Objective To explore the expressions of endothelial growth factor-D (VEGF-D) and urokinase-type plasminogen activator (uPA) in colon carcinoma and to reveal their correlation with the tumor invasion and metastasis. Objective To explore the expression of endothelial growth factor-D (VEGF-D) and uroki-nase-type plasminogen activator(uPA) in rectal carcinoma and to reveal their correlation to the tumor invasion and metastasis.Methods The expression of VEGF-D and uPA in 30 cases with rectal carcinoma and normal tissue was detected by immunohistochemistry and Western blot.Results The expression of VEGF-D and uPA was de-tected both in tumor tissue and normal tissue, but significantly higher in tumor tissue (P < 0.01), they expressed mostly in endochylema.The expression of VEGF-D or uPA was significantly higher in Dukes' stage C + D than that in Dukes' stage A + B(P <0.01), was also higher in tissues with lymph node metastasis or distant metasta-sis than those without metastasis(P <0.05 ,P <0.01).They were found to be lower in cancer tissue along with the differentiation degree increase(P < 0.05) .Condnsions The overexpression of VEGF-D and uPA in rectal carcinoma may play an important part in the tumorigenesis and progression of rectal carcinoma.
4.Distribution characteristic of MTA 1 and its relationship with MMP-9,VEGF-C protein expression in human laryngocarcinoma

Objective To investigate the expression characteristic of metastasis associated 1(MTA1) protein in laryngocarcino-ma tissue and to analyze its relationship with matrix metalloproteinase-9(MMP-9) and vascular endothelial growth factor C(VEGF-C) proteinin .Methods The Western blot method was adopt to detect the expression levels of MTA1 ,MMP-9 and VEGF-C protein in the specimens of normal laryngeal mucosal tissue ,paracarcinoma tissue and tumor tissue from 40 cases of laryngocarcinoma .Re-sults Compared with the groups of laryngocarcinoma without lymph node metastasis ,paracarcinoma and normal tissue ,the expres-sion level of MTA1 protein in the laryngocarcinoma with lymph node metastasis group was significantly increased (P<0 .05) .The expression of MTA1 protein in the laryngocarcinoma tissue was significantly correlated with the lymph nodes metastasis ,tumor staging and differentiation degree of laryngocarcinoma (P<0 .05) and was positively correlated with the expression of VEGF-C and MMP-9 (P<0 .05) .Conclusion MTA1 may participate in the development ,progress and metastasis of laryngocarcinoma .Moreo-ver the regulating effect may exist among MTA1 ,MMP-9 and VEGF-C .
5.Expression and location of YB-1 protein in hepatocellular carcinoma and their clinical significance

ObjectiveTo study the expression levels of YB-1 protein in hepatocellular carcinoma (HCC) tissues,tissues adjacent to tumour and normal liver tissues,and to explore the clinical significance.MethodsTo study the expression levels of YB-1 protein,immunochemistry was carried out on 58 HCC tissues and their corresponding adjacent tissues to tumour and on 20 normal liver tissues.The YB-1 protein with nuclear translocation was detected by Western blot in HCC tissues,tissues adjacent to tumour,normal liver tissues,HCC cell lines including QGY-7701 and SMMC-7721,and a normal hepatic cell line LO2.ResultsPositive signals of YB-1 protein were detected at a high level in HCC tissues (72.4%,42/58) when compared to tissues adjacent to tumour (41.4%,24/58) and to normal liver tissues (35 %,7/20) (P>0.05).The nuclear expression of YB-1 (31%,18/58) was significantly correlated with the pathologicalgrade,tumour size and portal venous invasion (P<0.05).Using Western blot,the YB-1 protein with nuclear translocation was expressed at a higher level in HCC tissues (0.474±0.107) than in tissues adjacent to tumour and in normal liver tissues (P<0.05).Similar results were obtained in HCC cell lines and the normal hepatic cell line (P<0.05).ConclusionThe high expression of YB-1 in HCC and its nuclear translocation may be involved in human HCC progression.YB-1 may be a potential target for HCC treatment in the future.
6.Clinical significance of MRP8 and cypA expression in laryngeal squamous cell carcinoma tissues.

