotrophomonas maltophilia,Acinetobacter and Pseudomonas aeruginosa ratio in severe group were significantly higher than that of common group (P < 0.05),while the Haemophilus influenzae and Streptococcus pneumoniae in common group was significantly higher than the proportion of severe group (P < 0.05).Stenotrophomonas maltophilia,Acinetobacter,Pseudomonas aeruginosa ratio in ventilation group was significantly higher than that of non ventilation group (P < 0.05),and in the invasive ventilation group was significantly higher than the proportion of noninvasive ventilation group (P < 0.05).Multi drug resistant bacteria detection rate in severe group was significantly higher than the common group (P < 0.05).Conclusion Both pediatric intensive care ward and respiratory ward,bacterial lung infections are dominated by Gram negative bacteria,but bacteria spectrum and drug resistance have obvious difference,multi drug resistant bacteria detection rate in pediatric intensive care ward significantly higher than that of respiratory ward.Mechanical ventilation especially invasive mechanical ventilation may easily lead to drug-resistant strains.

Recent studies show that the abnormalities of dickkopf-1 (DKK-1)are closely related to the bone metastasis and therapeutic effect of breast cancer,prostatic carcinoma,multiple myeloma,neuroblastoma and non-small cell lung cancer.Clinical researches of large sample on the relationships between serum DKK-1 concentration and malignant tumors bone metastases have important clinical significance for evaluating the value of DKK-1 as a clinical biological biomarker of bone metastases and understanding the tumor mechanisms of bone metastases.
8.A classification for hepatic venous outflow obstruction after piggyback liver transplantation and its clinical significance

Objective A classification for hepatic venous outflow obstruction after piggyback liver transplantation (PBLT) and its clinical significance.Methods We conducted a retrospective study on 248 patients who underwent liver transplantation from May 2000 to August 2006.The aims were to elucidate the causes and treatment of postoperative venous outflow obstruction.Results Venous outflow obstruction occurred in 38 patients after transplantation.Among those,2 (5.26%) had superior hepatic inferior vena cava (IVC) stenosis,13 (34,21%) had the hepatic vein anastomosis twisted at an angle,7 (18.42%) had IVC stenosis at the posthepatic segment,and 16 (42.10%) had outflow obstruction at the hepatic veins.In these 38 patients,34 underwent PBLT,2 underwent APBLT,and 2 COLT.Most patients with hepatic venous outflow obstruction improved with surgical treatment and interventional therapy.Conclusions Hepatic vein outflow obstruction was associated with the technique of hepatic vein anastomosis,the type of cavocaval anastomosis and graft size mismatch between the donor and the recipient.Performing piggyback liver transplantation according to the classification of hepatic vein and appropriate treatments could improve the prognosis of venous outflow obstruction in clinical practice.
9.Mediating role of activities of daily living and self-rated health in chronic disease-caused depressive symptoms among the elderly

Objective:
To investigate the mediating role of activities of daily living (ADL) and self-rated health in chronic disease-caused depressive symptoms among the elderly, so as to provide insights into depression control among the elderly.
Methods:
Basic features, depressive symptoms, number of chronic diseases, ADL and self-rated health of the elderly at ages of 60 years and older were collected from the China Health and Retirement Longitudinal Study 2018. Multiple linear regression model was created using the Process program to examine the mediating role of ADL and self-rated health in number of chronic disease-caused depressive symptoms, and the significance of the mediating role was tested using the Bootstrap test.
Results:
A total of 5 892 elderly participants were enrolled, with a mean age of (68.42±6.23) years and including 2 744 men (46.57%). The participants had a median depressive symptom score of 8 (interquartile range, 10) points, a median number of chronic diseases of 1 (interquartile range, 1), a median ADL score of 12 (interquartile range, 3) points, and a median self-rated health score of 3 (interquartile range, 1) points. Mediation analysis showed that number of chronic diseases affected depressive symptoms via the independent mediating role of ADL (β=0.163, 95%CI: 0.120-0.206) and self-rated health (β=0.303, 95%CI: 0.259-0.351), and affected depressive symptoms via the chain mediating role of ADL and self-rated health (β=0.057, 95%CI: 0.043-0.074) among the elderly, and the gross mediating role consisted of 58.44% of total effects.
Conclusions
The number of chronic diseases may incease the risk of depressive symptoms through aggravating the impairment of ADL and decreasing self-rated health levels among the elderly.
10.Establishment of fingerprint profiles of polysaccharides extracted from Guifu Dihuang Wan

Objective To find a new method for quality control of polysaccharides by establishing the fingerprint profiles of polysaccharides extracted from Guifu Dihuang Wan. Methods The polysaccharides extracted from Guifu Dihuang Wan were hydro-lyzed partially under appropriate hydrolysis condition with acid,and the fingerprint profiles of the hydrolyzates was obtained by HPLC. Microsoft Excel 2002 was applied for calculating the similarity of fingerprint profiles,and informatton analysis of these profiles was performed by a software for evaluating fingerprint profiles of traditional Chinese medicine. Results Fingerprint profiles of the hydroly-zates of polysaccharides from 10 of Guifu Dihuang Wan products,produced by different manufacturers,were obtained. It was observed that the similarity of eight products was greater than 0.99,while the similarity of the other two was 0.92 and 0.94,respectively. Con-clusion This method is technically feasible for the quality control of polysaccharides from Guifu Dihuang Wan.
