Objective:To establish the quality standard for Tangkening Granule.Methods:Gardenia,Gold Theragran and batatatis in Tangkening Granule were identifi ed by TLC,the content of geniposide was determined by HPLC.Result:The spots in TLC was clear,the negative control had no interference,the average recovery was 99.9%,RSD was 2.9%(n=6).Conclusion:The method was accurate,reproducible and specifi c.It can be used for quality control of Tangkening Granule.

Objective To evaluate the diagnostic values of location and qualitation of tendinitis of rotator cuff. Methods One hundred and sixty-two shoulders with shoulders pain or functional incapacity were performed ultrasonography and the sonograms were classified. Results Ultrasonic findings of these tendinitis were tendinitis of caput longum musculi bicipitis brachii in 39 cases, tendinitis of caput longum musculi bicipitis brachii in 33 cases, supraspinatus tendinitis in 51 cases, calcific supraspinatus tendinitis in 19 cases and bursitis of shoulder in 20 cases,especially. Conclusions Ultrasonography is a useful tool for diagnosis of tendinitis of rotator cuff in location and qualitation.

AIM:To clarify the impact of heart shock factor 4b (Hsf4b) K65R mutation on the regulation of downstream protein expression .METHODS:Non-functional Lys mutant plasmid pWZL-blast-HA-Hsf4b/K65R was genera-ted by replacing single , homologous amino acids using KOD-Plus-Mutagenesis-Kit.Mouse lens epithelial mLEC stable cell lines expressing Hsf4b or Hsf4b/K65R were constructed by lentivirus infection .The expression of Hsf4b in the mutant and the wildtype mLEC cells was confirmed by Western blotting .The expression of Hsf4b downstream proteins such as heat shock protein ( Hsp)70, Hsp90, Hsp27 and CryAB was examined by Western blotting and real-time PCR.RESULTS:The results of PCR and DNA sequencing confirmed the successful construction of mLEC Hsf 4b/K65R mutant.The K65R mutation didn’t influence Hsf4b expression in the mLEC cells.After K65R mutation in Hsf4b, the expression levels of Hsp27 and CryAB were down-regulated and the expression of Hsp 70i and Hsp90a upregulated.CONCLUSION: pWZL-blast-HA-Hsf4b/K65R can be used to construct a stable cell line by infecting with lentivirus .Hsf4b/K65R mutation influ-ences the regulation of downstream heat shock proteins .

In order to strengthen the management of disinfection quality of endoscopes, Quality Control Center of Digestive Endoscopy and Nosocomial Infection Control Center of Yunnan Province investigated the diagnosis, treatment, cleaning and disinfection conditions and disinfection quality of digestive endoscopes in some medical institutions of Yunnan Province by web questionnaire from April to May in 2019, and 277 valid questionnaires were finally obtained. SPSS 19.0 statistical software was used to analyze the influencing factors of cleaning and disinfection process and the infection control implementation of digestive endoscopes in 227 secondary and tertiary hospitals. The results showed that the number of decontamination people who had received systematic training in Yunnan Province was significantly lower than that in other areas of China. The hospital level, the number of decontamination personnel, and decontamination methods affected the implementation of cleaning and decontamination process and infection control, while the allocation of decontamination supplies had no effects. It is important to establish an effective mechanism for the normalized implementation of cleaning and disinfection of digestive endoscopes.

Objective To establish a method for rapidly detecting the G-protein ?3 subunit (GNB3) 825 site single nucleotide polymorphism (SNP) and to analyse the relationship between GNB3 825 site gene SNP and obesity. Methods The real-time fluorescent PCR was employed to analyse the GNB3 825 site gene SNP of 420 samples from 21 provinces and the the frequencies of genotypes were compared with those detected by gene sequencing. GNB3 825 site genotype, body weight, body mass index (BMI) and fat content were examined from 207 subjects and the correlation between GNB3 825 site gene SNP and obesity was analysed. Results The result by real-time fluorescent PCR showed that the frequencies of 825T and 825C haploid were 46.90% and 53.10%, respectively, and the frequencies of 825TT, 825TC and 825CC genotype were 22.38%, 51.42% and 28.10%, respectively, with no other genotype detected, which was consistent with the result by gene sequencing. BMI and fat content were significantly higher in subjects with GNB3 825TT than in subjects with other genotypes. Body weight was much higher in subjects with GNB3 825TT genotype than in subjects with 825CC genotype, but not significantly different with 825CT genotype. Conclusion A new rapid method for the detection of GNB3 825 site SNP has been successfully established. There existed significant correlation between GNB3 825TT genotype and obesity.

Objective To observe the clinical role of Qiliqiangxin capsules uniting conventional western medicine interfering with chronic heart failure(CHF).Methods 61 cases of CHF were randomly recruited into a control group(31 cases)and a treatment group(30 cases).The control group was treated with conventional anti-hean failure treatment,and the treatment group was added Qiliqiangxin capsules on the treatment of the control group.After having been treated for 2 courses (fonr weeks),such indexes as clinical efficacy,body weigbt,urine volume,6 min walking distance(6MWD),electrocardiogram,and cardiac ultrasound were measured for both groups.Results The overall efficiency of cardiac function was 93.33%for the treatment group and 74.19%for the control group.There was significant difierence between the two groups(χ~2=4.07985,P＜0.05).The enhance rate of left ventricular ejection fraction(LVEF)was(16±4)%in the treatment group as compared to(11±3)%in the control group,also showing statistical difierence between the two groups(t=5.509,P＜0.001).Conclusion Qiliqiangxin capsules combined with conventional western medicine could effectively improve the cardiac function,LVEE clinical symptoms,and quality of life in the patients with ehronic heart failure.

A total of 1507 children aged 7 ～ 12 years in Qinhuangdao were incruited to evaluate the diagnostic accuracy of different obesity indices in screening hypertension.Blood pressure and obesity indices were positively correlated(P＜0.01).Area under the receiver operating characteristic (ROC) curve were 0.744 ～ 0.835 in boys and 0.704 ～ 0.796 in girls (P＜0.01).Obesity indices could be used as parameters for detecting hypertension in children.

Objective: To study bidirectional binding ability and immunoactivity of anti-CD3/anti-glioma bispecific antibodies(BsAb) , and to study its effect on T lymphocytes cells. Methods: Elution from standardized gel column and immnuohistochemical method were used to testify the relative molecular mass of the compound. ELISA method was used to detect its bidirectional immunoactivity and potential T lymphocytes' cytotoxicity. Phenotype and shape of T lymphocytes were also observed. Results: The relative molecular mass of the compound was 110 000, and it combined with both T lymphocytes and glioma cells.Immunoactivity assay showed the compound had good bidirectional immunoactivity. It also enhanced T lympocytes anti-tumor activity. Immunoactivity of BsAb combining with T lymphocytes was 1∶32 000, and combining with glioma cells was 1∶8 000;In phenotypes, CD3 + cells were the major one when cultivated with BsAb/IL-2, CD8 + cells increased gradually with cultivation time. Conclusion: These compounds are bispecific antibodies with bidirectional combining activity to T lymphocytes and glioma cells, they also have bidirectional immunoactivity and can activate T lymphocytes
