1.Determination of ganciclover in human plasma by reversed-phase high performance liquid chromatography

Objective To develop a method for the determination of ganciclover in human plasma by RPHPLC.Methods Plasma containing ganciclover was extracted with methanol and methylene chloride,qualitative and quantitative analysis was carried out directly.Working curve,linear range,recovery,precision and so on was obtained according to the sample pre-processing method and analysis state.The HPLC method has been taken to investigate the plasma concentration of ganciclover for 12 volunteers.Results The relationship of the peak area of ganciclover concentration in plasma linear within the range of 0.05 μg/mL~1.60 μg/mL(r=0.9999).The lowest detection limit was 0.01 μg/mL(S/N≥13).The intra and inter-day RSD were less than 5.1%respectively.The recovery is about 90.0%~95.4%.Conclusion The established method in the article was shown to be sensitive,accurate and simple for the determination of ganciclover level.It is suitable for clinical detection of ganciclover and forensic medicine and toxicology analysis.
2.Impact of the image quality of conebeam CT on the accelerator for the Bowtie filter

Objective To estimate the impact of the image quality of conebeam CT on the accelerator for the Bowtie filter. Methods This study used the system of conebeam CT on the accelerator,and scaned the phantom of Catphan 500 in the multiplicate scanning conditions with and without the Bowtie filter. The scanning conditions were : voltage :120 kV, 100 kV, 80 kV, electricity:25 mA, 40 mA, 64 mA,time of exposure :40 MHz, collimator: M20, L20, scanning angle :358°. We transmitted the scanning data in the treatment planning systems of Philips Pinnacle 8. 0, and estimated the influence of the image quality.Results With and without the Bowtie filter, all of the images had high geometric precision, and the maximum difference of the nominal and actual distance at the horizontal, vertical and arrowy directions in the transection were 0. 50% and 0. 56% ( t = 0. 44, P > 0. 05 ), 0. 44% and 0. 48% ( t = 1.01, P > 0. 05 ),0. 50% and 0. 56% (t = 1.06,P >0. 05 ), respectively. The spatial resolution was 0. 167 cm, immutably.The ununiformity with the Bowtie filter was decreased of 28.0% -76.0% (t=2.53,P<0. 05)、2.8%-50.0% (t= 3.02,P<0.01 ) than those without the Bowtie filter, and the contrast-to-noise ratio was increased of 4.0% -41.6% (t=2.70,P<0.05)、4.0% -50.0% (t=2.14,P<0.05), using the collimators of M20 and L20, respectively. The Bowtie filter could display clearly the edge of scanning phantom, and improve the CT number of the image edge of phantom. And the linearity of CT number was also improved. Couclusions With the Bowtie filter, the image uniformity, the precision and linearity of CT number and the contrast-to-noise ratio were improved obviously, the edge artifacts were decreased, and the geometric precision and the spacial resolution did not change.
3.Effects of enteral nutrition added with glutamine on gastrointestinal functions and prognosis of acute severe traumatic brain injury

Objective To investigate the effects of enteral nutrition added with glutamine on the incidences of gastrointestinal complications,intestinal mucosal barrier function and inflammatory responses in patients with acute severe traumatic brain injury (sTBI).Methods A prospective case control study was made on 107 patients with sTBI hospitalized from January 2016 to June 2017.The patients were divided into experimental group added with glutamine (n =54) and control group without glutamine (n =53) according to the random number table.The general data of the patients were recorded.After treatment,the incidences of gastrointestinal complications in both groups were compared.The serum levels of intestinal mucosal barrier function indices,namely,diamine oxidase (DAO),Dlactate acid,and intestinal fat acid binding protein (I-FABP) were evaluated by enzymology spectrophotometer method.Meanwhile,the serum levels of C-reactive protein (CRP),tumor necrosis factor-α (TNF-α),and interleukin-6 (IL-6) were also tested with enzyme-linked immunosorbent assay (ELISA).Glasgow coma scale (GCS),acute physiology and chronic health evaluation Ⅱ (APACHE Ⅱ),and hospital stay in both groups were compared.Results The two group were comparable with respect to gender,age,injury reasons,body mass index,preoperative GCS,preoperative APACHE Ⅱ,injury type and injury time (P > 0.05).The experimental group had lower incidences of stress ulcer,gastric retention and diarrhea compared with the control group 14 days after treatment (P < 0.05).Within 14 days after treatment,the serum levels of DAO,D-lactate acid and I-FABP were significantly decreased in the experimental group at days 7 and 14 after treatment (P < 0.05).The serum levels of CRP,TNF-α and IL-6 in the experimental group were significantly decreased after treatment (P < 0.05).The experimental group had better prognosis compared with the control group (P < 0.05),with higher GCS scores [(9.3 ± 0.7) points vs.(8.2 ± 0.7) points],lower APACHE Ⅱ scores [(15.3 ± 1.1) points vs.(17.7 ± 1.2) points] at day 14,and shorter hospital stay [(19.1 ± 2.2) days vs.(25.3 ± 2.4) days] (P < 0.01).Conclusions Enteral nutrition added with glutamine can effectively reduce the incidence of gastrointestinal complications,as well as alleviate the intestinal mucosal barrier function damage and the inflammatory responses at early stage after sTBI,which possibly improves prognosis.
