1.Study of Periplaneta americana L.reversing multi-drug resistance of hepatocellular carcinoma

Objective To explore the effect of Periplaneta americana reversing multiple drug resistance (MDR) of hepatocellular carcinoma in vitro. Methods Drug resistance of Hepatocellular carcinoma cell line HepG2/ADM was established by Adriamycin ( ADM ) increasing concentration gradient method;determined cell growth curve, doubling time change using Trypan blue staining method;detected sensitivity of 4 kinds of chemotherapeutic drugs of HepG2 and HepG2/ADM by methyl thiazolyl tetrazolium method; detected the non toxic dose of Periplaneta americana CⅡ-3 and the skim cream of HepG2/ADM by MTT method; detected the inhibition rate differences of CⅡ-3, skim cream, ADM, CⅡ-3 and ADM, skim cream and ADM of hepatocellular carcinoma drug resistance cell line HepG2/ADM by MTT method.Results Drug-resistant cell line doubling time was extended obviously compared with parent cells,HepG2/ADM showed resistance to variety of chemotherapeutic drugs, ADM resistant index was especially high,that was 15.25 times.When the concentration of CⅡ-3 and skim cream were 28.76 and 45.08μg/mL respectively, the growth inhibition rate of HepG2 and HepG2/ADM two cell lines were<10%,no obvious toxic effect.The inhibition rate of the drug-resistant cell of 7.5,15,30μg/mL concentration CⅡ-3 joint with ADM concentration (1/2 IC50 ) were (24.52 ±2.05)%,( 34.78 ±3.08)%,( 45.09 ±3.56)% respectively; The inhibition rate of the drug-resistant cell of 12.5,25,50μg/mL concentration skimmed cream joint with ADM concentration (1/2 IC50 ) were (19.28 ±2.56)%,(35.08 ±2.13)%,(43.85 ±3.05)%respectively.Conclusion The cell line HepG2/ADM established has basic multidrug-resistant biological characteristics.Periplaneta americana extract can inhibit the growth of HepG2/ADM,and has good effect to reverse drug resistance at the same time.
2.Diagnostic significance of PCT for neonatal sepsis

Objective To determine the special diagnostic superiority of PCT test over CRP and WBC count test for neonatal sepsis .Methods 46 newborn inpatients diagnosed neonatal sepsis in the neonatal department of Dalian Central Hospital from Janu‐ary 2013 to January 2014 and contemporaneous 50 uninfected neonatal inpatients as control group were collected .The PCT and CRP concentrations and WBC count were simultaneously measured at admission .Results The WBC count had no statistical difference between the two groups ,but the PCT and CRP detection showed the statistical difference between the two groups (P<0 .01) ,the sensitivity of PCT test was 87% ,which was significantly higher than 63% of CRP test ,but the specificity of PCT test was 87% , which was slightly lower than 90% of CRP test .The Youden index of PCT test was obviously higher than that of CRP test and WBC count .Conclusion PCT is superior to CRP and WBC count in diagnosing neonatal sepsis .
3.Studies of Peripheral Nerve Electrophysiology in Uremic Patients

Objective To analyze electrophysiology of peripheral nerve in uremic patients. Methods 24 uremic patients and 19 normalvolunteers were included, surface electrodes were used to observe the electrophysiology performance, including tibial nerve, peroneal nerve,sural nerve and skin sympathetic response. Results The motor conduction, sensory conduction and skin sympathetic response were significantlydifferent between uremia patients and normal volunteers (P<0.05). The electrophysiology performance showed that F wave latencyprolonged, nerve conduction slowed, distal latencies prolonged, sensory fibers damaged more serious than motor fibers. Conclusion Peripheralneuropathy is one of the most common complications in uremia patients, neural electrophysiological abnormalities are significantly earlierthan clinical symptoms, nerve electrophysiological examination is of great significance in diagnosis and evaluation uremic peripheralneuropathy.
4.Mediating role of activities of daily living and self-rated health in chronic disease-caused depressive symptoms among the elderly

