1.MEASUREMENT OF LACTOFERRIN IN HUMAN MILK AND NEONATAL FAECES AND EXPLORATION OF RELATION BETWEEN THEM

The concentrations of lactoferrin (LF) in colostrum during the first 8 days and mature milk during the first 4 months of postpartum were determined by electroimmunodiffusion. Meanwile a sensitive enzyme linked imm-unosorbent assay for measurements of LF in neonatal faeces were established. The results showed that human milk, especially colostrum, was rich in LF. The colostrum contained 6.88 ? 3.10mg/ml (M?SD) aad mature milk 3.18 ? 0.37mg/ml LF. LF in human milk was the highest during the first 3 days of lactation with a decline in concentration as lactation proceeds although a wide variation in concentration was noted among different individuals.LF in neonatal faeces showed significant difference in different feeding manners. LF level in the breast-fed infants was 19 times higher than that in the bottle-fed infants. The LF percentage detected in the former was 100% and the latter only 60%. The dynamic variations of LF in faeces of breast-fed infants during the first day of life were found to be relative to LF level in mother'milk.These data demonstrated the influence of high LF level in mother'milk on the concentrations of LF in breast-fed infants' intestine.Furthermore a few amounts of LF was found in meconium and its percentage detected was 35%. This suggested that the production of endogenous LF began from foetus period, but was limited during the first day of life. LF ingested by the breast-fed infants were not completely destroyed and thus may supplement the insufficiency of the endogenous LF in the newborn infants' intestinal tract.
2.RAM-HPLC vs.FPIA for Determination of Plasma Concentration of Carbamazepine

OBJECTIVE:To develop a RAM(Restricted-access media)-HPLC for the determination of plasma concentration of carbamazepine,and to compare the determination results with those determined by FPIA method. METHODS: The plasma samples were centrifugalized,then the venous plasma samples were determined by FPIA and RAM-HPLC,respectively,while the fingertip plasma sample was determined by RAM-HPLC alone. The chromatographic column was Hisep shielded hydrophobic phase analytical column,and the mobile phase consisted of ammonium acetate buffer-acetonitrile (88∶12) at a flow rate of 1.5 mL?min-1. The detective wavelength was set at 280 nm;the sample size was 20 ?L and the column temperature was kept at room temperature. RESULTS: A good correlation was noted between the carbamazepine concentration in venous plasma sample determined by FPIA and carbamazepine concentrations in either venous or fingertip plasma determined by RAM-HPLC,with the correlation coefficients being 0.989 and 0.995,respectively,but the differences were significant(P0.05). CONCLUSION: Both RAM-HPLC and FPIA are applicable for the determination of carbamazepine concentration,but FPIA is more suitable for routine monitoring while RAM-HPLC is more suitable for the related research and monitoring of specific cases. Both methods shows good reproducibility and in which the samples don't have to be pretreated.

6.Therapeutic observation of Tui-pushing Wujing plus Chinese patent medicine for infantile diarrhea due to spleen deficiency

Objective: To observe the therapeutic efficacy of pediatric tuina plus Chinese patent medicine for preschool kids with diarrhea due to spleen deficiency. Methods:Ninety-two eligible kids were divided into a tuina plus Chinese patent medicine and a Chinese patent medicine group by the random number table, with 46 cases in each group. The Chinese patent medicine group was intervened by oral administration of Qi Wei Bai Zhu San (Seven-ingredientAtractylodes MacrocephalaPowder); the tuina plus Chinese patent medicine group was additionally given pediatric tuina therapy once a day, 20 min each time. Efficacy and safety were evaluated after 7-day treatment in both groups, and a 14-day follow-up was conducted. Results:After treatment, the symptom scores all changed significantly in the two groups (allP<0.05). After intervention, the symptom scores in the tuina plus Chinese patent medicine group were significantly different from those in the Chinese patent medicine group (allP<0.05). The total effective rate was 78.3% in the Chinese patent medicine group, versus 89.1% in the tuina plus Chinese patent medicine group, and the between-group difference was statistically significant (P<0.05). Three cases got a relapse in the Chinese patent medicine group, versus one case in the tuina plus Chinese patent medicine group, and the between-group difference was statistically insignificant (P>0.05). The two groups each had one case of vomiting, which were treated properly and turned better. No other adverse reactions occurred. Conclusion: Tuina plus Chinese patent medicine and Chinese patent medicine alone both are effective for diarrhea due to spleen deficiencyin kids, but the combined therapy can produce more significant efficacy.
7.Progress of artificial ureter

Artificial ureter has been studied for nearly 60 years, however it is still in experimental stage. So far there are no commercially available products for clinical use except autologous intestines loop even though many progresses have been made. Each approach for making artificial ureter has its own inherent advantages, disadvantages and limitations. But some approaches have shown great potential to be used as ureteral substitutes. Preventing the conduit from salt depositing and ensuring a watertight anastomoses with native tissues are the key factors in designing and manufacturing the prosthetic ureter. Along with scientific and technical improvement, artificial ureter will be successfully used in clinical therapeuticsin in the very near future.
8.Research on quality of life in children with immune thrombocytopenia

Immune thrombocytopenia (ITP) is a common bleeding disease in children.Recent advances has indicated that such factors as decreased platelet,fear of bleeding,prognosis,therapy associated side effect and restricted activities,usually decreased quality of life of patients and their parents.This review focuses on domestic and overseas researches on health-related quality of life (HRQoL) in children with ITP,in order to attract more scholars' attention.HRQoL should be recommended as an important indicator in treatment and management of ITP.
9.Adhesion of mice lymphocytes to extracellular matrix in delayed-type hypersensitivity

AIM: To investigate the adhesion between lymphocytes and extracellular matrix (ECM) in delayed-type hypersensitivity (DTH) in mice as well as the regulatory effect of several cytokines on it. METHODS: DTH was induced by the application of picryl chloride (PCl) to mice that had been sensitized twice by PCl, followed by isolating the spleen lymphocytes at different times. The adhesion of spleen lymphocytes to fibronectin or typeⅠcollagen were examined. In some cases, spleen lymphocytes obtained at 18 h were incubated with various concentrations of IL-2, IFN-?, and TNF-? for 4 h at 37℃ before the adhesion assay. T cells purified from the spleen lymphocytes were also used for the assay in the presence of TNF-?. RESULTS: The adhesion of spleen lymphocytes to ECM began to increase at 6 h after PCl challenge, reached a peak at 18 h, then decreased and returned to normal level at 36 h. IL-2 significantly increased the adhesion of lymphocytes to ECM at a dose of 1?10 5 U/L and TNF-? enhanced the adhesion in a concentration-dependent manner. The combined use of TNF-? and IFN-? showed a significantly enhanced adhesion. TNF-? was more effective to accelerate the adhesion of spleen T Cells than that of total spleen lymphocytes. CONCLUSION: In the process of DTH, the adhesion of lymphocytes to ECM was regulated by several cytokines and associated with the development of inflammation.