OBJECTIVE:
To study differential expression of MRP8, CypA protein in the patients of laryngeal squamous cell carcinoma (LSCC) and the relationship in the development of LSCC.
METHOD:
Immunohistochemistry was used to detect the expression of MRP8,CypA protein in LSCC tissues of 41 cases and matched paraneoplastic normal tissues of 41 cases,with results compared to the clinical data to determine significance.
RESULT:
The expression of MRP8, CypA protein in carcinoma and normal tissues and composition of different positive grades were in statistical significance (P < 0.01). The expression levels of MRP8 were no significant correlations were identified against any parameter (age,sex and cervical lymphatic metastasis) examined (P > 0.05), but related to pathological stage (P < 0.05).
CONCLUSION
MRP8 protein is on intimate terms with different pathological differentiation stage of LSCC. MRP8, CypA protein may play an important role in the development and progression of LSCC.

7.Construction of a caries diagnosis model based on microbiome novelty score.

OBJECTIVES:
This study aimed to analyze the bacteria in dental caries and establish an optimized dental-ca-ries diagnosis model based on 16S ribosomal RNA (rRNA) data of oral flora.
METHODS:
We searched the public databa-ses of microbiomes including NCBI, MG-RAST, EMBL-EBI, and QIITA and collected data involved in the relevant research on human oral microbiomes worldwide. The samples in the caries dataset (1 703) were compared with healthy ones (20 540) by using the microbial search engine (MSE) to obtain the microbiome novelty score (MNS) and construct a caries diagnosis model based on this index. Nonparametric multivariate ANOVA was used to analyze and compare the impact of different host factors on the oral flora MNS, and the model was optimized by controlling related factors. Finally, the effect of the model was evaluated by receiver operating characteristic (ROC) curve analysis.
RESULTS:
1) The oral microbiota distribution obviously differed among people with various oral-health statuses, and the species richness and species diversity index decreased. 2) ROC curve was used to evaluate the caries data set, and the area under ROC curve was AUC=0.67. 3) Among the five hosts' factors including caries status, country, age, decayed missing filled tooth (DMFT) indices, and sampling site displayed the strongest effect on MNS of samples (P=0.001). 4) The AUC of the model was 0.87, 0.74, 0.74, and 0.75 in high caries, medium caries, low caries samples in Chinese children, and mixed dental plaque samples after controlling host factors, respectively.
CONCLUSIONS
The model based on the analysis of 16S rRNA data of oral flora had good diagnostic efficiency.

8.Impact of LDB3 gene polymorphisms on clinical presentation and implantable cardioverter defibrillator (ICD) implantation in Chinese patients with idiopathic dilated cardiomyopathy.

OBJECTIVE:
Mutations in LIM domain binding 3 (LDB3) gene cause idiopathic dilated cardiomyopathy (IDCM), a structural heart disease with a complicated genetic background. However, the association of polymorphisms in the LDB3 gene with susceptibility to IDCM in Chinese populations remains unexplored as dose the impact on clinical presentation.
METHODS:
We sequenced all exons and the adjacent part of introns of the LDB3 gene in 159 Chinese Han IDCM patients and 247 healthy controls. Then we detected the distribution of polymorphisms in the LDB3 gene in all participants and assessed their associations with risk of IDCM. Additionally, we conducted a stratified genotype-phenotype correlation analysis.
RESULTS:
The A allele of rs4468255 was significantly associated with IDCM (P<0.01). The rs4468255, rs11812601, rs56165849, and rs3740346 were also associated with diastolic blood pressure (DBP) and left ventricular ejection fraction (LVEF) (P<0.05). Notably, a higher frequency of rs4468255 polymorphism was observed in implantable cardioverter defibrillator (ICD) recipients under a recessive model (P<0.01), whereas the significant association disappeared after adjusting for potential confounders. However, in the dominant model, notable correlations could only be observed after adjusting for multi parameters.
CONCLUSIONS
The rs4468255 was significantly correlated with IDCM of Chinese Han population. A allele of rs4468255 is higher in IDCM patients with ICD implantation, suggesting the influence of genetic background in the generation of this response. In addition, rs11812601, rs56165849, and rs3740346 in LDB3 show association with brain natriuretic peptide, DBP, and LVEF levels in patients with IDCM but did not show any association with IDCM susceptibility.