Objective:
To investigate the mediating role of activities of daily living (ADL) and self-rated health in chronic disease-caused depressive symptoms among the elderly, so as to provide insights into depression control among the elderly.
Methods:
Basic features, depressive symptoms, number of chronic diseases, ADL and self-rated health of the elderly at ages of 60 years and older were collected from the China Health and Retirement Longitudinal Study 2018. Multiple linear regression model was created using the Process program to examine the mediating role of ADL and self-rated health in number of chronic disease-caused depressive symptoms, and the significance of the mediating role was tested using the Bootstrap test.
Results:
A total of 5 892 elderly participants were enrolled, with a mean age of (68.42±6.23) years and including 2 744 men (46.57%). The participants had a median depressive symptom score of 8 (interquartile range, 10) points, a median number of chronic diseases of 1 (interquartile range, 1), a median ADL score of 12 (interquartile range, 3) points, and a median self-rated health score of 3 (interquartile range, 1) points. Mediation analysis showed that number of chronic diseases affected depressive symptoms via the independent mediating role of ADL (β=0.163, 95%CI: 0.120-0.206) and self-rated health (β=0.303, 95%CI: 0.259-0.351), and affected depressive symptoms via the chain mediating role of ADL and self-rated health (β=0.057, 95%CI: 0.043-0.074) among the elderly, and the gross mediating role consisted of 58.44% of total effects.
Conclusions
The number of chronic diseases may incease the risk of depressive symptoms through aggravating the impairment of ADL and decreasing self-rated health levels among the elderly.
5.Willingness to pay integrated medical and elderly care services among the elderly in Hohhot City

Objective:
To investigate the willingness to pay integrated medical and elderly care services and identify the influencing factors among the elderly in Hohhot City, Inner Mongolia, so as to provide insights into promoting integrated medical and elderly care services.
Methods:
The elderly at ages of 60 years and older were sampled using a convenient sampling method from public venues in four districts of Huimin, Saihan, Yuquan and Xincheng in Hohhot City from June to December 2021. Basic characteristics, health and disease burdens, social support and willingness to pay integrated medical and elderly care services were collected, and factors affecting the willingness to pay integrated medical and elderly care services were identified among the elderly using a multiple linear regression model.
Results:
A total of 1 008 valid questionnaires were recovered, with an effective recovery rate of 96.74%. The respondents included 519 men (51.49%) and 489 women (48.51%), and had a mean age of (69.47±6.42) years. The monthly average fees of willingness to pay integrated medical and elderly care services were (2 076.49±36.79) Yuan, and there were 636 participants with 2 000 Yuan and less monthly average fees of willingness to pay integrated medical and elderly care services (63.10%). Multiple linear regression analysis showed that place of residence (β=180.832), satisfaction with housing (satisfied, β=-140.760), physical self-care ability (completely self-care: β=-238.244; mostly self-care: β=-254.557), burdens of disease diagnosis and treatment (able to afford: β=452.488; partly afford: β=228.626), monthly income (β=347.144), expenses of medications (β=0.019) and total score of social support (β=17.116) were factors affecting the willingness to pay integrated medical and elderly care services among the elderly.
Conclusions
The willingness to pay integrated medical and elderly care services among the elderly in Hohhot City is associated with place of residence, monthly income, satisfaction with housing, physical self-care ability, burden of disease diagnosis and treatment, expenses of medications and social support.
6.Effect of GSK-3βtargeting RNAi recombinant adenovirus on proliferation of human umbilical vein endothelial cells

Objective To observe the effect of Wnt/β-catenin pathway on proliferation of human umbilical vein endothelial cell (HUVEC) using the glycogen synthase kinase 3β (GSK-3β)-targeting RNAi recombinant adenovirus vector. Methods Homologous recombination and cloning techniques were used to construct RNAi recombinant adenoviral expressive vectors specific to GSK-3β. Then, the adenovirus plasmids was transfected into HEK 293A cells to produce adenovirus and amplify the adenoviral stock. Plaque forming assay was used to titer the adenoviral stock. The GSK-3β and β-catenin protein expressions were detected by Western blot and immunohistochemistry. The proliferation of HUVEC was detected with MTr assay. Results The RNAi adenovirus vectors specific to GSK-3β were successfully produced with high titer. The expression of GSK-3β protein in HUVEC could be down-regulated efficiently by the RNAi adenovirus, along with increased β-catenin protein expression. The proliferation of HUVEC was significantly increased (P < 0.05 or P < 0.01) after infected with GSK-3β RNAi recombinant adenovirus for 3, 5, 7 days. Conclusion RNAi adenovirus is an important tool that can inhibit the expression of GSK-3β efficiently, along with increased β-catenin protein expression. Up-regulating of the Wnt/β-catenin pathway might play an important role in the proliferation of HUVEC.
7.Effects of patient participation-based dietary intervention on nutritional status for patients with gastrectomy

