1.A study on the mechanisms of apoptosis of gastric cancer line induced by Nimesulide

Objective Non steroidal anti inflammatory drugs(NSAIDs)can induce apoptosis in gastric cancer cell and the mechanism is not clear. We aimed to study the mechanism of selective COX 2 inhibitor Nimesulide induced apoptosis of human gastric cancer line SGC 7901 by detecting the expressions of COX 2 at mRNA level, c myc, Bcl 2 and caspase 3 at protein level. Methods Apoptosis was determined by electronic microscopy, Annexin V FITC staining and flow cytometry. The mRNA of COX 2 was detected by RT PCR. The protein expressions of c myc, Bcl 2 and caspase 3 were examined by immunohistochemistry. Results Nimesulide of 50 ?mol/L at 48 and 72 h, and of 100 ?mol/L and 200 ?mol/L at 24, 48 and 72 h induced apoptosis of gastric cancer cells in a dose and time dependent manner.Their apoptotic rates were 7.51%, 9.86% and 11.58%, 12.45%, 16.66% and 12.21%, 15.38%, 20.28% respectively. It increased c myc and caspase 3 expression and decreased Bcl 2 expression and COX 2 mRNA expression. The positive protein expression rates of Bcl 2, c myc and capase 3 were (20.2?7.6)%,(49.2?15.1)% and (34.6?12.9)% respectively with Nimesulide of 200 ?mol/L at 72 h,while the controls being (44.6?12.1)%, (24.7 ?9.5)% and (14.8?6.4)% the three comparative P

3.Diagnosis and treatment of secondary methylmalonic aciduria due to maternal vitamin B_(12) deficiency

Objective To investigate the causes, diagnosis and treatment of infants with secondary methylmalonic aciduria due to maternal vitamin B 12 deficiency. Methods The clinical, laboratory data and treatment of 7 children with secondary methylmalonic aciduria and their mothers′ health and nutrition were retrospectively reviewed. Results All children were presented to the hospital with vomiting, seizures or mental retardation during 1 to 13 months after birth. Varied degrees of anemia was found in 6 infants and 5 with metabolic acidosis and liver dysfunction. Methylmalonic aciduria was found in all subjects. Four of the mothers had chronic gastritis, one with gallstones in liver and gallbladder and one was a vegetarian. Decreased serum level of vitamin B 12 and folic acid and elevated homocystine concentrations were found in all mothers. After vitamin B 12 and folic acid supplementation, significant improvement was observed in all patients. Six infants showed normal development and one had mild mental retardation. Conclusions Chronic gastritis, liver/gall bladder diseases or being a vegetarian might lead to maternal vitamin B 12 deficiency and then infantile secondary methylmalonic aciduria. Early diagnosis and vitamin B 12 and folic acid supplement are crucial in improving the prognosis.
4.The effect of mifepristone on natural killer subsets of peripheral blood and decidua in early pregnancy

AIM: To investigate the effect of mifepristone on natural killer(NK) subpopulations of peripheral blood and decidua in early pregnancy. METHODS: Flow cytometry was used to detect the expression of CD56 and CD16 on lymphocytes of decidua and peripheral blood in early pregnancy, mifepristone-treated pregnancy. RESULTS: The percentages of different natural killer subsets in peripheral blood between early pregnancy and mifepristone-treated pregnancy were almost identical. The serum levels of estradiol and progesterone in mifepristone-treated pregnancy were slightly higher than in early pregnancy. The percentage of decidual CD56+NK cell in early pregnancy was significantly higher than in mifepristone-treated pregnancy. The CD56+NK cells were predominant lymphocyte population of decidua in mifepristone-treated pregnancy, but in which the CD56+CD16+ and CD16+NK cells were major lymphocyte subpopulations of peripheral blood. CONCLUSION: Mifepristone acted principally on feto-maternal interface, it blocked the proliferation and differentiation of decidual CD56+NK cells and induced embryo immune rejection.
5.Percentage change on natural killer subsets of peripheral blood and decidua in between normal early pregnancy and spontaneous abortion

AIM: To determinate the percentage change on natural killer(NK) subsets of peripheral blood and decidua in between normal early pregnancy and spontaneous abortion. METHODS: Flow cytometry was used to detect the expression of CD56 and CD16 on lymphocytes of decidua and peripheral blood in normal early pregnancy and spontaneous abortion. RESULTS: In peripheral blood, the percentage of CD56 + of spontaneous abortion tended to decrease in comparison with normal early pregnancy, and the percentage of CD56 +CD16 + in spontaneous abortion was significantly less than that in normal early pregnancy. The percentages of different natural killer subsets in decidua of spontaneous abortion were significantly lower than those in decidua of normal early pregnancy. CONCLUSION: The decrease of CD56 +NK cells in decidua may be one of causes for abortion, the loss of CD56 + and CD56 +CD16 +NK cells in peripheral blood could have a diagnostic value for spontaneous abortion.
6.Effect of PEG-IFN alpha -2a combined with ribavirin in type 1b chronic hepatitis C virus

