1.Influence of lower myocardial infarct patients appeared Ⅲ atrio-ventricular block treatment with thrombolytic recanalization

Objective To investigate the influence of patients suffered from acute lower wall cardiac infarct appeared Ⅲ atrio-ventricular block tneatment with thrombolytic recanalization for the early recovery of sinus rhythm.Methods From October 01 till October 06, a total of 25 hospitalized patients in our hospital with acute lower cardiac infarct, chest pain persist over thirty minutes, but less than one hour, immediately treatment with urokinase thrombolytic therapy after hospitalization, among them 17 cases reinfusion therapy success ful and 8 cases failed. Divided into reinfusion therapy success group and failed group, observate thromholytic therapy 6 hours recanalization success and fail group the situation of sinus rhythm recovore. Results Among 17 cases recanalization success patients and 14 cases recover sinus rhythm within 6 hours ,and 8 case fail in recanalization 2 cases recover sinus rhythm in 6 hours(25%)compared these two groups statistical are meaningful ( P<0. 01 ). Conclusion Acute lower cardiac infarct patients with Ⅲ atrio-ventricular block must use thrombolytic therapy as soon as possible in order to let oclusive coronary artery recanalize, help to recover sinus rhythm as soon as possible and decrease mortality rate.
2.Treatment and prognostic predictor of 28 cases with malignant melanoma of the head and neck

Purpose:To determine the treatments and prognostic predictors of malignant melanoma of the head and neck. Methods:Surgical treatment and outcome was analyzed for 28 patients with malignant melanoma of the head and neck at department of the head and neck of our hospital (1984-1994). Results:23 patients underwent local excised as the first treatment, and local recurrence was demonstrated in 8 (34.8%). Margin of resection of primary melanoma was divided into narrow excision (≤2 cm) and wide excision (>2 cm), and 5-year survival was 58% and 40%(P<0.01). According to surgical methods, 5-year survival of WE, WE/END and WE/TLND were 80%, 80% and 39%. Conclusions:An excision with 2 cm margin is safe procedure for primary tumor of malignant melanoma of the head and neck. Neck dissection may not be practiced until neck lymph node is positive. Neck dissections differ from the positions of the primary tumors, and the flaps must be raised preferably superficial to the platysma.
3.mRNA Expression of c-myc Gene in the Hyperacute Rejection of Xe notransplantation

Objective:To research expression and function of c-myc gene in hyperacute r ejection of xenotransplantation. Method:Through the model of hyperacute rejection of xenotransplantation wit h isolated rat heart perfused with human plasma,we detected c-myc mRNA expressi on and cell localization with in situ hybridization in hyperacute rejection. Result:c-myc mRNA increased expression in the endothelium cell of heart.Th e value of signal is 450.09±409.99.The value of signal of group of control is 1 74.40±51.50(p<0.05)。 Conclusion:From these study findings,it would appear that hyperacute rejecti on is associated with the elevation of c-myc mRNA expression.
5.Effect of curcumin in inducing apoptosis of MDA-MB-213 cells by activating endoplasmic reticulum stress.

OBJECTIVETo explore the possible mechanism of curcumin in inducing the apoptosis of breast cancer cell MDA-MB-231.
METHODCurcumin of different concentrations at 0, 10 25, 50, 100, 150, 200 micromol x L(-1) were used to intervene breast cancer cells MDA-MB-231 for 24 hours. MTT was used to observe its effect on the proliferation of breast cancer cells. The flow cytometry was used to detect its effect on the cell apoptosis. The real-time quantitative PCR and Western blot was used to assess the expression levels of GRP78 and CHOP in breast cancer cells.
RESULTCurcumin could inhibit the proliferative ability of breast cancer cells by inducing them in a concentration-dependent manner. Curcumin could significantly increase the expression levels of GRP78 and CHOP in breast cancer cells.
CONCLUSIONCurcumin could induce the apoptosis of breast cancer cells MDA-MB-231 by activating endoplasmic reticulum stress.

6.Effects of xihuangwan in assistant treatment of patients with advanced breast cancer.

In order to explore the possibility of Xihuangwan (XHW)'s application in assistant therapy in patients with breast cancer, short- and long-term clinical efficacy were assessed in this study. Eighty and four patients with advanced breast cancer were selected in this study. They were divided into control group and treatment group randomly and evenly. All patients received surgical treatment followed by chemotherapy regimen composed of PTX + EPI + CTX (TEC regimen). Treatment group received additional assistant treatment of XHW. Short-term clinical efficacy was assessed by KPS, lesion stabilizing rate and side effects in 3-month follow-up study. Long-term clinical efficacy was assessed by overall survival (OS) and free-progression survival (FPS). KPS increased significantly after treatment in all patients (P < 0.05), more significantly in treatment group than in control group after treatment (P < 0.05); lesion stabilizing rate in treatment group increased significantly in treatment group than in control group (92.86% vs. 85.71%, P < 0.05); there was no significant difference between control group and treatment group in occurrence of side effects. Compared with control group, OS and FPS increased significantly in treatment group. Data in this study showed that XHW was suitable in treatment of advanced breast cancer due to its satisfactory short-term and long-term therapeutic effects.

