Objective To optimize and verify an in vitro detection method for bioactivity of recombinant human bone morphogenetic protein-2(rhBMP-2)based on ASTM F2131-02 standard test.Methods Based on the method established by the American Society for Testing Material(ASTM),the detection cell lines(W-20-17 and C2C12 cells),dilution mode of rhBMP-2(2 and 3 times serial dilution),culture time(24,36,48 and 72 h),and freezing temperature(-20 and-80 ℃)were optimized,and the specificity,intermediate precision,linearity and durability of the optimized method were verified. The specific activity of rhBMP-2 from different manufacturers was detected using the optimized method.Results C2C12 cells were determined as detection cells;The best dilution method of rhBMP-2 was 2 times series dilution;The optimum cell culture timewas 48handtheoptimum freezingtemperatureofcellswas-80℃. C2C12 cells responded specifically to rhBMP-2 induction when adding heteroprotein(bovine serum albumin)to rhBMP-2;The CV of 12 detections by 2 experimenters at 2 time points was11. 7%. The theoretical titer of rhBMP-2 showed a good linear relationship with the measured titer in the range of 70%-142%,and the fitting linear regression equation was:y = 0. 983 1 x-0. 010 3,R~2= 0. 996 4;The CV of the detected specific of rhBMP-2 using 4,11 and 21 passages of C2C12 cells was 5. 2%. The CV of specific activity of rhBMP-2 samples from different manufacturers detected by the optimized method repeatedly for two times was 1. 3%-7. 5%.Conclusion The optimized method showed good specificity,intermediate precision,linearity and durability,and might be used to detect the bioactivity of rhBMP-2 in vitro,which provided a reliable analytical method for its research,production and application.

Large number of studies showed that NO play an important role in the pathogenesis of septic shock. Some other investigators reported that NOS inhibitors could have the potential therapeutic value in septic shock treatment.

[Objective] Have a research on therapeutic methods of phlegm syndrome of Li Ziyi in Qing dynasty, to sum up his therapeutic experience of such syndrome.[Methods]Analysis on the related states and cases on the base of researches on the contents of “Approaches of phlegmatic scrofula ”which was written by Mr. Li, elaborating his characteristics in treating such syndrome. [Results] Mr. Li considered that phlegm syndrome can be classified into milder phlegm and scrofula according to its level of the patient ’s condition and therapeutic effect. He also believed that several aspects should be taken into consideration through the therapeutic process:treat by internal combing with external treatment ,pay attention to seven emotions and smoothen the temper, emphasize nursing care and obey to the contraindication strictly. [Conclusion]There are features in Mr. Li ’s view on phlegm and his treating method of phlegm syndrome. His treating method has a good therapeutic effect and it is worth popularizing.

Intraperitoneal. injection of the essential oils of Asarum himalaicum and A. heterotropides var. mand shuricum can inhibit the proliferation of granuloma induced by implantation of cotton pellets in rats, and change the distribution of Copper and Zinc,the two necessary trace elements for rats,in rat serum and liver.They reduce concentration of Zinc but increase that of Copper in the serum,while elevate the concentration of Zinc but reduce that of Copper in liver. The oil of A. himalaicum collected from Ningxia Region showed even stronger effects.

At present, selective 5-hydroxytryptamine (5-HT) reuptake inhibitors (SSRIs) and 5-HT and noradrenaline (NA) re-uptake inhibitors ( SNaRI) are the first choice for the clinical treatment of depression, however, the clinical adverse effects of the drugs have become a prominent problem affecting the clinical efficacy. Chinese medicines have some experience in the treatment of depres-sion, and the combined use with the above antidepressants can enhance the efficacy and / or reduce the adverse effects. The article re-viewed the status of the combination of SSRIs and Chinese medicines in order to provide reference for the clinical treatment of depres-sion.

