1.Relationship between FAT10 expression and biological behaviors in infitrating ductal carcinoma of breast

Objective: To study the relationship between FAT10 expression and biological behaviors in inif trating ductal carcinoma of breast. Methods: The expressions of diubiquitin (FAT10), estrogen receptor (ER), progesterone receptor (PR) and c-erbB2 in 50 cases of inif trating ductal carcinoma of breast were detected by immunohistochemistry. Western blot was used to detect FAT10 expression in MB-MDA-435, MB-MDA-435-transfected with FAT10 siRNA expression plasmid, MCF-7 and MCF-7-transfected with FAT10 expression plasmid, respectively. Transwell was used to detect invasion capability of MB-MDA-435, MB-MDA-435-transfected with FAT10 siRNA expression plasmid, MCF-7 and MCF-7-transfected with FAT10 expression plasmid. Results: hTe expression intensity of FAT10 was signiifcantly correlated to patho-grading, lymph nodes metastasis, distant metastasis and TNM staging (P<0.01), but not to age of patients and tumor sizes in iniftrating ductal carcinoma of breast (P>0.05). hTe expression intensity of FAT10 in receptor- negative group was obviously stronger than that in receptor- positive group (P<0.01). hTe expression intensity of FAT10 in triple-negative breast cancer was signiifcantly stronger than that in non- triple-negative breast cancer (P<0.01). hTe survival rate of patients with FAT10 positive expression was significantly lower than negative ones (P<0.05). Western blot results showed that FAT10 intensity in MB-MDA-435 significantly higher than that in MCF-7. Up-regulation expression of FAT10 could obviously increase the invasion capability of MCF-7, and down-regulation of FAT10 could signiifcantly decrease the invasion capability of MB-MDA-435 (P<0.01). Conclusion: FAT10 might increase the invasion capability of breast cancer cells by direct or indirect ways, and play an important role in invasion and metastasis of breast cancer. FAT10 might be an independent index for evaluation of breast cancer prognosis, and a potential target for breast cancer therapy, especially for triple-negative breast cancer.
2.Application of Multivariate Statistics Analysis in HPLC Fingerprint of Shenmai Injection

Objective: To establish an effective fingerprint analysis. Methods: Cluster analysis and mahalanobis distance analysis were adopted to analyze data of HPLC fingerprint of shenmai injection. Results: Precise classification of 18 samples was done by cluster analysis and mahalanobis distance analysis. Conclusion: Authors believe that multivariate statistics analysis applied to fingerprint can be recommanded in the quality control of shenmai injection.
3.Isolation and culture of neural stem cells in ventral midbrain of a rat embryo

ObjectiveTo establish a method of isolation and culture of neural stem cells(NSCs). MethodsTissues of ventral midbrain were isolated from a rat embryo,and the NSCs were cultured stimulated with basic fibroblast growth factor(bFGF)in vitro.The cells were identified by immunocytochemistry.ResultsNSCs proliferated into neurosphere in symmetric and non-symmetric cleavage ways,and differentiated into neuron, astroglia and oligodendrosyte. ConclusionsThe method has been established to isolate and culture the neural stem cells.
4.Effect of Yiqi Xiaozheng granules on quality of life in patients with advanced malignant tumors