Objective To explore the effects of patient participation-based dietary intervention on nutritional status for patients with gastrectomy.Methods Fifty-two patients with gastrectomy were randomly assigned to the intervention group(n=26) and the control group(n=26).All patients were given general dietary guidance,and patients in the intervention group were provided patient participation-based dietary intervention(PPDI) as well.Height,body weight and body mass index(BMI) were recorded at baseline,2 weeks and 12 weeks after intervention,using PatientGenerated Subjective Global Assessment(PG-SGA),Diet-Related Symptom Scale,Adherence to Dietary Scale and a 3-day food diary.Results There was no obvious change in body weight and BMI before and after 12-week intervention,while outcomes in the control group gradually decreased.For time effect,there was significant difference in PG-SGA between two groups(P<0.05).For between-group effect,there were significant differences in diet-related symptoms(P<0.05),dietary intake(P<0.05) and adherence to dietary(P<0.05) between two groups.Conclusion Patient participation-based dietary intervention can improve nutritional status for patients with gastrectomy.
8.Expressions of Nampt in glucose metabolism related tissues of rats with tyge 2 diabetes mellitus

Objective To study the expressions of nicotinamide phosphoribosyl transferase (Nampt)in main energy metabolism organs (liver, pancreas, skeletal muscle, and kidney ) of the rats with type 2 diabetes mellitus (T2DM),and to explore the correlation between the expression and distribution of Nampt and the occurrence of diabetes.Methods The SD rat diabetes model was established by injecting with streptozotcin (STZ).The SD rats were randomly divided into diabetes group and control group. Immunohistochemical Envision staining assay was used to detect the distribution and protein expressions of Nampt in liver,pancreas,muscle,and kidney tissues of the rats,at the same time the blood glucose and serum insulin levels were also be detected. Results The blood glucose level of the rats in diabetes group was significantly higher than that in control group (P<0.01),and the fasting insulin level was lower than that in control group (P<0.01).The Nampt expression in the liver tissue of the rats in diabets group was significantly increased,which distributed near the hepatic sinus in diabetes rats,and the Nampt expression was also increased in skeletal muscle in which the whole cell was thick dying;the Nampt expression mainly distributed in the renal tubular epithelial cells. Compared with control group, the positive expression rates of Nampt in liver,skeletal muscle,and kidney tissues of the rats in diabets group were significantly increased (P<0.05 or P<0.01).There was nearly no Nampt expression in pancreas tissue of the rats in diabetes group and the Nampt expression level was significantly lower than that in control group (P<0.01). Conclusion The Nampt expressions are much different in main energy metabolic organs of the rats with diabetes.It is suggested that Nampt may be used as a specific indicative marker in the process of diabetes.
9.Inflnence of benzyl propionate nandrolone in Nampt expression in NIT-1 cells and insulin secretion

