Objective:To detect the expression of focal adhesion kinase(FAK) and vascular endothelial growth factor(VEGF)in human laryngeal squamous cell carcinoma to study their correlation in invasion and metastasis.Methods:Immunohistochemical S-P method was used to detect the expression of FAK and VEGF in 42 case of human laryngeal squamous cell carcinoma and 40 cases of polyp of vocal cords.All the samples were stained by HE method to confirm their pathologic characteristics.Positive rates of FAK and VEGF were evaluated by microscopy.Results:The positive rates of FAK,VEGF were 79% and 71%,respectively in human laryngeal squamous cell carcinoma.While in polyp of vocal cords,13% and 18%,respectively.The difference was significant(P
2.Establishment of librarian performance appraisal index system in academic library

Objective To establish the scientific, rational and standard librarian performance appraisal index system in academic library in order to adjust their working behaviors, improve their service quality and increase the overall performance of library.Methods The current librarian performance appraisal situation was investigated according to the literature and on the field inspection.The appraisal indexes and their weight were established by Delphi and step analysis.Results The 3-scale appraisal index system was established, which include 4 No.1 indexes, 10 No. 2 indexes, and 59 No.3 indexes with their weight and score displayed.Conclusion The performance appraisal index system we established can effectively arouse the enthusiasm of librarians in scientific research innovation, manage-ment or social work.
3.Role of podocyte autophagy in passive Heymann nephritis

Objective To investigate the role of autophagy in podocyte damage,and the intracellular mechanism of autophagy activation through passive Heymann nephritis (PHN) animal model.Methods Male Sprague-Dawley rats (n=40) were studied on day 0,2,4,7,and 21 after induction of PHN by injection of anti-Fx1A.Podocyte morphology and autophagosomes were observed by transmission electron microscopy.Podocyte numerical density was estimated by Weibel-Gomez =method.Cell apoptosis was detected by TUNEL assay and caspase-3 immunohistochemical staining.Expressions of autophagy markers and endoplasmic reticulum stress (ERS)-associated proteins were analyzed by Western blotting.Results (1) In PHN rats,immunohistochemical staining showed that C5b-9 deposited along glomerular basement membrane on day 4 to day 21.Small subepithelial electron -dense deposits and a part of foot process fusion were detected in the glomerulus of PHN rats on day 4 by transmission electron microscope,and podocyte damage was aggravated on day 21.Furthermore,compared with control,the urinary protein levels of PHN rats began to increase on day 3,and reached the top on day 21 [(50.6±6.0) mg/24 h].(2) The number of podocytes significantly decreased in PHN rats compared with control group on day 21(P < 0.05).(3) In PHN rats,apoptotic podocytes were found by caspase-3 immunohistochemical staining and TUNEL assay on day 21.(4) The expression of autophagy marker LC3 Ⅱ was markedly increased on day 7 and 21,but down-regulated on day 21 compared with day 7.Moreover,accumulated autophagosomes in podocytes were detected on day 7 and 21 by transmission electron microscope.(5) The level of GRP78 was significantly increased on day 2 and 7 but reduced to baseline on day 21.At the same time,the downstream pathways (ATF6α,p-PERK and p-JNK) of unfolded protein response were also up-regulated in the early process of PHN and down-regulated later.Conclusions Autophagy is an important way to protect against immunemediated podocyte injury in membranous nephropathy.Autophagy activation is mainly related to endoplasmic reticulum stress induced by complement attack.This provides an important basis for a thorough understanding of the role of autophagy in the process of podocyte damage and the pathogenesis of membranous nephropathy.
4.Effects of amphotericin B and voriconazole on ultrastructure of a clinical isolate of Penicillium marneffei

Objective To observe ultrastructural changes in a clinical isolate of Penicillium marneffei(PM) before and after treatment with amphotericin B or voriconazole by using scanning electron microscopy (SEM)and transmission electron microscopy (TEM). Methods A microdilution method was performed to determine the minimum inhibitory concentration (MIC)of amphotericin B and voriconazole against a clinical isolate of PM. Then, the PM isolate was treated with amphotericin B or voriconazole at their MICs and 10-fold MICs for 24, 48 and 72 hours. The ultrastructural changes in this isolate before and after the treatment were observed by using SEM and TEM. Results After the treatment with amphotericin B, SEM showed that the conidia or yeast cells of the PM isolate were gradually damaged, and their outer layers experienced detachment, shrinkage, breakage and adhesion with the increase in treatment duration and concentrations of amphotericin B; TEM also showed degenerated mitochondria, broken nuclei and cell walls, and shrunken cytoplasmic membrane with disappearance of cytoplasmic organelles. Similarly, the damage, shrinkage, shriveling and collapse of PM cells were seen by using SEM, and TEM showed many high-density electron-dense granules in cytoplasm, degeneration of mitochondria, roughening of cell wall surface, damage and shrinkage of cytoplasmic membrane, and disappearance of cytoplasmic organelles after voriconazole treatment. Conclusions Amphotericin B and voriconazole both had a strong antifungal effect on PM, and could induce evident ultrastructural changes, which were positively associated with treatment duration and concentrations. Moreover, amphotericin B caused more severe damage to PM compared with voriconazole.
5.An evaluation of the effectiveness of prevention and control measures on coal-burning-borne endemic fluorosis in Xishui County of Guizhou Province