BACKGROUND: Progressive body mass losses and physical strength decrease are common complications of malignant tumors, and they are important factors that affect the quality of life of the patients.OBJECTIVE: To probe into the curative effect of a Chinese herbal compound Yiqi Xiaozheng granules combined with antitumor drugs in the treatment of malignant tumors and its effect on the quality of life.DESIGN: A randomized controlled observation.SETTING:Clinical Tumor College of Peking University.PARTICIPANT: Totally 61 patients with malignant pulmonary carcinoma,breast or intestinal carcinoma hospitalized in the Wards of Integrated Traditional Chinese and Western Medicine of Beijing Cancer Hospital from June 1999 to March 2002 were recruited.METHODS: Altogether 61 cases of malignant tumor were randomized into the treatment group (treated with Chinese herbal compound Yiqi Xiaozheng granules supplemented with chemotherapy)(n=34) and the control group (treated with only chemotherapy was used)(n=27). The difference in curative effect, survival period with tumor and the quality of life of the two groups were observed. The comparison of the curative effects of the two groups was performed according to Clinical Curative Effect Criteria for Solid Tumors stipulated by World Health Organization: completely response, partly response, stable disease, and progressive disease. X-ray examination was performed on the patients before the therapy and two courses of treatment after the therapy to evaluate the curative effect. Physical strength grading was made according to the evaluation criteria of Eastern Oneology Cooperative Group of the United States. The two groups were graded before and after the treatment. The group with decreased grade after the treatment was considered to be improving, but the group with increased grade after the treatment was considered to be worsening.symptom before and after therapyparison of the curative effect of the two groups: The cure rate of the treatment group was higher than that of the control group (partial response rate,stability rate and progression rate of the two groups was 36% ,7% ;52%,survival with tumors: The mean period of survival with tumors of the treatment group was longer than that of the control group until March 2002ing: Physical strength grading status was better after the therapy in the treatment group than that in the control group [ In the treatment group, 9cases (26%)were ameliorated, 25 (74%)appeared stable , none was aggravated; in the control group, 1 case (4%) was ameliorated, 25(93%) appeared of qi-deficiency symptoms in traditional Chinese medicine before and after the treatment between the two groups. Symptoms such as sweating and qi deficiency manifested as disclination to talk, listlessness, debility, etc. were significantly reduced in the treatment group (The numbers of cases before and after the treatment were 20, 9; 17,9; 20,13 respectively). In the control group, the cases either were stable or showed an increase in symptoms (There were 13, 13; 14, 17; 18, 20 respectively before and after treatment).CONCLUSION: Compared with the control group, in the treatment of malignant tumors, Yiqi Xiaozheng granules supplemented with chemotherapy not only enhanced curative effect, but also improved the quality of life and prolonged the period of survival with tumors.
5.HPLC Determination of Paeoniflorin, Hesperidin and Glycyrrhizic Acid in Yiwei Capsules

Objective: To monitor the quality and production procedure of Yiwei Capsules from various aspects. Methods: The method of HPLC gradient elution analysis was adopted to determine paeoniflorin, hesperidin, and glycyrrhizic acid of Yiwei Capsules at the same time. Results: Good results were obtained in recoveries and coefficient of variation of the three substances. Conclusion: In this method the pretreatment is simple and the operation is easy which is helpful in quick and accurate determination.
6.Safety of Ultrasound-guided Multifetal Pregnancy Reduction in the Second Trimester

PurposeTo evaluate the safety and feasibility of transabdominal ultrasound guided multifetal pregnancy reduction performed in the second trimester.Materials and Methods Forty-three women receiving embryo implantation were divided into two groups according to whether they have received multifetal pregnancy reduction including 16 in the reduction group in the second trimester to reduce multiple fetuses to twins, and 27 with twin pregnancy and no reduction. The pregnancy outcome was compared.Results In the reduction group, 1 patient had miscarriage because of intrauterine infection; 1 patient lost one of the retained fetuses in two days after reduction and delivered the remaining fetus. In the other 14 cases, 8 delivered and 6 were still in pregnancy. This group had 17 newborns, all without neonatal death or birth defects. In the non-reduction group, there was 1 spontaneous abortion. In 2 patients, 1 of the twin embryos stopped development. 1 case had a spontaneous abortion, the other 24 cases delivered uneventfully. There were 50 newborns in this group with 3 birth defects and no neonatal death. There was no statistically significant difference in gestational age at delivery, birth weight, and the incidence of maternal and neonate complications (P>0.05).Conclusion Multifetal reduction improves pregnancy outcomes, similar to non-reduction twin pregnancy. It is safe and feasible to perform multifetal pregnancy reduction in the second trimester.
7.Effects of Naofucong grain on brain NMDA receptor in experimental dementia mice

