1.Clinical Observation of Aciclovir and A-Polyresistin Theraphy on Children with Recurrent Oral-Ulceration

Objective To observe the effect of immunity condition and clinical manifestations of children with recurrent oral-ulcer(ROU)treated with aciclovir(ACV) and a-polyresistin.Methods Forty-eight patients with ROU were randomly divided into whole treated group(25 cases)and partial treatment group(23 cases).The changes of their peripheral blood lymphocyte subgroups,before and after treatment,were measured and contrasted to analyze the relationship with curative effect.Results The ratios of CD3,CD4,and CD4/CD8,after the treatment,increased obviously,while CD8 lowered obviously in treatment group(P0.05).The total clinical efficacy rate was 47.82%,moreover the patients were recurrent after 1 year.Conclusions ACV and a-polyresistin,through antivirus and regulating the immunity function get the 96% total clinical efficacy rate,and that controle the recurrent rate.It is superior to the traditional method.

5.In vitro inhibition of trichosanthin-monoclonal antibody conjugate on human melanoma cells

We have constructed an immunotoxin(Ng76-TCS),which was composed of a monoclonalantibody directed against human melanoma and trichosanthin(TCS)——a single chain ribosomeinactivating protein.The cultured human melanoma cells(M21)were inhibited effectively byNg 76-TCS.The cytotoxicity of Ng76-TCS to M21 cells was 2,000-fold higher than that of free TCS and Ng76 mixture.A conjugate,which was prepared with normal mice immunoglobulinand TCS(NIgG-TCS),was 160-fold less cytotoxic to M21 cells.Meanwhile Ng76-TCS was125-fold less cytotoxic to nontarget cells Hela.These results showed that the immunotoxinNg76-TCS was a potent and specific anti-human melanoma agent.
6.A clinical study on integrated traditional Chinese medicine(TCM)and western medicine in treatment of acute exacerbation of chronic obstructive pulmonary disease combined with respiratory failure,TCM syndromes of spleen-kidney-yang deficiency and phlegm-dampness syndrome

Objective To evaluate the efficacy of integrated traditional Chinese medicine(TCM)and western medicine in treatment of acute exacerbations of chronic obstructive pulmonary disease(AECOPD)combined with respiratory failure,TCM syndromes of spleen-kidney-yang deficiency and phlegm-dampness by comparison between the integrated therapy and simple western therapy in treatment of the disease. Methods 160 patients with AECOPD combined with respiratory failure,spleen-kidney-yang deficiency and phlegm-dampness syndrome in the intensive care units(ICU)of Affiliated Hospital of Chengdu University of TCM and other four hospitals were randomly allocated into two groups in this double-blinded,multicenter,prospective,randomized,controlled trial. In the control group (78 cases),western medicine and placebo were given to the patients,and in the treatment group(82 cases), conventional western medicine plus fei-shuai mistura 25 mL were administered,four times per day,the therapeutic course lasting for 2 weeks in both groups. The all-cause mortality,respiratory failure-cause mortality,improvement of modified Medical Research Council(mMRC)Dyspnea Scale grades,6 minutes walk distance(6MWD),the forced expiratory volume in 1 second/forced vital capacity(FEV1/FVC)were observed in the 28 days after the end of treatment. Results In the comparisons between the control and treatment groups,there were no statistical significant differences in the all-cause mortality〔54.87%(45/82)vs. 64.10%(50/78)〕and the cases of FEV1/FVC(both P>0.05)in the 28 days after the end of treatment;the 28 day respiratory failure-cause mortality was significantly decreased〔19.51%(16/82)vs. 33.33%(26/78),P<0.05〕,the number of patients with mMRC Dyspnea Scale grades (1-2)was obviously increased(22 cases vs. 7 cases,P<0.05),and the number of patients with 6 MWD grades (4-6)was markedly enhanced in the treatment group(21 cases vs. 8 cases,P<0.05). Conclusions The integrated TCM and western medicine has better therapeutic results in improvement of the patients' degree of dyspnea, 6 MWD and respiratory failure mortality than simple treatment with western therapy for treatment of patients with AECOPD combined with respiratory failure, spleen-kidney-yang deficiency and phlegm-dampness syndrome. However,in regard to the effect on pulmonary function and all cause mortality,the integrated therapy for treatment of such patients in short term has no significant effect.

