1.Effect of Initial Periodontal Therapy on Eradiation Rate of Gastric Helicobacter Pylori in Patients with Gastric Ulcer

Objective:To investigate the adjuvant effect of initial periodontal therapy by observing the eradiation rate of gastric helicobacter pylori( Hp) in the patients with gastric ulcer treated by the therapy. Methods:Totally 88 patients with stomach ulcer and chronic periodontitis were enrolled and randomly divided into two groups. The control group was treated by triple therapy,the experimental group was treated by initial periodontal therapy additionally with the treatment course of one month. The changes of Hp in stomach and periodontium were observed and the eradiation rate of Hp was compared between the two groups respectively after 1-, 3-, 6-and 12-month treatment. Results:After 3, 6 and 12 months, the eradiation rate in the experimental group was significantly higher than that in the other group (P < 0. 05). Conclusion: Initial periodontal therapy combined with triple therapy can obviously improve the eradication rate of gastric Hp in the patients with gastric ulcer and chronic periodontitis.
2.Tenascin(TN) expressions and its effect in salivary gland tumor

Objective:To detect tenascin(TN) expressions and its effect in salivary gland tumor. Methods:The expressions of TN was examined in 8 chronic inflammations of salivary glands, 50 benign adenomas and 57 malign adenomas by Envision immunohistochemical technique. Results:Expression level of TN in the malign adenomas was higher than that in benign adenoma and chronic sialadenitis; higher in low differentiation than that in high differentiation and higher in lymphatic metastasis groups than that without lymphatic metastasis groups. No obviously difference was found between soakage inside the blood vessel and nerves and without soakage. Conclusion: TN might regulate cell attachment of epithelial cells in adenomas, and involve in adenomas' histogenesis and development.
3.Effect of compound anisodine on fundus bIood circuIation after vitrectomy with face-down posture

· AlM:To investigate the effect of compound anisodine on fundus blood circulation after vitrectomy with face-down position.
· METHODS: Sixty patients ( 60 eyes ) with rhegmatogenous retinal detachment received vitrectomy with silicone oil tamponade operation, who were randomized divided into treatment group ( 30 eyes ) and control group ( 30 eyes ) .The patients in the treatment group received the subcutaneously injection of compound anisodine hydrobromide by the superficial temporal artery once daily for 14d since postoperative first day.Retinal microcirculation blood flow parameters were recorded with Heidelberg retinal flowmeter postoperative 1d, 1 and 2wk, and were compared between two groups.
·RESULTS: The blood flow parameters ( Vol, Flw, Vel) of control group postoperative 1 and 2wk were significantly less than those postoperative 1d.Otherwise the parameters of treatment postoperative 1 and 2wk were significantly more than those postoperative 1d. The parameters between two groups were significant different ( P<0.01) .
· CONCLUSlON: Facing down after vitrectomy with
silicone oil tamponade may reduce retinal blood supply, consequently lead to retinal ischemia; compound anisodine can effectively improve the retinal and choroidal microcirculation after vitrectomy with face-down posture, reduce retinal ischemia, and enhance the visual function.

5.Effects of experiment-related factors on hematological parameters in SD rats

Objective To study the effects of experiment-related factors on hematological parameters in SD rats, analyze the data difference and causes, understand the effects of anesthetics and stress responses on the physiological aspects of animals, and to provide a reference for the standardization of animal welfare and compound toxicity testing methods.Methods According to gender (A), fasting time (B), anesthesia (C) and blood collection mode (D), SPF SD rats were divided into 24 groups.Blood samples were collected from each group.Then, red blood cell count, hemoglobin levels, white blood cell count and classification indicators were measured.Results The primary and secondary order of the factors affecting the white blood cell count was D > C > A > B, and the levels of white blood cell count of each factor were male rats > female rats, and venous blood > arterial blood, chloral hydrate > pentobarbital sodium > no anesthesia.The primary and secondary order of the factors affecting the white blood cell classification was C > D=A=B, and factors affecting the levels of white blood cell classification were chloral hydrate > pentobarbital sodium > no anesthesia.The primary and secondary order of the effects of the factors on the red blood cell count and hemoglobin level was C > D=A=B, and the levels of red blood cell count and hemoglobin level were pentobarbital sodium > chloral hydrate> no anesthesia.There was no significant difference in the blood indexes between the different fasting time groups.Conclusions There is no effect of fasting on hematological parameters, but there are differences in the blood parameters between arteries and veins.The effect of chloral hydrate anesthesia on the count and classification of white blood cells is greater than that of pentobarbital sodium.The effect of chloral hydrate anesthesia on the red blood cell count and hemoglobin level is greater than that of pentobarbital sodium.The two kinds of anesthesia methods have their own advantages and disadvantages.
6.Risk stratification and analysis of prognostic factors of acute myeloid leukemia with t(8;21)

