1.Analysis of pulmonary infection and susceptible factors following orthotopic liver transplantation

Objective To investigate the characteristics of pulmonary infection and susceptible factors following orthotopic liver transplantation (OLT). Methods Clinical data of 128 patients who underwent OLT from Feb. 1999 to Dec. 2004 were studied retrospectively in order to analyze primary pathogens, infectious time and susceptible factors.Results Forty-eight ( 37.5 %) of 128 patients had pulmonary infections and 27 ( 56.3 %) of them developed within postoperative 7 days. Thirty-four ( 70.8 %) cases suffered from mixed infection and 6 ( 12.5 %) died in the hospital after OLT. The primary pathogenic germs included Pseudomonas aeruginosa, Acinetobacter baumannii/haenolyticus, Golden staphylococcus, Aspergilosis and so on.Conclusion Pulmonary infection can be caused by various pathogens and associated with patients' constitution, mechanical ventilation, immunosuppressive drugs and so on.
2.Apoptosis of breast cancer cell line induced by small direct-current electric field and its mechanism

ObjectiveTo research the apoptosis of breast cancer cells by application of small direct-current electric field(EF) and its related possible mechanism.MethodsSmall direct-current EFs were applied in human breast cancer cell line MCF7 in vitro to observe the effects on apoptosis of cancer cells by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling(TUNEL)and to detect the mRNA expression of p53, Rb and E2F1 genes by reverse transcriptase-polymerase chain reaction (RT-PCR).ResultsA lot of cancer cells were observed slaughed from culture dish by 200mV/mm of the field strength and the number of the apoptotic cancer cells was gradually significantly increased with the application of EFs(P<0.01). mRNA expression level for p53 and Rb genes was elevated but that for E2F1 gene was decreased overtly in cancer cells exposed to EFs.ConclusionApoptosis of breast cancer cells can be induced by small direct-current EFs and the change for expression of p53, Rb and E2F1 genes may be involved in the mechanism.

ObjectiveTo observe the apoptosis of breast cancer cells induced by small direct-current electric field (EF) and its related possible mechanism in expression of p21 and p16 genes.MethodsSmall direct-current EF was applied in human breast cancer cell line MCF7 in vitro and the effect of EF on apoptosis and expression of p21 and p16 important genes of breast cancer cells was observed.ResultsThe small direct-current EF had an obvious apoptosis inducing effect on breast cancer cells. A lot of cancer cells were sloughed from culture dish by 200 mV/mm of the EF strength. The number of the apoptotic cancer cells was gradually significantly increased and mRNA expressions of p21 and p16 genes were raised following the EF strength being up (P<0.01).ConclusionThe small direct-current EF has an obvious apoptosis inducing effect on breast cancer cells. It may be related with the expression of p21 and p16 genes.
4.Piceatannol inhibits prostate cancer cell proliferation, migration and invasion

AIM:To investigate the effect of piceatannol on the viability, and the abilities of migration and invasion in the prostate cancer cells.METHODS:DU145 cells were treated with piceatannol at different doses (0, 5, 10, 20, 40 and 80 μmol/L) for different time (12, 24, 36 and 48 h) as indicated.The cell viability was assessed by CCK-8 assay.The migration and invasion abilities of the cells were analyzed by wound healing assay and Transwell assay, respectively.The protein levels of p-JAK2 and p-STAT3 were detected by Western blot.RESULTS:Piceatannol dose-dependently decreased the cell viability.After treatment with piceatannol, the abilities of migration and invasion of the cells were significantly inhibited.Moreover, treatment with piceatannol resulted in marked decreases in the protein levels of p-JAK2 and p-STAT3.CONCLUSION:Piceatannol inhibits the viability, migration and invasion of the prostate cancer cells via regulating the JAK2/STAT3 signaling pathway.
5.Co-transplantation of acellular allogeneic dermis and autologous split-thickness skin for burn repair at functional parts

BACKGROUND:Autologous split-thickness skin grafting is the main therapy for burn repair at functional sites, which has achieved certain effects, but there are stil some deficiencies, such as poor texture, stiffness and poor toughness, as wel as severer hyperplasia that is easy to result in contracture deformity and poor functional recovery. OBJECTIVE: To analyze the clinical efficacy of skin co-transplantation on burn repair at functional sites. METHODS:Sixty patients with burns at functional sites (n=84) were randomized into two groups: co-transplantation of acelular dermis and autologous split-thickness skin in experimental group and autologous split-thickness skin graft in control group. Survival rate of skin flap and rate of secondary operation were compared between two groups. At 1 month after transplantation, Vancouver Scar Scale was used to assess skin color, thickness, blood vessel distribution and flexibility, and meanwhile, the severity of scar was determined. RESULTS AND CONCLUSION:The survival rate of skin flap was significantly higher in the experimental group than the control group (93%vs. 70%,P < 0.05), and the rate of secondary operation was significantly lower in the experimental group compared with the control group (0vs. 13%,P < 0.05). At 1 month after transplantation, scores on the skin color, thickness, blood vessel distribution and flexibility were al lower in the experimental group than the control group (P < 0.05), but the incidence of mild hyperplasia in the experimental group was significantly higher than that in the control group (52% vs. 29%,P < 0.05). These findings indicate that co-transplantation of acelular alogeneic dermis and autologous split-thickness skin for burn repair at functional sites can effectively enhance the survival rate of skin flap, reduce the rate of secondary operation, contribute to wound healing and reduce the severity of hyperplasia.
6.Effects of adiponectin siRNA on the glucose transport in 3T3-L1 adipocytes

