1.Intervening effect of Fleece Flower Root Granule on the expression of proliferating cell nuclear antigen in testicle of aging rat

BACKGROUND: Fleece Flower Root Granule, which consists of Fleece Flower Root, Desert-living Root, Achyranthes Root, and so on,possesses the effects of nourishing the liver and kidney, supplementing essence and blood, and prolonging life.OBJECTIVE: To investigate the effect of. Fleece Flower Root Granule, a Chinese remedy, on the content of serum testosterone and the expression of proliferating cell nuclear antigen of testicular spermatogenic cell in subacute aging rat induced by D-galactose.DESIGN: A controlled study with completely randomized grouping design.SETTING: Institute of Basic Medicine of Chengde Medical College.MATERIALS: The experiment was conducted in Institute of Basic Medicine of Chengde Medical College from April to July 2003. Totally 32 adult male SD rats were at random divided into 4 groups: normal control group, model group, drug group and negative control group,with 8 in each group.METHODS: [1] The rats in model group, drug group and negative control group were intraperitoneally injected D-galactose, 200 mg/kg ,once a day for consecutive 40 days to establish subacute aging rat models. After model was set up, no treatment was given further for rats in model group; while rats in drug group were intragastrically given Fleece Flower Root Granule (The granule was made from sex Chinese medicinal herbs of Fleece Flower Root, Achyranthes, Root Desertliving Root, Poria, Red Sage Root and Epimedium, the concentration was 1.5 g/L in raw drug after decoction and preparation); rats in negative group were intragastrically given normal saline, in the same volume as that of the Granule and the time as the same with that for the rats in drug group. Rats in normal control group, with no model establishment, were given the same intervention with that for rats in negative group. [2]Enzyme-linked immunosorbent assays (ELISA) was used for the level of serum testosterone, and inmmunohistochemistry was used for the condition of expression of proliferating cell nuclear antigen, with the karyon in orange brawn yellow being taken as positive. [3]Measurement difference was analyzed by t-test.MAIN OUTCOME MEASURES: The content of serum testosterone and the expression of proliferating cell nuclear antigen of testicular spermatogenic cell of rats in each group.RESULTS: All 32 rats entered the result analysis. [1] The level of serum testosterone:the level of testosterone in model group was obviously lower than that in normal control group [(0.52±0.14),(1.26±0.32)μg/L, P< 0.05]; the level in drug group was obviously higher than that in model group [(1.16±0.32)μg/L, P < 0.05]. [2] The positive rate of testicular proliferating cell nuclear antigen in rats: The rate in model group was obviously lower than that in normal control group [(23.26±3.38)%,(39.15±3.48)%, P < 0.05]; and the rate in drug group was obviously higher than that in model group [(37.47±3.18)%, P < 0.05]. [3]rhe distribution and change of proliferating cell nuclear antigen in seminiferous tubule: The positive staining of proliferating cell nuclear antigen was found in slices of all groups, the immunological positive products were located at karyon, usually in brown yellow granules, only a small part of them in diffuse state; the immunological positive cells were mainly the spermospores, besides a small quantity were spermiocyte, supporting the cell and myoid cell nuclear to be stained.CONCLUSION: Fleece Flower Root Granule can increase the content of serum testosterone and reduce the level of expression of proliferating cell nuclear antigen of testicular spermatogenic cell in aging rat, thus delaying degeneration of genital function, possessing a certain role in delaying aging.

Mind map is a kind of thinking tool, which draws the mind′s thoughts through words and pictures and can be applied instudy,life and work.Through biblio-metrological analysis, this study finds that domestic application of mind map in nursing focuses on fields such as nursing teaching, clinical nursing management and health education. The study and application of mind map in domestic nursing field are only in the beginning and exploration stage, lacking experimentation and evaluation study under the guidance of scientific methods, both the depth and breadth of research are yet to be improved.