7.The safety and efficiency of precise liver resection for patients with primary liver cancer

Objective To evaluate the the safety and efficiency of precise liver resection for patients with primary liver cancer. Methods 86 patients with primary liver cancer were randomized to receive conventional routine hepatectomy (n=44) or the precise liver resection (n=42). Outcomes were compared between the precise hepatectomy group and the routine group, including, the blood loss, operation time, morbidity and mortality. Results There were significant differences in morbidity rates (7.1% vs. 20.5%; P<0.001), the blood loss [(320±315) ml vs. (613±526) ml;P<0.001) , postoperative alanine aminotransferase (ALT) value (in postoperation 7 d, 82.7 U/L vs.321.7 U/L; P<0.001) and length of hospital stay (12.3 d vs. 18.6 d; P<0.001) between precise hepatectomy and routine groups. The 1 year tumor recurrence rate and 1 year survival rate were 26.2%(11/42) and 78.6% (33/42) in precise liver resection group, 38.6% (17/44) and 65.9%(29/44) in routine liver resection group,with significant difference (P=0.010;P=0.018). Conclusion Precise liver resection is safe and effective in the treatment of liver tumor without much injury to patients.
8.Association of killer cell immunoglobin-like receptor gene with bone marrow failure syndromes

Objective To investigate the correlation of killer immunoglobulin-like receptors(KIR)gene polymorphism with bone marrow failure syndromes(BMFS). Methods SSP-PCR was used to examine the genotypic makeup of KIR in patients with aplastic anemia( AA), myelodysplatic syndrome (MDS) and healthy controls in our department. Results All the 16 KIR genes which had been prescribed were identified. The frequencies of KIR-2DS1, 2DS2, 2DS3, 2DS5 and 3DS1 genes were showed increased in patients with AA, MDS than in healthy controls. The patients with AA had lower frequency of KIR-2DS5 than the patients with MDS. Conclusion The increased frequencies of these activated KiRs in patients with MDS and AA suggest that the abnormal immunogenetic might be related to the pathogenesis of BMFS.
9.protective effect of Enalapril and weight reduction on podocyte lesions of obesity-related gLomerulop-athy rats

Objective To investigate the expression of WT1 in pedecyte of obesity- related glomerulopathy (ORG) rats and the mechanism of enalapril and weight reduction-induced renal protection. Methods After the ORG model was established by high lipid forage for 24 weeks, the rata were divided into control rats (group A), ORG rats (group B), enalapril-treated ORG rats (group C), ORG rats with weight reduction (group D) and enalapril -treated ORG rats with weight reduction (group E). At the 8th week, 24 hour urine albumen (24hUA1b) was assayed, and the pathological changes of kidney were detected by optics microscopy and transmission electronic microscopy. Immunohistochemistry was used to detect the expression of WT1, a marker of pedocyte injury. Results In ORG rats, pedocyte number and density were declined remarkably compared with group A (respectively, P<0.01), and 24 hour UA1b and obvious kidney damage were sig- nificantly higher than those of group A (respectively, P<0.01 ). After treatment, pedocyte number and density in group (C, D, E) were significantly increased compared with group B (respectively, P<0.05, P<0.01). Meanwhile, 24honr UA1b was reduced and kidney dam- age was alleviated ( respectively, P <0. 01 ), and therapeutic alliance effect in group E outweighed than that in group C and D. Conclusion Changes of pedocyte in ORG correlated with degree of proteinuria and renal function impairment. Enalapril and weight reduction can re- strain the expression of WT1 in ORG, which can reduce proteinuria and lessen kidney damage.
10.Surface instillation of stem cell culture in repair of severe skin trauma in guinea pigs

BACKGROUND: Stem cells, cells with special function in animals and humans, exist in various tissues. Most of stem cells differentiate into special tissue organs and some of them remain in the status of stem cells for tissue repair. Mesenchymal stem cells were transplanted to burn wounds in some researches for inducing the proliferation and activation of skin stem cells so as to cure burn.OBJECTIVE: To observe the effect of stem cell culture medium cultured in vitro instilled locally into the severely traumatic skin in guinea pigs on healing time and healing degree of the wound.DESIGN: Random grouping and blank control trial.SETTING: Department of Toxicology, School of Public Health, Jilin University.MATERIALS: Totally 14 adult healthy guinea pigs of either gender weighing 300 to 350 g were recruited.METHODS: The experiment was conducted in the Laboratory of Toxicology Department, School of Public Health, Jilin University, from March to September 2003. Ten guinea pigs were put to death by bloodletting on the neck. The bone marrow was extracted and cultured in unicellular supematant fluid for use. The 14 guinea pigs were made into models of bilateral severe skin trauma.Ten of the guinea pigs were chosen randomly, stem cell culture was instilled into one side of the animals (stem cell group), while the culture medium was instilled into the other side of the animals (culture medium group). The remaining 4 guinea pigs that received no treatment were blank control group.Three days later, transparent lucite was put on the wound every other day for drawing the shape and observing the wound. After the shape was copied onto the transparent lucite, the wound area was worked out on the rectangular coordinate paper and the speed of wound healing was calculated.MAIN OUTCOME MEASURES: Gross observation was performed on the healing status of the wound and average healing time and speed of the guinea pigs in each group.wound healing status of the guinea pigs in each group: At day 3, the wound in stem cell group was dry without obvious exudation. There was a layer of membrane at the bottom of the wound and it stuck to the pledget closely so that the dressing could not easily be removed. The wound status in the culture medium was practically the same as that of the stem cells, but no membrane-ike material was formed and the dressing was easily removed. Inflammation appeared obviously in blank control group.nificantly less in stem cell group than in culture medium group and blank control group [(12.45±2.18) days vs (26.29±1.38) days and > 30It was obviously faster in stem cell group than in culture medium group and blank control group [(40.42±2.14) mm2 per day vs (15.53±5.22) mm2 per day and(10.27±4.57) mm2 per day,P < 0.05].CONCLUSION: The stem cells of homologous animals were instilled on the skin surface to repair injury wound. Stem cells can grow on the wound and transform into skin cells, which promotes the recovery of wounded skin. Therefore, it is feasible to treat severe skin defect with stem cells.