Objective To investigate the effect of peginterferon alfa-2a injection (PEG-IFNα-2a) combined with ribavirin (RBV) in the treatment of chronic hepatitis C virus (1b) in patients with chronic hepatitis C. Methods The clinical data of patients with type 1b chronic hepatitis C in our hospital from May 2010 to October 2015 were retrospectively analyzed. According to the method of treatment, the patients were divided into IFNα-2a combined with RBV group and PEG-IFNα-2a combined with RBV group. The rapid virologic response (RVR), early virologic response (EVR), and sustained virologic response (SVR) rate were observed in the two groups. The liver function, expressions of CD4+T and CD8+T cell of peripheral blood and the incidence of adverse reaction were compared between the two groups before and after treatment. Results The RVR, EVR, SVR of two groups was no significant difference (χ2=0.641, 0.946, 0.154, P=0.423, 0.331, 0.694). After treatment, the levels of ALT , AST, DBIL and TBIL in PEG-IFNα-2a combined with RBV group were lower than those in IFNα-2a combined with RBV group (P<0.05). Of the PEG-IFNα-2a combined with RBV group, the CD4+T level was lower and the CD8+T level was higher than that in PEG-IFNα-2a in the combined Leigh Bhave Lin group were lower than that in the IFNα-2a level was lower IFNα-2a combined with RBV group(P<0.05). There was no significant difference in the incidence of influenza-like symptoms, marrow suppression, somnolence and abnormal laboratory indexes between two groups. Conclusion PEG-IFN alpha -2a combined with RBV has a good therapeutic effect on type 1b chronic hepatitis C virus, and has good safety and clinical application value.
7.Influence of BCG-PSN on Th3,Th2 and Th1 cells numbers in PBMC and airway inflammation status in patients with asthma

Objective To explore the effect of BCG-polysaccharide nucleic acid(BCG-PSN) on the numbers of Th3,Th2 and Th1 cells in peripheral blood mononuclear cell(PBMC) and airway inflammation status in patients with asthma.Methods Twenty-six patients with moderate persistent asthma were enrolled into the study and randomly divided into simple medication group(n=13,accepted Fluticasone/salmeterol alone) and combined medication group(n=13,accepted Fluticasone/salmeterol+BCG-PSN together).The numbers of Th3,Th1 and Th2 cells in PBMC, the serum levels of TGF-β,IFN-γ,IL-4 and IgE and the clinical effect and airway inflammation status were observed at three time points: before,1.5 months after and 3 months after treatment. Results (1)At 1.5 months after treatment,the Th1/Th2 number was significantly higher in the combined medication group than the simple medication group(1.20±0.57 vs 0.79±0.39,t= 2.129,P<0.05),and the asthma control test scale(ACT) showed similar difference(18.31±1.75 vs.15.54±2.40,t=3.359,P<0.01) between the two groups.(2)At 3 months after treatment,the Th1/Th2 number was significantly higher in the combined medication group than the simple medication group(1.73±0.74 vs1.16±0.48,t=2.327,P<0.05),and the ACT also showed the same kind of difference(22.46±1.13 vs.20.23±2.59,t=2.851,P<0.01) between the two groups.In addition,at this time point the PEF variability was significant lower in the combined medication group than the simple medication group([9.88±2.18]% vs.[12.05±2.74]%,t=2.235,P<0.05).(3)We found no significant differences in the comparisons of the numbers of T helper cell subsets in PBMC ,the serum levels of cytokines and IgE,the numbers of inflammatory cells in induced sputum between the two groups(Ps>0.05).(4)No correlation were found between Th3 cell numbers in PBMC and the serum levels of IgE in the two groups.Conclusion The combined use of Fluticasone/salmeterol and BCG-PSN can further correct the imbalance of Th1/Th2 cells,alleviate clinical symptom of asthma and airway hyper-responsiveness.The therapeutic efficacy improves along with the therapy period,better in the combined medication group than the simple medication group.BCG-PSN has no significant effects on the numbers of Th3 in PBMC in patients with asthma.
10.Effects of Litsea cubeba oil on gene expression profile of Candida albicans

Objective To analyze the effects of Litsea cubeba oil on gene expression profile of Candida albicans by comparing the differential gene expression profile after exposure to Litsea cubeba oil using genome-wide gene expression array.Methods Candida albicans ATCC90028 was exposed to Litsea cubeba oil for 90 min.Then RNA was isolated and gene expression profiles were compared to identify the differential gene expression profile using cDNA microarray analysis.Results A total of 491 geneswerefoundtoberesponsivetoLitseacubebaoil,accountingforabout11% ofthetotalnumberofgenesinCandida albicans (491/4 634),of which 216 genes were up-regulated and 275 down-regulated.These differentially expressed genes included genes encoding the key target enzyme in ergosterol biosynthesis pathway,genes in stress response,DNA replication and repair,molecular transport,and energy metabolism.Conclusions Litsea cubeba oil has significant effect on the expression of about 1 1% genes of Candida albicans genome.We presume that the genes encoding the key target enzyme in ergosterol biosynthesis pathway may contribute to the action of Litseacubeba oil on Candidaalbicans,which is similar to azole antifungal drugs.However,the role of other differentially expressed genes in the action of Litseacubeba oil on Candidaalbicans remains unclear,which deserves further study to characterize their potential association with the antifungal effect of Litsea cubeba oil.
