1.Terpinen-4-ol inhibits proliferation of VSMCs exposed to high glucose via regulating KLF4/NF-κB signaling pathway.

This study aimed to observe the effect of terpinen-4-ol(T4O) on the proliferation of vascular smooth muscle cells(VSMCs) exposed to high glucose(HG) and reveal the mechanism via the Krüppel-like factor 4(KLF4)/nuclear factor kappaB(NF-κB) signaling pathway. The VSMCs were first incubated with T4O for 2 h and then cultured with HG for 48 h to establish the model of inflammatory injury. The proliferation, cell cycle, and migration rate of VSMCs were examined by MTT method, flow cytometry, and wound healing assay, respectively. The content of inflammatory cytokines including interleukin(IL)-6 and tumor necrosis factor-alpha(TNF-α) in the supernatant of VSMCs was measured by enzyme-linked immunosorbent assay(ELISA). Western blot was employed to determine the protein levels of proliferating cell nuclear antigen(PCNA), Cyclin D1, KLF4, NF-κB p-p65/NF-κB p65, IL-1β, and IL-18. The KLF4 expression in VSMCs was silenced by the siRNA technology, and then the effects of T4O on the cell cycle and protein expression of the HG-induced VSMCs were observed. The results showed that different doses of T4O inhibited the HG-induced proliferation and migration of VSMCs, increased the percentage of cells in G_1 phase, and decreased the percentage of cells in S phase, and down-regulated the protein levels of PCNA and Cyclin D1. In addition, T4O reduced the HG-induced secretion and release of the inflammatory cytokines IL-6 and TNF-α and down-regulated the expression of KLF4, NF-κB p-p65/NF-κB p65, IL-1β, and IL-18. Compared with si-NC+HG, siKLF4+HG increased the percentage of cells in G_1 phase, decreased the percentage of cells in S phase, down-regulated the expression of PCNA, Cyclin D1, and KLF4, and inhibited the activation of NF-κB signaling pathway. Notably, the combination of silencing KLF4 with T4O treatment further promoted the changes in the above indicators. The results indicate that T4O may inhibit the HG-induced proliferation and migration of VSMCs by down-regulating the level of KLF4 and inhibiting the activation of NF-κB signaling pathway.

2.Preliminary clinical research on the afterloading brachytherapy inverse intensity-modulated radiotherapy of gynecological tumor

Objective To evaluate the clinical value of the afterloading brachytherapy inverse intensity-modulated radiotherapy of gynecological tumor.Methods Twenty patients with cervical cancer,were randomly divided into A and B groups,10 cases for each group.Group A received the afterloading brachytherapy inverse intensity-modulated radiotherapy.Group B received the three-dimensional comformal afterloading brachytherapy.The target volume dose distribution,organs at risk (rectum,bladder),shortterm curative effect and radioactive complications were analyzed on both groups.Results The dose homogeneity index of the target volume of group A was 52.43-± 0.45,better than that of group B (46.37 ± 1.45) (t =0.92,P < 0.05).The maximum dose of rectum and bladder of group A were about 37%,35%,less than that of group B (t =1.34,1.39,P < 0.05).The 75% prescription dose irradiated volume of rectum and bladder of group A were about only 1/2 of group B (t =1.23,1.13,P < 0.05).The local control rate of 96% for group A was better than 93% for group B (t =1.25,P < 0.05).Conclusions Afterloading brachytherapy inverse intensity-modulated radiotherapy technique could be better than the three-dimensional comformal afterloading brachytherapy.It should be recommended for gynecological tumor.

Based on the Internet information services ( IIS) 6. 0 6. 0 build system server service components,IE browser is used to implement the client access server capabilities. Results The information system including system Settings module,physics teacher module,the doctor module,technician module,data statistics and analysis, data download, seven modules such as video teaching. Conclusions The Radiotherapy information system is real?time performance, data security, stable operation, is the key construction for efficient utilization of resources in radiotherapy.

Objective:To study the effects of 1?,25-dihydroxyvitamin D 3 and UVB on cell proliferation and melanin synthesis of normal human melanocytes. Methods:Melanocytes of foreskins of healthy men were cultured and treated with various concentration of 1?,25-dihydroxyvitamin D 3 or UVB (55 mJ/cm 2),or both.Cell proliferation was measured with MTT assay .The synthesis of melanin was determined by chromatography. Results: 1?,25-dihydroxyvitamin D 3 and UVB could promote the proliferation of cultured melanocyte,and 1?,25-dihydroxyvitamin D 3 could promote the melanin synthesis.The significant concentration of 1?,25-dihydroxyvitamin D 3 ranging from 10 -7 to 10 -10 mol/L.Conclusion:1?,25-dihydroxyvitamin D 3 and UVB irradiation could promote the proliferation of melanocyte,which indicates that they might be effective in the treatment of vitiligo.

