Academic Journal of Second Military Medical University 2010;31(10):1109-1113
Objective: To screen for differentially expressed genes associated with the development and progression of human growth hormone adenoma, so as to lay a foundation for future study. Methods: The whole genome oligonucleotide microarray (Affymetrix 133 plus 2.0) was used to examine gene profiles of 8 growth homone adenoma samples and 2 normal pooled pituitary samples. Differentially expressed genes were subjected to bioinformatics analysis. Real-time qPCR was used to verify the microarray result of a randomly selected candidate gene. Results: A total of 187 up-regulated genes and 899 down-regulated genes associated with growth hormone adenoma were screened out, with their functions mainly associated with molecular binding, apoptosis/tumor, metabolism, signal transduction, cell cycle, and transportation activities. Conclusion: Microarray technology can be used for preliminary screen of growth hormone adenoma associated genes. The development and progression of growth homone adenoma are complex processes involving multiple genes, molecules, and pathways.

Objective To study the influence of bFGF gene transfected bone marrow-derived mesenchymal stem cells (BMSCs) on the inflammatory cytokines of COPD rat. Methods The BMSCs were separated from SD rat and cultured and then bFGF gene was imported to BMSCs by liposome transfection method. The samples were prepared into six groups: normal control group, COPD group (A), BMSCs group (B), pcDNA3.1-BMSCs group (C), bFGF-pcDNA3.1-BMSCs group (D), and bFGF group (E). The expressions of TNF-α and IL-1β by QRT-PCR were detected. Results Compared with COPD group, TNF-α and IL-1β genes from groups B to D dropped significantly (P < 0.05). The changes of TNF-α and IL-1β genes among groups B to D showed no significant difference (P > 0.05). Conclusion BFGF transfected BMSCs, sample BMSCs and pcDNA3.1 transfected BMSCs can inhibit the expression of inflammatory cytokines of TNF-α and IL-1β, but there is no obvious advantage in comparison to bFGF transfected BMSCs and sample BMSCs in respect of inhibiting the expression of inflammatory cytokines of TNF-α and IL-1β.

7.Therapeutic instrument for brain circulation function on the basis of direct digital synthesis technology.
Journal of Biomedical Engineering 2005;22(1):143-146
Electricity can stimulate cerebral fastigial nucleus to increase regional cerebral blood flow (rCBF) and narrow the volume of infarct, therefore its effect on ischemic brain disorder may be helpful. The therapeutic instrument for brain circulation function based on direct digital synthesis (DDS) technology uses the domestic newest scientific findings and others in foreign countries. Adopting the DDS technology directly, it institutes safe and efficacious treatment. Through pasting the electrodes on body surface, the non-invasive instrument introduces electric current to cerebral fastigial nucleus (FN) and hence it improves the state of brain blood supply and nerve conduction, increases cerebral blood flow, reduces the number of necrotic neurons in the district of penumbra, and mitigates brain edema. Consequently, it improves the function of brain circulation.