Objective To investigate the utilization of liver enhancement in hepatobiliary phase of gadolinium ethoxybenzyl diethylenetriamine pentaacetic acid (Gd-EOB-DTPA)-enhanced MRI for liver function evaluation.Methods Fifty-five patients who received enhanced MRI with Gd-EOB-DTPA were retrospectively analyzed.Images were obtained before injection and in hepatobiliary phase (5,10,and 20 minutes after Gd-EOB-DTPA injection).The patients were assigned into two subgroups according to individual liver function (n =35 in Group 1:normal liver and Child-Pugh class A; n =20 in Group 2:Child-Pugh class B and C).The relative liver enhancement (RE) was calculated at different time point.The general data (age,sex) and relevant laboratory results were recorded.Independent sample t-test was conducted to compare the RE between two groups at different time point.ROC curve was used to determine the best time point and RE threshold that can reflect the differences between two groups.Univariate analyses was performed to analyze the relationship between RE at the best time point and laboratory results.Multivariate analyses was performed to screen the independent influencing factor for RE at the best time point.Results The differences of RE between two groups were statistically significant at all time points (P ＜ 0.0001).10 minutes was the best time point for detecting the differences of liver function between two groups.When an RE cutoff value (＞ 1.52) was applied,normal or Child-Pugh class A could be predicted with sensitivity of 74.3％ and specificity of 90％.RE at the best time point was significantly related with total serum bilirubin level (TBil),serum albumin level (Alb) and prothrombin time (PT).And TBil was an independent influencing factor.Conclusion RE can be used to evaluate the liver function,and 10 minutes is the best time point that can be used to differentiate patients with normal or mild liver damage from those with moderate or severe liver damage.

[Objective]To discuss the diagnosis and surgical treatment of spinal myeloma.[Method]To make a retrospective analysis of cliniacal history and operation results of 13 patients with spinal myeloma from 1999 to 2004.There were 5 males and 8 females with average age of 47.67 years.They were diagnosed by X-ray,CT,MRI,ECT and biopsy.Resection of tumor was performed in all cases,of which 7 cases was through anterior approach,4 cases through posterior approach,2 cases through combined approach.[Result]The pain was decreased and neurological function was improved in all cases after the surgery.Three death and one recurrence was found during the follow-up time.[Conclusion]X-ray,CT and MRI are essential for diagnosis,ECT may help found multiple lesions,biopsy is reliable diagnosable method.Surgery can relived the symptoms obviously,but should be associated with adjuvant treatments.

[Objective]To approach the clinical characteristic,diagnosis and method of treatment of malignant fibrous histiocytoma of bone.[Method]The clinical data of 52 patients of malignant fibrous histiocytoma of bone were analyzed retrospectively,including sites of tumor,preoperative imageology,puncturing biopsy,surgical staging of tumor,methods of treatment,results of follow-up,and so on.[Result]In all 52 cases of malignant fibrous bistiocytoma of bone,32 cases were in periphery of knee joint,14 cases in periphery of shoulder joint,3 cases in the proximal femur,1 case in calcaneus,1 case in lumbar and 1 case in ilium.The preoperative imageelogy and puncturing biopsy were made as usual.There were 20 cases in staging of ⅡA and 32 cases in in staging of ⅡB according to Ennekings surgical staging of tumor.All cases were performed operation except of 1 case.The operation were made salvage limb mosfly and the ratio of limb salvage was 74.5 %(38/51).The preoperative and postoperative chemotherapy and radiotherapy were performed at the same time.Thirty-six of all cases were in long-time follow-up and the ratio of follow-up was 69.2%(36/52).Nine cases were recurred in the region,6 cases had distant metastasis and survival rate of 5 years was 61.1%(22/36).[Conclusion]Preoperative puncturing biopsy and imageology can help the diagnosis of malignant fibrous histiocytoma of bone.Surgery is the primary method and radiotherapy and chemotherapy can prevent the recurrence of the tumor.The resection of the tumor thorough is the key point of healing of malignant fibrous histiocytoma of bone.
