1.Platelet-activating factor induced bronchoconstriction and possible mechanisms

The effect of platelet activating factor (PAF) on bronchial smooth muscleboth in vitro and in vivo in guinea-pigs were observed and its mechanism explored. Theresults showed: 1. Bronchoconstriction can be induced by PAF both by i. v. and via aerosoladministration and was accompanied with a fall of the circulating platelet count. 2. Thecontractile effect of PAF on guinea-pig airway smooth muscle preparations is modest invitro. Yet this effect of PAF can be enhanced by platelet. 3. TXA_2 receptor antagonist(AA-2414) can inhibit bronchoconstriction induced by PAF with platelet significantly.These results suggested that PAF might be a potent endogenous bronchoconstrictor and itseffect was related to platelet. The activated platelets can induce bronchoconstriction byreleasing TXA_2.
2.Determination of the Contents of Neferine and Liensinine in Plumula Nelumbini by TLC Scanning

OBJECTIVE:To establish the method for determining the contents of neferine and liensinine in plumula nelumbini.METHODS:Neferine and liensinine in plumula nelumbini were extracted by ultrasound method and determined by TLCs.RESULTS:The average recovery and relative standard deviation were(97.91?2.49)% and (99.28?3.22)%,respectively.The correlation coefficient were 0.996 and 0.999,respectively.CONCLUSION:The method,operated simply and attained the reliable results with good reproducibility,can be used to determine the contents of bioactive alkaloid in plumula nelumbini or other compound preparations.

4.Applied anatomical and clinical application of the lateral approach to the vertebral lamina for treating extreme lateral lumber dsic herniation

[Objective]To provide microsurgery anatomical data for the surgical operation in treating of extreme lateral lumbar dsic herniation through the observations of the regional anatomic structure about the post-median approach tolateral vetebral lamina(PMALVL).[Method]Thicknesses of intertransverse ligament,the distribution of anterior branches of lumbar nerve and vessel were measured at the adult cadavas specimens.Retrospectively analyzed with 12 cases of extreme lateral lumbar disc herniation from January 2004 to January 2006,including 7 cases were treated by the lateral approach to the vertebral lamina,5 cases treated by PMALVL.[Result]The thickness of intertransverse lingament is 0.6~1.3 mm.More than 90 percent of anterior branches of lumbar segmental arteries and the accompanying veins from L_3 to S_1 were located ventrally in the superior portion of the intertransverse ligament.A branch from Lumbar artery permanently locates in the hfiddle of post-lateral intertransverse ligament.The angle between the anterior branch of Lumbar nerve root and midline sagittal plane is 18.9?~39.2?.The distance from the root of the tranverse to the anterior branch of lumbar nerve root and from the lateral margin of the pars interarticul aris to the anterior branch of lumbar nerve root(5.6~8.0)mm and(1.7~3.6)mm respectively.The period of follow up in 12 cases was from 3 months to 24 months,with an average of 10 months.The resultwas excellentin 8 cases,good in 3 cases,total fine rate according the classification of Nakal was 91.67%.[Conclusion]Intertransvarse ligament is a reliable landmark for PMALVL and treating extreme lateral lumbar disc herniation through PMALVL may get microtraumatic and excellent effect.

Objective To study the stability of potassium dehydroandrographolide succinate solution in different conditions.Methods The classical thermostatic test was adopted to investigate the stability of potassium dehydroandrographolide succinate in phosphate buffer solutions of different pH values,in different buffer solutions of the same pH value and in citrate buffer solutions of different concentrations.Results The stability of potassium dehydroandrographolide succinate solution was different at different pH values and was the best at pH 6.5.In various buffer solutions of pH 6.5,the hydrolyzation of potassium dehydroandrographolide succinate solution was delayed by citrate buffer solution,and the stability of the solution had no correlation with the concentration of citrate buffer solution.Conclusion The stability of potassium dehydroandrographolide succinate solution varies in different conditions.This will supply a reference for the study on potassium dehydroandrographolide succinate solution and its similar preparations.

