Objective To explore the reasons of the failure of primary arteriovenous fistula (AVF)surgery and the preventive measures in patients with end-stage renal disease. Methods A total of 819patients with end-stage renal disease who accepted primary AVF surgery were selected. The data of hemoglobin (Hb),albumin (ALB),cholesterol (CHOL),parathyroid hormone (PTH),fasting blood glucose and blood pressure were collected before surgery, while the diameters of radial artery and cephalic vein were measured by upper extremity vascular ultrasound. Results Seven hundred and forty-two patients with AVF surgery were successful, and 77 patients were failed. The age,the ratio of diabetes, hypertension and blood pressure < 120/70 mm Hg (1 mm Hg =0.133 kPa) in AVF failed patients was higher than that in AVF successful patients[(61.3 ± 13.4) years vs.(45.6 ± 11.2) years,46.8%(36/77) vs.31.7% (235/742),26.0% (20/77) vs.19.5% (145/742),36.4% (28/77) vs.9.2% (68/742)],and there was significant difference (P<0.05 or <0.01).The diameters of radial artery and cephalic vein in AVF failed patients was lower than that in AVF successful patients [(1.35 ± 0.64) mm vs.(1.98 ± 0.47) mm,(2.13 ± 0.81)mm vs.(2.47 ± 0.74) mm],and there was significant difference (P< 0.01).The level of PTH in AVF failed patients was higher than that in AVF successful patients [(782.39 ± 423.85) ng/L vs.(378.83 ± 352.21) ng/L],and there was significant difference (P < 0.05).Logistic regression analysis showed that blood pressure, diabetes,PTH,the diameters of radial artery and cephalic vein was the risk factor of AVF failed. Conclusion AVF surgery failed is highly correlated with the patient s blood pressure,th primary disease, the vessel diameter,
3.The effects and mechanisms of Gankyrin silencing on reversing the cisplatin resistance of human ovarian cancer SKOV3/DDP cell line
China Oncology 2014;(1):35-40
Background and purpose: Ovarian cancer is the common gynecological cancer, and the drug resistance of anti-tumor drug was one of major reasons for therapy failure, some studies considered that there is a closed relationship between Gankyrin and drug resistance. In this study, we investigated the effects and mechanisms of Gankyrin silencing on reversing the cisplatin resistance of ovarian cancer drug-resistant SKOV3/DDP cell line. Methods:The expression of Gankyrin in SKOV3 and SKOV3/DDP cells was measured by real-time PCR assay, MTS assay was employed to determine the effect of Gankyrin on SKOV3/DDP sensitivity to cisplatin, apoptosis rate and intracellular concentration of rhodamine-123 (Rh-123) were determined by lfow cytometry, the expression of multi-drugs resistant protein MDR1, Caspase-3/8, Survivin and Bcl-2 were determined by Western blot and real-time PCR. The phosphorylation of AKT and expression of p53, NF-κB and PTEN were analyzed by Western blot assay. Results:The expression of Gankyrin was increased in SKOV3/DDP cells, Gankyrin silencing was able to increase the cisplatin sensitivity of SKOV3/DDP. Before and after gene silencing, the reverse folds (RF) to cisplatin were 1.81 and 2.45, respectively, the intracellular levels of Rh-123 were 1.73 and 2.42 fold, the apoptosis rates were 2.23 and 4.23 fold, the expressions of MDR1, Survivin and Bcl-2 were downregulated, the mRNA expressions of MDR1 were 62.8%and 21.6%, the mRNA expressions of Survivin were 24.5%and 10.3%, the mRNA expressions of Bcl-2 were 47.5%and 18.4%, the levels of Caspase-3/8, p53 and PTEN were elevated, phosphorylation of AKT and expression of NF-kB were downregulated compared with control group. Conclusion:Gankyrin silencing was able to reverse the cisplatin resistance of SKOV3/DDP cells by inhibiting the drug eflfux and promoting cell apoptosis, the PTEN/AKT/NF-κB/p53 may be the key pathway.

Chinese Journal of Nuclear Medicine 2011;31(4):237-240
Objective To evaluate 18F-FDG PET/CT imaging in primary adrenal lymphoma (PAL). Methods Three male patients with clinically diagnosed PAL, aged 59 to 77 years old, were examined by18F-FDG PET/CT before histopathologic examination. The SUVmax ratio of tumor to liver (T/L)was calculated on 18F-FDG PET images. All patients underwent R-CHOP ( rituximab-cyclophosphamide,doxorubicin, vincristine, and prednisone) chemotherapy regimens. One patient had serial follow-up 18F-FDG PET/CT post therapy. Results The final diagnosis of all 3 cases was bilateral diffuse large B-cell lymphoma. The pathologic results of bone marrow biopsy were negative. Case 1 was in disease remission based on four times of follow-up 18F-FDG PET/CT. The patient's clinical follow-up was also negative and he was currently alive for more than 19 months. Case 2 was of late stage and suffered from adrenal insufficiency. He died 6 months after the diagnosis. Case 3 (the eldest patient) was treated with partial excision followed by the chemotherapy. He died of lung infection 12 months after treatment. Conclusion 18 F-FDG PET/CT may be of important value for maximizing patient management and survival for patients with PAL, a rare and highly malignant disease.

Chinese Journal of Biotechnology 2014;30(10):1515-1521
When mature adipocytes are subjected to an in vitro dedifferentiation strategy referred to as ceiling culture, these mature adipocytes can revert to dedifferentiated fat (DFAT) cells. DFAT cells have many advantages compared with adipose-derived stem cells (ASCs) and bone marrow mesenchymal stem cells (BMSCs). For example, DFAT cells are homogeneous and could be obtained from donors regardless of their age. Furthermore, DFAT cells also have the same multi-lineage potentials and low immunogenicity as ASCs. As an excellent source of seed cells for tissue engineering and stem cell transplantation, DFAT cells have better prospects in the treatment of many clinical diseases, such as bone defects, neurological diseases, ischemic heart disease and kidney disease. It is necessary to make more intensive studies of DFAT cells. This article summarizes progresses in the immunological characteristics, differentiation ability and potential clinical applications of DFAT cells.
