1.Contents Determination of Ginsenoside Rg1,Re,Rb1 and Geniposide in Panax ginseng Processed by Gardenia jasminoides by HPLC

OBJECTIVE:To establish a method for the contents determination of ginsenoside Rg1,ginsenoside Re,ginsenoside Rb1 and geniposide in Panax ginseng processed by Gardenia jasminoides. METHODS:HPLC was performed on the column of Agi-lent TC18 with mobile phase of acetonitrile-0.1% phosphoric acid,(gradient elution,ginsenoside Rg1,ginsenoside Re,ginsenoside Rb1)and acetonitrile-water(13∶87,V/V,geniposide)at a flow rate of 1.0 ml/min,the detection wavelength was 203 nm for ginsen-oside Rg1,ginsenoside Re,ginsenoside Rb1 and 238 nm for geniposide,column temperature was 40 ℃,and injection volume was 20 μl. RESULTS:The linear range was 0.418-3.762 μg for ginsenoside Rg1(r=0.999 8),0.270-2.430 μg for Re(r=0.999 8), 0.398-3.582 μg for ginsenoside Rb1(r=0.999 7)and 0.606-3.030 μg for geniposide(r=0.999 7);RSDs of precision,stability and reproducibility tests were less than 2.0%;recoveries were 97.86%-101.47%(RSD=1.29%,n=6),96.21%-100.11%(RSD=1.42%,n=6),96.24%-100.48%(RSD=1.57%,n=6)and 97.76%-102.44%(RSD=1.71%,n=6). CONCLUSIONS:The meth-od is simple with good precision,stability and reproducibility,and can be used for the contents determination of ginsenoside Rg1, ginsenoside Re,ginsenoside Rb1 and geniposide in P. ginseng processed by Gardenia jasminoides.
2.Ultrasound-guided percutaneous puncture 125 I radioactive seed implantation in treating hilar hepatic tumor:one case report and literature review

Objective:To report one case of hilar hepatic tumor treated by ultrasound-guided percutaneous puncture 125 I radioactive seed implantation,and to review the associated literatures.Methods:One patient’s CT and ultrasonic examinations were taken,and the plan of TPS was taken.The patient was treated by ultrasound-guided percutaneous puncture 125 I radioactive seed implantation according to the plan of TPS.The needles were parallel, and the distance between the seeds were 1.0 to 1.5 cm. Results:The operation was successfully completed.No postoperative complications were encountered during 1 month follow up.The volume of tumor was obviously reduced 1 month after operation detected by ultrasound and CT. The local control effect of tumor was good.Conclusion:Ultrasound-guided percutaneous puncture 125 I radioactive seed implantation is a safe and effective method in treating hilar hepatic tumor.
4.Interleukine 1 receptor antigonist inhibits proinflammatory factor production by human retinal pigment epithelial cells in vitro

Objective To investigate production of interleukine 6 (IL 6) and interleukine 8 (IL 8) by retinal pigment epithelial (RPE) cells and its inhibition by interleukine 1 receptor antigonist(IL 1ra). Methods Cultured human RPE cells was treated with interleukine 1 ? (IL 1?, 10 ng/ml) and/or IL 1ra ( IL 1ra, 1、10、100 ng/ml). IL 6 and IL 8 mRNA and protein expression were detected by ELISA, immunohistochemistry and in situ hybridization (ISH) assay. 5.Study on the Quality Standard for Maikang Mixture

OBJECTIVE:To establish the quality standard for Maikang mixture. METHODS:TLC was used for the qualitative identification of Paeonia lactiflora,Angelica sinensis,Ligusticum chuanxiong,Astragalus membranaceus,Schisandra chinensis and Ophiopogon japonicus. HPLC was used for the content determination of schisandrin:the column was Kromasil C18 with mobile phase of methanol-water (60∶40,V/V) at a flow rate of 1.0 ml/min,detection wavelength was 254 nm,column temperature was 40 ℃,and the infection volume was 20 μl. RESULTS:P. lactiflora,A. sinensis,L. chuanxiong,A. membranaceus,S. chinensis and O. japonicus showed clear spots and well separated. The linear range of schisandrin was 1-50 μg/ml (r=0.999 9);RSDs of precision,stability and reproducibility tests were lower than 2%;recovery was 97.01%-98.58%(RSD=0.56%,n=6). CONCLU-SIONS:The established standard can be used for the quality control of Maikang mixture.
6.Tissue-engineered nerve for repair of peripheral nerve injuries

