1.miR-15a-5p inhibits proliferation and migration of human hepatocellular carcinoma SMMC-7721 cells

AIM:To observe the influence of high expression of miR-15a-5p on the proliferation and migration of human hepatocellular carcinoma SMMC-7721 cells.METHODS: The miR-15a-5p oligonucleotide , which was recon-structed with additional restriction sites of EcoRⅠand HindⅢ, was chemically synthesized and confirmed by sequencing . The miR-15a-5p eukaryotic expression system was constructed by pcDNA 6.2-GW/Em-GFP-pre-miR-15a-5p plasmid.The miR-15a-5p was transfected into the SMMC-7721 cells transiently by plasmid , and quantified by quantitative real-time PCR at the mRNA level.The cell viability was measured by CCK-8 assay, and the living cell counting was performed by the method of Trypan blue exclusion .The migration ability of the SMMC-7721 cells with high expression of miR-15a-5p was de-tected by wound healing test .RESULTS: The sequence of miR-15a-5p oligonucleotide 100% matched the designed se-quence.Compared with control group , the miR-15a-5p expression was increased significantly (P<0.05).The viability, the living cell number and the migration ability of the SMMC-7721 cells were decreased in high expression of miR-15a-5p group with statistically significant difference (P<0.05).CONCLUSION: The abilities of proliferation and migration in human hepatocellular carcinoma SMMC-7721 cells are decreased by high expression of miR-15a-5p.
2.Antiviral effect of Fufang Yemazhui Capsule in vitro and in vivo

AIM: To study the antiviral effect of Fufang Yemazhui Capsule(Eupatorium lindleyanum DC) in vitro and in vivo. METHODS: The influences on life span,death rate and lung index and lung histopathology of mice infected by influenza virus were observed. RESULTS: Fufang Yemazhui Capsule obviously inhibited influenza virus in vitro.Fufang Yemazhui Capsule could remarkably prolong life span,decrease lung index and inhibite the lung consolidation. CONCLUSION: Fufang Yemazhui Capsule shows significantly antiviral effect both in vitro and in vivo.
3.Experimental study on TCM symptomatological and developmental characteristics of rats with radiation-induced lung injury

Objectives:To explore the possibility of using experimental animal models as a new means for studying the pathogenesis and symptomatologic development of radiation-induced lung injury,and provide theoretical and experimental evidences for early prevention and treatment with the traditional Chinese medicine.Methods:80 female Wistar rats were divided randomly into the model group and control group with 40 rats in each group respectively.The rats in model group were anesthetized and fixed,and a 2cm?3cm area of the right lungs received 6mV X-ray radiation at 3Gy?2/w and a maximal 10/5w.Eight rats randomly selected from each group were sacrificed at the end of weeks of 3,5,8,12 and 26.Diagnostic techniques of TCM were employed to analyze symptomatological and developmental characteristics of TCM in radiation-induced lung injury during exposure by dynamic examination and comparison of living rats,gross lung samples and tissue sections.Results: Signs and symptoms including redness around nose and eyes,yellowish secretion,tachypnea,dry stool,lethargy,sluggishness, inactiveness were observed in model group in early stage of radiation exposure,which consistent with dominant heat in the interior and primordial qi exhaustion syndrome.Swollen lungs,bright redness on surface and red bleeding patches were observed in early stage of radiation;microscopic examination showed interstitial tissues,and readily visible congestion,hemorrhage and edema of pulmonary interstitial tissues,consistent with damaged blood collaterals by heat and blood escaping circulation syndrome.In the later stage of radiation exposure,the lungs showed dark appearance,apparent purple petechia,hard texture and poor elasticity.Microscopic examination showed remote hemorrhage foci,pulmonary capillary stasis or closure,and focal fibrotic clumps,consistent with obstruction of collaterals by blood stasis syndrome.No abnormal changes were observed in the control group.Conclusions:Animal experiments can redeem the clinical inadequacy of traditional inspection,auscultation and olfaction,interrogation and feeling pulse and palpation diagnostic techniques,modern pathological methods can be utilized to study symptomatology of traditional Chinese medicine.Symptomatological and developmental characteristics of'heat toxin impairment in radiation-induced lung injury characterized by heat damaging pulmonary collateral syndrome in early stage and pulmonary collateral stagnation syndrome in the advanced stage'can be revealed at living organism,gross lung sample and microscopic pulmonary tissue levels,thus providing theoretical basis for early prevention and treatment of radiation-induced lung injury using'cooling blood to remove apthogenic heat'.