Objective To investigate the cytogenetic and immunological phenotypes of acute myeloid leukemia (AML) with t(8;21),and explore the risk stratification and risk-adapted treatments.Methods The chromosomal karyotype of bone marrow was detected and analyzed in 22 newly diagnosed patients with t(8;21) AML by direct culture and G banding technique.Patients were divided into two groups according to the chromosomal karyotypes.Clinical characteristics and immunological phenotypes were compared between patients with isolated t(8;21) and those with additional aberrations.A follow-up study with median time 30 months (4-68 months) was conducted to analyze prognostic factors.Results 13 cases (59.1%) were isolated t(8;21) AML,while 9 (40.9 %) had additional aberrations.Loss of sex chromosome was found in 3 cases and complex variant translocation in 2.The 10q-,9q-,-18 and +10 were found in single cases.Overall survival of patients with additional aberrations was significantly poorer than those with isolated t (8;21) (P =0.0176).Analysis of prognostic factors showed that t(8;21) chromosomal karyotype,initial white blood cells at diagnosis,and treatment regimen (chemotherapy alone or plus hematopoietic stem cell transplantation) had effects on overall survival.Conclusion Patients with t (8;21) AML are frequently associated with additional chromosomal aberrations.The latter indicates a poorer outcome and can be one of the bases of risk stratification.Hematopoietic stem cell transplantation might help to improve the overall survival.
7.Pro-Apoptosis Effects of Danhong Injection on the SMMC-7721 Hepatoma Cells

Objective To investigate the effects of danhong injection on the apoptosis of SMMC-7721 heptoma cells.Methods The hepatocellular carcinoma cell line SMMC-7721 was incubated with different concentrations(0,2,4,8 μL·mL-1) of danhong injection, and cell morphology was studied under the microscope. The apoptosis index was detected by flow cytometry at 0,6,12,24 h after cultured with 2 μL·mL-1danhong injection. Cell proliferation was measured by MTT assay;Gene expressions of p53,Bax and Bcl-2 were detected by RT-PCR at different drug concentrations. Results The morphology of the hepatoma cells changed obviously,and the apoptosis index increased by danhong injection in a dose-dependant manner. The danhong injection decreased gene expressions of Bcl-2,and obviously increased p53 and Bax. Conclusion Danhong injection can dose-and time-dependently promote apoptosis of SMMC-7721 cells.

The specific fragment of Pneumococcal surface protein A(PspA)and Pneumococcal Surface Adhesin A(PsaA)gene was amplified by PCR from Streptococcus pneumonia 5 and Streptococcus pneumonia 19.The amplified fragnent of PspA and PsaA gene was ligated into pET-27b(+)vector and transformed into BL 21 E.coli for expression and obtain the expressive production of PspA and PsaA.Induced by IPTG,the expression level was as high as 75 % of the total disolube protein.The result showed that the recombinant plasmid could express a specific 75 kDa and 37 kDa fusion protein in E.coli BL 21,which showed the good immunogenicity and a broadly cross reactivity with the other serotypes.

Objective To investigate the effect of polymorphism of TNF-?gene G308A on sever- ity of myocardial damage during cardiopulmonary bypass(CPB).Methods Sixty-three congenital ca- ses with ventrieualr septal defect(VSD)were divided into groups TNF1 and TNF2 after TNF-?gene pol- ymorphism was identified by polymerase chain reaction-restriction fragment length polymorphism(PCR- RFLP)before surgery.When the right atrium was opened and closed,specimens of myocardium were ob- tained to study the ultrastructural changes by electron microscope and determine expression of TNF-?mR- NA.Concentration of TNF-?in plasma was measured by radio-immunity after anesthetic induction(T1), 20 minutes of CPB(T2),at the end of CPB(T3) and six hours after CPB(T4)in all cases,respective- ly.Results (1)TNF-?level and the expression of TNF-?mRNA in myocardium were significantly increased during CPB(P<0.05)in both groups.(2)TNF-?mRNA level of group TNF2 was significant- ly higher than that of group TNF1(P<0.05).The myocardial CK-MB in group TNF2 was significantly higher than that in group TNF1 at 24 hours postoperatively(P<0.01).The electron microscope showed more severe ultrastructural changes of myocardium in TNF2 group compared with that in group TNF1(P<0.05).(3)The concentration of TNF-?in blood plasma in group TNF2 was significantly higher than that in group TNF1 at time points of T2-T4(P<0.05).Conclusion Polymorphism of TNF-?gene G308A may influence the transcription and production of TNF-?in vivo and hence affect severity of myo- cardial damage.