Objective To study the effects of benzyl propionate nandrolone (BPN ) on the nicotinamide phosphoribosyl transferase (Nampt), insulin receptor substeate-2 (IRS-2 )and pancreatic duodenal homeobox-1 (PDX-1)expressions, cell cycle changes as well as insulin secretion in pancreatic islet cell NIT-1 lines, and to explore the influence of BPN in the Nampte xpression in NIT-1 cells and insulin signaling molecules in high glucose oxidation stress.Methods The NIT-1 cells were cultured with different concentrations (5.6,11.1,16.7,and 27.6 mmol·L-1)of glucose,then they were treated with 10 mg·L-1 BPN for 48 h with no BPN treatment as corresponding control groups.The expression levels of Nampt,IRS-2,and PDX-1 were tested by Western blotting assay.The changes of cell cycle were determined by FCM and the cell insulin secretion levels were measured with radioimmunoassay.Results Compared with corresponding control groups,the expression levels of Nampt,IRS-2, and PDX-1 proteins in the NIT-1 cells in various BPM groups were increased (P<0.05 or P<0.01).The G0/G1 phase arrest was relieved (P<0.01)when the cells was cultured in low glucose (5.6 mmol·L-1 )condition,and the G2/M block was remitted significantly in high glucose (27.6 mmol·L-1 )condition (P<0.01),furthermore, the cell insulin secretion was promoted compared with control groups except 1 1.1 mmol· L-1 glucose group (P<0.01).Conclusion BPN can promote the expression levels of Nampt,ISR-2 and PDX-1 proteins in NIT-1 cells. There is close relationship between the Nampt expression in NIT-1 cells and insulin signaling pathway and BPN prevents the cells from insulin resistance.
10.The effect and analysis of retinoic acid on the peripheral blood Pin1 and T helper cell 17/regulatory cells of rheumatoid arthritis patients

Objective:To explore the expression of peptidyl prolyl cis-trans isomerase (Pin1) activity in peripheral blood of patients with rheumatoid arthritis (RA) and the value and correlation of T helper cell 17/regulatory cells (Th17/Treg) cells, and to analyze the effect and influence of all-transretinoic acid (ATRA) on it.Methods:① Comparing the difference of Pin1 expression and absolute counts of Th17 and Treg between RA patients before and after treatment and healthy control group, Kruskal-Wallis rank sum test was used for analysis. ② To analyze the correlation between the expression of Pin1 and its general data, activity indicators [such as erythrocyte sedimentation rate (ESR), C-reactive protein (CRP) and disease activity score 28 joints (DAS28) scores], Th17, Treg and some cytokines in RA patients, and to use Pearson and Spearman correlation tests. ③ To analyze the difference of Pin1 expression and Th17/Treg in peripheral blood of RA patients treated with low-dose all-trans retinoic acid (10 mg twice a week) and traditional immunosuppressants such as hydroxychloroquine for 3 months respectively. Mann-Whitney U test was used for comparison between the two groups, and the difference was statistically significant with ( P<0.05). Results:① The activity of Pin1 in peripheral blood of the newly treated group of RA was [13.62(9.16, 19.42)] higher than that of the healthy control group [8.97(7.62, 11.45)]( Z=42.82 , P<0.05), and Th17 was [18.28(12.76, 24.08)] higher than that of the healthy control group [6.04(4.96, 4.96)]( Z=48.83 , P<0.05). Treg [11.06(5.31, 21.87). It was lower than that of healthy control group [40.41(24.33, 48.52)]( Z=42.21 , P<0.05). ② the activity of Pin1 in peripheral blood of RA patients was positively correlated with CRP, the number of involved joints, DAS28 score, interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) ( r=0.396, P<0.05; r=0.683, P<0.05; r=0.466, P<0.05; r=0.315, P<0.05; r=0.416, P<0.05). ③ Compared with the newly treated RA group, the activity of Pin1 [6.94(5.96, 8.77), Z=42.82 , P<0.05] and Th17 7.38 decreased [7.38(3.85, 11.21), Z=48.83 , P<0.05], while Treg [40.41 (17.77, 33.47)] increased ( Z=42.21 , P<0.05). ④ Compared with the traditional medicine group, Treg [28.9(21.73, 37.36)] was higher in the retinoic acid group, and the difference was statistically significant ( Z=-2.683 , P<0.05). The activity of Pin1 was [6.23(5.58, 8.75)], but there was no statistical significance ( Z=-1.622 , P=0.104). Conclusion:Pin1 in peripheral blood of RA patients is over-expressed. Th17 is increased and Treg is decreased. ATRA combined with other traditional drugs can reduce Pin1 activity, promote Treg growth and improve disease activity of RApatients to a certain extent.