8.Protective effects of astragalus root on hippocampal nerves and learning and memory ability of newborn rats after anoxic-ischemic brain injury

BACKGROUND: Astragalus root can inhibit apoptosis through reducing the release and interstitial accumulation of excitatory amino acids, alleviating calcium overloading and antioxidative effect.OBJECTIVE: Astragalus root was used to treat anoxic-ischemic brain injury in immature brain. We evaluated the effect of astragalus root on caspase-3 mRNA expression, and meanwhile, labyrinth test was employed to investigate the intervention of astragalus root on learning and memory function of mature rats after anoxic-ischemic brain injury.DESIGN: Randomized and controlled study.SETTING: Pediatric Department, Zhongda Hospital Affiliated to the Medical College of Southeast University; Pathological Department, the Basic Medical Sciences Institute of Southeast University.MATERIALS: From October 2002 to June 2003, this study was conducted at the Experiment Center of the Medical College, Southeast University.A batch of 114 seven-day-old SD rats were selected from the same brood and divided into 3 groups, namely, sham-operation group (n=18), model group (n=48) and astragalus root group (n=48). Astragalus injection was produced by Chengdu DIAO Pharmaceutical Factory, with 10 mL astragalus injection corresponding to 20 g raw material.METHODS: Animal model of anoxic-ischemic brain injury was established in model group and astragalus root group, but was not established in sham-operation group. In astragalus root group, immediately after establishing anoxic-ischemic model and at the same time point each day, 0.08 mL astragalus injection was administered intraperitoneally until the 7th postoperative day. In model group, 0.08 mL normal saline was administered at the same time points. In sham-operation group, no treatment was given. In astragalus root group and model group, animals were decollatedat 24 hours and 5 days postoperatively to take out the brains. In sham-operation group,animals were decollated and their brains were taken out at 24 hours postoperatively. In all the groups, hippocampal brain injury was detected using histopathological method combined with semi-quantified RT-PCR methods for detecting caspase-3 mRNA. Adult rats aged 90 days were used in modified y maze to examine their learning and memory functions. All these three experiments were independent.MAIN OUTCOME MEASURES:① Hippocampal brain injury in each group was evaluated using pathological method.② Caspase-3 mRNA in the ligated side of hippocampus was detected.③ Results of modified Y maze test were analyzed.RESULTS:All of the 114 rats entered the statistical analysis.① Assessment ofhippocampal brain injury in each group with pathological method:In sham-operation group, the bilateral hippocampus showed no swelling or necrosis, and neural cells in this area had normal morphological features with a density of (87.7±0.6) × 103 per high amplification field. In model group, the ligated side of hippocampus was swollen with a widened spatium and the cell density decreased to (68.8±3.0) × 103 per high amplification field, which significantly differed from that in sham-operation group (P < 0.01). At the fifth day, the volume of ligated side of hippocampus reduced with pyramid layer disorganized and neural cells sparse at a density of (48.7±2.2) × 103 per high amplification field. These changes were significantly different from those of sham-operation group and the same side at 24 hours (P < 0.01). At 24 hours the ligated side of hippocampus was less swollen in astragalus root group than in model group.At day 5, the whole hippocampus was observed. At these two time points,cell death rate in astragalus root group was significant lower than that in model group(P < 0.01).②Caspase-3 mRNA in the ligated side of hippocampus in all the groups: In sham-operation group, the expression of caspase-3 was low, with an absorbency value of 0.220±0.009. In model group, after ischemia and anoxia its expression increased. At 6 hours, it was 11% higher than that in sham-operation group. In astragalus root group, mRNA level reached its peak, which was 260% higher than that in sham-operation group (P < 0.01). The peak of mRNA continued, decreased after 48 hours and returned to baseline at 5 days and 7 days. The fluctuation of mRNA was similar between astragalus root group and model group,but the peak value at 24 hours and 48 hours in astragalus root group was 44%-46% lower than that in model group (P < 0.01). ③ Results of modified Y maze test: As compared to model group, in astragalus root group, the number of training times for meeting the standard made by the Association was significantly smaller [(45.7±2.7), (16.1±2.5) times, P < 0.01] and at 24 hours after anoxia and ischemia, memory retention was significantly higher [(48.3±11.7), (80.0±9.0)%, P < 0.01].CONCLUSION: Astragalus root can effectively inhibit the apoptosis of neural cells in hippocampus in immature brain after anoxia and ischemia and enhance the survival rate of them. This protective effect may be related to its inhibitory effect on the expression of caspase-3. Meanwhile, astragalus root can dramatically improve learning and memory function of the immature brain after anoxia and ischemia.
9.Expression of Ezrin and its significance in giant-cell tumor of bone