BACKGROUND:Recent development of bioengineering technology and tissue-engineered nerve brings a new hope for the treatment of peripheral nerve injuries, which has gradual y become a research spot. OBJECTIVE:To review the new progress in the repair of peripheral nerve injuries using seed cells, biomaterials and tissue-engineered nerve construction technology. METHODS:PubMed and CNKI were searched by the first authors for articles concerning nerve tissue engineering and repair of peripheral nerve injuries published prior to July 2013. The keywords were“tissue engineering, peripheral nerves, nerve injuries, stem cells, Schwann cells, scaffold, growth factor”in English and Chinese, respectively. The articles published recently or in the authorized journals were preferred in the same field. Final y, 63 articles were included in result analysis. RESULTS AND CONCLUSION:Up to now, there is a great advance in the tissue engineering technology for the repair of peripheral nerve injuries. However, most studies are stil in experimental step. For the clinical application of nerve tissue engineering, some problems to be solved include:(1) source and ethics of seed cells;(2) immunological rejection fol owing cellproliferation and transplantation;(3) stability and oncogenicity of transplanted cells;(4) degradation rate, optimal porosity, tube thickness and shape;(5) repair timing for in vitro tissue-engineered nerve construction;(6) local release and regulation of various neurobiological factors. With the development of science, many patients with nerve injuries can profit from the solve of these problems.
7.The impact of manual small-incision cataract surgery and phacoemulsification on corneal regularity

Background Cataract surgery can induce the occurrence of dry eye because of the influence of incision on corneal nerve fibers and the change of corneal regularity derived by surgery.Objective This study was to investigate the change of regularity of anterior corneal surface,tear film and astigmatism caused by the process of manual small-incision cataract surgery (manual SICS) and phacoemulsification (Phaco).Methods A prospective non-randomized controlled study was designed.Fifty eyes of 46 age-related cataract patients were included in this clinical study.Thirty eyes of 30 patients received the manual SICS,and other 20 eyes of 16 patients underwent Phaco.Corneal irregularity measurement (CIM),shape factor (SF) and corneal astigmatism were assessed by the corneal tomography examination,the subjective symptoms were scored and tear film break up time (BUT),Schirmer Ⅰ test (S Ⅰ t) were examined before surgery and 1 day,1 week,1 month after surgery.The examined parameters were compared among different time points and two groups.This study was approved by Ethic Committee of Affiliated Eye Hospital of Wenzhou Medical College,and written informed consent was obtained prior to operation.Results The CIM values were gradually increased with the prolong of time after operation,showing a statistically significant difference dependent on time (Ftime =22.864,P =0.000),and CIM value of 1 day,1 week and 1 month after operation was higher than that of before operation (P<0.01).However,no significant difference was found in CIM value between two groups in different time points (Fgroup =0.062,P =0.804).There was no significant difference in SF values with time prolong (Ftime =4.600,P =0.428) and different groups (Fgroup =0.009,P =0.925).The BUT was gradually reduced with the time prolongs with the considerably difference (Ftime =39.384,P=0.000).Obviously decreases of BUT values were seen in 1 week and 1 month in comparison with 1 day after operation (both P=0.000).But no significant change was found in BUT between two groups in various time points (Fgroup =0.000,P=0.983).In the manual SICS group,the difference was found in S Ⅰ t values between before operation and 1 week or 1 month after surgery(P<0.01).Surgery associated astigmatism (SIA) was existed in the operative eyes in various time points after operation.With time lapse,SIA value was declined with a significant difference (Ftime =21.479,P =0.000),the difference also was found in different groups (Fgroup =8.709,P =0.005),and SIA value in the Phaco was significantly lower than that in the manual SICS group (P<0.01).Conclusions In the present study,in the early stage after manual SICS and Phaco,SIA appears due to the irregular change of the anterior corneal surface increases and stability of tear film worsens.
8.Research on retinoic acid system in retina of guinea pig eyes with form deprivation myopia

