1.Calculation of Optimal Pharmacokinetic-equation Parameters in Drugs by Intravenous Administration Using Excel Programming Solution

OBJECTIVE:To establish a simple method for calculating the optimal pharmacokinetic-equation parameters in drugs by intravenous administration using Excel programming solution.METHODS:The pharmacokinetic parameters and compartment model parameters after intravenous injection drugs were computed by means of Excel programming solution,and the results were compared with those computed by DAS method or residual method.RESULTS:By means of Excel programming solution,the optimal pharmacokinetic parameters of one-and two-compartment models were obtained,which were completely in line with those computed by DAS program.CONCLUSION:Excel programming solution is applicable for the computation of the pharmacokinetic parameters of one-and two-compartment models following intravenous injection drugs.
2.Design of Individualized Dosage Regimen of Multiple Oral Dosing Theophtlline Based on Excel Function

OBJECTIVE:To establish a simple individualized dosage regimen of multiple oral dosing of Theophyllin. METHODS:Based on pharmacokinetic parameters,Excel function was used to design the dosage regimen of multiple oral dosing extravascular administration of one-compartment model with Theophylline as an example. RESULTS:The following parameters such as plasma drug concentration at any time (t) since administration of Theophylline,peak time,maximum steady plasma-drug concentration,minimum steady plasma-drug concentration,accumulation coefficient,fluctuation percentage,fluctuation amplitude,and dosage for children and the old could be obtained and the concentration-time curves were able to be drawn from the input data including physical and pathological parameters,dosage (X0) of Theophylline,interval ?,absorption rate constant (Ka),drug clearance rate (CL),absorption fraction (F) and apparent volume of distribution (Vd). CONCLUSION:The method adopted in our study is simple,reliable and intuitive,and it is applicable for the design of the individualized dosage regimen of Theophyllin.

6.Study on long-term toxicity of ketoconazle in dogs

Ketoconazole(KET) is a new imi-dazole derivative with broad antimycotic spectrum. In order to verify the clinical toxic and side effect and its properties in animals, we made a long-term toxicity test for 30 days. Dosages of 70, 35 and 17. 5 mg?kg-1?d-1(e-quivalent to 21, 10. 5 and 5. 2 times of the clinical dosage) were given ig to dogs. The salivation , vomiting, anorexia, decrease in heart rate and loss of weight occurred in the large dosage group. Half of the dogs died from toxicosis within ig 15 days. Laboratory examination showed that the activities of ALT, LDH and ALP, the content of T-BIL, BUN in serum in-creased in this group. Pathological examination revealed that there were some pathological changes in the liver, kidneys, adrenal glands and sex gland in the group. There were no significant changes in other dosage groups compared with the normal control group. After withdrawal of KET, all toxic symptoms disappeared and the abnormal indexes were restored. The results indicated that toxic target organs of KET were liver, kidney, adrenal gland and sex glands. The safe dosage for dogs was about 17. 5 mg?kg-1?d-1.
7.Optimized formulation of alkanin liposome by a combination of orthogonal design with central composite design/response surface method

AIM:To select an optiomal formulation of liposome entrapped alkanin prepared with ethanol injection in the combination of orthogonal design and central composite design/Response surface method.METHODS:Five influencing factors,including ratio of alkanin to liposome and soyabean lecithin to cholesterol,volume ratio of ethanol phase to water phase,soybean concentration and mannitol concentration were chosen as indices by orthogonal design.Then the method utilized the central composite design,which fitted multivariate linear and second/third order nonlinear regression equation,to find out the optimal formulation.RESULTS:Third order regression equation gained made the good prediction about entrapment(85.4%)and alkanin load(5.16%),and exhibited no significant difference between theoretic value and measured value.CONCLUSION:The combination of orthogonal design with central composite design is a feasible and convenient method to optimize the prescription of alkanin-liposome.

10.Treatment and medical follow-up of three cases with acute radiation-induced skin injury

Objective To discuss diagnosis,treatment and late effects of three patients with acute radiation-induced skin injury after an accident.Methods Medical history collection and physical examination were made in a manner of one or several doctors to one patient.The general health condition was evaluated through laboratory studies,including complete blood count,liver and kidney function,thyroid function and humoral immunity measurement.The genetic analysis of radiation damage was performed by using chromosomal aberrations and micronucleus assay (CB method).The psychological status of the patients was evaluated during medical follow-up.Results Among the three patients,two were diagnosed with degree Ⅲ acute radiation-induced skin injury and one with degree Ⅱ.The medical follow-up showed that two patients had syndrome of neurasthenia.The two cases with degree Ⅲ acute radiation-induced skin injury then became chronic radiation-induced skin damage.Conclusions Acute radiation-induced skin injury could be persistent and become chronic radiation skin damage.It is important for long-term medical follow-up to be taken for patients with acute radiation-induced skin injury.