Aim To investigate the lipid-lowering mechanism of Chinese Medicine curcumin.A sterol regulatory report system of LDLR gene expression in HEK-293 cell line was established.Method A green fluorescent protein(GFP) gene was constructed downstream of the sterol regulatory element-1(SRE-1) and the constructed plasmid was introduced into HEK-293 cell line.This SRE-GFP transfected 293 cells were treated with different concentration of curcumin for 2 days.The expression of GFP in the cells was measured by a fluorescence microscope and Flow Cytometry.Results The expression of GFP in SRE-GFP transfected 293 cells was obviously induced by curcumin.Conclusion Curcumin may positively affect the expression of LDLR gene by its influence on SRE-1.
5.Study of proximal femoral locking plate internal fixation after dynamic hip fixation intertrochanteric fracture

Objective To study the mechanics situation of proximal femoral locking plate internal fixation after dynamic hip fixation in-tertrochanteric fracture. Methods Totally 10 couple of elderly proximal femur specimens were collected and intertrochanteric fracture model were prepared. Fixation material was removed after dynamic hip screw fixation. The left sides were collected as control group and given anti-rotation intramedullary nail internal fixation, while the right side were collected as observation group and given proximal femoral locking plate internal fixation. Then vertical displacement, axial stiffness and rotational stiffness under different loads were compared. Results Under dif-ferent loads, femoral bone vertical displacement and femur tuberosity vertical displacement in the observation group were both significantly shorter than those in the control group (P<0. 05), and femoral bone and femur tuberosity axial stiffness and rotational stiffness in the observation group were significantly higher than those of the control group (P<0. 05). Conclusion Proximal femoral locking plate internal fixation can improve stress load and enhance axial stiffness and rotational stiffness, and it's an ideal material for refracture fixation model after dynamic hip fixation intertrochanteric fracture.
6.Content Determination of Aciclovir Cream by Centrifugal Partition Chromatography

OBJECTIVE:To establish a method for the content determination of Aciclovir cream. METHODS:Centrifugal parti-tion chromatography(CPC)was conducted. The solvent system was hexane-acetonitrile-water(2∶1∶1,V/V/V),the injection vehicle was an aqueous solution of 5%Tween 80 and the volume was 1.0 ml;the flow rate was 5 ml/min;the wavelength was 254 nm. RE-SULTS:There was a good linear relationship between quality concentration and peak area in the range of 0.012 7-0.126 7 mg/ml (r=0.998 7). The RSD of precision,stability and reproducibility tests was all no more than 2.0% and the average recovery was 97.34%(RSD=0.90%,n=9). CONCLUSIONS:The method is with high precision and accuracy,and can be used for the content determination of principal components of Aciclovir cream.
7.Effects of soft contact lenses on corneal diseases

Objective To evaluate the therapeutic efficacy of soft contact lenses on corneal diseases. Methods A total of 70 cases (82 eyes) of corneal diseases in the treatment group were treated with therapeutic soft contact lenses combined with topical antibiotic eye drops and artificial tears, but 64 cases (81 eyes) suffering from corneal diseases in the control group were treated with eye drops and eye ointments. ? 2 test was used to compare the cure rate of corneal epithelial defects and corneal ulcers caused by chemical and thermal burns between the two groups. Double sample t test was used to compare the average treatment time between the two groups. Results The cure rates of the corneal epithelial defects and corneal ulcers caused by chemical injury and thermal burn in the treatment group and the control group were 83% and 57%, respectively. In the treatment group, 100% patients with bullous keratopathy and 90% patients with dry eye disease were free from symptoms after wearing the lenses, but in the control group, the percentages were 50% and 63.6%, respectively. In the treatment group, 3 cases (4 eyes) of corneal epithelial defects caused by explosive injury and 3 cases of corneal epithelial defects after lameller keratoplasty were all cured, and 4 cases(4 eyes) of corneal perforations were healed. In the control group, 1 out of the 2 cases of corneal epithelial defects caused by explosive injury was cured, and 4 cases of corneal epithelial defects after lamellar keratoplasty surgery were cured, but 2 cases (2 eyes) of corneal perforation failed in the therapy, and corneal surgery was performed. 8.Patterns of failure in head-and-neck cancer of unknown primary:a study of 92 patientshead-and-neck cancer of unknown primary