8.Correlation of XPD gene with susceptibility to gastric cancer

Background and Objective: Mutations in DNA repair system are related to carcinogenesis.This study was to evaluate the correlations of polymorphisms and haplotypes of XPD gene with individual susceptibility to gastric cancer. Methods: Genomic DNA were extracted from peripheral blood leukocytes of 207 gastric cancer patients and 212 healthy controls. Genotypes at codon 312 and codon 751 polymorphic sites were identified by amplification refractory mutation system-polymerase chain reaction(ARMS-PCR) or polymerase chain reaction-restriction fragment length polymorphism (PCRPFLP), respectively. Results: At codon 312, the frequency of GA or AA genotype was higher in the gastric cancer patients than in the healthy controls (P<0.01,OR=3.41, 95% CI: 2.06-4.79; P<0.01, OR=3.47,95% CI:1.39-8.68). No significant difference was found in the distribution of the polymorphism at codon 751 between the two groups(P>0.05). By the haplotype AA (codon 312A-codon 751A) analysis, the frequency of heterozygote(-/AA) or homozygote (AA/AA) was higher in the patients than in the controls (P<0.01 ,OR=2.81, 95% CI:1.82-4.34;P=0.02,OR=3.92, 95%CI:1.31-11.70,respectively). Whereas there were no significant differences of the other three haplotypes between the patients and the controls (P>0.05).Conclusions: The polymorphism of XPD at codon 312 might contribute to the etiology of gastric cancer.The haplotype AA (codon 312A-codon 751A) would be a critical risk factor of the susceptibility to gastric cancer.

OBJECTIVEHuman papillomavirus 6 (HPV 6) causes genital warts, a common sexually transmitted disease. L1-capsids protein is a highly promising vaccine candidate that has entered phase II clinical trial. But the existing methodologies for producing L1-capsids in insect cells is expensive for use in developing countries. As methylotrophic yeast,the Pichia pastoris expression system offers economy,and high expression levels. Over-expression of HPV6-L1 protein in P. pastoris was the purpose of this study.
METHODSThe whole L1 gene with preferred codons for P. pastoris was rebuilt and A-T rich regions were abolished, Cloning into pPIC3.5K,electroporation of KM71, in vivo screen of multiple inserts by G418 resistance, PCR analysis of pichia integrants, BMGY/BMMY are used for induction and expression of L1 proteins.
RESULTSThree clones were found to produce L1 protein which can be identified with Western blot. Compared with L1 protein from E.coli, pichia-produced L1 has some glycosylation. Reacting strongly with MabH6B10.5 in indirect immunofluorescence assay indicated that L1 protein expressed in pichia cell holds its native conformational epitopes which is important for vaccine use. A total 125 microg pure L1 protein could be obtained from 1L cultures through ion-exchange and Ni-NTA chromatography.
CONCLUSIONHPV type 6 L1 protein expressed in Pichia pastoris will facilitate the HPV vaccine development and structure-function study.

10.Effect of Chinese drugs for Pi strengthening Shen benefiting on the immunity function of HIV patients' specific T cells.

OBJECTIVETo explore the effect of Chinese drugs for Pi strengthening Shen benefiting (CDPSSB) on the immunity function of HIV/AIDS patients' specific T cells.
METHODSTotally 20 patients were randomly recruited from the treated group [treated by CDPSSB combined highly active anti-retroviral therapy (HAART)] and 23 patients were randomly recruited from the control group (treated by HAART alone). All patients were follow-up infected persons form You'an Hospital between from June 2010 to June 2012. CD4+ T absolute counts and HIV viral load were detected. Meanwhile, HIV whole gene overlapping peptides were used as stimulating antigen. The response intensity of HIV specific T cells was detected in the two groups.
RESULTSThere was no statistical difference in CD4 T absolute counts or HIV viral load between the two groups (P > 0.05). The response intensity of HIV specific T cells was significantly enhanced in the treated group, when compared with the control group (P < 0.05). Along with elongation of treatment time (6, 12, 18, and 24 months) in the treated group, the response intensity of HIV specific T cells showed enhancing tendency, but there was no statistical difference among these time points (P > 0.05).
CONCLUSIONCDPSSB could enhance improve the immunity function of HIV specific T cells, which might be one of its mechanisms.