Alzheimer's disease (AD) is the most common type of dementi a.PET probes can specifically detect the in vivo amyloid β peptide(Aβ) and Tau protein in AD inpatients' brain.The application of PET probes in AD is important for its early diagnosis and early intervention.US FDA has approved three PET probes for Aβ imaging while probes for Tau are still under research and development.This review summarizes current development of Aβ and Tau PET probes used for AD.
6.Enhancement of Lycopene Production by Blakeslea trispora Using Oxygen-vectors and Surface Active Agents

Addition of oxygen-vectors(n-dodecane,n-hexane,hydrogen peroxide) to fermentation medium was recognized as a method of enhancing oxygen transfer and promoting lycopene yield by Blakeslea trispora fermentation.Higher lycopene production was observed in shake flask containing 1%(v/v) n-hexane,n-dodecane and 150 L/100mL hydrogen peroxide as compared to shake flasks without oxygen-vectors.The result of assays indicated that when oxygen-vectors (n-dodecane,n-hexane,1%,v/v) and hydrogen peroxide(50 L/100mL) were added to the 0-day and 1-day old culture of Blakeslea trispora the production of lycopene were 25.32%,72.84% and 40.35% higher than those without oxygen-vectors addition respectively.The production of lycopene increased 114.83% when n-dodecane and surface active agents were used at the same time.
7.The Study on Gene Engineering Strains Producing Lycopene

Lycopene—a kind of important active compound of caroteinoids, is greatly beneficial to human health with its diverse biological functions. With the elucidation of lycopene biosynthetic pathway and cloning genes of relative enzymes from microorganisms, it is possible to regulate lycopene biosynthesis via genetic engineering. The biosynthesis pathways of lycopene and gene cloning of lycopene biosynthetic enzymes in microorganisms were reviewed, and gene engineering strains documented in previous works including: E.coli and yeast constructed by genetic recombination, mold strains enhanced the ability of producing lycopene by gene manipulation were summarized. At last, compared with the present methods, the problems existed in the process of construction were pointed out.
8. Simultaneous determination of four effective components in Taraxaci Herba by UPLC-MS/MS

Objective: To establish an LCMS/MS method for simultaneous determination of pcoumaric acid, caffeic acid,luteolin and luteoloside in Taraxaci Herba. Methods: The LC analysis was performed on a UPLC-MS/MS instrument with a Dikma Endeavorsil UPLC C18 column(100mm×2.1 mm,1.8 μm),the mobile phase was 0.1% formic acid solution-methanol in a gradient elution,and the flow rate was 0.2 ml/min;the column temperature was set at 35℃; the injection volume was 2 μl. For the MS/MS analysis,the ESI source was operated in the positive-ion mode and the ions were monitored in multiple reaction monitoring(MRM)mode,the temperature of drying gas was 370℃,the sheath gas pressure was set at 10 Bar,the aux gas pressure was maintained at 30 Bar,and the spray voltage was controlled at 4000 V. Results Calibration curves showed good linearity for all 4 compounds in the concentration range of 0.1-10 μg/ml,and the correlation coefficient®was 0.9991,0.9996,0.9994 and 0.9999 for p-coumaric acid,caffeic acid,luteolin,and luteoloside,respectively. Conclusion: The present method could be used for the simultaneous determination of the above mentioned 4 components in Taraxaci Herba.
9.Relationship of epigenetic and Dao-di herbs.

Dao-di Herbs is specificity and locality, and its unique phenotypic features is closely related to the growth and development of medicinal plants. In addition to traditional genetic, epigenetic play an important role in formation of Dao-di herbs. This paper introduces the concept of epigenetic and the role of DNA methylation in the gene expression regulation. We further prospects epigenetic mechanism in study of Dao-di herbs formation from specific phenotype and regional analysis. And study on the relationship of epigenetic and Dao-di herbs will provide a basis for quality assessment and identification of Chinese drugs.

10.Research progress in roles of high-risk human papillomavirus E2 protein.

High-risk human papillomavirus (HPV) is the principal cause of various cancers including cervical cancer, anal cancer, vulvar cancer, and some head and neck cancers. In the viral life cycle, by interacting with both viral and host DNA and proteins, the HPV E2 protein plays a pivotal role in viral transcriptional regulation and DNA replication, and it is also associated with modification of various cellular processes, including host gene transcription, RNA processing, apoptosis, ubiquitination, and intracellular trafficking, to create a convenient environment for a replicative cycle of the virus and contribute to the HPV pathogenesis. Elucidating the roles of E2 protein throughout the viral life cycle will improve our understanding of the viral life cycle and pathogenesis and help us identify novel antiviral agents with therapeutic potential. This article reviews the research progress in the structure, roles, and activity of high-risk HPV E2 protein, particularly that of HPV-16.