Objective To detect the expression of Ezrin in giant-cell tumor of bone,and to investigate its cilincal significance. Methods 60 cases of biopsy which had been confirmed as bone giant-cell tumors in our hospital from January 2008 to December 2013 were set as observation group;tumor tissues from 8 cases of reactive new bone in nonmalignant bone diseases,12 cases of osteoid osteoma and 11 cases of osteoblastoma in the corresponding period were set as control group. Protein and gene levels of Ezrin were tested with Western blotting method and real-time PCR detection,simultaneously proceeded the corresponding analysis combined with the clinical data of patients;60 cases of bone giant-cell tumor patients accepted tumor resection and pros-thesis replacement,2 courses of preoperative chemotherapy;mitochondria morphological changes of tumor tissue and Ezrin protein and genetic changes were observed before and after chemotherapy. Results In the giant-cell tumors of bone,the Ezrin protein mainly located in the cytoplasm,and its expression positive rate was much higher than that in reactive new bone of nonmalignant bone diseases(19. 7% ),osteoid osteoma(21. 2% )and osteoblastoma(20. 7% );the difference was statistically significant(χ2 = 4. 18,P = 0. 024),but no statistical difference in the Ezrin expression among the groups of osteosarcoma,osteoid osteoma and osteblastoma(χ2 =6. 18,P = 0. 087). In the giant-cell tumors of bone tissue after chemotherapy,mitochondria pyknosis and the phenomenon of liquid cavitation was less than that before the treatment,and Ezrin protein expression decreased and gene levels reduced[(23. 99 ± 1. 49)vs(20. 11 ± 1. 11),t = 5. 03,P = 0. 018)]. Conclusion The expression of Ezrin in giant-cell tumor of bone is much higher than other benign bone tumor,and it could be a biological marker for differentiating benign and malignant bone tumor. Early intervention in Ezrin may be helpful for reatment of giant-cell tumor of bone.
10.Effects of baicalin on the expression of TAFI,NF-κB,and ACE2 in the atherosclerotic rats

Objective To explore the effects of baicalin on the expression of TAFI,NF-κB,and ACE2 in the atherosclerotic rats and discuss the anti-atherosclerosis mechanisms of baicalin.Methods The subjects were healthy male Wistar rats.The rats were divided into sham operation group,model group,baicalin high dose group (1 50 mg·kg-1 ·d-1 )and baicalin low dose group (100 mg·kg-1 ·d-1 ).Plasma TAFI activity were detected with enzymatic assays.The expression of NF-κB and ACE2 were detec-ted with immunohistochemical staining.Results Compared with sham group,the rest groups of TAFI and ACE2 activity were sig-nificantly higher (P <0.05),and the expression of NF-κB,had significant decreased (P <0.05).Compared with the model group, the level of TAFI and ACE2 in baicalin drug group was significantly lower than the model group (P <0.05),and the expression of NF-κB,had significant increased.Conclusion Baicalin can reduce TAFI and ACE2 level,and upregulate the expression of NF-κB to play its role in anti atherosclerosis.