Objective Development of eyeball is visually regulated by messengers that released from the retina, and research demonstrated that retinoic acid (RA) is the only extrinsic biochemical candidate that could act as a growth controller. Present study was designed to evaluate the RA system in retina of guinea pig eye with experimental form deprivation myopia. Methods The form deprivation myopia models were monocularly established in the 24 2-week-old guinea pigs by occluding the lateral eyes using white translucent hemispheres for two weeks, and the fellow eyes were as normal control. Refraction diopter was detected with streak retinoscopy after cycloplegia, and axial length and vitreous depth were calculated with Cinescan A/B ultrasonography before and 2 weeks after experiment. All animals were sacrificed and retina was dissected 2 weeks after experiment. The RA level in retina was delected by High Performance Liquid Chromatography (HPLC). The expressions of RA-binding proteins Ⅰ(CRABP-Ⅰ) and RA receptor-β(RAR-β) protein and mRNA were assayed by Western-blotting and Real-time PCR, respectively. The experiment and use of animals followed the Standard of Association for Research in Vision and Ophthalmology. Results The spherical equivalent refraction was (+ 3. 00 ± 0. 75) D in the model group and (+ 2. 88 ± 0. 67) D in control eyes (t= 0. 672, P > 0. 05), the ocular axial length was (7. 822 ± 0. 083) mm in model group and (7. 791 ±0.073) mm in control eyes before experiment (t = 0. 346, P > 0. 05). In 2 weeks after experiment, the spherical equivalent refraction was (- 3. 82 ± 0. 13) D versus (1. 99 ± 0. 58) D and axial length was (8. 346 ± 0. 047) mm versus (7. 888 ± 0. 042) mm between model eyes and control eyes (t = 8. 376, P < 0. 001; t = 3. 343, P <0. 05). No significant difference in the level of RA in retina before and after experiment (1. 394 ±0. 079 μg/g vs 1. 295 ±0. 023 μg/g) (t =0. 897, P >0. 05) but obviously elevated after experiment in model eyes compared with control eyes (2. 356 ± 0.098 μg/g vs 1.499 ±0.035 μg/g) (t =4. 934, P <0. 01). The expression of CRABP-Ⅰ and RAR-β mirrored these directional changes. Conclusion The RA system in retina is upregulated in the eye of guinea pig with form deprivation myopia. This findings suggests that RA may act as a messenger in the development of experimental myopia.
9. Improvement of quality standards of Arthritis Ointment

Objective: To improve the quality standards for Arthritis Ointment. Methods: TLC was applied to identification of Angelica dahurica in qualitative identification. The camphor and borneol contents were determined by GC. Results: The TLC spots of A. dahurica were distinct and reproducible, and negative without interference. The linear ranges of camphor and borneol were 0.401 41-4.014 1 and 0.393 34-3.933 4 μg. The average contents of camphor and borneol were 6.038 0 and 5.353 9 mg/g. The average recoveries of camphor and borneol were 99.32% and 99.82%. Conclusion: The established quality standards are accurate, simple, and feasible, and could be used for quality control of Arthritis Ointment.
10.Limit test for 5-hydroxymethylfurfural in Yueju Baohe pills

Objective: To establish an HPLC method to detect the limit of 5-hydroxymethylfurfural in Yueju Baohe pills.
Methods: A Zafex Acutfex PW-C18 column(4.6 mm×250 mm, 5 μm) was adopted with mobile phase consisted of acetonitrile-0.1% formic acid aqueous solution(2:98)at the flow rate of 1.0 mL·min-1. The column temperature was 40 ℃ and the detective wavelength was set at 284 nm.
Results: The linear range of 5-hydroxymethylfurfural was 0.491 9-122.98 μg·mL-1 (r=1.000). The average recovery(n=6) was 97.99% with RSD 0.61%. All of the RSDs for precision,repeatability and stability met the methodological requirements. According to the proposed limit, 17 of the 65 batches of samples did not meet the requirements, and the failure rate was 26.15%.
Conclusion: The established method is suitable for the limit test of 5-hydroxymethylfurfural in Yueju Baohe pills.There was a significant difference in the content of 65 batches of samples. It is necessary to investigate the limit of 5-hydroxymethylfurfural in Yueju Baohe pills.