Objective To retrospectively analyze the treatment outcomes and failure patterns in patients with head?and?neck cancer of unknown primary head?and?neck cancer of unknown primary, and to compare the efficacy between elective mucosal irradiation and ipsilateral neck treatment. Methods The clinical data of patients with head?and?neck cancer of unknown primary who were admitted from January 2007 to December 2013 were retrospectively collected. Thirty?one patients received elective pharyngeal mucosal irradiation and 61 patients only received ipsilateral neck treatment. The SPSS 19. 0 software was used for comparison of the survival and local control between the two groups. Results In the 92 patients, the median age was 57 years;79. 3% had metastasis to level Ⅱ lymph nodes;the median follow?up time was 36. 5 months;the 3?year overall survival, mucosal control, and neck control rates were 89. 0%, 86. 6%, and 82. 4%, respectively. Primary sites were found in 15 patients, containing nasopharynx in 4 patients, oropharynx in 3 patients, oral cavity in 3 patients, throat and hypopharyngeal part in 3 patients, maxillary sinus in 1 patient, and esophagus in 1 patient. The patients undergoing elective pharyngeal mucosal irradiation had significantly higher 3?year mucosal control and neck control rates than those undergoing ipsilateral neck treatment ( 100% vs. 74. 9%, P= 0. 040;87. 5% vs. 62. 2%, P= 0. 037 ) . There was no difference in the 3?year overall survival rate between the two groups ( 83. 5% vs. 88. 7%, P= 0. 910 ) . Conclusions For patients with head?and?neck cancer of unknown primary, elective pharyngeal mucosal irradiation can reduce the incidence of primary site and increase the neck control rate. A new standard for target volume delineation should be established as soon as possible for elective prophylactic pharyngeal mucosal irradiation.
9.Seroepidemiology of Mycoplasma pneumoniae infection and evaluation of antibiotics medication

ObjectiveTo analyze the seroepidemiologic of Mycoplasma pneumoniae infection and evaluate antibiotics medication of some positive patients by follow-up. Methods Serodia-MycolⅡ particle agglutination assay was used to detect serum antibodies against Mycoplasma pneumoniae in 3 134 clinically suspected infections. Mycoplasma pneumoniae infection was determined and seroepidemiologic was analyzed by results of the test, including positive antibody rates in whole subjects, in male or female groups, in different seasons or age groups as well as in different sources. Evaluate antibiotics medication of some positive patients by follow-up. The average days of medication were counted, different antibiotics medication and medication effect were analyzed. Results In 3 134 serum samples from clinically suspected Mycoplasma pneumoniae infections, 350 ( 11.2% ) were tested with positive antibodies. The positive antibody rate in female patients was 12. 3% ( 198/1 604), which was higher than 9. 9% ( 152/1 530) in males (X2 =4. 58,P <0. 05). The peak season was found in the fourth quarter (October-December) with 13.2% of positive antibody and the highest positive rate (32. 8%, 45/137 ) was found in school aged (5 -9 years old )children. Samples from pediatrics clinic and ward were tested to have highest positive rates ( 27. 9% and 26. 5%, respectively ), comparing that from other sources. Infection due to Mycoplasma pneumoniae was identified in 28% (7/25) of community-acquired pneumonia (CAP) patients, which is higher than other diseases. Based on the follow-up of 91 antibody positive patients, between 5 to 120 days ( mean 24. 2 days )were counted from appearance of clinical symptoms to clinic visiting/testing. 71 of 91 (78. 0% ) patients was medicated with macrolide antibiotics, 4 (4. 4% ) with quinolones, 4 (4. 4% ) with cephalosporin, and the rest 12 ( 13.2% ) patients were medicated with other antibiotics or only symptomatic treatment. The average period of antibiotics medication was between 3 to 21 days (mean 8. 2 days). Medication effect results by follow-up were cure in 35 ( 38. 5% ), improvement in 50 (54. 9% ), and poor responses in 6 (6. 6% ).ConclusionsMycoplasma pneumoniae positive rate in female patients was higher than in males, and peak rate was found in the fourth quarter and in school aged children. Samples from pediatrics clinic and ward were tested to have highest positive rates. Physicians could choose first line antibiotics according to laboratory test results of Mycoplasma pneumoniae, and gain good effect.
10.Establishment, evaluation, and determination of saliva glucose concentration by ion chromatography

Objective:To establish an analytical method for measuring the concentration of glucose in saliva by ion chromatography.Methods:The proteins in saliva were removed by thermal denaturation method, CarboPac PA20 (3×30 mm) was used as a protective column and CarboPac PA20 (3×150 mm) was used as an analytical column for ion chromatography analysis. Gradient elution was carried out with A: ultra-pure water, B: 250 mmol/L NaOH solution and C: 500 mmol/L NaAc solution. Pulsed ampere detector was used for detection.Results:This method had a good linear relationship in the range of 0.04 to 0.12 mg/L, with a linear relation coefficient of 0.9967. The detection limit of glucose was 2 μg/L, the mean value of the relative standard deviation (RSD) of the repeatability measurement was 0.75%, and the average spike recovery was 103.07%.Conclusion:This method is simple, sensitive, accurate and stable, and can be used for the determination of glucose concentration in saliva.