Objective To study the effects of endothelin-1 (ET-1) and adrenocorticotropic hormone (ACTH) on the proliferation of cultured normal human melanocytes. Methods Normal human melanocytes were obtained from the foreskins of healthy men, and cultured in minimum essential medium (MEM) with 10% bovine serum. Twenty-three patients with pain from osteoporotic vertebral fractures,aged 65-90 yr,weighing 51-78 kg,received an image intensifier-assisted nerve-root block with a 6-8 ml mixture of 0.5 % lidocaine,mecobalamine 0.5 mg and betamethasone sodium phosphate injection 5.26 mg in a prone or lateral position.The VAS scores before operation,at 0,1 week,1 and 3 months after operation were 8.6 ± 0.9,1.5 ± 0.7,2.8 ± 0.9,1.6 ± 0.5 and 2.5 ± 0.7,respectively.VAS scores were significantly lower at each time pint after operation than before operation (P < 0.05).According to modified MacNab standard,the effectiveness of treatment was rated as excellent/good in 87% of the patients.No complication such as bleeding,hematoma,infection,pneumothorax,hemopneumothorax,headache was found during or after operation.Selective nerve-root block is effective in the treatment of pain resulting from osteoporotic vertebral fractures in patients.
7.Effect of photodynamic therapy on acne

Objective To investigate the curative effect of photodynamic therapy on acnes and summarize the nursing measures.Methods Twenty-four acne patients were treated with photodynamic therapy from our hospital.The nursing experience was summarized.Result Nineteen had complete recovery,3 significant improvement,and 2 mild improvement,with a total effective rate of 91.7%.Conclusion Health education on acnes,keeping the patients away from strong sunlight and instructing them to clean skin in a right way and timely treatment of complications are critical for the improvement of curative effect.

Objective To explore the expression of high selective A receptor of endothelin-1 (ETA-R) in melanocytes from normal human beings. Methods Normal human melanocytes were cultured and mR-NA expression of ETA-R in melanocytes was measured by reverse transcription-polymerase chain reaction. Results The mRNA expression of ETA-R was seen in melanocytes. Conclusion This study combined with other experiments suggests that endothelin-1 may affect melanocytes by high selective ETA-R which may con-tribute to the pathogenesis of vitiligo.
9.Changes of CD34(+) and CD71(+)CD45(-) cell levels in bone marrow of MDS and AA patients.

This study was aimed to investigate the changes of CD34(+) and CD71(+)CD45(-) cell levels in MDS and AA patients. A total of 25 cases MDS and 43 cases of AA (18 cases SAA and 25 cases of NSAA) from January 2010 to October 2013 in the Department of Hematology, affiliated hospital of Hebei United University were enrolled in this study. The complete blood count, bone marrow smears, bone marrow biopsy, karyotype analysis and bone marrow blood cell immune genotyping (mainly the proportion of CD34(+) cells, CD71(+)CD45(-) cells in nucleated cells) were carried out for all patients; the changes of CD34(+) and CD71(+)CD45(-) cell levels in patients with MDS and AA (SAA NSAA) were compared; the differences of white blood cell count, platelet count and hemoglobin concentration in patients with count of CD71(+)CD45(-) ≥ 15% or <15% were analyzed. The results showed that the count of CD34(+) in MDS group was higher than that in AA (NSAA and SAA) group (P < 0.05). The count of CD71(+)CD45(-) cells in MDS group was higher than that in SAA (P < 0.05), there was no significant difference between NSAA group and MDS group. In MDS group with CD71(+)CD45(-) ≥ 15%, the platelet count was significantly higher than that in NSAA group (P < 0.05); and there was no statistical difference for leukocyte, platelet count and hemoglobin level between MDS and NSAA group with CD71(+)CD45(-) <15% (P > 0.05). It is concluded that the count of CD34(+) cells in MDS patients is significantly higher than that in AA and SAA patients. The count of CD71(+)CD45(-) cells in MDS group is significantly higher than that of SAA group. The platelet count in MDS patients with CD71(+)CD45(-) cells ≥ 15% is significantly higher than that of the NSAA group.