Objective To evaluate the effectiveness of comprehensive prevention and control measures on coal-burning-borne endemic tluorosis in Xishui,on residents healthy behavior and situation of the disease.Methods In 2013,five towns were selected according to their location of east,south,west,north and centre.Three villages and one elementary school in each town were chosen,and 20 families' related behavior and fluorine prevention knowledge questionnaire in each village were surveyed.All the students from one class aged 8-12 of grade 3-6 in each school were selected,according to the Diagnosis of Dental Fluorosis,dental fluorosis was examined and fluorine knowledge questionnaire was surveyed.At the same time,the detection rate of dental fluorosis of children aged 8-12,information of the knowledge about prevention and cure of fluorosis and related behavior of target groups in 2007,and the urinary fluoride contents of children aged 8-12 in 2000 were collected.Analysis and comparison of the data in 2000,2007 and 2013 were done.Results After the intervention,the witting rates of students and family householders 90.95% (11 773/12 945),84.27% (3 792/4 500),were significantly higher than those of before the intervention [19.29% (1 039/5 385),26.27% (394/1 500),x2 =9 279.14,1 794.22,all P < 0.01].Proper utilization rate of cooking stove was 100.00% (37/37),iron stove utilization rate was 99.64% (279/280),dry chili correct rate was 100.00% (299/299) and before the intervention,these values were 3.20% (43/1 345),90.28% (1 207/1 337)and 62.14% (855/1 376),respectively,and the differences were statistically significant (x2 =601.12,27.28,27.28,all P < 0.01).Children's dental fluorosis detection rate was 24.21% (206/851),compared with that of before the intervention,61.62% (2 150/3 489),the difference was statistically significant (x2 =385.93,P < 0.01).Children's urine fluoride was (0.51 ± 0.29) mg/L,and before the intervention [(1.55± 1.59) mg/L],the difference was statistically significant (t'=10.25,P < 0.01).Conclusions The effect of comprehensive prevention and control measures against coal-burning-borne endemic fluorosis in Xishui are remarkable.The residents health related behavior has been formed and the condition is reduced markedly.
6.Clinicopathological analysis on glomerulonephritis complicating with acute interstitial nephritis

Objective To identify the clinical characteristics and pathological changes of patients suffered from glomerulonephritis complicating with acute interstitial nephritis (AIN) . Methods Twenty one patients of glomerulonephritis complicating with AIN diagnosed by renal biopsy were retrospectively analyzed . Thirty-five pure AIN patients were selected as controls .Results Glomerulonephritis complicating AIN accounted for 37 .5% of all the AIN cases . Βlactam antibiotics and Chinese herbs were the major causes of AIN . 76 .2% of cases received further examinations due to the elevation of serum creatinine (Scr) during their follow-up of kidney injuries or during routine exams for all kinds of discomforts . Pathological features of AIN were also detected besides glomerular leisions . The impairments of renal interstitia were severe than those of the glomeruli . Eosinophil in the renal interstitia was an important indicator for the diagnosis of AIN .The renal function returned to normal or baseline in 64 .7% of the patients of glomerulonephritis complicating with AIN whose follow-up data were available . The median period for renal function restoration was 150 days (compared with 60 days in pure AIN) . But there were no significant differences between these two groups as for the rate of irreversible renal insufficiency during a follow-up period of 2 years . Conclusions Symptoms of AIN in patients of glomerulonephritis complicating with AIN tend to be masked by their glomerular diseases . Renal biopsy is of most importance for the diagnosis . Early diagnosis and treatment leads to satisfactory prognosis .

