Objective:To investigate the effect of RNA-mediated interference EGFR (epidermal growth factor receptor) expression on the apoptosis of muitidrug-resisitant ovarian cancer cell line SKOV3/DDP. Methods: Small hairpin RNA (shRNA) targeting EGFR was synthesized and recombinant plasmid containing pEGFR-shRNA was constructed, pEGFR-shRNA was tansfected into SKOV3/DDP cells by liposome system, untransfected cells and SKOV3/DDP cells tansfected with nonspecific-shRNA (Ctrl-shRNA) were used as control. Expression of EGFR mRNA and protein in SKOV3/DDP cells was examined by RT-PCR and immunocytochemistry after transfection, respectively. The apopototic rates and cell cy-cles of SKOV3/DDP cells were detected by flow cytometry. Results: Compared with Ctrl-shRNA-transfected cells, the ex-pression of EGFR mRNA and protein in pEGFR-shRNA-transfected SKOV3/DDP cells was significantly inhibited. Flow cytometry results showed that cell cycle distribution in pEGFR-shRNA-transfected SKOV3/DDP cells was dramatically changed, and the apoptosis rate was significantly increased after further treatment with cisplatin for 24 h. Conclusion: EGFR-targeted interference RNA can inhibit the expression of EGFR in SKOV3/DDP cells, thereby regulating the cell cy-cle and increasing apoptosis of multidrug-resistant SKOV3/DDP cells.

China Journal of Chinese Materia Medica 2015;40(21):4155-4159
The tuberous roots of Aconitum carmichaeli are largely used in traditional Chinese medicine and widely grown in Jiangyou, Sichuan, China. During the growth process, this medicinal plant releases a large amount of allelochemicals into soil, which retard the growth and development of near and late crops. Therefore, a pure culture experiment was thus carried out by seed soaking to study the allelopathic effects of extracts from tuberous roots of A. carmichaeli (ETR) on the seed germination and young seedling growth of Lolium perenne, Trifolium repens, and Medicago sativa, the late pasture grasses after cultivation of A. carmichaeli. The results showed that three pasture grasses varied significantly in seed germination and young seedling growth in response to ETR concentrations. Seed germination of M. sativa was stimulated by low ERT concentration (0.01 x g(-1)), while all of pasture grass seeds germinated poorly in solution with 1.00 g x L(-1). Seed soaking with 1.00 g x L(-1) also inhibited significantly the growth of pasture young seedlings, with M. sativa showing the highest seedling height reduction of 42.05% in seeding height, followed by T. repens (40.21%) and L. perenne with about 11%. Cultivation of L. perenne could thus be beneficial to increase whole land productivity in A. carmichaeli-pasture grass cropping systems. In addition, hydrolysis of protein, starch, and inositol phosphates was blocked and free amino acids, soluble sugars and phosphorus were decreased in seeds by seed soaking with ETR, which could be one of the reason for the inhibition of seed germination. There was a significant reduction in root vigor, nitrate reductase, and chlorophyll after the seed treatment with ETR, indicating the suppression of nutrient uptake, nitrate assimilation, and photosynthesis by allelopathic chemicals in ETR, which could lead to the slow growth rate of pasture grass seedlings.

OBJECTIVE:To investigate the early changes of ECG,cTnⅠ,LDH,α-HBDH and CKMB in patients with anthra-cyclines chemotherapy-induced heart damage after breast cancer surgery, and to explore their significances in the diagnosis of early heart damage. METHODS:Medical information of 152 cases of anthracyclines chemotherapy after breast cancer surgery in our hos-pital during Jan. 2015-Jun. 2016 were analyzed retrospectively. According to diagnosis criteria of drug-induced cardiotoxicity, the occurrence of heart damage was evaluated during hospitalization and 1-year follow-up. According to evaluation results, those pa-tients were divided into heart damage group(50 cases)and control group(102 cases). The early changes of ECG,cTnⅠ,LDH, CKMB and α-HBDH were analyzed before chemotherapy(T0),24 h after first chemotherapy(T1),24 h after second chemothera-py(T2),24 h after forth chemotherapy. RESULTS:At T1,T2,T3,the proprotion of ECG abnormalities in heart damage group was significantly higher than control group;the serum levels of cTnⅠ,LDH,CKMB andα-HBDH in heart damage group were signifi-cantly higher than control group, with statistical significance (P<0.01). CONCLUSIONS:For anthracyclines chemotherapy drugs-induced heart damage after breast cancer surgery, early regular monitoring of ECG,cTnⅠ,LDH,CKMB and α-HBDH can improve the efficiency of early diagnosis of heart damage, and improve prognosis.

Objective To find an ideal method of DNA isolation from blood and especially from clotted blood and to minimize the volume of blood collected for laboratory and clinical tests. Methods DNAs were isolated from antiagglutinated and agglutinated blood samples from auricular veins of 30 healthy subjects. The DNAs of these samples were obtained by a nonenzymatic, nontoxic procedure optimized by us and determinated by agarose gel electrophoesis and PCR. Results The yields of DNA isolated from clotted blood and antiagglutinated blood were (40.2?8.86)mg DNA/L and (39.1?10.2)mg DNA/L, and purities were 1.87?0.11 and 1.92? 0.12. The DNAs that we isolated from all samples had high molecular weight and by PCR the dimorphism of ALU alleles of the 8th intron of t-PA was easy to be obtained, so they were complete and reliable. Conclusion This method is rapid, easy, efficient and nontoxic for isolation of DNA from clotted and fresh blood and meets requirements for clinical testing and molecular biology study.

Chinese Journal of Laboratory Medicine 2001;0(05):-
Objective To identify and analyze the plasmid (pR_ ST98 ) encoding multi-resistance to anti-microbial agents in S.typhi presenting the Salmonella plasmid virulence gene (spv). Methods Plasmid pR_ ST98 ,which could mediate virulence to its host bacteria, was used as the templete. The spv-specific PCR and Southern blot were employed to identify the spv virulence gene on this plasmid. The amplified spv fragments (spvR and spvB) were cloned into pGEM-T EASY.Then the DNA sequences were analysed. Results The date of PCR and Southern blot showed that spv, which had been found in other pathogenetic Salmonella spp. except S. typhi was also presented on pR_ ST98 . The ORF of spvR and spvB of pR_ ST98 were 894bp and 1 776bp respectively. They had more than 99% homologus with that of spvR and spvB on virulence plasmid in S.typhmurium.Conclusion From the results of PCR,Southern blot and nuclei acid sequencing, we concluded that this is the first report of revealing a mosaic-like plasmid carrying genes encoding not only drug resistance but also virulence in S.typhi.