AIM:To construct the adenovirus vector with adiponectin (Acrp30) siRNA, and to observe its effect on the Acrp30 expression and glucose transport in 3T3-L1 adipocytes. METHODS: Mouse Acrp30 siRNA fragment was designed, synthesized and cloned into the adenovirus vector. 3T3-L1 cells were infected with the two recombinant adenoviruses, respectively. The mRNA expression and protein levels of Acrp30 in these cells were evaluated by RT-PCR and ELISA. Glucose transport was measured by 2-Deoxy-[3H]-D-glucose incorporation method. RESULTS: The recombinant adenoviruses were successfully constructed. 7.TACE combined with microwave ablation for the treatment of large-sized hepatic carcinoma:a preliminary study

Objective To evaluate the efficacy and safety of transcatheter arterial chemoembolization(TACE)combined with per-cutaneous microwave ablation(MWA)in treatment of large-sized hepatic carcinoma.Methods A total of 84 patients with large-sized hepatic carcinoma were randomly and equally divided into the study group(n=42)and control group(n=42).TACE combined with MWA was carried out in the patients of the study group,while only TACE was performed in the patients of the control group.Results The effective rate of the study group and the control group was 71.4% and 42.8% respectively,and the difference between the two groups was statistically significant(P <0.05).In the study group,the survival rates at 6,12,18,24 months after the treatment were 88.1%,73.8%,52.3% and 33.3% respectively,while in the control group the survival rates were 76.2%,57.1%,30.9% and 9.5%respectively.There was no statistically significant differences in the postoperative complications between the two groups.Conclusion TACE combined with MWA appears to be an effective approach for the treatment of large-sized hepatic carcinoma.The effect of combined therapy is obviously superior to the only TACE.
8.Protective effect of hydrogen-rich saline solution on lung injury in rats with severe acute pancreatitis

Objective To explore whether intravenous injection of hydrogen‐rich saline having the protective effect on sodium taurocholate induced severe acute pancreatitis(SAP) associated lung injury(APALI) in rats and its possible mechanisms .Methods Fifty‐four healthy male SD rats were randomly divided into sham‐operation group (Sham group) ,model group (SAP+ NS group) and hydrogen water treatment group (SAP + HRS group) ,and each group was subdivided into 6 ,12 ,24 h subgroups .Six rats were killed at each time point for collecting serum ,lung tissue and pancreas tissue .Serum TNF‐αand IL‐1βlevels ,lung wet /dry weight ratio ,expression of TNF‐αmRNA and IL‐1βmRNA in the lung tissue were detected .The pathological evaluation of pancreas and lung tissue injury was performed .Results (1)The levels of TNF‐α and IL‐1β in serum ,pancreas and lung tissue pathological scores ,TNF‐αmRNA and IL‐1βmRNA expression levels in the lung tissue and lung wet dry weight ratio at the time points of 6 , 12 ,24 h in the SAP+NS group and the SAP+ HRS group were higher than those in the sham group (P<0 .05) .(2) Compared with the SAP+NS group ,the levels of serum TNF‐α,TNF‐αmRNA expression level in the lung tissue and lung wet dry weight ra‐tio at all time points in the SAP+ HRS group were lower(P<0 .05);the levels of serum IL‐1β,pancreas and lung tissue pathologi‐cal score and IL‐1β‐mRNA expression at 6 h in the lung tissue had no statistical difference between the SAP+NS group and SAP+HRS group ,but which at time points of 12 ,24 h in the SAP+ HRS group were lower than those in the SAP+NS group(P<0 .05) . Conclusion HRS realize the protection on APALI possibly via its elective anti‐oxidation action for inhibiting oxidative stress injury related cytokines expression .
9.Synchronous papillary thyroid carcinoma and medullary thyroid carcinoma – a pitfall waiting to happen

Papillary thyroid carcinoma (PTC) is the most common thyroid carcinoma and is derived from
thyroid follicular cells. In contrast, medullary thyroid carcinoma (MTC) is rare and originates
from the parafollicular C-cells. Synchronous occurrence of these two carcinomas is uncommon and
occurs as either discrete lesions or as a mixed lesion. The current case report describes a 50-year-old
woman with synchronous multiple discrete MTC and PTC with lymph nodes metastasis. Pathologists
and treating physicians should be aware of the synchronous coexistence of these entities to avoid
possible misdiagnosis.
10.Study on the Effect of Dark Tea Fermentation Liquid with Eurotium cristatum on the Activity of Digestive Enzyme

The effect of dark tea fermentation liquid with Eurotium Cristatum on the activity of digestive enzyme was researched, as fellowing amylase, protease, lipase. The results showed that dark tea fermentation liquid with Eurotium Cristatum may increase remarkably the activity of ?-amylase and protease, but decrease efficiently the activity of lipase. The fermentation liquid improves the digestion and absorption of starch and protein, but inhibits the decomposition and absorption of fat, so it can explain the mechanism of Fu-brick tea's health functions.