The association of non-HDL-cholesterol and non-HDL-C-to-HDL-cholesterol ratio (non-HDL-C-to-HDL-C ratio) with early diabetic nephropathy in patients with type 2 diabetes mellitus was investigated.Non-HDL-C and non-HDL-C-to-HDL-C ratio were positively related with microalbuminuria (P<0.05 or P<0.01).Non-HDL-C-to-HDL-C ratio is an independent risk factor of early diabetic nephropathy in patients with type 2 diabetes mellitus.
8.A comparative study of using the CTP imaging to evaluation the hemodynamic changes before and after clipping of intracranial aneurysm

Objective To explore the change of cerebral microcirculation before and after clipping of intracranial aneurysm using CT perfusion (CTP)imaging.Methods CTP images of 70 patients diagnosed intracranial aneurysm by CTA were retrospectively an-alysed.They were divided into four groups according to have or not intracranial arterial spasm:A group:Preoperative CVS,postop-erative CVS (4 cases),B group:Preoperative CVS,postoperative nCVS (13 cases),C group:Preoperative nCVS,postoperative CVS (20 cases),D group:Preoperative nCVS,postoperative nCVS (33 cases).CBV/CBF/MTT were measured in four groups be-fore and after surgery,and analyzed statistically.Results (1)The postoperative CBV/CBF/MTT was compared with the preopera-tive data.There were statistically significant in CBVA/B/C/D/CBFA/B/D/MTTA/B during the surgery (P <0.05 ).There were no statistically significant in the CBFC and MTTC/D during the surgery (P >0.05).(2)There were statistically significant in the△CBVBC/BD/△CBFAB/BC/BD/△MTTAB/BD (P < 0.05 ).△CBVAB/AC/AD/CD/△CBFAC/AD/CD/△MTTAC/AD/BC/CD were no statistically significant (P >0.05).Conclusion CTP can early and accurately predict the change of cerebral microcircu-lation after surgery.
9.Effects of Matrine in Combination with 3 Antitumor Drug on Cell Cycle of KBV200 Cell Line

OBJECTIVE:To observe the effects of matrine in conjunction with vincristine(VCR),adriamycin(ADM)and diamminedichloroplatinum(DDP),respectively on the cell cycle of the KBV200drug-resistant cell line.METHODS:The cultured KBV drug-resistant cells were divided into control group,antitumor drug groups and the combination therapy groups(marine+antitumor drugs,respectively),FCM assay was used to detect and analyze the expression of cell cycle and apoptosis of each group.RESULTS:As compared with single antitumor drugs groups,no significant change was found in the proportion of cells and cell apoptosis in the combination therapy groups(matrine+VCR or matrine+ADM).But marine+DDD resulted in a reduction of the G 0 ~G 1 stage cell population but an increase of G 2 ~M stage cell population.CONCLUSIONS:It was assumed that matrine-induced reversal of KBV200cells'drug-resistant to VCR and ADM may not relate to cell cycle and cell apoptosis.Matrine can slightly enhance the cytotoxic effect of DDP.
10.The study of differential diagnostic value between pigmented villonodular synovitis and rheumatoid arthritis in active age with clinic and ultrasound

ObjectiveTo explore the value of differential diagnosis between pigmented villonodular synovitis (PVNS) and rheumatoid arthritis (RA) in active stage with clinic and ultrasound. MethodsWe analyzed retrospectively the characters of clinic and ultrasound of sixteen patients with PVNS and sixteen patients with rheumatoid arthritis inactive stage.ResultsIn aspects of patient’s age (T=185), pain scores (T=136) history of trauma (P=0.029), functionally uncompensated conditions (P=0.001) type (P=0.000) and sum (P=0.000) of involved joints、bright spot-echoes in hydrops (P=0.001) tendons nearby involved (P=0.001) and the Adler’s grade of Doppler ultrasound blood flow (P=0.009), the differences between PVNS and RA in active stage had statistical significance (P<0.05). Clinic characters: patients with PVNS were younger, often had a history of trauma. But patients with RA in active stage had a higher pain score and functionally uncompensated condition. Ultrasound characters: big and single joints were often involved in patients with PVNS, and tiny bright spot echoes could be detected in hydrops. Instead, big and small, multiple joints, tendons nearby were often involved in patients with RA, who had a higher ratio of gradeⅢ Doppler ultrasound blood flow. And in aspects of sex, the presence of hydrops and synovium thickness, the differences between the two had no statistical significance (P>0.05).ConclusionThe clinical characters including Patient’s age, pain scores, history of trauma, functionally uncompensated conditions, and the characters of ultrasound including type and sum of involved joints, bright spot echoes in hydrops, tendons nearby involved and the Adler’s grade of Doppler ultrasound blood flow, are helpful for the differential diagnosis of PVNS and RA in active stage.