ObjectiveTo observe effects of Naofucong grain on learning and memory in cerebral ischemic mice, and identify the mechanisms on N-methyl-D-aspartate (NMDA) receptor.MethodsExperimental dementia mice were induced by ischemia reperfusion injury, and treated with Naofucong grain or Dangguishaoyao powder (positive control medicine). Learning and memory were tested in the Morris water maze. Activities of NMDA receptor in brain cortex and hippocampus were detected ResultsIncubation periods of mice treated with Naofucong grain were significantly shorter than that of ischemic model mice (P<0.01) Activities of NMDA receptor in brain cortex and hippocampus showed no difference between Naofucong grain treated mice and normal mice (P>0.05)Activities of NMDA receptor in brain cortex and hippocampus in Naofucong grain treated mice were significantly lower than that in ischemic model mice(P<0.01) ConclusionThe improvement of Naofucong grain in the memory and learning in cerebral ischemic mice may be related to the inhibition of the activity of NMDA receptor.
8.Studies on the protective effect of drug serum taken from rats administered with Naofucong in PC12 cell ischemic model

ObjectiveTo examine effects of traditional Chinese herbs Naofucong grain on PC12 cells injured by H2O2 in vitro MethodsRats random divided into 2 groups: Naofucong grain treating group (administered with Naofucong grain) and control group (administered with distilled water) After 3 days administration, serums were obtained and added to cultured PC12 cells which were under different concentrations of H2O2 Morphological examinations in PC12 cells were detected by MTT measurement ResultsMTT measurement showed that Naofucong grain administered rats serums significantly increased the survival number of PC12 cells in different concentrations of H2O2.Conclusion Naofucong grain may have obvious protective effects on PC12 cells injured by H2O2.
9.Effects of β-amyloid peptide on the monoamine oxidase-B, interleukin-6 in the brain of mice and the interfering effect of traditional Chinese medicine

ObjectiveTo observe the changes of monoamine oxidase-B (MAO-B), interleukin-6(IL-6) in the cortex and hippocampus of Alzheimer's disease model mice and the effect of traditional Chinese medicine Naofucong.MethodsAlzheimer's disease model was induced in mice by β-amyloid(Aβ)25-35 icv. Space learning and memorial ability was tested in Morris water maze. The activity of MAO-B was measured by colorimetric method. IL-6 was observed with the immnuohistochemical stain.ResultsMice in the model group presented longer latent periods of Morris water maze(81.3±13.4)s, higher activities of MAO-B in brain cortex and hippocampus (120.12±10.15,83.60±5.29) compare with that of the control group, which was (34.2±10.9)s,(93.09±10.54) and (50.39±9.16)(P<0.05~0.01).There were many IL-6 positive cell in dentate gyrus of hippocampus of the model group. After administration with Naofucong grain, latent periods (43.7±12.7) s and activities of MAO-B (47.11 ± 6.57)in hippocampus were recovered(P<0.05~0.01), and the IL-6 positive cell in dentate gyrus decreased.ConclusionNaofucong grain can antagonize the Aβ25-35 toxicity by decreasing the overactivition of MAO-B and the excretion of inflammatory medium by microglia, as well as improve the memory function.
10.Survival and differentiation of transplanted neural stem cells in the brain of MPTP-induced Parkinson's disease model mice

ObjectiveTo determine survival and differentiation of cultured neural stem cells (NSCs) into viable and functional neurons upon transplantation into the brain of MPTP-induced Parkinson's Disease (PD) mode micel.MethodsMice model of PD was established with intraperitoneal (i.p.) injections of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP,40mg/kg) twice interspersed by 16 hr. Tissues isolated from an embryonic rat midbrain were cultured in clonal density. After labeled with 5-bromo-2′-deoxyuridine (BrdU), the NSCs were transplanted into the uni- or bilateral striatum of PD mice. Tyrosine hydroxylase (TH) immunofluorescence was used to evaluate the toxicity of MPTP on the neural cells in the substantia nigra. Immunohistochemistry and laser confocal were used to detect the survival and differentiation of transplanted NSCs.ResultsTH positive neural cells were significantly reduced in the substantia nigra after MPTP injection. Immunohistology showed that the uni-or bilateral transplanted NSCs could survive in the brains of PD model mice.Some transplanted NSCs could properly differentiate into targeted TH positive neural cells in vivo.ConclusionsThe transplanted multipotent NSCs could survive, differentiate into functional dopamine neurons in the brains of PD model mice.
