Academic Journal of Second Military Medical University 1982;0(02):-
Objective:To study the protective effect of self-designed Shanghai multi-organ preservation solution(SMO) on mitochondria function in canine kidney cortex during cold storage. Methods: Canine kidneys were perfused and stored with cold(0-4℃) SMO,HC-A and UW solution (the later 2 as control) separately.Ultrastructure alteration of mitochondria was observed by electron microscopy.Mitochondria were isolated from cortex homogenization by differential centrifugation and their respiratory function was measured polarographically with Clark oxygen electrode. The four-state curve of mitochondria respiration was drawn and the state Ⅲ and Ⅳ oxygen consumption rate, the respiration control proportion(RCR) and phosphatasel/oxygen ratio(P/O) were calculated. Results: We found that the mitochondria swelling and membrane disruption in HC-A group were more severe than those in SMO and UW group. The rate of oxygen consumption in state Ⅲ,RCR and P/O were obviously decreased in all groups with the prolongation of cold storage time. The rate of oxygen consumption in state Ⅲ in HC-A group was lower compared to that in SMO group during 1-3 d cold storage(P
2.Multi-organ preservation solution: progress in laboratory research and prospects of clinical application
Academic Journal of Second Military Medical University 1985;0(06):-
UW solution has bean considered the gold standard for abdominal organ preservation. Celsior solution and HTK solution, with low potassium and low viscosity, are becoming more widely used in the preservation of the kidney, liver and pancreas, and are gradually accepted as an ideal alternative for UW solution. Recent studies have found that addition of trophic factors can attenuate hypothermic ischemic injury to the donor organ, thus improving the preservation outcome. This paper reviews the progress in laboratory research and looks foward to the clinical application of multi-organ preservation solution.

?METHODS: Seventy cases ( 70 eyes ) whose the main symptoms was epiphora induced by CCh were divided into II, Ⅲ and Ⅳ in 3 groups according to Zhang’s grading standards, meanwhile, all the patients were treated with the eye drops of Pranoprofen for 2wk. After that, the curative effect of each group was analyzed before and after treatments according to the symptoms, dye disappearance test, bitter taste test and tear break-up time.
?RESULTS: After 2wk treatment of Pranoprofen, the symptom of patients at grade I and II improved obviously grade Ⅲ improved more, grade Ⅳ did little. The positive rate of dye disappearance was the highest in grade II, the second in grade Ⅲ, and the third in gradeⅣ. There was significant difference among the 3 groups (P<0. 05). The time of bitter taste test was shortened (P<0. 05) and the tear break-up time was prolonged (P<0. 05) in grade II after treatment, however, the time of those in gradeⅢandⅣhad not changed than before ( P>0. 05).
?CONCLUSlON: Pranoprofen has good effects on tear dynamic abnormity induced by grade II of cch.
5. Influence of a self-designed multiple organ preservation solution on apoptosis of renal cortical cells during hypothermic storage
Academic Journal of Second Military Medical University 2006;27(10):1059-1062
Objective: To observe the influence of a self-designed multiple organ preservation solution, namely Shanghai multi-organ preservation solution (SMO), on the apoptosis of canine renal cortical cells during cold storage and the related mechanism. Methods: Canine kidneys were freshly perfused and separately stored with cold (0-4°C) SMO, hypertonic adenine citrate solution (HC-A), and University of Wisconsin solution (UW solution) (the latter 2 as control). At defined time points during storage, the morphologic changes of renal cortical samples were observed; apoptosis of renal cortical cell was determined by TUNEL; changes of malonldialdehyde (MDA) in cortex homogenate were measured by thiobarbituric acid method; and the activity of superoxide dismutase (SOD) was determined by xanthine oxidase method. Results: Pathologic assessments indicated that the kidney damage was similar in 3 groups within 24 h. The kidney tubular cell swelling and necrosis in SMO group and UW group were slighter than those in HC-A group after stored for 2 days. With the prolongation of storage time, apoptosis increased in all groups, with the apoptotic index of SMO group significantly lower than that of HC-A group (P<0.05). The levels of cortical MDA increased with storage time, and that in SMO group was significantly lower than that in HC-A group (P<0.05). The activities of cortical SOD were similar in the 3 groups. Conclusion: SMO solution can improve the efficiency for protection of renal cortex, which might be associated with the decrease of cells apoptosis and inhibition of lipid peroxidation injury.
6. Protective effects of self-designed Shanghai multi-organ preservation solution on kidney cortex mitochondria function during cold storage
Academic Journal of Second Military Medical University 2006;27(2):196-199
Objective: To study the protective effect of self-designed Shanghai multi-organ preservation solution(SMO) on mitochondria function in canine kidney cortex during cold storage. Methods: Canine kidneys were perfused and stored with cold (0-4 °C) SMO, HC-A and UW solution (the later 2 as control) separately. Ultrastructure alteration of mitochondria was observed by electron microscopy. Mitochondria were isolated from cortex homogenization by differential centrifugation and their respiratory function was measured polarographically with Clark oxygen electrode. The four-state curve of mitochondria respiration was drawn and the state III and IV oxygen consumption rate, the respiration control proportion(RCR) and phosphatase1/oxygen ratio(P/O) were calculated. Results: We found that the mitochondria swelling and membrane disruption in HC-A group were more severe than those in SMO and UW group. The rate of oxygen consumption in state III, RCR and P/O were obviously decreased in all groups with the prolongation of cold storage time. The rate of oxygen consumption in state III in HC-A group was lower compared to that in SMO group during 1-3 d cold storage(P<0.05). The rate of oxygen consumption in state IV was slightly increased during the cold storage, but there was no statistical difference between HC-A and SMO group. RCR and P/O in SMO group were statistically higher than those in HC-A group (P<0.05) and were similar to those of UW group. Conclusion: SMO solution has similar protective effect on cortex mitochondria function impairment induced by hypothermia and ischemia during cold storage as UW solution, and the effect is more potent than that of HC-A solution.

Objective To study the characteristics of plasma adiponectin levels exhibiting in gestation women and newborns, as well as the relationships between adiponectin levels and fetal birth weight. Methods Totally 98 subjects have been considered in this study, in-cluding venous blood samples of 36 healthy non-pregnant women (control group), 31 uncomplicated pregnant women (pregnancies group) and the cord blood samples taken at delivery of their singleton infants born at term (newborns group). The concentrations of adiponectin and insulin were determined by radioimmunoassay technique. And SPSS was used for statistical analyses. Results The adiponectin level in women of late period of gestation (WLPG) (11.10±5.72)g/ml was lower than that of other two groups, and the newborns average cord plasma level of adiponectin was (30.71±12.77) g/ml significantly higher than that of the control group (16.52±6.87) g/ml. The adipone-ctin level for WLPG was correlated with FINS and HOMA-IR (r = -0.411, -0.393, P <0.05). The adiponectin level in cord plasma was positively correlated with birth weight(r = 0.416, P < 0.05). The adiponectin level in WLPG was correlated neither with the adiponec-tin level nor body weights of their newborns. Conclusions The average cord plasma adiponectin level of newboms is significantly higher than that of the control group and WLPG. It is positively correlated with birth weight, which suggests that adiponectin may be involved in reg-ulating fetal growth. The adiponectin level in WLPG is correlated neither with the adiponectin level nor body weights of their newborns.
